Abstract

Twenty percent of primate species is found in Indonesia1. Proboscis monkey (Nasalis larvtaus) or bekantan is one of endemic species in Kalimantan2,3. This primate is belonging to Colobinae subfamily and distributed in Kalimantan (Indonesia), Sabah and Serawak (Malaysia), and Brunei. Proboscis monkey is belonging to endangered species based on International Union for the Conservation of Nature and Natural Resources (IUCN) assessment4. Indonesian Ministry of environment and forestry established Proboscis monkey is belonging to one of twenty five species which has been incrased in population.Blood cells have important role in body regulation such as maintaining body temperature, carriying oxygen, nutrition, hormone and metabolism product to the whole body5. Blood examination is important for body health evaluation6,7, especially in non-human primate where lived in zoo, animal captive or rehabilitiation center6. The aim of this study was to investigate the profile and morphology of female-juvenile proboscis monkey blood cells at rehabilitation center (Sahabat Bekantan Indonesia Foundation) in Banjarmasin, South Kalimantan.

Abstract

Mesenchymal stem-cell is a pluripotent cells that can be differentiated into many kinds of cells (1) and have abilities to differentiated into mature cell, secreted cytokines, and releasing many kinds of protein functioned in body body regulation (2). However, the therapeutic effect of stem cell arise from their secreted factor such as growth factors, cytokines, chemokines, and metabolits which acts as biologic regulator in the autocrine and paracrine body function (3). Secreted factors of mesenchymal stem cells known as secretome or mesenchimal stem cell conditioned medium (MSC-CM) have therapeutic effect for antiapoptosis, angiogenic, immunomodulatoric, and chemoattraactive activity (4). Mesenchymal stem cell conditioned medium used as the replacement agent for cell therapy because it contains no cells and save for used in different individual (5).Type 1 diabetes mellitus (DMT1) is a disorder characterized by destruction of the insulin-producing pancreatic ?-cell that progressively leads to insulin de?ciency and resultant hyperglycemia. Insulin de?ciency can lead to progressive metabolic derangement, with worsening hyperglycemia, ketoacidosis, starvation, and death (6). Glucagon is a hormone produced by pancreatic ? cells. Glucagon releases by the pancreas to raise the concentration of glucose in the bloodstream when the concentration of glucose in the bloodstream too low (7). Glucagon worked in opposite with insulin, which lowers the extracellular glucose level, while insulin works to increase the extracellular glucose level (8). The aim of this study was to investigate the role of MSC-CM on the structural and functional regeneration of pancreatic ? and ? cells in Wistar rat (Rattus norvegicus) induced with type 1 diabetes mellitus. The datas in this study will completes the information about the effect of MSC-CM on pancreatic cells regeneration, which has been presented in previous study (9).

Abstract

Secretome is a factor found in the stem cell culture medium. This factor may repair the tissues of the organs that were damaged by various degenerative disorders1. A previous study has reported that secretome derived from the fetal human umbilical cord mesenchymal stem cell (HUC-MSC) may be an effective regenerative agent for ?-cell pancreatic regeneration in Type 1 diabetes mellitus and for skin regeneration in incisional and burn wound healing.The administration of cisplatin as a chemotherapeutic agent is limited because it causes side effects, such as reproductive toxicity and progresses to testicular dysfunction2. As a degenerative disorder, testicular dysfunction is characterized by failure in the synthesis of reproductive hormones and spermatogenesis. Cisplatin-induced testicular cell damage would promote vimentin and cytokeratin localization in the spermatogenic, Sertoli, and Leydig cells.Studies on the use of secretome in promoting the recovery of various degenerative disorders and its effects on the spermatogenesis of rats with cisplatin-induced testicular dysfunction have not been conducted. The aim of this stud was to investigate the effect of secretome derived from HUC-MSC on cisplatin-induced testicular dysfunction in rats.

Abstract

Lumba-lumba hidung botol (bottlenose dolphin) adalah spesies lumba-lumba yang paling umum danpaling dikenal orang. Gangguan sistem respirasi pada lumba-lumba sering dijumpai, sementara jenisbakteri yang sering menyerang lumba-lumba dari perairan Laut Jawa belum pernah diteliti. Penelitianini bertujuan untuk mempelajari pemeriksaan klinis sistem respirasi lumba-lumba dan melakukan isolasibakteri serta jamur pada sistem respirasi bottlenose dolphin (Tursiops aduncus) dari Perairan Laut Jawa.Penelitian dilakukan dengan studi pustaka, pemeriksaan klinis serta isolasi sampel sistem respirasi. Swabdeep blowhole dilakukan terhadap sepuluh ekor bottlenose dolphin di PT. Wersut Seguni Indonesia.Sampel dikirim ke laboratorium Mikrobiologi FKH UGM untuk isolasi dan identifikasi terhadap bakteridan jamur. Hasil penelitian menunjukan bahwa pemeriksaan klinis lumba-lumba hanya dapat dilakukansecara inspeksi. Hasil isolasi ditemukan bahwa 5 dari 10 lumba-lumba (50%) positif Staphylococcus aureus,sedangkan jamur tidak ditemukan. Berdasarkan hasil penelitian, disimpulkan bahwa pemeriksaan klinislumba-lumba dapat dilakukan dengan cara inspeksi terhadap blowhole, tingkah laku hewan, adanya?chuff? atau ?honk?, frekuensi respirasi, sosialisasi hewan dan posisi hewan saat berenang. BakteriStaphylococcus aureus dijumpai pada sistem pernafasan lumba-lumba hidung botol dari perairan LautJawa di PT. Wersut Seguni Indonesia.

Abstract

AbstractThe two sub units of intermediate filaments, desmin and vimentin, are found in most cells of mesenchymal origin. The content and distribution of desmin in mature skeletal muscle have been known, but for vimentin has been a subject of disagreement. The present study was aimed to clarify the presence of desmin and vimentin immunoreactivities in the skeletal muscle fibers of Kedu Cemani chickens (Gallus gallus domesticus) by immunohistochemistry methods of avidin-biotin-complexs. Samples of muscle tissues were obtained from pectorales mayor, biceps brachii, and biceps femoris of five adult Kedu Cemani chickens (Gallus gallus domesticus). Immunohistochemical staining results showed that desmin immunoreactivities were detected in Z disc of myofibers, but not vimentin. The results stimulate further exploration on characteristics of Kedu Cemani chickens, especially in terms of the distribution of other intermediate filaments and neuroendocrine cells in a variety of organs system.Key words: Kedu chicken, skeletal muscle, desmin, vimentin, immunohistochemistryAbstrakVimentin dan desmin merupakan subunit filamen intermedia dan dapat ditemukan dalam sebagian besar sel mesenkimal. Keberadaan dan distribusi desmin dalam sel otot skelet dewasa/masak telah diketahui, namun keberadaan dan distribusi vimentin pada sel yang sama masih menjadi perdebatan. Penelitian ini bertujuan mengklarifikasi keberadaan immunoreaktifitas desmin dan vimentin dalam serabut otot skelet ayam kedu cemani dengan metode immunohistokimia komplek avidin-biotin. Penelitian dilakukan menggunakan jaringan otot yang meliputi otot pectorales mayor, biceps brachii, dan biceps femoris dari 5 ekor ayam kedu cemani (Gallus gallus domesticus). Pengamatan terhadap immunoreaktifitas desmin dan vimentin dalam serabut otot normal ayam kedu cemani dewasa dengan menggunakan antibodi monoklonal terhadap desmin dan antibodi poliklonal terhadap vimentin memperoleh hasil bahwa immunoreaktifitas desmin terdeteksi pada diskus Z serabut otot skelet, namun immunoreaktifitas vimentin tidak terdeteksi. Hasil penelitian memunculkan dorongan untuk menggali lebih lanjut ciri-ciri khusus ayam kedu cemani, terutama dalam hal distribusi filamen intermedia dan jenis neuroendokrin lainnya pada berbagai sistem organ tubuh.Kata kunci: Ayam kedu, otot skelet, desmin, vimentin, immunohistokimia

Abstract

The one the influence factors to the normal functions and testes growth is thyroid gland status. The knowledge about the influences of euthyroid, and hypenhyroid to thetesies growth will help to give more understanding of the normal lestes growth. The new born male rats were used todetect the influence of hypothyroidism to the testosterone level during prepubertal periode. Twenty primigravida pregnant rats (Rattus norvegicus albinus) were used. After birth, the male pups were divided inio controls and treatments group. Mothers of the treatment group were given 0,1% 6-n-prapyl-2-thiauracil (PTU), via drinking water, ad libitum, after birth until on day 8. On dary 27, 30, 37, 44, and 51, five control pups and 5 treatment pups were sacrificed. The blood from each pup were collected to detec serum testosteron hormone level using immunoradLometric assay (IRMA) method. The research showed that serum testosteron level ofthe; treated group increased on day 30 (P<0,05) and day 37(P<0,05). Theresultof This research showed that PTU induced hypothyroidism produced increasing serum testosterone level.

Abstract

Glucose Transporter 4 (GLUT 4) merupakan protein transporter glukosa yang diatur oleh insulin, ditemukan terutama di jaringan adiposa dan otot lurik (baik otot skelet maupun otot jantung). Ayam kedu cemani merupakan ayam endemik Indonesia yang terdapat di wilayah Kedu, Temanggung, Jawa Tengah. Penelitian ini diperlukan untuk melengkapi dokumentasi data anatomi mikroskopik kekayaan hayati asli Indonesia. Penelitian ini bertujuan untuk mengklarifikasi distribusi GLUT 4 dalam serabut otot skelet ayam kedu dengan metode imunohistokimia avidin-biotin-peroxidase complex (ABC). Penelitian dilakukan dengan menggunakan jaringan otot yang meliputi otot pectorales mayor, biceps brachii, dan biceps femoris dari 5 ekor ayam kedu cemani. Hasil penelitian menunjukkan bahwa immunoreaktivitas GLUT 4 terutama terdeteksi di sarkolema dan komponen miofibril pada otot pectorales mayor, biceps brachii, maupun biceps femoris. Intensitas imunorekatifitasnya meningkat dari intensitas lemah pada otot pectorales mayor, menjadi intensitas sedang pada otot biceps brachii, dan kemudian intensitas kuat pada otot biceps femoris. Hasil penelitian memunculkan dorongan untuk menggali lebih lanjut ciri-ciri khusus ayam kedu cemani lainnya, untuk mendokumentasi data anatomis kekayaan hayati asli Indonesia.

Abstract

Glucose transporter (GLUT 4) is glucose transporter protein regulated by insulin, found in adipose tissue and striated muscle (skeletal and cardiac muscle). Kedu cemani chicken is one of Indonesia endemic animal, found in Kedu, Temanggung regency, Central Java. This study was required to complete microscopic documentation of  Indonesia?s native biodiversity. The objective of this study was to clarify GLUT 4 distribution in skeletal muscle fibers of kedu cemani chicken by using avidin-biotin-peroxidase complex (ABC) immunohistochemistry method. This study was conducted by using pectorales major, biceps brachii, and biceps femoris muscle tissue from 5 kedu cemani chicken. The result showed that GLUT 4 immunoreactivity were detected in sarcolemma and myofibrils component of pectorales major, biceps brachii, and biceps femoris muscle tissue. Intensity of GLUT 4 immunorectivites increased from weak intensity in pectorales major muscle tissue, moderate intensity in biceps brachii muscle tissue, then strong intensity in biceps femoris muscle tissue. This result might motivate to further exploration about the other kedu cemani chicken specific features to complete microscopic  documentation of  Indonesia?s native biodiversity.

Abstract

Obesity is one of major nutritional problems in the world. Obesity is very dangerous, especially when concentrated in the abdomen, because it is closely linked to various diseases such as diabetes, hypertension, heart disease, which can to causing death. This study aims to identify the nature and distribution of ghrelin on gastric endocrine cells in the obese rat (Rattus norvegicus) by using immunohistochemical techniques. The results will strengthen the understanding of the role and function of ghrelin as an alternative therapeutic target on obesity. The research used gastric tissues of ten obese and control rats which were stained with avidin-biotin-peroxidase complex method of immunohistochemistry. The results showed the existence of two types of ghrelin-producing cells (open and closed types) on the gastric mucosa of control rats, and only one type of ghrelin producing cells (open type) in obese rats. The intensity of ghrelin immunoreactive positive cells was detected weak in obese rats, but very strong in control rats. Ghrelin endocrine cells mainly distributed in the basal part of the gastric mucosa of the fundus parts, with a very small number in obese rats, but highly abundant in control rats. This study confirmed the decrease of the ghrelin synthesis and secretion in obese rat (Rattus norvegicus) at the cellular level. The decrease of ghrelin synthesis is characterized by a reduction on the number of ghrelin producing cells, the disappearance of the close type of ghrelin producing cells, and the low activity of protein synthesis in the ghrelin producing cells. Ghrelin endocrine cells distributed mainly in the basal part of the gastric mucosa, especially in the fundus parts.

Abstract

Di laut Jawa banyak populasi lumba-lumba yang sering tersangkut jala para nelayan saat mencari ikan dilaut. Di Indonesia, lumba lumba hidung botol merupakan mamalia air yang dilindungi. Penelitian ini bertujuanuntuk mengetahui profil eritrosit bottlenose dolphin (lumba-lumba hidung botol) sebelum dan setelah penangkaran. Tujuh ekor lumba-lumba digunakan dalam penelitian ini. Sampel darah diambil melalui vena superfisialis sirip ekor guna pemeriksaan eritrosit, meliputi jumlah, morfologi, kadar hemoglobin dan hematokrit. Pengambilan sampel dilakukan saat lumba-lumba diangkat dari perairan Laut Jawa dan setelah lumba-lumba mengalami penangkaran di PT. WSI, Kendal, Jawa Tengah. Data sebelum dan sesudah penangkaran dibandingkan dan diuji dengan analisis statistik t-test berpasangan pada tingkat signifikasi 95%. Hasil penelitian menunjukkan adanya peningkatan jumlah eritrosit, kadar hemoglobin dan nilai hematokrit secara signifikan. Dari hasil penelitian ini dapat disimpulkan bahwa penangkaran yang dilakukan oleh PT. WSI,Kendal, Jawa Tengah meningkatkan profil eritrosit lumba-lumba hidung botol.Kata kunci : lumba-lumba hidung botol, captivity, profil eritrosit, vena superficial ekor, uji t-berpasangan
