Abstract

The aim of the study was to assess the viability of vitrified embryo using cryoloop as a carrier ofembryo. The blastocyst stage embryos were collected from superovulated mice. Embryos were frozenusing vitrification method and vitrified embryos were loaded on copper filament cryoloop before dipped inliquid nitrogen. The viability of vitrified embryos was assess in vitro by medium cultered and in vivo bytransfered them to recipient mice. The result shows the viability of vitrified embryos was 85,7% after 24hours cultured and the embryos were born from two pregnant recipient mice out of nine (22%) or fouroffspring out of 63 trasfered embryos (6%). In conclusion, vitrified blatocyst stage embryos using cryoloopas a carrier could keep the viability of the embryos and they could be transfered to the recipient mice andwere born normally.

Abstract

Oocyte vitrification is the important part of gamete preservation for further purpose. The objective of this study was to evaluate the response or development of vitrified-mice oocyte following activation using various concentrations of Strontium Chloride (Sr Cl2). Oocytes were collected from superovulation-induced female mice. Oocytes vitrification was then performed using a gradual equilibration of 2 M Ethylene Glycol in 0.25 M sucrose and 7 M Ethylene Glycol on 0.5 sucrose. Subsequently, the vitrified oocytes were thawed and activated using various Strontium Chloride concentration in each group. Control 1 is unvitrified oocyte and without Sr Cl2. Control 2 is unvitrified oocyte then activated by 20 mM Sr Cl2. Zero (0) mM  Sr Cl2 is vitrified oocyte without Sr Cl2. Group Ten (10) mM is vitrified oocyte then activated by 10 mM Sr Cl2. Group Twenty (20) mM is vitrified oocyte then activated by 20 mM Sr Cl2. The viability of vitrified-thawed oocytes was observed based on ooplasm integrity. Whereas the oocytes respond to artificial activation was observed based on pronucleus formation after 10 hours of activation. The result showed that 39% of oocyte degenerated following vitrification. The respond of vitrified-thawed oocytes following artificial activation using Strontium Chloride was significantly lower compared to fresh oocytes (p<0,05). Interestingly the highest percentage of activated oocytes (36.36%) was present in a group achieved 20 mM Strontium Chloride. As conclusion is Strontium Chloride 20mM has a best result (36,36%) to activate vitrified oocyte than 0 mM and 10 mM of Strontium Chloride.

Abstract

Osteoarthritis (OA) is the most common joint disease that cause of pain and disability by various factors such as advanced age, obesity, trauma, and arthritis disease. These factors affect by degeneration of the cartilage surface, leading to loss of matrix include proteoglycan osteophyte formation, subcondral and synovial membrane affected. In the healthy joint, meniscus, articular cartilage, subchondral bone, and synovial membrane provide support to the joint. The meniscus is an important load bearing structure and has nutritive as well as lubricating properties in the knee joint as well (Little et al. 2010).Animal models are research materials that can be used in studying potential pathogenesis and therapy in various diseases in humans. Sheep are commonly large animal model of OA because of the availability, ease of handling, and have a similarities with humans in size and structure of joint. In the development of science, sheep can be used as an animal model in studying the pathogenesis of diseases in human orthopedics studies such as joints, ligaments, and bones. Garut sheep is an Indonesian germplasm indigenous that has the structure, density, and size of joint anatomy that are similar in human joints rather than other small animals. This is the basis of the utilization of Garut sheep as an animal model in human orthopaedic. (Little et al. 2010; Gregory et al. 2012).The aim of this study was to identify and analyze the determining surgical method by meniscectomy induction on Garut sheep with 8 weeks post meniscectomy observation for early stage of OA.

Abstract

Parthenogenesis is an artificial oocytes activation process without paternal contribution so that embryos will develop without fertilization [3]. The process of parthenogenesis as a reproductive strategy occurs in species of insect, pisces, or amphibian, which not require any implantation. Naturally, parthenogenesis is not common in mammals, but by understanding cellular mechanism during fertilization, it is possible to artificially activate mammalian oocytes.Blastocyst, derived from parthenogenesis, can be used for developmental study, embryo reconstruction, and one of potential source for pluripotent stem cells. Unfortunately, previous studies reported that parthenogenetic embryo did not achieve exhilarating blastocyst rate.One of the component that has been predicted to inhibit parthenogenetic embryo development is phosphate. Haraguchi et al. (1996)    reported that phosphate caused a negative effect on in vitro culture of AKR/N mice fertilized embryos, removal of phosphate elements was significantly improved the blastocyst rate up to 42.6% [1]. The effects of phosphate also became an interesting finding in the study that reported mouse fertilized embryos could well developed in modified medium rat 1 cell embryo medium (MR1ECM) which not contained any phosphate [2].The effect of phosphate on in vitro culture of mouse parthenogenetic embryo has not been clear. The aim of this research was to analyze inhibitory effect caused by phosphate in the medium and compare the development pattern between parthenogenetic and fertilized embryos in order to reach optimal production of parthenogenetic blastocyst for further purposes.

Abstract

Peningkatan penggunaan telepon genggam diiringi dengan peningkatan kepedulian masyarakatterhadap bidang kesehatan dan keamanan terkait emisi gelombang elektromagnetik yang berasaldari telepon genggam. Penelitian ini bertujuan mengetahui tingkat keamanan paparan gelombangelektromagnetik telepon genggam melalui pengamatan terhadap jumlah implantasi dan anak lahirdengan menggunakan mencit sebagai hewan model. Dua puluh empat ekor mencit betina disinkronisasidan dikawinkan dengan mencit jantan dengan perbandingan jantan dan betina 1:1 (single mating).Pemaparan dilakukan dengan gelombang berfrekuensi 900 MHz selama 7 hari pertama setelah mencitkawin. Mencit betina dibagi ke dalam empat kelompok berdasarkan lamanya waktu paparan. Waktupaparan adalah satu, dua, dan empat kali per hari dengan masing-masing lama paparan 15 menit untukkelompok pertama, kedua, dan ketiga, sementara kelompok keempat sebagai kelompok kontrol tidakdiberikan paparan. Hasil penelitian menunjukkan tidak adanya pengaruh nyata lama paparan gelombangelektromagnetik telepon genggam terhadap jumlah implantasi dan anak lahir dari induk yang terpapar(P>0,05). Untuk seluruh kelompok, jumlah implantasi berkisar antara 8,66 sampai dengan 10,00 danjumlah anak lahir berkisar antara 10,00 sampai dengan 12,33; tidak berbeda nyata dengan nilai padakelompok kontrol. Berdasarkan hasil penelitian, lama paparan gelombang elektromagnetik yang berasaldari telepon genggam dalam penelitian ini masih berada dalam tingkat aman untuk mencit bunting.

Abstract

Bone is a tissue that has a density of extracellular matrix structures and composed by organic and inorganic components. Decalcification is a stage that plays an important role in bone histology using various types of solutions. The sample used in this study was lateral condyle from tibia of three garut sheeps (Ovis aries) which had been fixed with 10% Neutral Buffered Formalin (NBF) for 24 hours. Sample were cut into pieces ranging from 1 cm x 1 cm x 1 cm in size, the decalcification using three solutions; 10% nitric acid, 10% EDTA (pH 7.4) and 10% EDTA (pH 7.4) + TBD-1®. The aim of this study was to evaluate the effectiveness of three solutions for the decalcification process of lateral tibial condyle of garut sheep. Observation parameter in this study includes: duration of decalcification, sectioning process of ribboning formation, structural integrity and absorption of hematoxylin-eosin (HE) staining. The results shows that 10% EDTA (pH 7.4) solution provides a long duration of decalcification which is ease sectioning process ribboning, the best structural integrity of lateral tibial condyle. In hematoxylin-eosin (HE) staining shows that 10% nitric acid solution does not absorb the optimum color, opposite in 10% EDTA and 10% EDTA + TBD-1® solution, the color intensity between hematoxylin and eosin in the tissue shows the best results. Based on these results, it can be concluded that the 10% EDTA (pH 7.4) is the best decalcification solution for lateral condyle from tibial of garut sheeps.

Abstract

Pada penelitian ini dikaji kemampuan spermatozoa domba hasil pengeringbekuan untuk melakukan dekondensasi dan membentuk pronukleus setelah diinjeksikan ke dalam oosit dengan menggunakan teknik intracytoplasmic sperm injection (ICSI). Metode pewarnaan aceto lacmoid digunakan untuk mengevaluasi kejadian dekondensasi dan pembentukan pronukleus pada oosit setelah ICSI. Hasil penelitian menunjukkan bahwa spermatozoa hasil pengeringbekuan dapat melakukan dekondensasi (2%) dan mendukung pembentukan 1PN (34%) tetapi belum mampu mendukung pembentukan 2PN setelah diinjeksikan ke dalam oosit. Sedangkan injeksi dengan menggunakan spermatozoa segar mampu mendukung pembentukan 2PN (30%) dan 1PN (40%). Sebagai kesimpulan dapat dikemukakan bahwa spermatozoa hasil pengeringbekuan mampu melakukan dekondensasi dan mendukung pembentukan 1PN setelah ICSI

Abstract

The purpose of this research was to study the changes in uterine support capability power due to increase of litter size in Kacang goat. In this study, 9 pregnant female Kacang goats were divided into 4 different groups based on litter size, namely 1, 2, 3, and 4. At 7 weeks of pregnancy, the experimental Kacang goats were sacrificed to observe the macro and micro parameters of the uterus. The results showed that the litter size had a quadratic relationship with micro parameters of uterus but it had a linear relationship with macro parameters of the uterus. The variables of uterine glands area, glands lumen area, and cytoplasmic area reached maximum condition at the litter size of 2.5. Litter size had a linear relationship with the volume, weight, and dimension of the uterus. It can be concluded that the optimal number of litter size in Kacang goat was two offsprings which was proven by the optimal function of the uterus to support fetal development.

Abstract

Penelitian ini bertujuan mengetahui motilitas dan kemampuan memfertilisasi sperma kucing yang berasal dari epididimis yang disimpan pada suhu 4° C. Epididimis disimpan dalam media phosphate buffer saline (PBS) pada suhu 4? C selama 1, 3, dan 6 hari. Viabilitas spermatozoa diamati dengan pewarnaan hoechst-propidium iodine. Fungsi biologis spermatozoa dievaluasi melalui teknik kultur in vitro dengan fertilisasi mikro dan perkembangan embrio di dalam media kultur CR1aa. Hasil penelitian menunjukkan persentase spermatozoa hidup pada hari ke-1, 3, dan 6 penyimpanan masing-masing adalah 81,0; 71,7; dan 70,7% (duktus deferens), 84,0; 81,2; dan 63,2% (kauda epididimis), 84,0%; 75,0; dan 74,7% (korpus epididimis). Persentase pronukleus (PN) yang didapat dengan teknik intra cytoplasmic sperm injection (ICSI) menggunakan spermatozoa epididimis pada hari ke-1, 3, dan 6 hari penyimpanan masing-masing adalah 8,0; 10,0; dan 5,9%. Preservasi epididimis pada suhu 4? C dalam PBS dapat digunakan untuk fertilisasi dan menghasilkan embrio kucing secara in vitro.
