Abstract

Trigonella  foenum-graecum  (TFG)  is  one  of  medicinal  plants  contains  some steroidal sapogenin  such as diosgenin, yamogenin, gitogenin, tigogenin and trigoneoside, also  alkaloid  trigonellin,  which  is  have  many  activity  as  antidiabetic,  estrogenic  and  also as  anti  cancer.    This  experiment  was  done  to  explore  the  activity  of  some  extract  of TFG on some cell lines such as MCF7 (Human Breast Cancer Cell-line), T47D (Human Breast  Cancer  Cell-line),  PC3  (Human  Prostate  Cell-line)  and  SKOV  (Human  Ovarian Carcinoma Cell-line).  This assay was done using MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium)  methods.    Results  showed  that  ethyl  acetate  fraction  gives  the lowest  IC50  than  another  extracts.  IC50  for  PC3  is  66.24  ppm,  IC50  for  MCF7  is  41.81 ppm,  IC50  for  T47D  is  58.63  ppm.    These  datas  can  be  used  for  further  research  to isolate the active compound from TFG. Abstract

Phytoestrogen is the chemical compound contains in plant which has estro-genic like effect. Estrogen has important function on woman’s sexual organ, such as proliferation of uterine and vaginal cornification. Phytoestrogen are weak agonistsfor estrogen and illicit statistic significantly increases in uterine wet weight, at definite dose, in uterothrophic bioassay. Biji Klabet or Fenugreek seed (Trigonella foenum-graecum L.) contains steroidal sapogenins such as diosgenin, tigogenin,gitogenin, yamogenin and trigoneoside, that suspected having an estrogen-like ef-fect or as phytoestrogen. The aim of this research was to investigate the estrogenic effect of fenugreek’s ethanolic extract in ovariectomized and immature rats models.These models represent the climacteric/menopause phase, where estrogen level is very low because ovary produces no estrogen. The testing animals were divided intonormal group, ovariectomized control group, estradiol control group and three level doses of fenugreek extract (30mg/200gBW; 60mg/200gBW and 120mg/200gBW).The result indicated that start on 60mg/200gBW, fenugreek extract significantly could increasing mammary gland proliferation. Empirically fenugreek containingdiosgenin, that caused breast enhancement. This research showed that treatment with fenugreek extract can caused proliferation of mammary gland, both on imma-ture and ovariectomized rats.Key word : fenugreek, phytoestrogen, mamae duct, ovariectomized rat, im-mature rat, Trigonella foenum graecum L.

Abstract

Biji Klabet or Fenugreek (Trigonella foenum-graecum L.) contains steroidal sapogenins such as diosgenin, tigogenin, gitogenin, yamogenin and trigoneoside, that are suspected having an estrogen-like effect or as phytoestrogen. The objective of this study was to evaluate the estrogen-like effect of biji klabet alcohol extracts. This research used immature Wistar female rats, age of 19 days, which were represent the climacteric/menopause phase. The testing animals were divided into five groups, namely normal group, estradiol control group and three level dose groups of fenugreek extract (30 mg/200 gBW; 60 mg/200 gBW and 120 mg/200 gBW). The result showed that on 120 mg/200 gBW fenugreek extract significantly (a=0.05) increased estradiol and FSH level on immature rat. Analysis of vaginal smear showed vaginal lubrication effect, surprisingly on 120 mg/200 gBW could induce estrous cycle.

Abstract

Recombinant protein Fim-C S.typhi is a potential protein that can be used as an alternative typhoid vaccine and produced on a large scale. However, before entering into a large-scale stage, laboratory optimum data on the factors that affect the production process of Fim-C Salmonella typhi proteins are required. This study aims to obtain information regarding the effect of host cell number E.coli BL21 (DE3) pLysS and overexpression time on production of recombinant protein Fim-C Salmonella typhi as the basis for developing candidate of typhoid vaccine. The optimization process of overexpression was done by adding 0.5 mM IPTG (Isopropyl-?-D-thiogalactopyranoside) inducer into bacterial cultures of 2%, 4%, and 6% with 4, 5, and 6 hours over expression. The measurement of the concentration of Fim-C recombinant protein extracted samples were done by a spectrophotometric method used BCA Kit Assay Thermo ScientificTM with wavelength 590nm. The characterization of those protein extracts was performed using SDS-PAGE method. The results from the study concluded that the number of 4% E.coli bacterial cells and four-hour overexpression time are the optimum condition of Fim- C Salmonella typhi characterized by the presence of high-intensity bands at ± 31 kDa.

Abstract

The evaluation of toxicity of a mixed herbal extract containing mahkota dewa fruits (Phaleria macrocarpa Scheff. Boerl) and pegagan leaves (Centella asiatica L. Urban) on Wistar-strain rats had been carried out by the determination of the urea and creatinine content in urine and plasma after feeding. Oral doses of 100 mg, 500 mg, and 2500 mg of the mixed extract/kg body Weight were administered for 16 consecutive weeks to three groups of rats. Each treated group consisted of 15 males and 15 females, and the control group was represented by 10 males and 10 females. Samples of urine and plasma of the treated groups were taken at the time right before treatment (Week zero) and at Sm, 16th, 18th Week, while those of the control were taken at zero Week, 8th, and 16th Week, respectively. The result showed that theurea and creatinine contents among the treated and control groups were not signihcantly different. It could be concluded that oral administration ofthe mixed extract by a dose up to 2500 mg/kg body weight for 16 Weeks did not influence the urea and creatinine contents both in urine and plasma of the treated animals. Based on this result, it could be assumed that the use of the mixed extract is safe.

Abstract

Biji klabet (Trigonella foenum-graecum L.) adalah salah satu tanaman obat Indonesia yang telah dikenal memiliki aktivitas sebagai antidiabetes, antikolesterol, fitoestrogen dan antikanker. Biji klabet mengandung sejumlah sapogenin steroid (diosgenin, yamogenin, tigogenin dan trigoneosida), alkaloid trigonellin dan flavonoid (vitexin, orientin). Penelitian ini dilakukan untuk mengetahui aktivitas sitotoksik dan potensinya menginduksi apoptosis pada galur sel kanker payudara MCF-7. Pengujian aktivitas sitotoksik dilakukan menggunakan metode MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium) terhadap sel MCF-7. Sampel biji klabet dipreparasi melalui proses ekstraksi (ekstrak metanol dan ekstrak etanol) serta proses fraksinasi menggunakan perbedaan kepolaran pelarut (fraksi heksan, etilasetat, butanol dan air). Sedangkan analisis potensi apoptotik terhadap sel MCF-7 dilakukan dengan analisis siklus sel menggunakan flowcytometer dengan marker propidium iodida (20μg/mL) pada konsentrasi sampel 100μg/mL. Potensi apoptotik diukur berdasarkan % populasi sel pada fase subG0/G1. Hasil pengujian menunjukkan bahwa fraksi etilasetat memberikan aktivitas sitotoksik yang terbaik dengan IC50 = 41,81ppm dan secara signifikan (p<0,05) meningkatkan % populasi sel pada fase subG0/G1sebesar 20,87% pada konsentrasi 100ppm. Dari hasil tersebut dapat disimpulkan bahwa fraksi etilasetat biji klabet mempunyai aktivitas sitotoksik dan dapat menginduksi apoptosis sel kanker MCF-7.

Abstract

Buah adas (Foeniculum vulgare Mill.) merupakan salah satu tanaman obat tradisional yang dikenal memiliki efek sebagai anti-infl amasi, analgesik dan antioksidan. Dikenal juga untuk meningkatkan sekresi susu, mempromosikan menstruasi, memfasilitasi kelahiran, meringankan gejala klimakterik laki-laki dan meningkatkan libido. Buah adas banyak mengandung minyak esensial, seperti anethole, serta mengandung lignan dan fl avonoid yang diprediksi memiliki efek seperti estrogen atau sebagai fi toestrogen. Fitoestrogen adalah agonis lemah untuk estrogen dan dapat menimbulkan peningkatan yang signifi kan dalam berat basah uterus, pada dosis tertentu, dalam bioassay uterothrophic. Telah diketahui bahwa estrogen memiliki efek pada organ seksual wanita, seperti proliferasi uterus, kelenjar susu dan ovarium. Tujuan dari penelitian ini adalah untuk menyelidiki efek estrogenik ekstrak etanol buah adas pada tikus Wistar yang belum dewasa berusia 19 hari. Model-model ini mewakili faseklimakterik / menopause, di mana tingkat estrogen sangat rendah karena pada usia ini, estrogen belum diproduksi oleh ovarium. Hewan uji dibagi menjadi lima kelompok, yaitu kelompok normal, kelompok kontrol estradiol dan tiga tingkat dosis ekstrak adas (30 mg/200 g BB; 60 mg/ 200 g BB dan 120 mg/200g BB). Hasilnya menunjukkan bahwa mulai dari 60 mg/200 g BB, ekstrak adas secara signifi kan (α =0,05) meningkatkan berat basah uterus. Ekstrak adas juga memperpanjang uterus dan meningkatkan proliferasi uterin, seperti diameter uterus, diameter endometrium dan ketebalan uterus pada dosis 60mg/ 200gBW dan 120mg / 200gBB. Analisis pulasan cairan vagina menunjukkan efek lubrikasi vagina, secara mengejutkan dimulai pada 60 mg/ 200 g BB dapat menginduksi siklus estrus.

Abstract

Trigonella foenum-graecum (TFG) is one of medicinal plants containing several steroidal sapogenins, such as diosgenin, yamogenin, gitogenin, tigogenin and trigoneoside, and also alkaloid trigonellin and some flavonoids such as vitexin, isovitexin, orientin, isoorientin, which has many activities, such as antidiabetic, estrogenic, and also anticancer.  As phytoestrogen, TFG was predicted to have potency as Selective Estrogen Receptor Modulators (SERMs) which is used for hormonal-dependent breast cancer treatment. This experiment was carried out to investigate interaction of some sapogenin steroids and flavonoids in TFG to estrogen receptor alpha (ERα) and its activity to breast cancer cell line as confirmation. In silico prediction was carried out to investigate their estrogenic activity by analyzing their binding affinity to ERα using AutoDock Vina program. In vitro activity confirmation of TFG extract and its fractions were carried out using MTT assay on Erα-positive human breast cancer cell line, MCF-7.  Results showed that free binding energies of diosgenin and yamogenin were -6.4 kcal/mol, estradiol was -6.0 kcal/mol, and tamoxifen was -5.1 kcal/mol.  While cytotoxicity assay showed that ethyl acetate fraction gave the lowest IC50 of 41.81 ppm, with total steroid content of 20.03 ppm.  From these results, we can conclude that diosgenin and yamogenin have greater binding affinity to ERα comparing to estradiol and tamoxifen.  In vitro assay confirmation showed that ethyl acetate fraction has cytotoxic effect on MCF-7 cells.Keywords: Trigonella foenum-graecum, sapogenin steroids, MCF-7, estrogen receptor alpha, binding affinity

Abstract

Indonesian medicinal herbs have so many biological activity, including anticancer activity. This assay aims to evaluate three Indonesian medicinal plants (Trigonella foenum-graecum L. (TFG), Aglaia elliptica Blume. (AE) and Foeniculum vulgare Mill. (FV)) which have potency in anticancer activity. The experiment was done by MTT assay to observe the cytotoxicity activity on breast cancer cell lines MCF7.  Apoptotic potency was evaluated based on % population of MCF7 cell in subG0/G1 phase.  The results showed that IC50 from ethanolic extract of TFG was 241.24 ppm, ethanolic extract of AE was 19.442 ppm and  ethanolic extract of FV was 69.41 ppm.  Apoptotic potency for TFG was 6.46%,  AE was 11.94% and for FV was 8.73%. Thus, it is concluded that ethanolic extract of AE gives the lowest IC50 and the higher potency of apoptotic by significantly (p<0.05) increase % population of cells in subG0/G1 phase on 100ppm concentration.Keywords :  Trigonella foenum-graecum L., Aglaia elliptica Blume. and Foeniculum vulgare Mill., MCF-7, Apoptotic, Cytotoxic
