Abstract

The objectives of this study was to observe antibacterial activities of water lily seed, against diarrhea-causing pathogenic bacteria as well as lactic acid bacteria; phytochemistry components in water lily seed and to evaluate each component?s activities against pathogen bacteria. Extraction of antibacterial components in the seed was done by fractional extraction methods using solvent based on its polar level, i.e. hexane, ethyl acetate and ethanol. The activities of each extract was tested by using diarrhea-causing bacteria, Enteropathogenic Escherichia coli K.1.1 (EPEC K1.1) and S. typhimurium with agar well diffusion method. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values were calculated with plate counting. Qualitative phytochemical tests were performed on all extracts. Fractionation was performed on extract with the largest antimicrobial activity by using thin-layer chromatography. The activities of each fraction were tested qualitatively by bio-autography method on thin layer chromatographic plates. The water lily seed had an antibacterial activity against EPEC K.1.1 and Salmonella typhimurium, especially in ethyl acetate extract. Ethanol extract had the same, yet lower activity. Ethyl acetate and ethanol extract of the seed did not show inhibition against the growth of lactic acid bacteria (Lactobacillus sp) and Bifidobacterium bifidum. The MIC and MBC values of the ethyl acetate extract on EPEC K1.1 were 0.89 (mg/mL) and 1.33 (mg/mL), respectively, while similar values of that on S. Typhimurium were 1.11 (mg/mL) and 1.33 (mg/mL¬), respectively. Phytochemistry components within ethyl acetate extract were alkaloids, flavonoids, tannins, glycosides, saponins, and triterpenoids. All fractions in the ethyl acetate extract had antimicrobial activities against EPEC K.1.1 and S. Typhimurium. These fractions were thought to inhibit the growth of the test-microbes by synergic action of each component.Key words: water lily seed, antibacterial, ethyl acetate extract

Abstract

High level of blood glucose is an indicator for diabetes mellitus (DM) condition. The condition iscaused by low level of insulin secretion or impairement of insulin receptor. The number of DM patientincreases every year. The World Health Organization reported that the number of DM patient in Indonesiawas the 4th highest in the world, after following China, India, and the United States of America, respectively.This study was conducted to analyze the effect of sea cucumber (Holothuria scabra J) on the profile ofantioxidant copper, zinc-superoxide dismutase (Cu, Zn-SOD) in the pancreatic tissues of diabetic rats. Atotal of 25 male white rats (Sprague Dawley) were used in this study. They were divided into five groups;(1) negative control (KN), (2) positive control, diabetic rats (KP), (3) diabetic rats treated with hydrolyzatedprotein of sea cucumber (HDL), (4) diabetic rats treated with concentrated protein of sea cucumber (KST),and (5) diabetic rats treated with isolated protein of sea cucumber (ISL), respectively. Diabetic conditionwas obtained by alloxan injection 110 mg/kg bw. The treatments were done for 28 days. At the end oftreatment period, the rats were sacrificed and pancreatic tissues were collected and fixed in Bouin solution and then processed to paraffin embedding standard method. The tissues were then stained withimmunohistochemical staining techniques using monoclonal antibody of Cu, Zn-SOD. The results showedthat treatment of HDL, KST, and ISL of sea cucumber (Holothuria scabra J) increased the content ofantioxidant Cu, Zn-SOD either in Langerhans islets and acinar cells of pancreatic tissues-diabetic rats.The HDL of sea cucumber treatment gave the best effect in increasing the antioxidant content of Cu, Zn-SOD in pancreatic tissue of diabetic rats.

Abstract

ABSTRACTThis study was conducted to evaluate the effect of tempe that were made from Genetically Modified (GM) and non-GM soybean on protein quality, malondialdehide (MDA) levels, intracellular antioxidant superoxide dismutase (SOD) activity in the liver and kidneys, as well as spermatozoa profile of experimental rats. Fourty five Sprague Dawley rats divided into eight treatment grups and one control, fed with tempe and soybean from GM and non-GM at 10% and 20% concentrations for 90 days. The results showed that there was no significant difference in term of protein quality, tempe made from GM soybean is substantially equivalent with tempe made from non-GM soybean. Results showed that group which was given ration of 10% protein from conventional soybean had lower liver and kidney MDA levels as compared to GM tempe 10% and 20% groups, but was not significant compared to conventional soybean 20% and casein 10% groups. While the value of liver and kidney SOD activity were not significantly different (p>0.05) between the groups of rats. There was no significant differences among the spermatozoa profiles treatment groups and control and they were within normal condition. Results of protein quality, MDA, SOD, and spermatozoa profile showed that tempe made from GM soybean was substantially equivalent with the non-GM soybean.Keywords: GM soybean, MDA, non-GM tempe, SOD, spermatozoa profileABSTRAKTujuan penelitian ini adalah untuk mengevaluasi pengaruh tempe Pangan Rekayasa Genetik (PRG) dan non-PRG meliputi kualitas protein, kadar malondialdehida (MDA) hati dan ginjal, aktivitas superoksida dismutase (SOD) hati dan ginjal, dan profil spermatozoa pada tikus percobaan. Sebanyak 45 tikus terbagi ke dalam delapan perlakuan dan satu kontrol yang diberikan perlakuan dengan ransum tempe dan kedelai, baik PRG maupun non-PRG, dengan konsentrasi 10% dan 20% selama 90 hari. Hasil percobaan menunjukkan bahwa tidak ada perbedaan yang signifikan dalam kualitas protein, tempe PRG memiliki nilai yang sama dengan tempe non-PRG. Tikus yang diberi ransum 10% protein kedelai non-PRG mempunyai nilai MDA hati dan ginjal yang lebih rendah dibandingkan dengan tempe PRG 10% dan 20%, tetapi tidak signifikan dengan kelompok kedelai non-PRG 20% dan kasein 10%. Sementara itu, nilai SOD hati dan ginjal tidak signifikan antar grup perlakuan (p>0,05). Tidak ada perbedaan yang signifikan pada profil spermatozoa antar perlakuan. Hasil dari kualitas protein, MDA, SOD, dan profil spermatozoa menunjukkan bahwa kedelai dan tempe PRG memiliki kesamaan substansial dengan kedelai dan tempe non-PRG.Kata kunci: MDA, profil spermatozoa, SOD, tempe kedelai PRG, tempe kedelai non-PRG

Abstract

ABSTRACTThe objective of this study was to analyze the influence of blondo, snakehead fish flour and brown rice flour-based biscuit on the improvement of the nutritional status of anthropometric and serum albumin in malnourished children aged 3-5 years old. The design of this study was Randomized Controlled Trial (RCT) with single blind pre-post study. The total subjects were 50 underfive children and divided into two groups. They were given the biscuits for 90 days. Before intervention, the body weight and serum albumin level of subjects were measured. The analysis method were paired t-test. The result showed that the difference between baseline and endline in control group for energy was 20.8±3.9 kcal, treatment group was 45.6±8.6 kcal, endline in control group for protein was 9.5±1 g, and treatment group was 16.6±2.4 g. The changing of weight in the control group from 11.7±0.8 kg increase to 13.3±1.1 kg, the weight of treatment group from 11.7±0.7 kg increase to 13.6±1.0 kg. The increasing of albumin level in the control group was 0.3±0.1 g/dL, and treatment group was 1.0±0.2 g/dL. The increase of weight for age and albumin level after consuming nutritious biscuits can increase and normalize the nutritional status of the malnourished children. It is concluded that intervention of the formulated biscuits for 90 days could increase the nutritional status (weight for age and weight for height) and the serum albumin level in the 3-5 years old malnourished children.Keywords: albumin, biscuit, blondo, brown rice flour, malnutrition, snakehead fish flourABSTRAKTujuan penelitian adalah menganalisis pengaruh biskuit berbasis blondo, ikan gabus (Channa striata), dan beras merah (Oryza nivara) terhadap perbaikan status gizi antropometri dan serum albumin anak gizi kurang usia 3-5 tahun. Desain penelitian Randomized Controlled Trial (RCT) Single Blind Pre-post Study. Jumlah subjek 50 balita terbagi menjadi dua kelompok yaitu kelompok perlakuan dan kontrol. Kelompok perlakuan diberikan biskuit selama 90 hari. Sebelum dan setelah intervensi dilakukan pengukuran berat badan, tinggi badan, dan kadar serum albumin. Metode analisis menggunakan paired t-test. Hasil penelitian menunjukkan perubahan asupan energi antara awal dan akhir masa perlakuan pada kelompok kontrol adalah energi 20,8+3,9 kkal, kelompok perlakuan 45,6+8,6 kkal, perbedaan asupan protein kelompok kontrol 9,5+1 g dan kelompok perlakuan 16,6+2,4 g. Perubahan berat badan kelompok kontrol 11,7+0,8 kg menjadi 13,3+1,1 kg, kelompok perlakuan 11,7+0,7 kg menjadi 13,6+1,0 kg. Peningkatan albumin kelompok kontrol 0,3+0,1 g/dL, kelompok perlakuan 1,0+0,2 g/dL. Peningkatan berat badan dan albumin setelah mengonsumsi biskuit mampu memperbaiki status gizi anak gizi kurang menjadi normal. Dengan demikian, dapat disimpulkan bahwa intervensi formula biskuit selama 90 hari dapat meningkatkan status gizi BB/U, BB/TB, dan kadar serum albumin anak gizi kurang usia 3-5 tahun.Kata kunci: albumin, biskuit, blondo, gizi kurang, tepung beras merah, tepung ikan gabus

Abstract

Tempe is a potential source of protein for 250 million people in Indonesia. Indonesian soybean demand reached 2.2 million tons per year, and 1.3 million tons of which are used for tempe production. Approximately 70% of the soybeans are imported from different countries. To improve national food security, it is important to use local soybean as a raw material. Therefore, this study used Grobogan local soybean as raw material. The purpose of this study was to evaluate the impact of the consumption of tempe (from local soybean Grobogan) in the long term, ie over a 90-day subchronic test using rats as a model. Parameters measured were serum biochemical profile, hematology, malonaldehyde (MDA), and superoxide dismutase (SOD) in liver and kidney organs of rats. This research was conducted using three groups of rats were fed tempe, boiled soybean, and casein as a source of protein in the ration. Data feed convertion efficiency showed tempe of Grobogan soybean had a better quality protein than that of boiled soybean and casein. As compared to the groups of rats fed boiled soybean and casein, the tempe group had higher HDL cholesterol, but not significantly different in hematological parameters, MDA and SOD in liver and kidney organs. This suggests consumption of tempe made from Grobogan soybean in the long term does not have a negative impact on health.

Abstract

The objective of this study was to observe the effect of functional synbiotics yoghurt (made ofindigenous probiotic + oligofructosaccharides) in rats that were infected with Enteropathogenic Escherichiacoli (EPEC), and to detect its effect on hematological status. A total of 25 male Sprague Dawley rats wereused in this study and divided into five treatment groups: (i) negative-control; (ii) positive-control; (iii)synbiotic yoghurt; (iv) synbiotic yoghurt + EPEC; and (v) conventional prebiotic yoghurt. Yoghurt (109 cfu/mL lactic acid bacteria) was given orally using feeding tube at day one until day 4. At day 8 ? day 14animals were infected with EPEC (107 cfu/mL) orally using feeding tube; at day 22 blood samples werecollected for hematology analysis (erythrocytes, hematocrit (Hct), hemoglobulin (Hb), platelets, andleucocytes). Post infection with the EPEC diarrhea was developed in rats both in the control positivegroups and in the synbiotic yoghurt + EPEC group. Animals in the positive-control group had a significantlyhigher thrombocytes and leucocytes counts and Hct compared to that in animals in the other groups(P<0.01). Whereas there were no significant effect on the erythrocytes counts and Hb (P>0.05). The treatmentwith synbiotic yoghurt, in both synbiotic yoghurt only and synbiotic yoghurt + EPEC groups significantlyreduced the platelets and leucocytes counts and Hct of rats

Abstract

Aim This study aimed to elicit the protective effect of Sargassum echinocarpum extract on endothelial dysfunction in thoracic aorta of streptozotocin-induced diabetic rats.Methods The animals were divided into 5 groups. The first was normal, the second was diabetic non treated animals. The third to fifth groups were the diabetic animals which given Sargassum echinocarpum extract (150; 300, and 450 mg kg-1 body weight, respectively) by oral gavage and extract treatment was given for 12 weeks. Diabetes was induced by single administration of streptozotocin (45 mg kg-1, i.p.), dissolved in freshly prepared 0.1 M citrate buffer, pH 4.5. Diabetes was confirmed ten days latter in streptozotocin induced animals showing blood glucose levels > 200 mg dL-1 (11.1 mmol L-1) as monitored in the blood from tail vein using glucometer. After the treatment period, the blood serum acquired was used for antioxidant enzymes assays and the thoracic aorta was used for vasorelaxation assay.Results There was a significant decrease in the activity of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-px) in diabetic rats (3.31 ± 0.12;67.17 ± 0.62;35.10 ± 0.83) comaped to control rats (9.97 ± 0.12;185.31 ± 0.23;116.38 ± 0.88). Administration of Sargassum extract increased the activity of SOD, CAT, and GSH-px. The diabetic rats exhibit endothelial dysfunction as shown by loss of vasodilatory response to acethylcholine (ACH). This was restored by administration of Sargassum extract.Conclusion Sargassum echinocarpum extract ameliorates oxidative stress and reverses the endothelial dysfunction associated with diabetes. Abstract

Telah dilaporkan bahwa probiotik dapat memelihara keseimbangan mikroflora usus dan meningkatkan sistem imun usus. Penelitian ini bertujuan untuk menganalisis pengaruh pemberian probiotik indigenous Lactobacillus fermentum dan Lactobacillus plantarum pada profil kesehatan usus halus tikus yang diinfeksi Enteropathogenic E. coli (EPEC). Penelitian ini dilaksanakan pada bulan Agustus?November 2011 di Institut Pertanian Bogor. Sebanyak 90 ekor tikus jantan galur Sprague Dawley telah digunakan dan dibagi menjadi enam kelompok perlakuan, yaitu kelompok kontrol negatif (A), perlakuan Lactobacillus plantarum (B), perlakuan Lactobacillus fermentum (C), perlakuan Lactobacillus plantarum dan EPEC (D), perlakuan Lactobacillus fermentum dan EPEC (E), dan perlakuan EPEC (F). Perlakuan dilaksanakan selama 21 hari. Setelah tikus dikorbankan, usus halus diproses menggunakan metode embedding standar dengan parafin dan diwarnai memakai hematoksilin eosin. Data hasil penelitian dianalisis secara statistik dengan rancangan acak lengkap analysis of variance (ANOVA). Hasil penelitian menunjukkan bahwa perlakuan L. fermentum dan L. plantarum dapat menurunkan persentase kerusakan vili usus halus 14?62% dan meningkatkan tebal mukosa usus halus 14?29%. Simpulan, perlakuan probiotik indigenous L. fermentum dan L. plantarum dapat meningkatkan kesehatan usus halus. L. fermentum lebih baik dalam menghambat kerusakan usus halus tikus yang diinfeksi EPEC. The objective of this study was to analyze the influence of indigenous probiotic Lactobacillus fermentum and Lactobacillus plantarum on the morphological profile of small intestine of Enteropathogenic E. coli (EPEC)- infected rats. This study was done from August to November 2011 at Bogor Agricultural University. A total of 90 male rats Sprague Dawley were used in this study. They were devided into 6 groups; negative control (A), Lactobacillus plantarum treated (B), Lactobacillus fermentum treated (C), Lactobacillus plantarum and EPEC treated (D), Lactobacillus fermentum and EPEC treated (E), and EPEC infected group (F). The treatments were done for 21 days. Small intestine tissues were processed using paraffin embedding standard method and stained with haematoxylin eosin. The data were analyzed using analysis of variance (ANOVA). The results showed that L. fermentum and L. plantarum treatments decreased percentage of small intenstine villi damage (14 to 62%) and increased the thickness of small intestine mucosa (14 to 29%). In conclusions, treatment of indigenous probiotic L. fermentum and L. plantarum increase the health of small intestine. L. fermentum give better effect in inhibiting the damage of small intestine of rats, which under EPEC infection. Abstract

Diabetes mellitus is a spectrum of inherited and acquired disorderscharacterized by elevating blood glucose levels. Diabetesis an abnormal carbohydrate metabolism, therefore, diet therapyfor diabetics plays a key role in the management of thedisease. Most Indonesian people consume rice as source ofenergy and protein. Generally, diabetics consume very limitedrice because they believe that rice is the one of hyperglycemicfood, even though different rice varieties have large range ofglycemic index. The study aimed to evaluate hypoglycemicproperties by using rat assay and chemical characteristics of 10Indonesian rice varieties, i.e. Pandan Wangi, Rojolele, BengawanSolo, Cenana Bali, Memberamo, Celebes, Ciherang, BatangPiaman, Cisokan, and Lusi. Taj Mahal, an herbal ponni importedrice, was used as a comparison. Male Sprague Dawley rats (150-200 g body weight) were used for hypoglycemic assay. The ratswere fasted overnight before the blood glucose was measured inthe morning. The rats were then feed with 4.5 g rice per kgbody weight by oral administration, followed by 1 ml of 10%glucose solution in the next 30 minutes. The blood glucose wasmeasured for the next 30, 60, 90, and 120 minutes. Changes inblood glucose concentrations (mg dl-1) before and after the oraladministrations were calculated for each rice variety tested.Results showed that Cisokan and Batang Piaman were categorizedas low glycemic responses and Ciherang as high glycemic response,while the other varieties (Memberamo, Cenana Bali,Lusi, Bengawan Solo, Pandan Wangi, Celebes, and Rojo Lele)showed moderate glycemic responses. As the best hypoglycemicactivity, Cisokan contained high amylose (27.6%), fat (0.87%),total dietary fiber (6.24%), resistant starch (2.02%), and loweststarch digestibility (52.2%), which are ideal for diabetic’s consumption.Ciherang as the worst hypoglycemic activity had lowresistant starch (1.78%), low total dietary fiber (4.52%), and mediumamylose (23.0%). This study implies that Cisokan varietyis suitable for diabetic’s consumption.

Abstract

Sea cucumber (Holothuria scabra J.) is one of fishery commodities whichcontain high nutritional and economic value. This study aims were 1) thephysical characteristics of sea cucumber, 2) nutrient content (proximate) seacucumbers fresh meat, and 3) the yield and nutrient content (proximate) seacucumber powdery fresh meat. The study was conducted in four stages of theexperiment: 1) Preparation of meat of sea cucumber, 2) analysis of nutrientcontent (proximate) sea cucumber fresh meat, 3) The making of powder of seacucumber meat , and 4) analysis of yield and nutrient content (proximate) seacucumber meat powder. Results showed the proportion between the parts of thebody meat: offal and gonads: the skin: water and dirt were 4:3:2:1 (w / w). Freshsea cucumber meat has a high nutrient content that is 87.03% water, 9.94%protein, 0.54% fat, 1.86% ash, and 0.64%. carbohydrate. The average yieldproduced approximately 10.16%. While the nutrient content (proximate) powderysea cucumber meat that is 9.13% water, 61.31% protein, 3.68% fat, 12.52% ash,and 13.36% carbohydrate.
