Abstract

Pada tahun 1997, dunia dikejutkan oleh suatu film dokumenter dalam program Tomorrow`s World serial BBC. Di dalam tayangan BBC menyiarkan tentang Vacanti Mouse, seekor tikus dengan daun telinga yang tumbuh di punggungnya. Sekalipun penelitian aslinya (seperti yang diakui oleh kelompok Cao dan Vacanti) yang berlangsung di laboratorium MIT (Massachussets Institute of Technology) tidak se-spektakuler seperti tayangan BBC, sejak saat itu istilah rekayasa jaringan (tissue engineering) menjadi istilah yang dikenal oleh jutaan masyarakat dunia. Harapan manusia akan adanya The Body Shop (yang setiap waktu dapat menyediakan organ tubuh pengganti) menuai titik terang.

Abstract

Reconstruction of large bone defects caused by trauma, excision of tumors, and congenital malformations can be very difficult to perform. Bone engineering offers an option to improve bone reconstruction procedures. This interdisciplinary field applies the principles of biology and engineering to the development of functional substitutes for damaged bone. Our research aimed to find the ideal scaffold for bone regeneration, focusing on Calcium and Phosphate combination. In this study, Plaster of Paris (POP) was combined with CHA and implanted in femoral condyles of rats. According to the experimental result, it can be concluded that there was no significant difference in response to the implantation of POP and POP-CHA in Sprague Dawley rat femur condyle (p<0.05). It can be stated that both POP-CHA and POP shows similar trait in bone healing.

Abstract

The field of life sciences moves forward at a rapid pace. Many of us do not fully realize that this acceleration is relatively new phenomenon in the history of mankind. It was in 1990 before the era of wireless phones and laptop computers when J. Craig Venter flew over the ocean and thought inside the airplane about the future of genome projects (as it was written by Ted Anton on his book entitled “Bold Science, Seven Scientists Who Are Changing Our World”).However, just over a short period of time, genome projects, which in 1997 required several years to complete, nowadays take only days. J. Craig Venter stated, as reported by Albert Sasson about the achievements, prospects, and perceptions of medical biotechnology, “For the past 15 years at ever faster rates we have digitizing biology. By that I mean going from the analog world of biology through DNA sequencing into the digital world of the computer. The human genome is perhaps the best example of digitizing biology. Our computer databases are growing faster per day than during the first 10 years of DNA sequencing. Thedatabases have been filling even faster with the results of J. Craig Venter’s global ocean sequencing project. As a result, we have now over 10 million genes in the public databases.” Indeed life sciences progress rapidly.In other areas, nanotechnology, for example, offers new solutions for biomedical sciences and provides a broad technological platform for application in industry. Such applications include bio-processing, molecular medicine, biotechnology, bio-fluidics for handling DNA, and other molecules, etc.Moreover, in another area, regenerative medical therapy reaches the interaction between scaffold, signaling molecules, and cells to enable neo-tissue formation and enhance healing potentials.All the above progresses in life sciences and biomedical sciences improve possibilities for patients to be helped. Meanwhile, ethical problems also arise by the fast science progresses.What about in oral and dental sciences? In some countries, the prevalence of oral and dental diseases is still considered very high. In some other countries the prevalence of dental caries is reducing by the application of fluoridation in societies. Although science has made human beings wealthy and prolonged life span, but to some extent scientists realize the limitation of sciences to cope health problems. In term of oral and dental sciences, opinions arise from research results and the confrontation of them also happens.In view of the overall mapping, The Indonesian Journal of Dental Research is published by The Faculty of Dentistry, Universitas Gadjah Mada, in collaboration with AFDOKGI, The Indonesian Association of The Faculty of Dentistry. The Indonesian Journal of Dental Research is an international journal dedicated to the latest advancement of dental research in Indonesia and Asia Pacific regions. Abstract

Background: The increasing of osteoblast activities during bone formation will be accompanied with the increasing expression of alkaline phosphatase enzyme (ALP). ALP can be obtained from clear fluid excreted by gingival crevicular fluid (GCF). Bone turnover, especially bone formation process, can be monitored through the expression of ALP secreted by GCF during orthodontic treatment. Thus, retention period is an important period that can be monitored through the level of bone metabolism around teeth. Purpose: This research were aimed to determine the relation of distance change caused by tooth relapse and ALP activities in gingival crevicular fluid after orthodontic; and to determine ALP as a potential biomarker of bone formation during retention period. Methods: Lower incisors of 25 guinea pigs were moved 3 mm to the distally by using open coil spring. Those relapse distance were measured and the gingival crevicular fluid was taken by using paper points to evaluate ALP levels on days 0, 3, 7, 14 and 21 respectivelly by using a spectrophotometer (405 nm). t-test and ANOVA test were conducted to determine the difference of ALP activities among the time intervals. The correlation regression analysis was conducted to determine the relation of distance change caused by the relapse tooth movement and ALP activities. Results: The greatest relapse movement was occurred on day 3 after open coil spring was removed. There was significant difference of the average of distance decrease among groups A1-A5 (p<0.05). It was also known that ALP level was increased on day 3, but there was no significant difference of the average level of ALP among groups A1-A5 (p>0.05). Finally, based on the results of correlation analysis between the ALP level decreasing and the relapse distance on both right and left of mesial and distal sides, it is known that there was no relation between those two variables (p>0.05). Conclusion: It can be concluded that relapse after orthodontic tooth movement occurs rapidly as the teeth are free from orthodontic force. ALP level can be detected through gingival crevicular fluid during relapse by using a spectrophotometer.Latar belakang: Peningkatan aktivitas osteoblas selama pembentukan tulang akan disertai peningkatan ekspresi enzim alkalin fosfatase (Alkaline Phosphatase/ALP). Sumber ALP dapat diperoleh dari cairan bening yang diekskresi celah gingiva gigi yang dikenal sebagai cairan krevikuler gingiva (gingival crevicular fluid/GCF). Bone turnover terutama proses pembentukan tulang dapat dimonitor melalui ekspresi ALP cairan krevikuler gingiva selama perawatan ortodonti. Periode retensi merupakan periode yang penting. Kesulitan memecahkan masalah retensi akan dapat ditangani dengan memonitor tingkat metabolisme tulang disekitar gigi. Tujuan: Penelitian ini bertujuan untuk meneliti perubahan jarak relapse gigi dan aktivitas ALP pada cairan krevikuler gingiva setelah digerakkan secara ortodonti dan potensi ALP sebagai biomarker pembentukan tulang pada periode retensi. Metode: Gigi insisivus bawah 25 ekor marmot digerakkan ke distal menggunakan opencoil spring sampai mencapai jarak ± 3 mm, diukur gerakan relapse gigi dan pengambilan cairan krevikuler gingiva menggunakan paper point untuk dievaluasi kadar ALP pada hari ke 0, 3, 7, 14 dan 21 menggunakan spektrofotometer (405 nm). Analisis t-test dan anova untuk mengetahui perbedaan aktivitas ALP antar interval waktu dan analisis regresi korelasi untuk mengetahui hubungan besarnya jarak relapse dengan aktivitas ALP. Hasil: Pergerakan relapse yang paling besar terjadi pada hari ke 3 setelah opencoil spring dilepas. Terdapat perbedaan rata-rata penurunan jarak antar kelompok A1-A5 yang signifikan (p<0,05). Kadar ALP mengalami peningkatan pada hari ke 3, namun tidak terdapat perbedaan rata-rata yang signifikan kadar ALP antar kelompok A1-A5 (p>0,05). Hasil uji korelasi antara penurunan jarak dengan kadar ALP pada mesial distal gigi baik kanan maupun kiri tidak menunjukkan adanya hubungan kedua variabel (p>0,05). Simpulan: Relapse pada perawatan ortodonti terjadi secara cepat ketika gigi terbebas dari gaya ortodonti. Kadar ALP dapat terdeteksi dari cairan krevikuler gingiva pada pergerakan relapse gigi menggunakan spektrofotometer.

Abstract

The clinical success of current generation of synthetic bone substitute relies on bio-inspired design which has a performance level close to that of natural one. In this context, biomedical approaches are considered very important to result bio-functional hybrid for bone substitution purposes. In this study, effect of cross-linking to the mechanical properties of apatite gelatin hybrid has been investigated. Cross-linking was employed by 1-ethyl-3-3-dimethylaminopropyl carbodiimide (EDC) agent. The EDC agent creates a peptide bond between gelatin molecules inside the hybrid to the cross-linked structure. Cross-linked structure of gelatin increases physical property of the hybrid since it can hold the outer forces longer than that of without cross-linking.

Abstract

Latar belakang: Kerusakan jaringan oleh patah jaringan, trauma, atau penyakit memerlukan restorasi untuk memperbaiki fungsinya seperti sedia kala. Beberapa penelitian telah dilakukan untuk menemukan bahan-bahan substitusi jaringan, termasuk mengembangkan bahan hidroxyapatite (HAp). HAp merupakan bahan bioaktif yang memiliki kemampuan osteokonduktif, bioaktivitas, dan biokompatibilitas dalam proses pembentukan tulang. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh substitusi tulang dengan hidroksiapatit (HAp) pada gambaran histologis tulang pada tibia tikus (Rattus Sprague Dawley).Penelitian dilakukan dengan 15 tikus jantan Sprague Dawley, usia 3 bulan, dengan membuat defek di tibia kanan dan tibia kiri. Setiap defek berukuran 3 mm x 1,5 mm x 1 mm. Sebagai perlakuan diberikan implantasi hidroksiapatit pada defek tibia kanan dan tibia kiri sebagai situs kontrol tanpa implantasi. Masing-masing subjek didekapitasi setelah 1, 2, 3, 4, dan 8 minggu. Daerah defek diambil dan dibuat gambaran histologi, kemudian dilakukan pengamatan dan penghitungan jumlah sel osteoblas, dan osteoklas menggunakan mikroskop kontras fase.Data dianalisis oleh SPS 2000 dengan metode Two Way Anava. Hasil penelitian menunjukkan bahwa substitusi tulang dengan hidroksiapatit tidak signifikan pada aktivitas osteoblas atau osteoklas ditunjukkan dengan p>0,05, tapi analisis antar waktu menunjukkan signifikasi pada osteoklas (p<0,05). Investigasi pada gambaran histologi menunjukkan peningkatan pada aktivitas pembentukan tulang baru pada defek tulang. Kata kunci: substitusi tulang; hydroxyapatite; sel-sel tulang; gambar histologi

Abstract

Background: The subject of dental health care has been receiving a lot of attention from Moslems in Indonesia, especially its accordance with Al-Qur’an and Sunnah. It has encouraged numerous researches which corelate dental health care through Islamic perspective, and ignored the perspective of dentistry. Consequently, this research is aimed to develop, formulate and validate the competence of moslem dentists.Method: Experts’ agreement was develop using Delphi method. The result then was developed into an instrument with 2 criteria was used to determine content validity index (CVI) and content validity ratio (CVR). The result was developed into a 5-point Likert scale instrument which would be used to determine internal consistency (Cronbach’s alpha) and corrected item–total correlation with Pearson’s correlation.Results: The qualitative method generated 4 domains. Domain shiddiq comprised 2 components, domain amanah 2 components, domain tabligh 2 components and domain fathonah 4 components with the value of agreement of each domain was approximately between 67% - 100%. The CVR value of the components of competence was between 0,69 – 1, while the CVI value was 0,877. The reliability test scored 0,923.Conclusion: The Competence Standards of Islamic Dentistry can be used as guidance for designing Islamic dental curriculum. Further research should be conducted to design assessment.

Abstract

This study aims to synthesize simvastatin hydrogel as drug delivery system with surfactant addition for improving solubility of simvastatin. Surfactants used in the study were the zwitterionic amino acid of arginine and nonionic surface-active agent of polysorbate 80. The solubility study was conducted by pouring of an excess mass of simvastatin into the solution of a surfactant in a conical flask. The sample was shaken up to 72 h in a mechanical water bath shaker at a varied temperature of 25, 40, and 50 °C. The amount of drug dissolved in solution was analyzed by UV/Visible spectrophotometer at 238 nm. The results showed that the simvastatin solubility is profoundly influenced by the surfactant type, surfactant concentration, and temperature. Polysorbate 80 exhibited as a better surfactant than arginine and an enhancement up to 1400 times, in respect of without any addition of a surfactant, was observed. Based on the solubility study, simvastatin-loaded gelatin hydrogel composite was formulated and the characterization (FTIR and SEM) showed the successful impregnation. The hydrogel microparticles of featured swelling indexes in the range of 2–6 for every patch and presented a sustained release profile.
