Abstract

Cysticercus bovis is the larval stage of Taenia saginata, the bovine tapeworm. The infection of thislarval in cattle musculature causes Bovine cysticercosis or Cysticercosis bovis.  Bovine cysticercosis is foundworldwide, but mostly in developing countries, where unhygienic conditions, poor cattle managementpractices, and the absence of meat inspection are common.  The adult Taenia infection in man is referredto as taeniasis.  Taenia saginata taeniasis is also found almost all over the world.  The prevalence ofTaenia saginata taeniasis has reported up to 27.5% in Gianyar Bali. In order to control the diseases,vaccination against the larvae stages in cattle of Taenia saginata may play an important role in controllingthe disease in the endemic regions.  The aims of the present study were to prepare and to investigate theimmunogenic protein as vaccine candidate for controlling  Cysticercus bovis infection in in Bali cattle.Cysticercus protein from the cyst fluid was firstly used to immunize mice and the mice sera were thencollected. Cysticercus proteins then analyzed using sodium dodecyl sulfate-gel electrophoresis (SDS-PAGE).All cysticercus proteins were then visualized by Commasie blue staining. The proteins were also transferredonto nitrocellulose membrane and the immunogenic proteins were visualized by Western Blotting usingimmune sera raised in mice.  By Commasie blue staining, a total of 17 proteins were detected with themolecular weight of 14,86 kDa -122,40 kDa from the smallest to the largest. As many as 7 immunogenicproteins with the molecular weights of 16.81 kDa; 19.22 kDa; 20.98 kDa; 27.41 kDa; 34.02 kDa; 38.31 kDa;and 54.94kDa were detected.

Abstract

Sag1 and bag1 is a gene specific-stage for Toxoplasma gondii tachyzoite and bradyzoite. The purposeof this study was analyze the sequences of sag1 and bag1 tachyzoite genes of local of Toxoplasma gondiiisolate as deoxyribonucleic acid probe candidate. Tachyzoite of local of Toxoplasma gondii isolate used onthis study. Gene sag1 and bag1 gene of Toxoplasma gondii were amplified by PCR, and then sequenced. Theresults showed sag1 fragment gene contained 612 bp and bag 1 contained 470 bp in length. BLAST analysisof sag1 and bag1 gene fragments as probe candidate showed that high specific for Toxoplasma gondii andno significant cross-reaction fragment with host and other parasites. The sequences 136 bp and 98 bpfragments as DNA probe candidate of Toxoplasma gondii sag1 and bag1 respectively.

Abstract

ABSTRAK Penelitian ini bertujuan untuk mengisolasi dan deteksi antibodi T. gondii pada itik lokal  di Bali. Pemeriksaan untuk mendeteksi antibodi T. gondii dilakukan secara serologisdengan uji Indirec Haemaglutination test (IHA) dan dilanjutkan dengan bioasaay pada mencit dan kucing.  Jumlah sampel yang dipergunakan sebanyak 188 ekor itik yang diambil serum, otot, otak dan jantung untuk mengetahui adanya infeksi T.  gondii. Hasil penelitian  pada itik  secara serologis ditemukan 47/188 sampel  terdeteksi adanya antibodi terhadap T. gondii, sehingga seroprevalensinya  sebesar 25 %. Seroprevalensi berdasarkan jenis kelamin itik pada jantan 27,8% dan betina 22,4 %, namun secara statistik tidak berbeda nyata (P>0,05).  Seroprevalensi pada itik yang dipemelihara dengan cara dikandangkan 18,7% dan dilepas 29,2%,  secara statistic tidak berbeda nyata (P>0,05).  Titer antibodi T.gondii yang ditemukan pada itik berkisar antara 1:64 sampai 1: 2048. Hasil Uji digesti dan bioassay pada mencit dan kucing dapat diisolasi kista bradisoit pada itik yang positif terdeteksi adanya antibodi terhadap T. gondii (IHA ? 1:64) dan ditemukan ookista T.gondii pada kucing.  Hasil uji patogenitas pada mencit menunjukkan bahwa isolat tersebut avirulen terhadap mencit. Hasil penelitian ini membuktikan bahwa daging itik dapat sebagai sumber potensial penularan T. gondii pada manusia.

Abstract

Toxoplasmosis is a zoonotic disease caused by the protozoa Toxoplasma gondii.Primary infection in pregnant women can cause abortion, neonatal death or abnormalityof fetus. Accurate diagnosis is needed to prevent infection especially related to thepresence of cyst in the tissuesThe aim of this research was to study sensitivity and specificity of ELISAmethod with 30 kDa protein antigen to detect T.gondii infection in pig with micebioassay as gold standard.. Samples were 171 pigs slaughtered at pig slaughter house inDarmasaba BadungThe result showed that sensitivity of ELISA method was 100% and 90,7% inspecificity.Research about sensitivity using ELISA test to predict cysts presence in tissuewere needed in the future.

Abstract

The aim of the study was to evaluate the bioactivity of the Tapakdara leaf extract (Catharanthus roseus) on the creatinine and blood urea levels in rat (Rattus norvegicus). Tweenty four male rats (250-260 gram body weight) were devided into three goups, with 8 animals in each group. Group 1 as a control group that was geven placebo, the group 2 was treated with 100 mg/kg body weight of leaf extract orally and the group 3 was treated with 100 mg/kg body weight of leaf extract orally.   The animals were treated during 8 days after one week adaptation period. The examination of ceratinin and Blood Urea Nitrogen (BUN) were done in the last day of treatment. The result of the study indicated that the creatinin level of rats treated with 100 and 200 mg/kg BW was high significantly higher (p<0.01) than control group, however the creatinin level was in range of normal value. The level of Blood Urea Nitrogen of rats were treated with leaf extract of Tapakdara 100-200 mg/kg BW was significantly hihger (p<0.05) than control group, it was also in normal value.

Abstract

Telah dilakukan penelitian perkembangan metacestoda Taenia sagianta pada sapi bali. Dua ekor sapi bali masing-masing diinfeksi 500.000 telur T. saginata yang diperoleh dari pasien orang Bali.  Nekropsi pada ke dua sapi dilakukan 103 dan 131 hari pasca infeksi. Pemeriksaan karkas dan organ viseral dikerjakan dengan cara insisi dan inspeksi mengikuti metode pemeriksaan rutin kesehatan daging.  Pada sapi yang dinekropsi 103 hari pasca infeksi ditemukan 534 metacestoda di beberapa otot skeletal, jantung dan diafragma yang sebagian masih berukuran 3x2 mm.  Sementara pada sapi yang dinekropsi 131 pasca infeksi, ditemukan 2.249 metacestoda menyebar di seluruh otot skeletal, jantung, diafragma, dan lidah dengan ukuran 4 x 3 mm.  Dari hasil pengamatan ini, dapat dibuat kesimpulan bahwa perkembangan optimal metacestoda pada sapi bali sekitar empat bulan pasca infeksi.

Abstract

The purpose of this study was to identify the nematode of the small intestine of pigs in Arfak Mountains and Baliem Valley in Papua, and determine the prevalence of the nematode infection. Small intestine content of 20 pigs originated from Arfak Mountains and 10 pigs from Baliem Valley were examined to identify the nematode species. Four species of nematodes were found in small intestine of pigs namely, Strongyloides ransomi, Ascaris suum, Macracanthorhyncus hirudinaceus, and Globocephalus urosubulatus. The result of the study showed that the prevalence of nematode infections in small intestine  were highly, in which the Baliem Valley had the prevalence of 90%, and 40% in Arfak Mountains.

Abstract

The study was couducted to determine the difference of heritability of Heterakis gallinarum in local and Lohman chicken. Sixteen local chicken and 16 Lohman chicken were inoculated with infected egg of Heterakis gallinarum for 250 egg in ech chicken. All chiken were necropsed 3 months after inoculated. The heritabilty of the worm was evaluated by determinan the number of infected chicken and number of worms. There were found 10 local chicken infected by Heterakis gallinarum and 2 local chicken infected by the worm. The result of the study indicated that the heratibility of Heterakis gallinarum is  higher in local chicken than Lohman chicken.

Abstract

  Penelitian ini bertujuan untuk mengetahui heritabilitas cacing Heterakis gallinarum asal Asia dan Eropa pada ayam lokal yang ditinjau dari tingkat infeksi, jumlah dan ukuran cacing. Sebanyak 32 ekor ayam lokal digunakan dalam penelitian ini, 16 ekor ayam lokal diinokulasi dengan telur cacing H. Gallinarum asal Asia dan 16 ekor diinokulasi dengan telur cacing H. Gallinarum asal Eropa dengan dosis masing masing 250 telur infektif dalam 0,2 ml. Nekropsi dilakukan setelah ayam berumur 3 bulan untuk mengevaluasi heritabilitas cacing tersebut. Hasil penelitian ini menunjukkan bahwa dari 16 ekor ayam lokal yang diinfeksi dengan telur infektif Heterakis gallinarum asal Asia, ditemukan 10 ekor (62,5 %) positif cacing Heterakis gallinarum. Sedangkan dari 16 ekor ayam lokal yang diinfeksi dengan telur infektif cacing Heterakis gallinarum asal Eropa ditemukan 15 ekor (93,8 %) positif Heterakis gallinarum. Rata- rata jumlah cacing Heterakis gallinarum asal Asia yang ditemukan pada setiap ayam lokal adalah 9,75 ± 12,96 ekor, sedangkan asal Eropa rata-rata jumlah cacing pada ayam lokal adalah 22,43 ± 20,45 ekor. Perbedaan tingkat infeksi dan intensitas infeksi antara ayam lokal yang diinfeksi dengan H. Galinarum asal Asia dan Eropa memberikan indikasi adanya perbedaan heritabilitas.

Abstract

Eighty percent of selected pig farm?s in two region?s in Papua Wet infected by Entamoeba spp, and also 32,4% from 102 pig?s infected by its parasite. The examined use SAF (Sodium Acetic Formaldehyde), to determine the prevalence of Entamoeba spp. The result of the study indicated of pig?s in Papua were infected by Entamoeba spp indicated that the prevalence was significatly higher in Baliem valley than Arfak Mountain.
