Abstract

ABSTRACT This study aimed to evaluate the effects of Zinc (Zn) supplementation on the quality and quantity of striped catfish sperm. Experimental design for this study was a complete randomized design with five treatments and five replications. Male broods fed with Zn supplementation for eight weeks. The Zn supplemented into the fish diet at different concentrations (0, 50, 100, 150 and 200 mg/kg of feed). The results showed that Zn supplementation could improve the quality and quantity of striped catfish sperm. The treatments also showed significant effects on semen volume, sperm motility, sperm viability, and sperm concentration (P<0.05). Zn supplementation at a dose of 200 mg/kg feed demonstrated the best result has indicated by enhancement of quality and quantity of striped catfish sperm, increasing 51% of the volume, 11.6% of motility, 5.81% of viability, 54.1% of concentrations. The results suggested that Zn played an important role in improving reproductive performances of male striped catfish reproduction. Keywords: quality of sperm, a quantity of sperm, striped catfish, supplementation zinc                                                                                            ABSTRAK Penelitian ini bertujuan untuk mengetahui pengaruh suplementasi Zinc (Zn) terhadap kualitas dan kuantitas sperma ikan patinPangasianodon hypophthalmus. Penelitian ini menggunakan rancangan acak lengkap dengan  lima perlakuan danlima ulangan. Induk jantan diberi pakan dengan suplementasi Zn selama 8 minggu. Zn disuplementasikan dengan dosis berbeda (0, 50, 100, 150 dan 200 mg/kg pakan). Hasil penelitian menunjukkan bahwa suplementasi Zn dapat meningkatkan kualitas dan kuantitas sperma ikan patin sehingga berpengaruh signifikan terhadap volume semen, motilitas, viabilitas dan konsentrasi sperma (P<0,05). Suplementasi Zn pada dosis pakan 200 mg/kg menunjukkan hasil terbaik yang ditunjukkan oleh peningkatan kualitas dan kuantitas sperma ikan patin 51% volume; 11,6% motilitas; 5,81% viabilitas; 54,1% konsentrasi sperma. Hasil penelitian ini menunjukkan bahwa Zn memainkan peran penting dalam meningkatkan reproduksi ikan patin. Kata kunci: kualitas dan kuantitas sperma, Ikan patin, suplementasi Zn

Abstract

ABSTRACT The present study is a preliminary research for producing mullet fry to aquaculture. The research aimed to evaluate stimulation of gonad maturation in mullet (Mugil dussumieri) using hormones. The method used for research was completely randomized design consisting of three treatments and each individual replication was repeated three times. Two experiments were conducted separately with different treatments. First experiment used 9-14.7 cm body length of fish treated with different hormone injections i.e. 4 mg/kg 17?-methyltestosterone (MT), 0.07 mg/kg estradiol-17? (E2), and 0.5 ml/kg 0.9% physiological solution as control. Second experiment used 10-31 cm body length fish treated with 750 IU/kg chorionic gonadotropin (hCG), 0.5 ml/kg Ovaprim, and 0.9% physiological solution as control. Gonadosomatic index (GSI) value of the first experiment on day 60 showed that GSI of E2 treatment (1.31±0.94%) was higher than both MT treatment (1.00±0.51%) and control (0.54±0.20%). On the other hand, the second experiment on day 60 showed that GSI of hCG treatment (7.18±0.59%) was higher than both Ovaprim treatment (3.29±2.66%) and control (6.72±0.32%). Egg diameter frequency distribution for control in the first experiment on day 30 showed that egg size ranged from 9?144 µm. Egg diameter for E2 and MT treatments on day 60 showed that egg size ranged from 9?243 µm and were higher than control. In the second experiment, egg diameter on day 30 for control showed that egg size ranged from 9-144 µm, hCG treatment showed egg size ranged from 9?441 µm, while Ovaprim egg size ranged from 9-111 µm. Blood glucose, blood cholesterol, testosterone and estradiol hormone level in the first and second experiment showed no significant difference. The results showed that estradiol-17? and 17?-methyltestosterone induction in 9-14.7 cm body length mullet increase gonad maturity to stage II while hCG induction in 10-31 cm body length mullet increase gonad maturity to stage III. Keywords : estradiol-17?, hCG, 17?-methyltestosterone, Mugil dussumieri, Ovaprim ABSTRAK Penelitian merupakan rintisan untuk menghasilkan benih ikan belanak dalam wadah budidaya. Penelitian ini bertujuan untuk mengevaluasi pematangan gonad ikan belanak Mugil dussumieri menggunakan hormon. Penelitian dilakukan menggunakan rancangan acak lengkap yang terdiri dari tiga perlakuan dan tiga kali ulangan individu. Terdapat dua percobaan dengan perlakuan berbeda dan dilakukan secara terpisah. Percobaan pertama menggunakan ikan berukuran 9?14.7 cm dengan hormon 17?-metiltestosteron (MT) 4 mg/kg, estradiol-17? (E2) 0.07 mg/kg dan kontrol larutan fisiologis 0.9% 0.5 ml/kg. Percobaan kedua menggunakan ikan berukuran 10?31 cm dengan human chorionic gonadotropin (hCG) 750 IU/kg, Ovaprim 0.5 ml/kg dan kontrol larutan fisiologis 0.9 % 0.5 ml/kg. Nilai gonadosomatic index (GSI) percobaan pertama pada hari ke-60 menunjukkan bahwa pemberian estradiol-17? (1.31±0.94%) lebih tinggi dibandingkan 17?-metiltestosteron (1.00±0.51%) dan kontrol (0.54±0.20%). Hasil percobaan kedua pada hari ke-60 nilai GSI menunjukkan bahwa pemberian hCG (7.18±0.59%) lebih tinggi dibandingkan Ovaprim (3.29±2.66%) dan kontrol (6.72±0.32%). Sebaran frekuensi diameter telur pada percobaan pertama untuk kontrol hari ke-30 menunjukkan kisaran 9?144 µm. Sebaran frekuensi diameter telur untuk estradiol-17? dan 17?-metiltestosteron hari ke-60 menunjukkan kisaran 9?243 µm lebih banyak dibandingkan kontrol. Sebaran frekuensi diameter telur pada percobaan kedua untuk kontrol hari ke-30 menunjukkan kisaran 9?144 µm, hCG 9?441 µm sedangkan Ovaprim hanya 9?111 µm. Kadar glukosa darah, kolesterol darah, hormon testosteron dan estradiol pada percobaan pertama dan percobaan kedua menunjukkan tidak terdapat perbedaan yang nyata. Hasil penelitian menunjukkan bahwa induksi hormon estradiol-17? dan 17?-metiltestosteron pada ikan berukuran 9?14.7 cm dapat meningkatkan kematangan gonad mencapai TKG II sedangkan induksi hormon hCG pada ikan berukuran 10?31 cm dapat meningkatkan kematangan gonad mencapai TKG III. Kata kunci : estradiol-17?, hCG, 17?-metiltestosteron, Mugil dussumieri, Ovaprim

Abstract

The objective of this research was to find the caryotype of the endemic ornamental fish, Celebes rainbow (Telmatherina ladigesi) from Maros River South Sulawesi, Indonesia. The experimental fish were caught from the branches of Maros River, those are Bantimurung River, Pattunuangasue River and Makkatoang River. Tissue collection and slide preparation were carried out using the solid tissue technique with slight modification. The amount of chromosome pairs found from those three rivers (three population) were 2n = 48. Abstract

Individu jantan ikan pelangi Iriatherina werneri lebih digemari sebagai ikan hias dibandingkan individu betina. Ikan jantan memiliki bentuk sirip punggung kedua dan sirip anal yang panjang seperti filament dan warna tubuh yang indah. Namun permasalahannya adalah secara alami populasi ikan jantan yang dihasilkan rendah. Oleh karena itu, maskulinisasi diperlukan untuk meningkatkan jumlah populasi ikan jantan. Teknik maskulinisasi menggunakan perendaman embrio fase bintik mata di dalam larutan hormon 17?-metiltestosteron (MT). Tujuan penelitian yaitu mengkaji persentase ikan jantan, tingkat penetasan telur, sintasan, abnormalitas dan pertumbuhan panjang total ikan pelangi melalui perendaman embrio pada dosis MT dan lama perendaman berbeda. Embrio yang digunakan pada fase organogenesis berumur 64 jam 40 menit setelah pembuahan. Perlakuan yang digunakan yaitu dosis MT 15, 30 dan 45 mg L-1, serta lama perendaman selama 6, 12 dan 24 jam. Hasil penelitian menunjukkan kombinasi perlakuan MT dosis 30 mg L-1 dan lama perendaman 6 jam merupakan kombinasi perlakuan terbaik yaitu menghasilkan ikan jantan 56,67%. Kombinasi perlakuan tersebut juga menghasilkan tingkat penetasan telur, abnormalitas dan pertumbuhan panjang total ikan pelangi pada kisaran normal. Pemberian MT dosis tinggi dan pemaparannya yang lama pada embrio dapat menurunkan performa penetasan telur dan kualitas larva ikan pelangi. Bentuk abnormal terlihat pada tulang belakang bengkok dan bentuk mulut yang tidak sempurna. Kajian maskulinisasi pada ikan pelangi masih perlu dilakukan untuk memperoleh sintasan yang lebih baik.

Abstract

This experiment was conducted to study the effect of giant gouramy larval immersion in triiodothyronine (T3) hormone solution on their growth and survival rate. One-day old larvae were treated with different concentrations of triiodothyronine (T3) i.e. 0; 0,001; 0,01; 0,1 and 1 ppm by immersion method for 24 hours. During eight weeks rearing period, larvae were fed to satiation with Tubifex three times daily. The media were aerated and changed 10-30% daily. At the end of experiment, there was no significant difference in yolk sac absorption between treated larvae and control. The highest dose of 1 ppm T3 resulted significant decreased in total length, average weight and survival rate of larvae. Abstract

ABSTRACT Marketed eel Anguilla bicolor bicolor is commonly produced from larvae rearing activity whose broodstocks and larvae are caught from the nature. Supply of eel broodstock is restricted by its life cycle and uncertain size variation of mature male and female. This study was aimed to evaluate the effect of hormonal induction through injection to enhance masculinization and gonadal maturation of eel at the weight of 100?150 g. The experiment used completely random design with the use of combination pregnant mare serum gonadotropin  (PMSG), antidopamine (AD), dan 17?-methyltestosterone (MT), which were (10 IU/kg PMSG + 0,01 mg/kg AD), P2 (20 IU/kg PMSG + 0,01 mg/kg AD), P3 (10 IU/kg PMSG + 0,01 mg/kg AD + 150 µg/kg MT), P4 (20 IU/kg PMSG + 0,01 mg/kg AD + 150 µg/kg MT), and P5 (control; without hormonal treatment). The result showed that an increasing of fish length along with fish weight were performed by treatment P4 and P3. The highest gonadosomatic index value was obtained by treatment P3 (1,3030±0,24262). Based on gonadal histology analysis, 2nd phase of spermatogonia development was found in P3 in week-8. The highest testosterone level was obtained by treatment P3, followed by P4, P2, and P1 in week-4. Combination of 10 IU/kg PMSG + 0,01 mg/kg AD + 150 µg/kg MT could enhance masculinization and gonadal maturation of eel in eight weeks of rearing period. Keywords: gonadal maturation, Anguilla bicolor bicolor, PMSG, AD, MT  ABSTRAK Ikan sidat Anguilla bicolor bicolor yang dipasarkan pada umumnya merupakan hasil usaha pembesaran yang benih dan induknya masih diperoleh dari alam. Penyediaan induk ikan sidat terkendala dengan siklus hidup dan variasi perbedaan ukuran induk ikan sidat jantan dan betina matang gonad yang belum pasti. Penelitian ini bertujuan untuk mengkaji peran induksi hormonal yang disuntikkan pada ikan sidat dalam mempercepat proses pematangan gonad ikan sidat ukuran 100?150 g. Metode penelitian yang digunakan adalah metode eksperimen rancangan acak lengkap dengan kombinasi pregnant mare serum gonadotropin (PMSG), antidopamin (AD), dan  17?-metiltestosteron (MT) sebagai berikut P1 (10 IU/kg PMSG + 0,01 mg/kg AD), P2 (20 IU/kg PMSG + 0,01 mg/kg AD), P3 (10 IU/kg PMSG + 0,01 mg/kg AD + 150 µg/kg MT), P4 (20 IU/kg PMSG + 0,01 mg/kg AD + 150 µg/kg MT), dan P5 (kontrol; tanpa perlakuan hormon). Hasil penelitian menunjukkan peningkatan pertambahan panjang seiring dengan pertambahan bobot diperoleh dari perlakuan P4 dan P3. Indeks gonadosomatik tertinggi diperoleh dari perlakuan P3 (1,3030±0,24262). Hasil histologi gonad ditemukan perkembangan spermatogonia fase 2 pada P3 di minggu kedelapan. Konsentrasi testosteron tertinggi didapat dari perlakuan P3 kemudian diikuti P4, P2, dan P1 pada minggu keempat. Kombinasi hormon 10 IU/kg PMSG + 0,01 mg/kg AD + 150 µg/kg MT dapat mempercepat pematangan gonad dan pertumbuhan pada ikan sidat selama delapan minggu pemeliharaan. Kata kunci: pematangan gonad, Anguilla bicolor bicolor, PMSG, AD, MT

Abstract

 ABSTRCT The phenomenon that often occurs in climbing perch culture (Anabas testudineus, Bloch) is a significant size difference between individuals of male and female those lead to high variation in culture yields. In addition to genetic factors, the difference possibly reflects the sexual dimorphism in this fish; female grows faster than male. This research was conducted to examine sexual dimorphism related to growth quantitatively. Sixty individuals of 45-days old juvenile derived from mating of five pairs broods were randomly picked up and reared individually in 20×20×100 cm3 net cages for 135 days. Fish were fed on commercial feed (30% protein) three times a day at 10% feeding rate. Body weight and body length were individually recorded every 15 days, sex was determined at the end of the experiment by surgering the fish and gonad was weighed to calculate gonadosomatic index (GSI). The results showed that growth and specific growth rate of female were 1.17 and 1.48 fold respectively higher than male. This result indicates that female monosex culture of climbing perch is highly promising. Keyword: dimorphism, growth, climbing perch, Anabas testudineus  ABSTRAK Salah satu fenomena yang sering dijumpai dalam budidaya ikan papuyu (Anabas testudineus, Bloch) adalah perbedaan ukuran yang mencolok antara individu jantan dan betina yang menyebabkan hasil panen ikan sangat bervariasi. Selain faktor genetik, perbedaan ini diduga disebabkan oleh adanya dimorfisme seksual terkait pertumbuhan. Penelitian ini dilakukan untuk menguji dimorfisme seksual terkait dengan pertumbuhan ikan papuyu secara kuantitatif. Benih ikan umur 45 hari hasil pemijahan lima pasang induk diambil secara acak sebanyak 60 ekor dan dipelihara selama 135 hari secara individu di dalam 60 unit hapa (20×20×100 cm3). Ikan diberi pakan komersial (protein 30%) tiga kali sehari sebanyak 10% bobot tubuh. Bobot dan panjang semua ikan dicatat setiap 15 hari sekali, jenis kelamin ditentukan di akhir penelitian dengan cara membedah, mengamati gonad, dan menimbangnya untuk menentukan indeks gonadosomatik (IGS). Hasil penelitian membuktikan adanya dimorfisme seksual terkait pertumbuhan pada ikan papuyu; pertumbuhan bobot mutlak dan laju pertumbuhan bobot harian ikan betina masing-masing 1,48 kali dan 1,17 kali lebih tinggi dibandingkan dengan ikan jantan. Hasil penelitian ini mengindikasikan bahwa pengembangan budidaya ikan papuyu monoseks betina prospektif untuk dilakukan. Kata kunci: dimorfisme, pertumbuhan, papuyu, Anabas testudineus

Abstract

ABSTRACT The objective of this study was to determine the effect of zeolite, active carbon, and clove oil on water quality (dissolved oxygen/DO, total ammonia nitrogen/TAN, temperature) and biological quality (glucose concentration, total protein, survival/SR) of giant freshwater prawn juvenile Macrobrachium rosenbergii in closed transportation system. The study was conducted in laboratory scale with a completely randomized design. The biota used was juvenile giant prawn with an average weight 0.407 ± 0.005 g/ind. The type and dose of additive used were A (20 g/L zeolite + 10 g/L active carbon + 14 µL/L clove oil), B (20 g/L zeolite + 10 g/L active carbon + 9.33 µL/L clove oil), C (20 g/L zeolite + 10 g/L active carbon + 4.67 µL/L clove oil), D (20 g/L zeolite + 10 g/L active carbon + 1.87 µL/L clove oil), K+ (20 g/L zeolite + 10 g/L active carbon), and K- (without material addition). The glucose concentration of treatment B and C significantly different with treatment A, D, K+, K-. Total protein of treatment A, B, C and K+ significantly different with treatment K-. DO, TAN, and temperature of the transportation media were still in the suitable concentration for living of giant prawn. The highest survival of the prawn was observed in group C. Konsentrasi glukosa perlakuan B dan C berbeda nyata dengan perlakuan A, D, K+, K-. Total protein perlakuan A, B, C, D, dan K+ berbeda nyata dengan perlakuan K-. DO, TAN dan suhu media transportasi masih sesuai dengan kehidupan udang galah. Tingkat kelangsungan hidup transportasi tertinggi yaitu pada perlakuan C. Abstract

Hatchery managers and maintaining genetic diversity and fitness population in endangered and threatened species in pond is a important and difficult thing to do.  But is must to do for conservation biology.  Fitness and structure population depends on effective breeding number (Ne) and population connectivity between each other. The second most important thing is gene flow and genetic drift. Ne is major role in the maintenance of genetic diversity as indicator for inbreeding depresion and genetic drift.  We sampled 6 hatchery in Jakarta  and Bekasi Indonesia region and used 12 polymorphic microsatellite loci to quantify population genetic structure. Result in this study are, each farm have different methode. The old farmers is bapak Gusi and bapak Hasan (+30 years).  Ne in bapak Hasan and bapak Gusi hatchery is 66,667 and F value is 0,00749 and loosing allele (P) is 0,26183.  Compared with younger farmers bapak Yahya (5 years) have Ne = 133,333, F = 0,00375 and P = 0,06855.  Even though hatchery bapak Hasan and bapak Gusi have the same value Ne, F and P but the result in population structure they are different founder populations. Hatchery bapak Hasan have unique structure and alleles composition compared with other hatchery.Keywords: breeding, management, Melanotaenia boesemani, hatchery, population.

Abstract

This study was conducted to examine activity of medaka (Oryzias latipes) â-actin promoter (mBP) in common carp(Cyprinus carpio) as the first step towards development of common carp transgenic in country. Gene constructpmBP-hrGFP that consists of mBA promoter and humanized Renilla reniformis green fluorescent protein gene(hrGFP) was injected into cytoplasm of one cell stage of common carp by using microinjector. PmBP-hrGFPconcentration used for microinjection was 50 ?g/mL aquabides. Parameters observed were survival rate of embryo(SRe), hatching rate (HR) and expression of hrGFP gene. SRe was calculated before eggs hacthed, while hatchingrate (HR) was after all of eggs hatched. The activity of mBA promoter was analyzed by observation of hrGFP genetransient expression using a fluorescence microscope. The results of experiment showed that SRe (87,5%) andHR (79.2%) of control was respectevily higher than that of injected treatment (75.0% & 61.7%). Expression of hrGFPwas observed firstly at blastula (12 hours after fertilization) to 1-day-old larval stages (24 hours after hatching)with higher gene expression at blastula to late gastrula stages. Percentage of micronjected larvae expressinghrGFP at 6 hours after hatching reached 71.6 ± 6.7%. Conclusion was that mBA promoter could drove hrGFPexpression in common carp, hence it can be used to produce common carp transgenic by changing hrGFP withgenes correlated with important traits in aquaculture.
