Abstract

ABSTRACT   Quantitative Structure-Activity Relationship (QSAR) study have been conducted to  antimalarial active compounds of cassane-type diterpenes isolated from seed kernels of Bagore (Caesalpinia crista Linn.), using the atomic net charges data resulted from AM1 method of semi-empirical calculations. The aim of this research is to determine the mathematic equation model of quantitative structure-activity relationship from antimalarial active compounds of cassane-type diterpenes isolated from seed kernels of Bagore (Caesalpinia crista Linn.) using the electronic parameter of AM1 method. The best QSAR equation model was determined by multiple linier regression analysis and tested by leave-one-out type of cross validation methods. The result indicates that the biological activity is represented by a linier function of activity versus atomic net charges of C1,  C2, C3, C4, and C6 which is expressed by : Log (1/C) = -7,362(+ 4,251) qC1 +3,258 (+ 4,590) qC2 + 11,038 (+4,485) qC3 + 29,421  (+20,542) qC4 + 19,849 (+6,171) qC6 + 8,541 (+2,256) The equation is significant at 95% level. The cross validation analysis give minimal value of PRESS=64,372 and SEP = 2,93; where Log (1/IC50) = biological activity and q = atomic net charges Key words : QSAR, Atomic Net Charges, Cross Validation Methods   ABSTRAK   Studi hubungan kuantitatif struktur-aktivitas (HKSA) telah dilakukan terhadap 42 senyawa aktif antimalaria turunan diterpen kassan hasil isolasi dari biji Bagore (Caesalpinia crista Linn.) menggunakan data muatan bersih atom hasil perhitungan semiempiris metode (Austin Model 1) AM1. Tujuan penelitian ini adalah menentukan model persamaan matematis hubungan kuantitatif struktur-aktivitas dari senyawa aktif antimalaria turunan diterpen kassan furano hasil isolasi dari biji Bagore (Caesalpinia crista Linn.) dengan parameter elektronik menggunakan metode AM1. Pemilihan model persamaan HKSA terbaik ditentukan dengan analisis regresi multilinier dan diuji dengan metode validasi silang leave-one-out. Hasil penelitian menunjukkan bahwa aktivitas biologis merupakan fungsi linier dari muatan bersih atom C1,  C2, C3, C4, dan C6 yang dinyatakan dalam persamaan : Log (1/C) = -7,362(+ 4,251) qC1 +3,258 (+ 4,590) qC2 + 11,038 (+4,485) qC3 + 29,421  (+20,542) qC4 + 19,849 (+6,171) qC6 + 8,541 (+2,256) Persamaan signifikan pada tingkat kepercayaan 95%. Hasil analisis validasi silang memberikan data parameter pengujian relatif minimal yaitu PRESS = 9,645; dimana Log (1/IC50) = aktivitas biologis dan q = muatan bersih atom   Kata kunci : HKSA, Muatan Bersih Atom, Metode Validasi Silang

Abstract

The native plants of East Borneo the Costus specious (Koening) J.E. Smith stem, Lagerstroema spesiosa Pers leaf, Cerbera mangans L leaf, Vitis trifolia L fruit., Scurrula atropurpurea (Blume) Danser root, Brucea javanica (L.) Merr. leaf, Lygodium microphyllum, Bidens Chinensis Willd., Sonneratia caseolaris L. peel, Sonneratia caseolaris L. stem is almost under-explored for their potensial benefits. They were extracted by the solvents of increasing polarity (n-hexane, ethyl acetate, and n-butanol) were tested for their free radical activity against DPPH (2,2-diphenyl-1-picrylhydrazyl). The ethyl acetate extract of Costus spesiosus (Koening) J. E. Smith antioxidant were screnned concentration of 68 ppm, similarly athyl acetate extract of Vitis trivolia L., showed antioxidant activity at 64,30 ppm. As the ethyl acetate extract of Scurrulla atropurpurea (Blume), Brucea javanica (L.) Merr, Lygodium microphyllum and Sonneratia caseolaris L. stem, showed antioxidant activity at 273,52 ppm, 91,12 ppm, 17,39 ppm and 7,03 ppm. N-butanol extract of Lagerstroema spesiosa Pers, Cerbera mangans L, Bidens chinensis Willd, and Sonneratia caseolaris L. peel showed 8,37 ppm, 128,59 ppm, 18,17 ppm and 54,29 ppm antioxidant activity using DPPH model systems. Owing to the property, the studies can be further extended to exploit them for their possible application for preservation of food products as well as their use as health supplements.

Abstract

The previous research showed that the fraction C from active extract of Garcinia latissima Miq. fruit was active against Bacillus subtilis. This study aims to isolate and identify the active compound as an antibacterial agent from the fraction C. Fraction C was purified by recrystallization using chloroform and n-hexane solvents and then isolated using preparative-thin layer chromatography-silica gel 60 GF254 to give a yellow compound. The antibacterial activity was determined using microdilution with thiazolyl blue tetrazolium bromide indicator against B. subtilis American Type Culture Collection 6633. The isolate was identified using UV-Vis, IR, MS, Proton Nuclear Magnetic Resonance (1H-NMR) and carbon NMR (13C-NMR), and NMR-2D techniques including HMQC and HMBC. Based on the spectroscopic analysis and literature review, the compound was identified as 6-deoxyjacareubin, which is a new compound from Garcinia latissima Miq. The 6-deoxyjacareubin showed antibacterial activity with MIC value of 156.25 ppm and was categorized as a weak antibacterial agent because the MIC value was more than 100 ppm. According to in silico approach to the docking study, 6-deoxyjacareubin showed similar hydrophobic interaction with several amino acid residues including C2565, C2589, G2484, U2590, and U5588 between a native ligand.

Abstract

  ABSTRACT   Research about active antioxidant compounds isolated from plants ant nests (Myrmecodia pendens Merr. & Perry) from Papua by the method of column chromatography has been done. This study aimed to explore the isolation of the active class of compounds as antioxidants from ant nest plant. Isolates obtained using the method of column chromatography and antioxidant activity of the ant nest plants isolates tested used spraying method with free radical DPPH (2,2-diphenyl-1-picrylhydrazyl) after eluted on thin-layer chromatography. At separation by column chromatography, eluate collected in vials with a volume of 10 ml. Based on the results of fractionation, separation of the components obtained by 74 vials. Identification using TLC by determining the Rf value of each component. The same Rf value is considered as a single fraction. Based on the results obtained seven fractions TLC. Active isolates (F6) obtained by spraying. The results of fraction of spot with DPPH staining showed that the isolates was considered the most active isolates as seen from the formation of a yellow color that is lighter and more dominant than the other fractions, this indicates that the fraction (F6) binds more free radicals (DPPH).   Key words: antioxidant, isolates, Myrmecodia pendens Merr & Perry, DPPH (2,2-diphenyl-1-picrylhydrazyl).   ABSTRAK   Telah dilakukan penelitian isolasi senyawa aktif antioksidan dari tumbuhan sarang semut (Myrmecodia pendens Merr. & Perry) asal Papua dengan metode kromatografi kolom. Penelitian ini bertujuan untuk mencari isolat golongan senyawa yang aktif sebagai antioksidan dari tumbuhan sarang semut. Isolat diperoleh dengan menggunakan metode kromatografi kolom dan aktivitas antioksidan isolate sarang semut diuji menggunakan metode penyemprotan dengan radikal bebas DPPH (2,2-diphenyl-1-picrylhydrazyl) setelah dielusi pada kromatografi lapis tipis. Pada pemisahan dengan kromatografi kolom, eluat ditampung dalam vial dengan volume 10 ml. Berdasarkan hasil fraksinasi, diperoleh komponen hasil pemisahan sebanyak 74 vial. Identifikasi menggunakan KLT dengan cara menentukan nilai Rf masing-masing komponen. Nilai Rf yang sama dianggap sebagai satu fraksi. Berdasarkan hasil KLT di dapatkan 7 fraksi. Isolat  aktif (F6) yang diperoleh dengan penyemprotan. Hasil pewarnaan spot fraksi dengan DPPH menunjukan bahwa isolat yang dianggap paling aktif adalah isolat yang dilihat dari terbentuknya warna kuning yang lebih terang dan lebih dominan dibanding fraksi yang lainnya, ini menandakan bahwa fraksi (F6) lebih banyak mengikat radikal bebas (DPPH).   Kata kunci: antioksidan, isolat, Myrmecodia pendens Merr & Perry, DPPH (2,2-diphenyl-1-picrylhydrazyl).

Abstract

Research on the effects of skin ointment dosage active ingredient ethyl acetate leaf extract Sungkai (Peronema canencens Jack.) has be done burns infection against pathogens bacteria. This study aims to determine the effective concentration of the ethyl acetate extract antimicrobial in skin ointment preparations against pathogens skin burns infection. Test material obtained by fractionation of the methanol extract of leaves Sungkai, further formulated into an ointment base, was tested to determine the effective concentration of activity. The results obtained by the concentration of effective skin ointment preparation with the active ingredient ethyl acetate leaf extract Sungkai is 4% for gram-positive bacteria: B. subtilis and S. aureus, dankonsentrasi 2% for gram-negative bacteria: P. aeruginosa and Str. mutans. Mutans   ABSTRAK Telah dilakukan penelitian efek sediaan salep kulit berbahan aktif ekstrak etil asetat daun sungkai (Peronema canencens Jack.) terhadap bakteri patogen penginfeksi luka bakar. Penelitian ini bertujuan mengetahui  konsentrasi efektif antimikroba ekstrak etil asetat dalam sediaan salep kulit terhadap bakteri patogen penginfeksi luka bakar. Bahan uji diperoleh dengan fraksinasi ekstrak metanol daun sungkai, selanjutnya formulasikan ke dalam basis salep, diuji aktivitasnya untuk menentukan konsentrasi efektif. Hasil penelitian diperoleh konsentrasi efektif sediaan salep kulit dengan bahan aktif ekstrak etil asetat daun Sungkai adalah 4 % untuk bakteri gram positif: B. subtilis dan S. aureus, dankonsentrasi 2% untuk bakteri gram negatif: P. aeruginosa dan Str. Abstract

Has conducted research on formulation and stability of sunscreen cream onion bulb extract dayak ( Eleutherine Americana L. Merr . ) . From the empirical data for the onions dayak has been used as an anti- cancer prevention and treatment by people of Borneo so it needs to be further investigated to obtain the data laboratory to support the empirical data . Several classes of secondary metabolites found in the bulbs of onions dayak alkaloids , glycosides , flavonoids , phenolics , steroids and tannins . The study was conducted as the basis for the formulation of sunscreen preparations aimed at the prevention of skin cancer . The study was conducted with bulb onions dayak extraction using Soxhlet method with solvent n - hexane , ethyl acetate , n - butanol , and ethanol as the active ingredient of the formula of making preparations . Tests conducted on formulations A and B with the concentration of garlic bulb extract dayak by 0.1% , consisting of physical stability test ( organoleptic , homogeneity ) and chemical ( pH and dosage ) in order to obtain a stable dosage formula . The test results obtained on the organoleptic test was no change in the observations made on each preparation before and after storage at room temperature on day - 7 , -14 , and -21 . Then the homogeneity test of the hedonic test results at 95% confidence level ( SNI 01-2346-2006 ) , concluded that the homogeneity of the preparation in all formulas produced in the category of smooth . Further testing on the pH stability and pH 5.5 to 8.0 to be produced that Formula A and Formula B to pH 7.0 to 7.7 with various concentrations of the extract , after 21 days of storage , inferred pH remains stable and safe dosage for used ( SNI 16-4399-1996 ) as formula sunscreen preparations made from onion bulb extract dayak . Key words : bulb onions dayak, sunscreen, formulas, test stability, skin cancer   ABSTRAK Telah dilakukan penelitian mengenai formulasi dan stabilitas krim tabir surya ekstrak umbi bawang dayak (Eleutherine Americana L. Merr.). Dari data empirik selama ini bawang dayak telah digunakan sebagai pencegah dan pengobatan anti kanker oleh masyarakat Kalimantan sehingga perlu diteliti lebih lanjut untuk mendapatkan data laboratorik untuk mendukung data empirik tersebut. Beberapa golongan metabolit sekunder yang terdapat pada umbi bawang dayak yakni alkaloid, glikosida, flavonoid, fenolik, steroid dan zat tannin. Penelitian tersebut dilakukan sebagai dasar pembuatan formulasi sediaan krim tabir surya yang ditujukan untuk pencegahan terjadinya kanker kulit. Penelitian dilakukan dengan ekstraksi umbi bawang dayak menggunakan metode soxhlet dengan pelarut n-heksan, etil asetat, n-butanol, dan etanol sebagai bahan aktif dari pembuatan formula sediaan. Pengujian dilakukan terhadap formulasi A dan B dengan konsentrasi ekstrak umbi bawang dayak sebesar 0,1%, terdiri dari uji stabilitas fisik (organoleptis, homogenitas) dan kimia (pH dan sediaan) agar diperoleh formula sediaan yang stabil. Hasil pengujian yang diperoleh adalah pada uji organoleptis tidak ada perubahan pada pengamatan yang dilakukan terhadap masing-masing sediaan sebelum dan setelah penyimpanan pada suhu kamar pada hari ke-7, -14, dan -21. Kemudian pada uji homogenitas dari hasil uji hedonik pada tingkat kepercayaan 95% (SNI 01-2346-2006), disimpulkan bahwa homogenitas sediaan pada semua formula yang dihasilkan masuk dalam kategori halus. Selanjutnya pada stabilitas pH dilakukan pengujian dan dihasilkan bahwa pH 5,5-8,0 untuk Formula A dan pH 7,0-7,7 untuk Formula B dengan berbagai konsentrasi ekstrak, setelah penyimpanan 21 hari, disimpulkan pH sediaan masih stabil dan aman untuk digunakan (SNI 16-4399-1996) sebagai formula sediaan tabir surya berbahan ekstrak umbi bawang dayak. Abstract

A study concerning the characteristics and Analysis of Drug Related Problems (DRPs) Patients with Tuberculosis Patients in Public Health Center (PHC) Temindung, Samarinda in East Kalimantan. The study was conducted using a prospective approach to the study of data sources in the form of primary data and secondary data. The primary data source is data from interviews with patients and through the provision of questionnaires and secondary data in this research is a Tuberculosis patient medical record data. Data collection was performed by analyzing the accuracy of the DRPs categories of accuracy of drugs, side effects and patient non-compliance. As well as the data recorded in the form of research support patient characteristics are age, sex, and education. Data were analyzed descriptively. Data obtained from patients with male gender 63.6% and women 36.4. Patient with age <15 years 4.5%, aged between 15-50 years and 68.2% aged> 50 years 27.3% . Based on the patient's level of education, no school 9.1%, graduated from elementary school22.7%, graduated from high school18.2%, graduated from high school40.1%, scholar 9.1%. Incidence of DRPs on drugs interactions 507%. Tuberculosis patient compliance rate of 81.8% in PHC Temindung adherent patients and 18.2% of patients do not comply Keywords: Tuberculosis, Drug Related Problems (DRPs) drugs interactions and patient compliance   ABSTRAK Telah  dilakukan penelitian tentang Karakteristik dan Analisis Drug Related Problems(DRPs) Pasien Penderita Tuberkulosis di Puskesmas Temindung, Samarinda Kalimantan Timur. Penelitian ini dilakukan dengan menggunakan pendekatan prospektif dengan sumber data penelitian berupa data primer dan data sekunder. Sumber data primer  adalah data hasil wawancara dengan pasien dan melalui pemberian kuisioner sedangkan data sekunder dalam penelitian ini adalah data rekam medik pasien Tuberkulosis. Pengumpulan data dilakukan dengan menganalisis DRPs kategori interaksi obat dan ketidakpatuhan pasien. Serta dicatat data pendukung penelitian berupa karakteristik pasien yaitu usia, jenis kelamin, dan tingkat pendidikan.Data dianalisis secara deskriptif.Dari data yang diperoleh pasien dengan jenis kelamin laki-laki 63,6% danperempuan 36,4%.Pasien yang berusia < 15 tahun 4,5%, usia antara 15-50 tahun 68,2% dan usia > 50 tahun 27,3%. Berdasarkan tingkat pendidikan pasien, tidak sekolah 9,1%,lulus SD 22,7%,lulus SMP 18,2%,lulus SMA 40,1%, sarjana 9,1%. Kejadian DRPs mengenai interaksi obat 50%.Tingkat kepatuhan pasien Tuberkulosis di Puskesmas Temindung 81,8% pasien patuh dan 18,2% pasien tidak patuh. Abstract

The Quantitatif Structure-Activity Relationship (QSAR) study has been established on 13 Carbazole analog compounds to their antibacterial activity using electronical descriptors. The development of computational chemistry, in particular quantum chemical calculation progression, has made the possibility of molecule modeling and pharmacochemistry exploration of molecule structures. This emerging semiempirical method can be implemented in QSAR using Hansch model. A research associated with QSAR antibacterial activity of Carbazole analog compounds which used semiempirical AM1 (Austin Model 1) method has been conducted. The geometrical optimation of 2D into 3D structures was done utilizing HyperChem program with the Polak – Ribiere algorithm. Results revealed that atom in the position C8, C9, C10 and C12 are necessary to predict biological activity of Carbazoleanalog. Keywords: QSAR, Atomic Net Charges, Cross Validation Methods

Abstract

Screening Ageratum conyzoides L. fraction for antibacterial activity using thin layer chromatography bioautography have been done. The method is based on thin-layer chromatography–direct bioautography of Ageratum conyzoides chloroform fraction. The aim of study to determine the antibacterial activity of chloroform Ageratum conyzoides L. fraction in TLC bioautography. Extraction was done by maceration using methanol solvent, followed by partition using chloroform solvent using liquid-liquid extraction method is done by chromatographic profile of each eluent comparison is then performed further fractionated using vacuum liquid chromatography method. Screening activity of the compound was done qualitatively raction results are antibacterial activity test using thin layer chromatography bioautography The results showed that the eluent to separate the components of a compound based on the chomatography profile is eluent n-hexane: ethyl acetate (2: 1), the results obtained by fractionation of the active fraction (51-53) Rf value of 0.59. Results of screening activity held chloroform fraction Ageratum conyzoides leaves are antibacterial activity which can be seen from the formation of inhibition zones in the region stain spot on the TLC plate were grown in glucose medium Nutrient. The results were analyzed descriptively, that is to describe the characteristics of the research which is obtained.   Keywords: TLC bioauttography, Ageratum conyzoides L., TLC profiles, antibacterial   ABSTRAK   Telah dilakukan penelitian dengan judul skrining aktivitas dari fraksi daun bandotan (Ageratum conyzoides L.) secara kromatografi lapis tipis bioautografi. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri fraksi kloroform daun bandotan secara KLT bioautografi. Ekstraksi dilakukan secara maserasi dengan menggunakan pelarut metanol yang dilanjutkan dengan partisi menggunakan pelarut kloroform dengan menggunakan metode ekstraksi cair-cair yang dilakukan dengan melihat profil kromatografi dari setiap perbandingan eluen selanjutnya dilakukan fraksinasi lebih lanjut dengan menggunakan metode kromatografi cair vakum. Skrining aktivitas senyawa hasil hasil fraksi dilakukan secara kualitatif yaitu uji aktivitas antibakteri menggunakan metode kromatografi lapis tipis (KLT) Bioautografi. Hasil penelitian menunjukkan bahwa eluen yang dapat memisahkan komponen senyawa dengan baik berdasarkan profil kromatografinya adalah eluen n-heksana : etil asetat (2 : 1), hasil fraksinasi diperoleh fraksi aktif pada (vial fraksi 51-53) nilai Rf sebesar 0,59. Hasil skrining aktivitas yang dimiliki fraksi kloroform daun bandotan yaitu aktivitas antibakteriyang dapat dilihat dari terbentuknya zona hambat pada daerah spot noda pada plat KLT yang ditanam di dalam medium Glukosa Nutrient Agar. Hasil penelitian yang diperoleh dianalisis secara deskriptif, yaitu dengan menjabarkan karakteristik hasil penelitian yang diperoleh.   Kata Kunci: KLT bioautografi, Ageratum conyzoides L.,  profil KLT,   antibakteri
