Abstract

Dengue hemorrhagic fever (DHF) is major public health problem in tropical and subtropical areas of the world with lack of approved vaccines and effective antiviral therapies. With no current treatment for illness attributed to dengue virus (DENV) infection other than supportive care, therapeutic strategies that use natural extract was developed. Indonesia have many plants that potential for antiviral drµgs such as Pterocarpus indicus Willd (P. indicus). The objective of this study was to determine the effect of P. indicus to inhibit DENV replication. We used a well-differentiated hepatocytes-derived cellular carcinoma cell line (Huh-7 it-1 cells) to determine and select antiviral activity. The toxicity effects were determined by MTT assay. Then, the suppression of DENV replication was determined by Focus assay. Dengue infected cells with DMSO were used as control. We found that crude extract (Pi), hexane (Pi.1) and ethyl acetate (Pi.2) extract showed strong inhibition with high selectivity index (SI) of 1,392; 285.36 and 168.56 respectively.  Sub fraction of Pi.1 and Pi.2 still showed strong inhibition with high SI.  Further sub-sub fraction of Pi.2 such as Pi.2.12 and Pi.2.12.1 still showed inhibition of DENV replication but there was reduction of SI value. The mechanism experiment of Pi.2.12, we found that Pi 2.12 more profound to inhibit in the post infection stage that entry or pre-infection. We conclude that the sub-fraction of Pi.2.12 has potential antiviral activity against DV infection in vitro. Further studies are still needed to investigate the pure compound of Pi.2.12 that inhibit and have advantages in the future as alternative for treatment of DENV infection.

Abstract

Apoptosis  is  an  important  target  on  anticancer  mechanism. The  purpose  of  this  research  is  to  investigate  apoptosis  induction of Selaginella plana Hieron active fractions on T47D cells. Absolute ethanol  was  used  to  extract Selaginella  plana powders.  Ethanolic extract  was  dilluted  by  methanol:water  (4:1)  and  then fractionated  by  hexane  (S_Hex),  methylene  chloride  (S_MTC), ethyl acetate (S_EA), and buthanol (S_BuOH). The proliferation of T47D  cell  line  was  detected  by  SRB  (Sulforhodamine  B) assay which  was  measured  at  a  wavelength  of  515nm.  Flowcytometry analysis  to  determine  apoptosis  was  examined  by  Propidium Iodide  (PI)  and  Annexin  V  assay  using  T47D  breast  cancer  cell line.  The  result  showed  that  the  IC50 value  of  S_Hex,  S_MTC, S_EA,  and  S_BuOH  on  T47D  cells  were  107  µg/mL,  4  µg/mL,  6 µg/mL,  and  17  µg/mL  respectively.  The  active  fractions  (S_MTC and  S_EA)  at  its  IC50 concentration  significantly  (P<0.05) increased  the  total  number  of  early  apoptotic  cells  in  the  T47D cells  3.39%  and  4.1%  respectively  compared  to  that  of  control (1.95%).  Based  on  the  result,  methylene  chloride  and  ethyl acetate  fraction  of Selaginella  plana induced  apoptosis  on  T47D cell.Keywords: apoptosis, breast cancer, Selaginella

Abstract

Peperomia pellucida L. Kunth known as “Katumpangan Air” is a potential medicinal plants used traditionally for uric acid, rheumatic, headache, stomachache, antimicrobial. This study aims to establish  the non-specific  and specific parameters of ethanol extract from Katumpangan air (EKA). EKA was collected from three different growth places: South Tangerang (6.833%), Bogor (7.8%) and Yogyakarta (13.125%). The results show that organoleptic extract characterization from three places were same: thick, blackish green brown, bitter and have a characteristic odor. The average content of water-soluble compounds was within range 7.39-13.29%, ethanol-soluble 15.33-16.68%, flavonoid total 3.807-4.244%. The range of moisture content was 12.25-16.34%, ash total 1.21-2.78%, acid insoluble ash 0.19-1.62%, drying shrinkage 21.62-24.98%, specific gravity 1.0010-1.0034 g/mL for the non-specific parameter. Microbial contamination testing results 0.61x103-1.13x103 coloni/g, the contaminant testing mold/yeast 0.1x102-1.7x102 coloni/g, while lead metal 0.15-0.18 mg/kg, cadmium 0-0.11 mg/kg and arsenic <0.005µg/kg.

Abstract

Background: Hepatitis C virus (HCV) is a leading cause of chronic liver diseases. Drug resistance to the regimen is also increasing. Hence, there is a need for new anti-HCV agents that are less toxic and more efficacious. The aim of this study is to evaluate the  possibility of A. pauciflorum extracts can be a antiviral drug.Methods: Huh-7it cells were infected with the HCV genotype 2a strain JFH-I in the presence of methanol extracts of Archidenron pauciflorum. The methanol extract further partition used n-hexane, ethyl acetate, n-butanol, and water showed in which butanol extracts exerted the strongest IC50 (6.3 g/ml). Further, the butanol fraction was fractionated and yielded into 13 fractions.Results: The methanol extract of the leaves of A. pauciflorum exhibited concentration dependent inhibition against the JFH1 strain of HCV genotype 2a with an IC50 is 72.5 μg/ml. The butanol fraction exhibited the highest anti-HCV activity with an IC50 is 6.3 μg/ml. The butanol fraction was fractionated which yielded 13 fractions. Fractions 5 and 13 exhibited high anti-HCV activities with IC50 is 5.0 μg/ml and 8.5 μg/ml and a time-of-addition study demonstrated that fraction 5 inhibited viral infection at the post-entry step, whereas fraction 13 primarily inhibited the viral entry step.Conclusion: The extract A. pauciflorum can be used as a herbal-based antiviral drug.

Abstract

Selaginella sp belongs to the Selaginellaceae family. It has been used in China and Indonesia as a traditional medicine. It hasseveral medicinal properties including antibacterial, anticardiovascular, and anticancer agent. The aim of the present study was to access the anticancer property of the ethanolic extracts of Selaginella plana Hieron. on T47D breast cancer cell line. The proliferation of T47D cell line was detected by SRB (Sulforhodamine B) assay which was measured at a wavelength of 515 nM. The result showed that the IC50 of the ethanolic extract was determined at 7.03 pg/mL. This significant activity was assumed due to its high total flavonoid content. The totalflavonoid content of the ethanolic extract was 23.04%. Flow cytometry analysis indicated that the extract may undergo the cell death via apoptosis pathway. In conclusion, the ethanolic extract of Selaginella plana Hieron: has considerable activity in inhibiting T47D cell line proliferation.Key words: Selaginella plana Hieron., SulforhodamineB, T47Dcells, anticancer flow cytometry

Abstract

The preparation of Brucea leaves have been done for specific parameters and non specific parameter laboratory worb refers to TheNational Agency of Drug and Food Control Regulation for extract quality control. The result for specific extract parameters were the rendement of extraction is 28%, the values of water- extractive and ethanol-extractive are 11.49% and 9.41%, respectively. The result of determination of the non specific parameters of the extract are; loss on drying is 18.95%, the water content is 15.06%, the total ashes is 16.12%. and the level of ashes not dissolved in acid is 11.14%. The analysis of chemical compound shows that the extract containedflavonoid, tannin, and glycoside. Abstract

The flavone glycoside was isolated from ethyl acetate fraction of ethanol extract of leaves Pterocarpus indicus Willd. The isolation was conducted by gravitation column chromatography and eluted successively with hexane, ethyl acetate and methanol by gradient, and purified by sephadex-LH20. The structure was elucidated base on spectroscopy data of NMR (1D and 2D), UV, LC-MS and FT-IR. Antioxidant was evaluated using 2,2-diphenyl-1-picrylhidrazyl (DPPH) radical scavenging. The isolation and identification led a stigmasterol as Compound 1 and a new flavonol-glycoside [(2R)-7-hydroxy-3-(3,4,5-trihydroxy-6-(hydroxymethyl)tetrahydro-2H-pyran-2-yloxy)-2-(3,4,5-trihydroxy phe nyl)chroman-4-one] or ptevon-3-D- glycoside as Compound 2. Antioxidant activity of Compound 2 showed IC50 for 18.53 mmol and blank of quercetin was 7.94 mmol and Vitamin C was 40.25 mmol. These compounds and antioxidant activities are the first time reported from this plant.

Abstract

Ketumpangan air (Peperomia pellucida (L.) Kunth), an Indonesian wild plant, is commonly found in every Indonesian region with moist shady place. People usually consume ketumpangan air as a fresh vegetables. Empirically, ketumpangan air is consumed to treat fever, stomach disease, kidney disorders, drug abscesses, boils acne, headaches, pain in arthritic and rheumatic gout, or other external treatment. In this research, we tested the acute toxicity of ethanol extract from ketumpangan air using experimental animals mice DDY strain. We also tested the Brine Shrimp Lethality Test (BSLT) using Artemia salina Leach larva shrimp. The main aim of this reseach was to determine the LD50 and LC50 values of ethanol extract from Ketumpangan air. The results showed that the ethanol extract was categorized as ‘mild toxic’. The calculation of LD50 with Reed and Munich method showed that the LD50 value for male and female group was 15.13 g/ kg and 11.87 g/kg respectively. The BSLT result showed that the LC50 values of Ketumpangan air was 81.28 ppm.

Abstract

Katekin merupakan salah satu senyawa bioaktif yang terkandung dalam gambir. Hasil studi pendahuluan menunjukkan bahwa katekin aktif sebagai antioksidan dan antibakteri, sehingga berpotensi sebagai oral nutraceutical pencegah plak gigi. Pada studi ini, akan dilakukan pembuatan oral nutraceutical dalam bentuk obat kumur dari ekstrak gambir dengan memvariasikan kandungan senyawa aktif katekin dari  ekstrak gambir sebagai parameter formulasi obat kumur, yaitu 0,01-0,1% (b/v). Obat kumur yang dihasilkan, dianalisa pH, aktivitas antioksidan, antibakteri, dan ukuran partikelnya. Hasil studi ini menunjukkan bahwa obat kumur dengan kandungan senyawa aktif katekin 0,01-0,08% (b/v) menghasilkan obat kumur berukuran nano.

Abstract

Dilakukan penelitian aktivitas anti agregasi platelet dan anti trombosis ekstrak etanol daun Cinco (Cyclea barbata Miers.) (Cb) secara in vivo pada mencit. Latar belakang dari penelitian ini karena telah dilaporkan sebelumnya bahwa Cinco mempunyai aktivitas anti agregasi secara in vitro. Tujuan penelitian ini untuk mengetahui efek inhibisi ekstrak etanol daun Cb terhadap agregasi platelet serta aktivitas anti trombosis, dan juga memperpanjang waktu pendarahan dan waktu koagulasi. Hasil penelitian menunjukkan bahwa ekstrak daun Cb secara signifikan dapat menghambat agregasi platelet, memperpanjang waktu pendarahan, memperpanjang waktu koagulasi, dan menghambat pembentukan trombus secara akut. Selain itu ekstrak etanol daun Cb juga memberikan perlindungan pada thrombus secara signifikan dengan efek menyeluruh yang tergantung pada dosis. Semua pengukuran diatas dianalisa kebermaknaannya dengan menggunakan program ANOVA Satu Arah. Ekstrak etanol daun Cb dengan dosis rendah (150 mg/kg) memperlihatkan perpanjangan waktu pendarahan yang signiiikan pada ekor mencit dibandingkan kontrol dengan efek tergantung pada dosis. Ekstrak etanol daun Cb secara signiiikan memperpanjang waktu koagulasi dibandingkan terhadap kontrol dengan efek tergantung pada dosis. Ekstrak etanol daun Cb pada dosis 150 mg/kg bb dan 300 mg/kg BB signiiikan menghambat agregasi dibandingkan dengan kontrol. Dapat disimpulkan bahwa ekstrak etanol daun Cb mempunyai aktivitas anti agregasi platelet dan antitrornbosis pada dosis 150 dan 300 mg/kg BB.
