Abstract

Cardiovascular disease is related to the high consumption of trans fatty acid. This issue has driven SEAFAST IPB Laboratory to develop a trans fatty acid analysis method for gas chromatography based on AOCS Official Method Ce 1h-05. This research was aimed to validate the method via three steps: preparation step, procedure orientation test, and method validation. During instrument performance testing in the preparation step, linearity, limit of detection (LoD), limit of quantitation (LoQ), range, and precision parameters were validated for FAME 9t-C18:1 and C18:2 isomer mixed standard solution. During the method validation step, specificity, precision and accuracy parameters were validated for 9t-C18:1 compound attached to food matrix, i.e. doughnut. The instrument validation result of 9t-C18:1 standard solution had fulfilled the criteria of linearity (r2=1, requirement r2 > 0.990) within the range of 0.0020 to 0.1200 mg/mL; precision (RSD) 0.24%; LoD 0.0007 mg/mL and LoQ 0.0020 mg/mL. The instrument validation result for each isomer in C18:2 standard solution also met the criteria of linearity (r2= 0.9999, requirement r2 > 0.990) with precision (RSD), LoD and LoQ respectively; 1) 9t,12t-C18:2: 1.40%; 0.0033 mg/mL; 0.0100 mg/mL; 2) 9t,12c-C18:2: 1.89%; 0.0015 mg/mL; 0.0045 mg/mL; 3) 9c,12t-C18:2: 2.39%; 0.0015 mg/mL; 0.0044 mg/mL; 4) C18:2 cis: 3.39%; 0.0006 mg/mL; 0.0019 mg/mL. The validation result for 9t-C18:1 in 100 mg fat extract of doughnut suggested that the modified method is validated in term of specificity (Rs = 1.2), precision (RSD fatty acid content (mg/g) = 1.78%; RSD percentage of fatty acid content towards total fatty acid = 0.72%) and accuracy (as percentage of recovery) 80.54-94.25%.

Abstract

Deterioriation of palm oil fortified with vitamin A and provitamin A could be caused by the presence of oxygen and light exposure. The purpose of this study was to determine the effect of the initial peroxide value (PV) in palm oil (1.99, 3.98, and 9.95 meq O2/kg oil) and light intensity (15000, 10000, and 5000 lux) on the rate of oxidation and the shelf life of palm oil fortified with Red Palm Oil (RPO) equal to 45 IU vitamin A. The RPO contained ?-carotene as provitamin A in the amount of 504.67 ppm. The PV and free fatty acid (FFA) content were observed as the parameters of oil deterioriation during storage. The results showed that the rate of PV was influenced by light intensity, while the rate of FFA formation were more influenced by the amount of initial PV in the oil. Based on the palm oil standard quality for PV (SNI 7709: 2012), the shelf life of palm oil with the lowest initial PV at ambient temperature was 9.5 days, while that with the highest PV was 1.32 hours. The deterioration rate of RPO fortified palm oil due to light exposure was also compared with its deterioration rate due to heat and the deterioration rate of palm oil fortified with vitamin A. The shelf life of vitamin A fortified palm oil stored in the dark was 90.67 days, while RPO fortified palm oil was 68.12 days. This shelf life results showed that RPO had a potency as provitamin A fortificant for palm oil as long as it is stored in a closed container in the dark.

Abstract

Mono and Diacylglycerol (MDAG) is one of the food additives that can be used for stabilizing food emulsion system. MDAG is commercially manufactured through glycerolysis reaction at high temperatures and catalyzed by inorganic alkaline such as NaOH. This process results in an equilibrium mixture containing partial glycerol esters and excess of glycerol. The present study describes the effect of reaction temperature and time in a 50 kg-pilot plant production on the chemical properties of the resulting MDAG emulsifier. Fully hydrogenated palm stearin (FHPS) and NaOH were used as the substrate and catalyst, respectively. The production was conducted following a factorial experiment in a completely randomized design with two variable combinations, i.e. temperatures at 170, 180, and 185°C; and times of 120 and 180 min. Each experiment combination was conducted in two replications. The glycerolysis reaction at 180°C for 180 min produced the highest yield of MDAG, i.e. 87.2±1.41%, which comprised of 48.78±2.28% monoacylglycerol (MAG), 30.92±1.23% diacylglycerol (DAG), 4.79±0.61% triacylglycerol (TAG), 1.97±0.04% free fatty acid (FFA), and 2.35±0.05% free glycerol.

Abstract

The study was conducted to analyze the association between food consumption and physical activity with blood cholesterol level. The study design was cross sectional located in the District and the Municipality of Bogor. Subjects in this study were 64 adult men and women aged 25?60 years. Stepwise regression test results indicate that the intake of protein, carbohydrate and cholesterol did not significantly affected blood cholesterol levels. Physical activity level and gender significantly affected blood cholesterol levels (p<0.05). Dietary fiber intake and fat intake significantly affected blood cholesterol levels (p<0.10).

Abstract

Anthocyanin is a natural pigment with color varying from red, purple, blue to yellow. The stability of its anthocyanin color is affected by pH, temperature and light. Purple sweet potato is rich in anthocyanin, particularly a stable acylated anthocyanin. This research was conducted to study the effect of pH on color and antioxidative activity of anthocyanin extracted from purple sweet potatoes harvested from Cilembu-Sumedang, Banjaran-Bandung, and Pakembangan-Kuningan. The experiments applied a completely randomized design with two replicates analyzed triplo.The results showed that the total number of monomeric anthocyanin in purple sweet potato harvested from Cilembu (3.78±0.08 mg cyanidin-3-glucoside/g dry weight, dw) was higher than that of Banjaran (3.18±0.01 mg/g) and Pakembangan (2.25±0.01 mg/g). The color of purple sweet potato anthocyanin extract was pH dependent. The color changed from red, faded red, purple, blue, green and yellow along with the increase of pH from 1 to 14. The content of anthocyanins from three locations of purple sweet potatoes differed from each other (p<0.05). Radical scavenging activity and reducing power of purple sweet potato anthocyanins extract at pH 1 was higher than that at pH 4.5 and pH 7.

Abstract

ABSTRACTWaterleaf shoot (Talinum triangulare (Jacq.) Willd) is consumed as vegetable that contains some metabolites. The purpose of this research was to investigate the effect of organic fertilizer on shoot production and its metabolites i.e. protein, vitamin C, flavonoid, and peroxidase enzyme (POD) activity at 10, 12, and 14 weeks after planting. This experiment was conducted at Leuwikopo Experimental Station, Bogor, Indonesia from November 2012-February 2013. The organic fertilizers applied were cow manure (CM) (12.3 ton ha-1), rock phosphate (RP) (1.5 ton ha-1), and rice-hull ash (RH) (5.5 ton ha-1). These organic fertilizers were combined into four treatments by using minus one test method and one control. Each treatment was repeated three times. The result showed that combination of organic fertilizer had the same effects on shoot production and metabolite content of waterleaf. It showed that the amount of organic fertilizers was not significantly sufficient to contribute nutrients to the plant. Keywords: flavonoid, POD activity, repeated harvesting, Talinum triangulare, vitamin C

During growth and development phase, body needs macro and micro nutrients even more than later phase. The process can not be catched up at later stage therefore it is very important to pay attention to food consumption during this phase. This study used secondary data of South East Asian Nutrition Surveys (SEANUTS) 2011-2012 which was covered recapitulation profile data of 300 children age 2-12 years old representative of 48 districts in Indonesia. The study showed that Indonesian children consuming less calcium and vitamin D-rich foods than the recommended daily allowance (RDA). It does not reflects on the result of anthropometry data, vitamin D on the blood and bone mass density since the dietary recall 24 hours shows only food consumption in that specific day while nutritional status shows a result of longer process. However, vitamin D in blood has a significant correlation with bone mass density of tibia bone (p<0.05). Deficiency of calcium and mainly vitamin D is a new finding at this time. Therefore, the study showed that fortification of calcium and vitamin min D in food become important to improve nutritional status of Indonesian children. 20% of RDA per serving size twice a day is recommended as well for fortification level of Calcium and Vitamin D.

Abstract

ABSTRACTBangun-bangun [Plectranthus amboinicus] is a functional vegetable that is used as lactagogue. This research was aimed to provide information of the effect of organic fertilizer on shoot, total phenolic, and anthocyanin production of bangun-bangun. This experiment was conducted at Bogor Agricultural University, Leuwikopo experimental station (Indonesia), from December 2012 to February 2013. The experiment was laid out in randomized block design with single factor with five combination of organic fertilizer treatments (combination of cow manure 12.3 t ha-1, rock phosphate 1.5 t ha-1, rice-hull ash 5.5 t ha-1) with three replications. The result showed that application of organic fertilizer increased of shoot production. Application of 12.3 t ha-1 cow manure + 1.5 t ha-1 rock phosphate + 5.5 t ha-1 rice-hull ash produced shoot dry weight ha-1 (57.33%) and metabolite production ha-1 (total phenolic 12.06%, anthocyanin 41.73%) higher than no fertilizing (P > 0.05). Application of cow manure + rock phosphate produced the lowest shoot dry weight ha-1 and metabolite production ha-1. The result of this research suggested that nitrogen, phosphorus, and potassium were needed on shoot production of bangun-bangun.Key words: Plectranthus amboinicus, phenylalanine ammonia lyase, secondary metabolite

Abstract

Sea snail gonggong is an icon of Tanjungpinang-Riau Islands Province. It is a favorite seafood item in Riau Islands Province, and is high economic value but not known widely yet. Until now, sea snail gonggong has been highly exploited but the research on this snail is very limited. The aim of this study was to morphology and molecular characterization of Bintan gonggong snail (Strombus sp.) as a species validation. Bintan gonggong snail included thick-shelled gonggong and thin-shelled gonggong. Morphology identifcation of species Bintan gonggong snail was based on morphometric variability. Molecular identifcation used partial Histone-H3, MEGA version 6.06, and bioinformatics analysis. The result showed that the morphological identifcation of thick-shelled and thin-shelled gonggong based on shell width, the lip thickness, and total weight signifcantly di?erent, but other variables (i.e shell length, shell depth, aperture length, and gonggong weight) were not signifcantly di?erent (p<0.05). Resulted of a molecular identifcation with phylogenetic analysis that the thin-shelled and thick-shelled Bintan gonggong snails were 1 species and a genetic distance of 1%. They were not species Strombus canarium, Strombus vitatus, and Strombus epidromis. Bintan gonggong snails were Strombus turturella (Leavistrombus turturella). DNA sequences of Bintan gonggong have been registered in Gen-Bank with registration numbers MH348131 (thinshelled gonggong) and MH348132 (thick-shelled gonggong).

Abstract

he objective of research was to get the chemical composition of red algal(E. spinosum) from different coastal of water that have the potensial as a sourceof carrageenan. The analysis was conducted to analyze the chemical content ofthe algal (E. spinosum) of three coastal water were moisture content, ash content,protein content, fat content, carbohydrate content and dietary fiber content. Theresults of analysis chemical composition of red algal from different waters hadshowed that content of moisture content 19.55-21.27% , ash content 18.55-18.95%, protein content 4.85-5.59% content, fat content of 0.06-0.1% andcarbohydrate content 53.44-55.52 % while the total dietary fiber content of redalgal (E. spinosum) was 12.78-15;92%. The chemical composition of red algal E.spinosum of three coastal water no real effect unless total dietary fiber contentwas significantly diffrence between the coastal water.
