Abstract

Cauda epididymal sperm could be used as an alternative of male gamete source for application in reproductive technology, because the sperm is motile and has ability for fertilizing oocyte. This sperm could be collected from life or death animal, then it might be processed for chilling or freezing. Epididymis from death animal could also be preserved at 5°C for several days before collecting and processing. Cauda epididymal sperm which has motility more than 40% can be applied for artificial insemination (AI) or in vitro fertilization (IVF) programs. Key words: Cauda epididymal spermatozoa, Al, IVF

Abstract

Ethylene diamine tetraacetic acid (EDTA) is a molecular structure that binds to heavy metals andother toxins which needed to protect the spermatozoa during the cryopreservation process. The objectiveof this research was to examine EDTA in maintain the quality of garut ram frozen semen. Semen werecollected from mature Garut ram by artificial vagina. Fresh semen were evaluated and divided in equalvolume into three tubes and diluted with Tris extender containing 20% egg yolk (TEY-20) as control, TEY-20 + 0.01% EDTA (EDTA0.01), and TEY-20 + 0.02% EDTA (EDTA0.02), respectively. Semen was loaded in 0.25 mL mini straw with the concentration of 100 million motile spermatozoa. Semen was equilibrated at5oC for three hours, then frozen and stored in liquid nitrogen container. Quality of spermatozoa includingpercentages of motile spermatozoa (MS), live spermatozoa (LS), and intact plasma membrane (IPM) wereevaluated after diluting, equilibrating, and thawing. Data were analyzed using completely randomizeddesign with three treatments and five replicates. Means were compared significant difference test at 0.05significant level. Results of this research showed that mean volume, color, consistency, pH, mass movement,spermatozoa concentration, percentage of MS, percentage of LS, percentage of abnormal spermatozoa,and percentage of IPM of garut ram fresh semen were 0.87 mL, cream, thick, 6.98, 3, 4,296 million cell/mL,75%, 86.33%, 4.4%, and 85%, respectively. Percentages of MS, LS, and IPM after thawing in control (42.5,52.5, and 52.17%) was not significantly difference with EDTA0.01 (44.17, 52.5, and 52.5%) and EDTA0.02(39.17%, 51%, and 48.33%). In conclusion, the addition of EDTA in Tris extender can maintain quality ofGarut ram frozen semen, and its suitable using in artificial insemination program. Addition of EDTA cannot enhancing quality of garut ram frozen semen.

Abstract

A research to get inheritance of Arabica coffee resistance to Radopholus similisnematode was done in screen house and laboratory of Indonesian Coffee and Cocoa Research Institute, also at endemic area of coffee plantation, using F1, F1 R and F2 crossing between BP 542 A(resistant) x Andungsari 1 (susceptible) with their reciprocal, and BP 542 A x Kartika 1. The purpose of this study that was conducted at seedling stage is to formulate a Strategy for Arabica coffee breeding to get resistant varieties to nematode. As the variables of resistance were weight of seedling biomass, percent of root weight deviation, number of root nematodes, number of soil nematodes, reproduction and percent of necrotic root. Using discriminant analysis and fastclus, those data variables were analyzed for genetic of resistance with Statistical Analysis System programme version 8. Genetic study on the inheritance of resistance to R. similiswas started with evaluation of homozigosity of BP 542 A was resistant parent. The result showed that BP 542 A was heterozygous. Therefore, segregation test could not be suggested with segregation pattern principals as Mendel proposed. Segregation test on BP 542 A showed that it was heterozygote and the resistance was controlled by single gene with complete dominant effect, so the progeny segregated in 75% resistant and 25% susceptible. The result of the test showed the absence of maternal effect for root weight deviation and percentage of necrotic root variables, which meant that no cytoplasmic inheritance was involved. Based on the test of segregation ratio, almost all of the resistance was not appropriate for monogenic and or digenic segregation pattern as expected due to non allelic gene interaction that caused epistasis. Abstract

Abstract: This study was aimed at describing the students’ semiotic progress on learning number by using traditional games of Bermain Satu Rumah (BSR) supported by a set of learning activities based on Indonesian Realistic Mathematics Education. The study was designed into three stages, that is, the preliminary design, the teaching experiment, and the retrospective analysis. Then, Hypothetical Learning Trajectory (HLT) was modified as the reference of this study as well as the research instrument. HLT was tested to 24 third graders. The data were collected by applying the interview technique, observation, and field notes. The findings of the study showed that this instructional design could motivate students to join learning activities and could stimulate them to conserve the context, to understand the symbol meaning and use it, and to communicate starting from the informal to the formal levels. The progress was on the ability of students to abstract by holding and using symbols as a part of plan to solve the problem. Keywords: semiotics, bermain satu rumah, hypothetical learning trajectory, learning numbers KEMAJUAN SEMIOTIKA DALAM PEMBELAJARAN BILANGAN BERBASIS PERMAINAN TRADISIONAL BAGI SISWA SEKOLAH DASAR Abstrak: Penelitian ini bertujuan untuk mendeskripsikan kemajuan semiotika siswa dalam pembelajaran bilangan dengan menggunakan permainan tradisional Bermain Satu Rumah yang didukung dengan serangkaian aktivitas belajar berdasarkan Pendidikan Matematika Realistik Indonesia. Penelitian didesain melalui tiga fase, yaitu desain awal, eksperimen mengajar, dan analisis retrospektif. Dugaan Lintasan Belajar dibuat sebagai acuan dalam pelaksanaan penelitian sekaligus menjadi instrumen penelitian. Dugaan Lintasan Belajar diujikan kepada 24 orang siswa kelas III SD. Pengumpulan data dilakukan dengan menggunakan teknik wawancara, pengamatan, dan catatan lapangan. Hasil penelitian menunjukkan desain pembelajaran ini dapat memotivasi siswa untuk mengikuti kegiatan pembelajaran dan dapat merangsang siswa untuk mengonservasi konteks, memahami makna simbol dan menggunakannya, serta mengomunikasikan mulai dari tahap informal sampai formal. Kemajuan yang ditemukan adalah kemampuan siswa untuk mengabstraksi dengan mengadakan dan menggunakan simbol sebagai bagian dari rencana untuk menyelesaikan masalah. Abstract

This study aims to produce a optimal and effective effort meet the availability of disaster logistics with minimum requirements, appropriate methods of undertaking disaster logistics supplies, and a model of disaster logistics inventory monitoring. This research using approach system method which consist of input element, process element, and element of ouput supported by primary data and secondary data obtained by in-depth interview and observation. The results of this study indicate that by evaluating the rainfall data in Aceh Tamiang District the appropriate time in logistic supply is in June. The calculation of economic order quantity method with general economic logistic rice standard for each order is 282 kilogram with frequency of ordering 12 times a year and total cost of economical inventory every time Rp 208.075, meanwhile, with economical rice logistic special standard every time the order is 416 kilogram with the frequency of ordering 12 times a year and the total cost of economical inventory every time Rp 208.158. The model of disaster logistic monitoring is formulated by evaluating several components on the capacity aspect of local government and is expected to be done routinely and periodically.

Abstract

Simpur (Dillenia indica Linn) merupakan tanaman yang digunakan secara empiris sebagai obattradisional. Daun simpur memiliki nilai LD50 lebih dari 5000 mg/kgBB yang dapat dikategorikanpraktis tidak toksik sehingga tanaman ini berpotensi untuk dikembangkan. Namun, belum tersediainformasi mengenai keamanan terhadap penggunaan ekstrak etanol daun simpur (EEDS) yangdigunakan secara berulang selama 28 hari. Tujuan penelitian ini adalah untuk mengetahuihubungan dosis dan toksisitas pada pemberian oral secara berulang ekstrak etanol daun bintangurselama 28 hari terhadap perubahan indeks organ jantung, hati dan lambung pada tikus putih (Rattusnorvegicus L.) galur wistar. Daun simpur diekstraksi menggunkan metode maserasi dengan pelarutetanol 96%. Hewan uji dibagi menjadi 6 kelompok yang terdiri dari 7 ekor tikus jantan dan tikusbetina. Induksi dan pengamatan dilakukan selama 28 hari untuk kelompok kontrol dan dosis,sedangkan kelompok satelit hanya dilakukan pengamatan yang dilebihkan 14 hari untukmengamati efek reversibilitas. Pengujian toksisitas subkronik dilakukan dengan pemberian dosiskontrol, dosis 100 mg/kgBB, dosis 400 mg/kgBB, dosis 1000 mg/kgBB, satelit dosis 1000mg/kgBB dan dosis satelit kontrol. Hewan uji dideterminasi dan dibedah untuk mengamati indeksorgan jantung, hati dan lambung. Hasil pengamatan data indeks organ tikus jantan menunjukkanbahwa adanya kenaikan indeks organ yang signifikan pada organ hati kelompok satelit dosis atas 1000 mg/KgBB sedangkan pada organ jantung dan lambung tidak terjadi perbedaan yangsignifikan. Hasil yang diperoleh pada data indeks organ tikus betina menunjukkan adanya kenaikanindeks organ hati secara signifikan pada kelompok dosis 1000 mg/KgBB yang diberikan ekstraketanol daun simpur terhadap kelompok kontrol, sedangkan pada organ jantung dan lambung tidakterjadi perbedaan yang signifikan. Kesimpulannya pemberian EEDS secara berulang selama 28hari relatif aman terhadap organ jantung dan lambung, akan tetapi pada indeks organ hati tikusjantan dan betina mengalami peningkatan pada dosis 1000 mg/KgBB bersifat irreversibel.
