Abstract

The aim of the research is to analyse germination percentage, effect of coconut water addition in the media to number of last stage embryos and development of final stage D. anosmum Lindl. embryo. Research was conducted at Plant Structure and Development of Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences University of Udayana and UPT. Balai Benih Induk Tanaman Pangan dan Hortikultura Provinsi Bali from February 2015 to July 2015. Three type of media were employed, i.e. PDA, Organic, MS. Three different concentration of coconut was added to media, i.e. 0 ml/L 50 ml/L, 100 ml/L. Results showed that seed D. anosmum Lindl. on PDA, Organic and MS has not germinated yet on all media, only swollen cells observed. Seed dormancy and maturity of seed were the main factors affecting seed germination.  Addition of 100 ml / L coconut water in MS showed that media is capable to increase the formation of final stages embryos (> 32 cells). Embryo development on PDA and Organic largely showed cell stage embryos 1 to 8 cells, while MS media already showed embryo development stages of > 32 cells.

Abstract

Broccoli (Brassica oleracea L. var. italica Plenck.) is a vegetable crop belongs to Brassicaceae family.  Broccoli has high nutrition, high in fiber and contains isotiacyanate that has anticancer activity.  Broccoli market in Indonesia, particularly in modern supermarkets, increases 15-20% per year, however, productivity was still low, therefore effort to increase broccoli production in Indonesia need to be investigated. Field trial was conducted at Batur Village, Kintamani District, Bangli Regency, Bali, to find out cultivars that were adapted in tropical region. Introduced cultivars being trialed were ?Alborada?,?Belstar?, ?Fiesta?, ?Sarasota?, ?Bay Meadows?, ?Castle Dome?, ?Liutenant?, ?Iron Man?, ?Marathon?, ?Green Gold?, ?Imperial?, ?Green Magic? and ?Lucky? as control.  Variable observed were vegetative growth, curding period, and productivity of each cultivar.  This study employed Randomized Completely Block Design with 3 replicates (plots) and 8 plants each plot.  Data were analyzed using ANOVA (Analysis of variance) on Costat program, followed by Duncan?s Multiple Range Test (DMRT) on 1% level.  Results show that each cultivar varied in adaptability in tropical region. ?Castle Dome? has the highest productivity, while ?Fiesta? was the lowest. Abstract

This study aims to determine the type of Hemerocallis fulva L. pollen, pollen viability and growth after storage at 280C, 40C, -200C, -800C temperature for 0-8 weeks. The study was conducted from January - March 2010. Pollen samples were taken from UPT. Botanical Gardens Plant Conservation 'Eka Karya? Bali. Pollen storage and observations was carried out at the Laboratory of Plant Biotechnology and Development Structure, Biology Department, Faculty of Science, Udayana University. Viability test were conducted using hanging drop media and agar media with GA3. The results showed that type of Hemerocallis fulva L. pollen were: monosulkat, Prolate spheroiodal, (54.25 X 47.47), P/E: 1.14, Zono, colpate, reticulate. Fresh pollen viability on hanging drop media was 8.33%, decreased to 4% after storage at 4°C for one week and -20°C for 5 weeks. Whereas on agar media, control pollen has viability of 3.17% and increased at -20°C to become 11.67%. Mature pollen can be observed at the level of uninukleat, binukleat, and trinukleat. The development of pollen with the media hanging drop on the control has a long reed 115.94 ?m and length increased at 4 ° C with storage duration ie 256.39 weeks to 1 ?m, whereas on an agar medium + GA3 to control reed 149.82 ?m in length and at 28 ° C, 4 ° C, -20 ° C and -80 ° C respectively 94.73 ?m; 142.1 ?m; 95.53 ?m; 118.67 ?m at 1 week storage time.

Abstract

The aim of the research was to determine pollen viability, pollen tube length and pollen development of white, red and super red dragon fruit after storage at different temperatures and times. The method used to test pollen viability was hanging drop technique and to observe the development of pollen used acetolysis techniques. The results showed viability and pollen tube length of white, red and super red dragon fruit after storage at temperature of 10° C and -20° C for 1, 2 and 3 weeks decreased (66% and 25%), tended to increase (2% ) after 4 weeks. Viability and pollen tube length decreased (100%) after storage at 30° C for 4 weeks. Pollen development of white, red and super red dragon fruit after storage at 30°C, 10°C and -20°C for 1 to 4 weeks showed the majority of pollen consists of uninucleat and binucleat.

Abstract

GiliRanggoFisheryReserve Areais one of conservation areaestablished throughthe Regional Regulationof East Lombok, West Nusa Tenggara number10year 2006 onRegionalMarine ProtectedArea Management(RMPAM). Sincethe enactment ofthereserve fisheries zone, managementsystemis not yet optimizedeitheronthe mangrove protected areasandstakeholdercommitmentto theconservationefforts ofthemangrove resource. The purpose ofthis study were 1) to analyzethe extent of mangrove damageinGiliRanggoFisheryReserve Area, 2) to find out perceptions of stakeholderson the management ofmangrovesin the area of GiliRanggoFishery at the reserve area, 3) to formulatea sustainablemanagementstrategyinvolvingallstakeholders. This study useda combination ofqualitativeandquantitativeapproaches. Data analysis methodsused include: vegetationanalysis, qualitativeanalysisandSWOT analysiskatagorik. The results showed that the destruction level of mangrove damage at GiliRanggoFishery Reserve Areaon seedlings and saplings phase are trees are relatively minor with density <50% and widespread closure> 1,500 trees/ha, whereas severely damaged represented by closure <50% and a density of <1,000 trees/ha. Mangrove vegetation in the GiliRanggoFishery Reserve Areais not classified as critical represented by the Total Value Scoring (TNS) = 370. Society supports the efforts to conserve the mangrove, as people realize the key benefits of mangroves. Violations that occurred during this due to people not knowing the rules of forbide on harvesting mangrove. The formulation of mangrove management strategies in GiliRanggoFishery Reserve Area consists of SO strategy: maximizing the primary function of mangroves and management principles tailored to the status of the area, ST strategies: increase public knowledge about environmental management and improve the economy of the community, WO strategies: increasing community involvement and increasing the studies and research in the management of mangroves in GiliRanggoFishery Reserve Area, WT strategies: joint management plans involving all stakeholders, improve supervision and monitoring

Abstract

Conserving natural resources are a necessity to ensure the sustainability of it is utilization. Throughthe determination of the suitability of the coastal villages on Nusa Penida as coral reef protected areas, it isexpected that all activities use of coastal resources will sustainable. The results showed Nusa Penida totalarea of coral reef ecosystems is 1.419 ha with fringing reef types. Based on live Nusa Penida?s coral reefcover conditions at a depth of 3 meters categorized as good condition (74%), for a depth of 10 meters theaverage condition of live coral cover categorized as good condition (64%). Depth of 10 meters showed interactionscoral communities are more stable than 3 meters. Suitability protected area of coral reefs was conducted in12 villages that have reef condition data at a depth of 10 meters. From 12 villages studied, 9 villages havesuitability as a protected area of coral reefs namely the Village of Pajukutan, Suana, Batu Nunggul, BatuMadeg, Sakti, Batu Kandik, Sekartaji, Toya Pakeh and Jungut Batu. From those villages, 4 villages have notake zone recomendation areas namely the Village of Jungut Batu, Toyapakeh, Batu Nunggul and Suana

Abstract

Dendrobium heterocarpum Lindl. is one of the orchid species which is often seeked. It is because this species has a mystique appearance and special fragrance. The research aimed to observe the growth and developing responses of protocorm like bodies (PLBs) of D. heterocarpum after 12 weeks in three different medium, i.e. Murashige and Skoog (MS), Kursor C and Western 3 (W3). The D. heterocarpum seeds were planted in MS, Kursor C and W3 medium. Qualitatively the orchid growth were observe including growth score and colour score. The result showed that growing time of MS was 4 weeks, a week faster than the growing time on Kursor C and W3 medium. The growing phase on MS medium and Kursor C medium reached phase 1 whereas on W3 medium reached phase 2. PLB?s colour after 12 weeks on MS medium reached phase E, strong yellowish green phase (142A). Meanwhile, on Kursor C and W3 was on phase D, moderate yellowish green (143D).

Abstract

Nino Coni Santana National Park had been suffered by illegal logging, and other activities that are not environmenknowleage. Lack of a security and knowledgeabout forest also contributeto increasethe damage of Nino Coni Santana National Park (1)The purpose of this research is to determine management strategy of Nino Coni National Park due to illegal logging in the area Tutuala and Mehara village at Tutuala sub-district,Lautem district, Timor Leste and determine management strategy of Nino Coni Santana National Park. Public perception obtained through questionnaires with purpose sampling method. The formula of management strategy for the damage in Nino Coni Santana National park use SWOT analysis to identify internal and external factors. The level of damage known by identifiy thearea due damaged by illegal logging.(2) The results showed that the damage by illegal logging at Nino Coni Santana national park caused because people are not involved in management of Nino Coni Santana National Park. The problems are illegal logging, encroachment,of forest, slash the wood harting wild animals, lack organization,andillegal grazing.(3) The management strategy of Nino Coni Santana National Park based on local culture, the establishment of a special UPTD that manage Coni Nino Santana forests, HKM and tourism programs, public development programs and cooperation of local people in order to utilize, surveillance and patrolling forests. In order to develop local knowledge as a base inNino Coni Santana National park,through HKM and tourism, Public socialization about the importance of forest. To measure the success of this management,should do monitoring, controlling andevaluation periodically by the government and involve the local people around Nino Cono Santana National Park.

Abstract

Artificial reefs are made to act as temporary substitutes for small areas of damaged original reef. The structures attract fish and other marine organisms, which can eventually be exploited (e.g. for human food), so it is expected that they will form the basis around which natural reefs can grow and recover. The purpose of this study is to document recruitment of organisms onto the artificial structures, evaluate the effectiveness of the structures in terms of shape and recruitment potential, and use the recruitment information to show which species occur in the surrounding reefs. This research was conducted from May-July 2010 in north Bali, in the Village District Gerokgak Patas, Kampung Baru Village in Buleleng District, and Penuktukan Village, Kecematan Tejakula. The research method used was a visual census method to determine the number of fish species and fish abundance on artificial reefs at three different locations. Three replications were performed at each location. 2481 individuals from 18 families (38 species) of reef fish were observed. The area with the greatest abundance of fish and the highest number of fish species was in Kampung Baru, with 1170 individuals from 21 species of fish. Dominant reef fish species were Plotosus lineatus (Cat Fish) - 510 individuals, Pomacentrus auriventris (Goldbelly damsel)- 256 individuals, Dascyllus trimaculatus (Three spot damsel) - 335 individuals, Pseudanthias squamipinnis (Lyretail Anthias) - 320 individuals. A Sorensen analysis showed that reef fish communities on the artificial reefs in the form of 'hexadomes' were not derived from the surrounding reefs.

Abstract

In vitro propagation study was carried out at Plant Tissue Culture Laboratory, Faculty of Agriculture, Udayana University. It was conducted using Completely Randomized Design with two factors.  The first factor was plant growth regulator (PGR), consist of nine concentration mixture of IBA (0, 0.5, 1 mg/L) and BAP (0, 1, 2 mg/L). The second factor was grape varieties, consisted of Prabu Bestari and Jestro Ag 86.  There were five replicates for each treatment combination. Variable observed included presence and absence of callus, percentage of callus growth, percentage of shoot growth, number of shoot per explants, emergence of shoots, percentage of root growth and number of roots per explants.  Data were statistically analyzed using Analysis of Variance (ANOVA).  When treatment showed a significant different (P<0.05) or highly significant (P<0.01) mean separation was conducted following Tukey Test. Data were analyzed using MINITAB Statistical Program. Results showed that PGR and varieties was highly significant for percent of callus growth.  PGR also showed significant effect on shoot growth percentage, while there was significant difference found on root growth percentage.  It can be concluded that combination of 1 mg/L IBA and 2 mg/L BAP was able to produce the highest percentage of callus growth. Addition of 2 mg/L BAP without IBA was able to produce the highest percentage of shoot growth. Percentage of root growth of Jestro Ag 86 was higher than Prabu Bestari.
