Abstract

ABSTRACTLysozymes play an important role in the first line of defense in fish and potentially used as an immunity status biomarker and immune responses evaluation in fish, which often found in two types, i.e. chicken-type and goose-type (c- and g-type, respectively). To recent, the information related to the sequences and the expression analysis of the c- and g-type lysozyme genes in African catfish is still limited. In the present study, we report a partial cloning and mRNA expression analysis of c-type and g-type lysozymes in African catfish Clarias gariepinus. We have successfully cloned and partially identify the c-type, and g-type lysozyme genes of C. gariepinus, which consist of 594 and 560 of coding sequences, respectively. Catalytic and other conserved residues were identified by multiple sequences alignment and they showed high similarity with other teleost fish species. mRNA levels of the genes were analyzed by using qPCR method and their expressions in the spleen, liver, and head kidney were rapidly modulated after Aeromonas hydrophila injection, with different patterns were observed in each organ. These results confirmed that c- and g-type lysozymes played an important role in non-specific immunity against A. hydrophila infection. This study provided valuable information that can be used to understand the African catfish immune systems for better disease and stress management in C. gariepinus culture.Keywords: lysozymes, gene identification, gene expression, bacterial infection, African catfish ABSTRAKLisozim berperan dalam sistem pertahanan dini pada ikan dan sangat potensial digunakan sebagai marka status imunitas dalam evaluasi respons imun. Lisozim umum ditemukan dalam dua tipe pada ikan: tipe-ayam (tipe-c) dan tipe-angsa (tipe-g). Informasi terkait sekuens gen dan ekspresi gen kedua tipe lisozim pada ikan lele dumbo sangat terbatas. Pada penelitian ini, kami melaporkan kloning gen secara parsial, dan analisis ekspresi gen dari kedua tipe lisozim pada ikan lele dumbo C. gariepinus. Sekuens parsial gen lisozim tipe-c dan tipe-g yang berhasil diidentifikasi adalah sepanjang 594 dan 560 pasang basa. Situs katalitik dan residu khas memiliki tingkat kesamaan yang tinggi dengan spesies ikan yang lain. Analisis mRNA dilakukan dengan metode quantitative PCR (qPCR). Ekspresi kedua gen di jaringan ginjal depan, limpa, dan hati dengan cepat terinduksi pasca infeksi bakteri A. hydrophila dengan pola yang berbeda. Hasil ini menunjukkan bahwa lisozim tipe-c dan tipe-g memiliki peran yang sangat penting dalam sistem imun ikan lele dumbo terhadap infeksi A. hydrophila. Penelitian ini menghasilkan informasi penting yang dapat digunakan untuk mempelajari sistem imun ikan lele dumbo dan sebagai acuan dalam penanganan dan manajemen penyakit pada budidaya ikan lele dumbo.Kata kunci: lisozim, identifikasi gen, ekspresi gen, infeksi bakteri, ikan lele dumbo

Abstract

Gonggong adalah sejenis siput laut yang merupakan makanan laut (seafood) favorit dan ikon Kota Tanjungpinang, ibukota Provinsi Kepulauan Riau (Kepri). Gonggong mengandung protein tinggi, yaitu sekitar 46,65%. Tidak ada laporan tentang produksi budidaya dan upaya konservasi gonggong. Teknologi produksi benih buatan gonggong belum berkembang. Tujuan penelitian ini adalah untuk mengevaluasi pemberian kombinasi hormon LHRH-a dan antidopamin untuk menginduksi proses pemijahan. Evaluasi pemberian hormon LHRH-a dan antidopamin pada pemijahan siput gonggong dilakukan dengan empat dosis: 0,5 ìLgι bobot badan lunak (BB) (P1); 0,7 ìLgι BB; dan 0,9 ìLgι BB (P2); dan tanpa suntikan (TS). Siput gonggong pascasuntikan dipelihara di akuarium selama 14 hari. Hasil penelitian menunjukkan bahwa (1) kombinasi hormon LHRH-a dan antidopamin mampu merangsang pemijahan gonggong. Dosis rendah hormon LHRH-a dan antidopamin (P-1) menghasilkan induk betina yang memijah paling banyak, yaitu 34,48%; lebih tinggi dari P-2 (27,59%), P-3 (20,69%); dan TS (17,24%); (2) jumlah telur yang dikeluarkan oleh induk betina berbeda secara signifikan antar perlakuan (P<0,05). Jumlah telur yang dikeluarkan oleh masing-masing induk berkisar antara 10.874-63.489 butir/ekor dengan rata-rata 39.347 ± 16.667 butir/ekor.The gonggong is a species of sea conch which is a favourite seafood and an icon of Tanjungpinang City, capital of Kepulauan Riau (Kepri) Province. Gonggong contains high protein, about 46.65%. There were no reports on aquaculture production and conservation effort of gonggong. The technology on artificial seed production of gonggong has not yet developed. The aim of this study was to evaluate the administration of LHRH-a hormone and anti-dopamine to induce the spawning process. Evaluation of the administration of LHRH-a hormone and anti-dopamine on the gonggong conch?s spawning was carried out with four doses: 0.5 ìLgι soft body weight (BW) (P-1), 0.7 ìLgι BW; and 0.9 ìLgι BW (P-2); and without injections (TS). The gonggong conchs after injection were reared in aquarium for 14 days. The results showed that (1) a combination of LHRH-a hormone and anti-dopamine was able to stimulate gonggong spawning. The lower dose of LHRH-a hormone and anti-dopamine (P-1) produced the highest number of spawned female broodstock, which was 34.48%, higher than P-2 (27.59%), P-3 (20.69%), and TS (17.24%); (2) the number of eggs released by female broodstock was significantly different among the treatments (P<0.05). The number of eggs released by each female broodstock ranges between 10,874-63,489 grains/ind. with an average of 39,347±16,667 grains/ind.

Abstract

Wilayah pesisir merupakan anugrah dari Allah SWT yang memiliki berbagai macam potensi yang wajib dikelola  untuk  kepentingan pembangunan pada umumnya dan bahkan untuk kelangsungan hidup manusia, sehingga setiap orang secara langsung atau tidak langsung dilarang untuk melakukan sesuatu, namun larangan tersebut diperbolehkan sepanjang mendapat izin dari pemerintah. Izin merupakan sesuatu yang dilarang tapi diperbolehkn. Bahkan  izin bertujuan  menghilangkan halangan, hal yang dilarang menjadi boleh.

Abstract

Honesty Based Selling Price Concept: Achieving Gain Reaching Goodness. This article aims to reconstruct honesty-based selling price concept in Islam. The research involved owners and management of YDT, Kopontren DT in Bandung, Charni’s Productions in Yogyakarta, as well as Kedai Assalamu’alaikum and Bismillah Restaurant in Malang. Analysis was conducted by using bayani, burhani and irfani epistemology. Honesty-based selling price concept could be classified as cost-plus pricing consistency, conditioned market mechanism, and balanced/equibrium market mechanism according to bayani, burhani and irfani epistemology respectively. Generally, honesty-based selling price concept in Islam is consistency market mechanism which is the determination of price at the very beginning until the complete consumption of products. Konsep Harga Jual Kejujuran: Meraih Keuntungan Menggapai Kemaslahatan. Makalah ini bertujuan menyusun konsep harga jual berbasis nilai kejujuran dalam Islam. Penelitian dilakukan pada pemilik dan manajemen YDT dan Kopontren DT di Bandung, Charni’s Productions di Yogyakarta, serta Kedai Assalamu’alaikum dan rumah makan Bismillah di Malang. Analisis dilakukan dengan menggunakan epistemologi bayani, burhani, dan irfani. Konsep harga jual berbasis nilai kejujuran dapat diklasifikasikan menjadi consistency cost-plus pricing, mekanisme pasar bersyarat serta mekanisme pasar keseimbangan dari epistemologi bayani, burhani dan irfani. Secara umum konsep harga jual berbasis nilai kejujuran di dalam Islam adalah consistency market mechanism, yaitu suatu konsep penetapan harga jual secara konsisten melalui mekanisme pasar sejak dari awal penetapan harganya hingga produk tersebut habis terjual.

Abstract

Accounting makrifat, determination of journey of accounting spirituality: ontological evaluation. Accounting is basically a product of ideol-ogy was form as an ideological instrument to construct of reality. Logic Contradic-tions in building basic ontology of accounting to reproduce the contradictions in reality be the form of contradictions that lead to inequality and marginalization. The presence of Islamic accounting to be deconstructing the power of capitalism, altough in fact impressed’’ barren’’. The spirit of Islam as liberation ideology is reduced in the grip of market power. Therefore, an ontological perspective review to reassess the basic philosophy of Islamic accounting. Efforts to redefine and re-parse the critical relations with Islamic as ideology of science in accounting as a doctrinal-theoretical framework. Creating patterns to be Islamic of ideology in accounting as a basic framework for understanding the scope Islamic accounting study, by positions’’ liberation’’ as the highlight of the discourse (makrifat) Islamic accounting.Abstrak: Makrifat akuntansi, determinasi puncak perjalanan spirituali-tas akuntansi: suatu tinjauan ontologism. Akuntansi pada dasarnya adalah produk ideologis yang lahir sebagai instrumentdalam mengkonstruk realitas. Kontradiksi logicdalam bangunan dasar ontologi akuntansi akan mereproduksi model-model kontradiksi dalam realitas berupa lahirnya pertentangan-perten-tangan yang berujung pada ketimpangan dan marginalisasi. Hadirnya ideologis akuntansi Islam dalam mengdekonstruksi kuasa akuntansi kapitalisme, dalam kenyataannya terkesan ‘’mandul’’. Semangat pembebasan Islam sebagai ideologi tereduksi dalam cengkeraman kuasa pasar. Oleh karenanya tinjauan ontolo-gis merupakan sudut pandang untuk menilai kembali filosofi dasar akuntansi Islam. Upaya untuk meredefinisi dan mengurai kembali dengan kritis hubun-gan Islam sebagai ideologi dengan akuntansi sebagai ilmu pengetahuan dalam kerangka doktrinal-teoritis. Abstract

ABSTRACT Outbreaks of infectious diseases due to Aeromonas hydrophila in catfish can cause high death rates (80?100%). Fish disease control can be done using phytopharmaceutical to prevent or treat diseases of fish. One of the phytopharmaceutical that known to prevent the fish diseases is ambon banana stem Musa cavendishii var. dwarf Paxton. This study was conducted to test the effectiveness of catfish immersion using banana stem juice as an immunostimulant against bacterial infections A. hydrophila. The observed parameters were a total performance of production, hematological test, and water quality. Immersion of catfish seeds with stem juice was performed at a concentration of 5 mL/L, 13 mL/L, and 21 mL/L for 30 minutes. Each treatment consisted of three replications. A  total of 15 fishes were immersed in 1.5 L water. At day-9 after immersion, fish was infected by A. hydrophila bacteria at a dose of 104 cfu/mL. The results showed that fish treated with banana stem juice at a concentration of 13 mL/L had the survival rate of 53.33±6.67% which is higher than positive control (33.33±6.67%). Based on hematology observations on the 3rd day post-challenge test (H12) with A. hydrophila, total erythrocytes, hemoglobin, total leukocytes, phagocytic activity, differential leukocyte, and lysozyme activity was highest in treatment13 mL/L. Lower feed conversion ratio (1.08±0.04) were also obtained in treatment 13 mL/L. Thus Immersion of seeds in stem juice can boost the immune system against infections A. hydrophila. Keywords: Aeromonas hydrophila, ambon banana stem juice, soaking, catfish, immune system  ABSTRAK Wabah penyakit akibat infeksi Aeromonas hydrophila pada ikan lele dapat menyebabkan tingkat kematian yang tinggi (80?100%). Penanggulangan penyakit ikan dapat dilakukan menggunakan fitofarmaka untuk mencegah ataupun mengobati penyakit ikan. Salah satu fitofarmaka yang dapat digunakan dalam upaya pencegahan penyakit ikan adalah batang pisang ambon lumut Musa cavendishii var. dwarf Paxton. Penelitian ini bertujuan untuk menguji efektivitas perendaman benih ikan lele dengan menggunakan air perasan batang pisang ambon sebagai imunostimulan terhadap infeksi bakteri A. hydrophila. Parameter yang diamati selama penelitian adalah kinerja produksi, uji hematologi dan pengukuran kualitas air. Perendaman benih ikan lele dengan air perasan  dilakukan pada konsentrasi 5 mL/L, 13 mL/L, dan 21 mL/L selama 30 menit. Pada hari ke-9 setelah perendaman, ikan diinfeksi bakteri A. hydrophila pada kepadatan 104 cfu/mL. Hasil penelitian menunjukkan bahwa perendaman ikan dengan air perasan pada konsentrasi 13 mL/L memiliki kelangsungan hidup yakni sebesar 53,33±6,67%, lebih tinggi bila dibandingkan dengan kontrol positif  (33,33±6,67%). Berdasarkan pengamatan pada uji hematologi hari ke-3 pascauji tantang (H12) dengan bakteri A. hydrophila, total eritrosit, hemoglobin, total leukosit, aktivitas fagositik, dan aktivitas lisozim tertinggi terdapat pada perlakuan konsentrasi 13 mL/L. Rasio konversi pakan yang rendah (1,08±0,04) juga terdapat pada perlakuan 13 mL/L. Hal ini menunjukkan bahwa perendaman benih ikan lele pada air perasan dapat meningkatkan sistem imun terhadap infeksi bakteri A. hydrophila. Kata kunci: Aeromonas hydrophila, air perasan batang pisang ambon, perendaman, ikan lele, sistem imun

Abstract

ABSTRACT This study was aimed to evaluate the dietary ambon lumut banana Musa cavendishii var. dwarf Paxton stem extract on the immune responses, growth, and survival rate of Pacific white shrimp against white spot disease. Ambon banana steam extract was obtained by maceration method using ethanol. The shrimps fed by pellet containing ambon banana stem extracts with different dosages for 29 days, i.e designated as 0.1 (A); 0.3 (B), and 0.5 (C) g/kg, the diet treatment without ambon banana stem extract without challenged test (K-), and diet treatment without ambon banana stem extract with challenged test (K+). Each treatment consisted of three replications. Feeding was conducted for 29 days of maintenance (four times a day). The results showed that the immune responses (average total hemocyte count: 45.15×106 cells/mL, phenoloxidase activity 1.03±0.08 OD, respiratory burst 0.95±0.04 OD, phagocytic activity 94.33±1.53%, growth (specific growth rate: 7.79±0.06%/day, feed ratio conversion was 52±0.01), and survival of treatment C (survival rate 100%) were higher compared with the treatment K+ (total hemocyte count: 3.83×106 cells/mL, phenoloxidase activity 0.04±0.01 OD, respiratory burst 0.18±0.06 OD,  phagocytic activity 5.67±0.58%, specific growth rate: 2.61±0.08%/day, feed conversion ratio 2.11±0.02, survival rate: 50%). Therefore, banana stem extract at a dose of 0.5 g/kg everyday diet can be used to improve growth and nonspecific immune system against white spot disease on Pacific white shrimp. Keywords: Pacific white shrimp, white spot disease, immune response, Ambon banana stem extract  ABSTRAK Penelitian ini bertujuan untuk menguji dosis optimal ekstrak batang pisang ambon melalui pakan dalam meningkatkan respon imun, pertumbuhan dan kelangsungan hidup udang putih terhadap penyakit white spot. Ekstrak batang pisang ambon lumut Musa cavendishii var. dwarf  Paxton menggunakan metode maserasi dengan menggunakan pelarut etanol. Pakan udang yang mengandung ekstrak batang pisang ambon pada dosis yang berbeda selama 29 hari 0,1 (A); 0,3 (B), dan 0,5 (C) g/kg, pakan tanpa pemberian ekstrak batang pisang ambon dan tidak diuji tantang (K-), pakan tanpa pemberian ekstrak batang pisang ambon dan diuji tantang (K+). Masing-masing perlakuan terdiri atas tiga ulangan. Frekuensi pemberian pakan empat kali sehari selama 29 hari pemeliharaan. Hasil penelitian menunjukkan bahwa perlakuan C respons imun (rata-rata jumlah total hemosit: 45,15×106 sel/mL, aktivitas phenoloxydase 1,03±0,08 OD, respiratory burst 0,95±0,04 OD, aktivitas fagositik 94,33±1,53%, pertumbuhan (laju pertumbuhan spesifik: 7,79±0,06%/hari, rasio konversi  pakan 0,52±0,01), dan kelangsungan hidup  (survival rate 100%) lebih tinggi dibandingkan dengan perlakuan K+ (jumlah total hemosit: 3.83×106 sel/mL, aktivitas phenoloxydase 0,04±0,01 OD, respiratory burst 0,18±0,06 OD, aktivitas fagositik 5,67± 0,58%, laju pertumbuhan spesifik: 2,61±0,08%/hari, rasio konversi pakan 2,11±0,02, nilai kelangsungan hidup: 50%). Jadi, ekstrak batang pisang pada dosis 0,5 g/kg setiap hari pada pakan dapat digunakan untuk meningkatkan pertumbuhan dan sistem kekebalan tubuh nonspesifik terhadap penyakit bintik putih pada udang putih.   Kata kunci: udang putih, white spot, respons imun, ekstrak batang pisang ambon

Abstract

The research study of cadmium (II) metal ion adsorption in a solution by 2-mercaptobenzotiazole modified cellulose and unmodified cellulose had been done. The aims of this research are to know the ability of 2-mercaptobenzotiazole modified and unmodified celluloses for adsorbing cadmium (II) metal ion with the composition of modification. The modification processes of cellulose were done by the composition variation between cellulose and MBT of 1:1, 1:2, and 1:3. The adsorption process of metal ions by 2-mercaptobenzotiazole modified cellulose were done by shaking at pH 3, 4, 5, 6, 7, 8, and 9 with the effect of metal ions concentration of 0.05; 0.1; 0,15; 0,2, and 0,25 mg/L. The analysis of functional groups of cellulose-MBT and cellulose unmodified was using FTIR Spectrophotometer. The analysis of cadmium (II) concentration metal ions as cadmium metals were done by atomic absorption spectrophotometer (AAS). The result of this research showed that on the functional groups which derived from FTIR spectrum, there was some similarity of functional groups between unmodified cellulose and MBT-cellulose which could be seen from the wave length of C-S stretching at 663.51 cm-1; functional group of C-H aromatic stretching at 3749.62 cm-1; functional group of C-N at 1319.31 cm-1, and functional group of C=N that came from diazole  functional group (-N=C-S) which could be seen at 1597.06 cm-1. The optimum adsorption metal ions cadmium by MBT-cellulose happened at 1:2 composition modification, pH 4, and at the concentration of 0.2 mg/L.   Key words: adsorption, modified, cadmium (II) metal ion, cellulose, 2-merkaptobenzotiazole

Abstract

The study was objective to describe students? sight on the subject delivery through the classroom-task-based on the subject of fine art learning evaluation. The subject of the study was all students of the Fine Art Program about 45 students. The study was utilized descriptive qualitative method.  The required data was collected by means of questionary, interview, and documentation schemes. Data has gathered to be utilizing questionary, observations sheet, interview sheet, and camera.  The data was analyzed qualitatively descriptive scheme. Result of the study was suggested that classroom-task-based subject delivery may applied in good manner for the subject of fine art learning, that is could be support students to decrease their problem in acquiring learning contents, to support student?s performance to finish all questions of the related subject, students is to be enjoyful, that is promotes their motivation and active in their class, students quickly comprehends delivered subject, students? is to be facilitated in finishing their assigned tasks.

Abstract

Deoxyribonucleic acid (DNA) vaccine has recently been developed as an alternative vaccine against virus infection.This study was the first step of DNA vaccine development to protect cyprinids including common carp (Cyprinuscarpio) and fancy koi (Cyprinus carpio) from KHV (koi herpesvirus) infection in Indonesia. One of KHV glycoproteingenes, i.e. glycoprotein (GP) was ligated with Japanese medaka (Oryzias latipes) â-actin promoter to generatepAct/GP as a DNA vaccine. Fourty fish in body weight of 10-15 g/fish were individually injected by pAct/GP intomuscle in different dosage of 2.5 ?g, 7.5 ?g and 12.5 ?g/100 ?l phosphate buffer saline. Total RNA was extractedfrom the 12.5 ?g of pAct/GP-injected fish muscle at 24, 48 and 67 hours post-injection to analyze GP expression byRT-PCR method. Potential of pAct/GP as DNA vaccine was examined by injecting KHV into the 30-days-vaccinatedfish. Both of possitive and negative control fish group were not vaccinated. Possitive control fish group wereinjected with KHV, but negative control fish group were not. KHV-challenged fish were reared for 1 month, and thedeath fish were calculated daily. Result of RT-PCR analysis showed that GP gene expression were detected at 3 dpost-injection. Expression of GP in the vaccinated fish groups helped to improve their survival rate after challengedby KHV. All of fish without DNA vaccination had dead 17 days after KHV injection. The results demonstrated thatpAct/GP had high potency to be used as a DNA vaccine against KHV infection in cyprinids.
