Abstract

Literacy could be a supporter of the forefront of education. Teachers have a crucial role in filtering and remodeling data on their students. The utilization of knowledge skill has to be trained by sorting and selecting the proper media for learning whose main purpose is to extend students' intelligence. Skill coaching is a way to boost the flexibility to use learning media, particularly Early Childhood Education and Kindergartens in Lubuk Begalung, Padang. The objectives to be achieved include: to increase the insight of Early Childhood Education and Kindergarten teachers about media, and the ability of media information literacy in learning. Training methods i.e. lectures, discussions, questions, and answers, and case simulations. The conclusion obtained from this activity is that early childhood and kindergarten teachers can choose the media responsibly and able to criticize media content more intelligent in the classroom.

Abstract

Separation of spermatozoa with x and y chromosome at boer goat and its application by artificial insemination for kid sex purposeABSTRACT. The purposes of this experiment are to investigate the separation of X and Y spermatozoa by measuring the spermatozoa quality, sex ratio between X and Y, capacity of fertility indicated by conception rate and sex ratio of goat boar kids. Samples, used in this experiment, are fresh semen from Boer goat with high quality consists of 4 group treatments with 6 replications 1) group of spermatozoa without separation (control), 2) group of spermatozoa separated by percoll gradient density centrifugation 3 levels (P1), 5 levels (P2) and swine up (P3). The observed parameters are spermatozoa quality, X and Y spermatozoa ratio, fertility?s capacity and sex ratio on the birth. Quality examination of spermatozoa and identify X and Y spermatozoa is based on the standard method of WHO. The conception rate was based on the ratio of pregnant goat after the first insemination. Data of spermatozoa quality and spermatozoa ratio were analyzed by using analisis of variance (ANOVA) and further analysis by LSD if there were differences between treatments. The results of this experiment showed that spermatozoa quality Boer goat significantly reduced (p 0,05) after separation with percoll gradient density centrifugation  and swim up. Percentage spermatozoa X after percoll gradient density centrifugation was significantly higher (P 0,05) compared to control and swim up. Meanwhile, the Y spermatozoa population was significantly higher (P 0,05) after swim up treatment compared to percoll gradient density centrifugation and control. The percentage of sex ratio (male: female) after insemination from percoll gradient density centrifugation produced more female than male. On the other hand, insemination from swim up produced more male than female. Sex ratio produced from separation of percoll gradient density centrifugation, swim up was difference from control semen (P 0,05). From this experiment, it was concluded that spermatozoa separation by percoll gradient density centrifugation and swim up can be used as one of the methods to separate   X and Y spermatozoa and further can be applied to get preferred sex animals.

Abstract

Abstrak.  Penelitian  ini  bertujuan  untuk  menganalisa senyawa  oksigen  reaktif  dan  pengaruhnya  terhadap integritas membran plasma spermatozoa kerbau lumpur basil sentrifugasi gradien densitas percoll. Penelitian ini menggunakan semen segar kerbau lumpur sehat yang diperoleh dengan cara penampungan menggunakan vagina buatan.  Sampel semen dibagi dalam tiga kelompok yaitu  tanpa  sentrifugasi (kontrol) sentrifugasi gradien densitas percoll selama 5  menit  dan sentrifugasi gradien densitas percoll selama 10 menit. Selanjutnya semenbasil  sentrifugasi  diamati  produksi  senyawa  oksigen  reaktif  dan  integritas  membran  spermatozoa.Hasil penelitian  menunjukan  bahwa  produksi  ROS  spermatozoa  setelah  sentrifugasi  gradien  densitas  percoll meningkat secara bermakna {p 0,05),  sedangkan persentase membran plasma utuh menurun secara bermakna{p 0,05). Tingkat produksi ROS dan penurunan persentase integritas membran plasma utuh spermatozoa kerbau lumpur setelah sentrifugasi gradien densitas percoll selama IO menit lebih tinggi secara bermakna (p 0,05) dibandingkan dengan sentrifugasi gradien .d1;nsitas  percoll selama 5 menit.. Tingkat produksi ROS berkorelasi negatif'dengan persentase integritas membran palsma utuh spermatozoa. (JKS 2007; 2:69-80) Kata  Kunci:  Sperma kerbau lumpur; sentrifugasi gradien densitas percoll,  ROS, integritas membran plasma utuh Abstract  The objective of this research was to find out the effect of preparation of sperm by using percoll gradient density centrifugation on the of reactive oxygen species (ROS) production and the demage integrity membranre of sperm swamp buffalo.The research is a laboratory study using a true experimental design,  used swamp buffalo  of  sperm divided into  3   groups i.  E  (  no  centrifugation/control;  percoll  density gradient centrifugation for  5  minutes  and percoll  density gradient centrifugation  for  IO   minutes.  Subsequently a determination of ROS production and· demage membrane integrity of sperm. The colleted data in this reseacrh were analysed varians and regression analysis. The result showed that ROS production after preparation with percoll density gradient centrifugation was highest of significant {p 0,05) and the procentage intact membrane of sperm a significant decreased (p 0,05).   The increase of ROS concentration and decrease. of procentage plasma membrane intact were after percoll density gradient centrifugation for IO minutes increase significantly (p 0,05)  compared with percoll  density gradient centrifugation for  5  minutes. The ROS  concentration of spermatozoa has correlated with the prosentage of membrane plasma intact of spermatozoa after percoll density gradient centrifugation. Abstract

Abstrak.  Penelitian   ini  bertujuan   untuk  rnengetahui   karakterisasi   protein   PAS  serum  sapi  bunting sebagai  kandidat  bahan  aktif  untuk  test  kebuntingan   dini  pada  sapi  Aceh, Penelitian   ini  merupakan penelitian    eksperimental     laboratorium    dengan    menggunakan     serum   darah   sapi   bunting    yang diperoleh   dari  petemakan   masyarakat   disekitar   Darussalam-Banda    Aceh.  Preparasi   dan  ekstraksi protein  PAS dilakukan   dengan  mengunakan   metode  sonikasi  dan identifikasi   protein  PAS dilakukan dengan   elektroferosis     rnelalui   SDS-PAGE    dan   Western   bloting.    Sedangkan    uji   kadar   protein dilakukan   rnelalui metode biuret.  Hasil  penelitian  menunjukan  bahwa  serum  darah  sapi bunting  dapat memproduksi   epitop  protein  PAS,  dengan  berat  molekul  protein  PAS    sebesar  65, 15 kDa.   Protein PAS  serum  darah  dapat   menginduksi   antibodi   terhadap   PAS  yang  efektif  sebagai   bahan  bioaktif deteksi  kebuntingan  dini pada sapi Aceh.  (JKS  2006; 2:59-68)  Kata Kunci: kotiledon  sapi aceh,  protein  PAS,  anti-PAS,  deteksi  dini kebuntinganAbstract.    This research  was aimed  to study  the characteristic   of PAS protein  of serum as a candidate of active  material  for early  detection   of cow  pregnancy.   This  study  was  conducted  as a  laboratory experimental   by using  serum  of pregnant  cow,  which  was  obtained  from  livestoxk  in Darussalam- Banda   Aceh.   Preparation   and  extraction    of  PAS   protein  ··were  performed    using  sonication   and identification    of  PAS   protein,   by  SDS-PAGE   electrophoresis    and  Western   blotting.   The  results showed   that  serum  is able  to produce   the epitope  of PAS protein.  The  weight  of molecule  of PAS protein  epitope  was  6~, 15 kDa.  PAS  protein  of serum  could  cause  an induction  of antibody  to PAS, which   is effective  as a bioactive  material  for early detection  of cow pregnancy.  (JKS 2006;  2:59-68)Keywords:  serum Aceh  bovine, protein  PAS, Anti PAS  and early pregnant

Abstract

Abstract.    Phytochemistry and biological    activity investigation  of medical  plants was an important step in exploring of their potency. The objectives  of this research were to determine the antioxidant activity of the extract and purified isolate of C. alata  leave and also,  to  elucidate of  the molecular structure of  the  responsible molecule. The methods used were extraction,  isolation and purification by means of chromatography, DPPH radical scavenging and spectroscopy. Result of the research provided antioxidant activity was (ED5o)   28.50±1.86 µg/mL for  the extract and 9.99±0.28  µM  for  the  purified  isolate.  Elucidation toward  the  purified  isolate indicated   kaemferol   structure   as   the   most   active   antioxidant   compound. Abstract

ABSTRAK. Penelitian bertujuan mengetahui pengaruh konsentrasi pengencer AdroMed® terhadap kualitas semen sapi aceh setelah proses pembekuan. Penelitian ini menggunakan rancangan acak lengkap (RAL) dengan 4 kelompok perlakuan pengenceran AndroMed®. Kelompok A1: AndroMed® 10% (5 ml AndroMed® + 45 ml Aquadestilata), A2: AndroMed® 15% (7,5 ml AndroMed® + 42,5 ml Aquadestilata), A3: AndroMed® 20%(10 ml AndroMed® + 40 ml Aquadestilata) dan A4: AndroMed® 25% (12,5 ml AndroMed® + 37,5 ml Aquadestilata). Masing-masing kelompok diulangi sebanyak 6 kali. Variabel yang di amati pada penelitian ini adalah Motilitas spermatozoa yang diamati tiap kelompok setelah pembekuan yang selanjutnya dianalisis dengan analysis of variance (ANOVA) pola satu arah yang dilanjutkan dengan uji Duncant. Rata-rata persentase motilitas spermatozoa pada kelompok A1, A2, A3 dan A4 secara berturut-turut adalah 30,69±3,68%, 39,79±2,44%, 45,22±3,17% dan 42,42±4,24%. Hasil penelitian menunjukkan bahwa konsentrasi pengencer AndroMed® berpengaruh secara nyata (P 0,05) terhadap persentase motilitas spermatozoa sapi aceh. Persentase motilitas spermatozoa pada kelompok A1 tidak berbeda secara nyata dengan A2, dan keduanya berbeda secara nyata (P 0,05) dibandingkan dengan kelompok A3dan A4. Konsentrasi AndroMed® 20% lebih baik dari pada 10%, 15% dan 25% dalam mempertahankan motilitas spermatozoa sapi aceh setelah pembekuan. (Analysis of Aceh Cattle spermatozoa motility after freezing using Andromed® with different concentration)ABSTRACT. The study aims to determine the effect of diluent concentration of AndroMed® against the motility of aceh cattle spermatozoa following freezing process. This study uses a completely randomized design (CRD) with 4 treatment groups. Group 1 used diluent 15%; AndroMed®, Grroup 2 diluent 15%; Group 3 with AndroMed® 20%; and, Group 4 with AndroMed® 25%. Each group was repeated 6 times. Motility of spermatozoa assessed which each group observed after freezing.The motility data obtained were analyzed by one way analysis of variance (ANOVA) followed by Duncant test. The average percentage of motility after freezing were found in the group A1, A2, A3 and A4 respectively are 30,69 ± 3,68%, 39,79± 2,44%, 45,22± 3,17%, and 42,42± 4,24%. Statistical analysis showed that the concentration of diluent AndroMed® significantly affected (P 0.05) the motility of aceh cattle spermatozoa. There is no significant difference of sperm motility percentage, between A2 with A1 treatment but  both are significantly different (P 0.05) compared to group A3. AndroMed® concentration affects the motility of aceh cattle spermatozoa after freezing. The treatment of 20% AndroMed® concentration were improved the quality of aceh cattle spermatozoa following freezing compared to those with AndroMed® 10%, 15%, and 25%.

Abstract

The purpose of this research was find out the effect of age and ejaculation frequency on quality of Aceh bull spermatozoa.. This research was conducted at Frozen Semen Prossesing Laboratorium, Saree, Aceh Besar from Januari to April 2013. Six healthy aceh bulls were divided into two groups of age, those were 3.0-4.0 years old; and 4.0-5.0 years old. Three numbers of bull at each group were assumed as the replication. Semen was collected once a week using artificial vagina and collected until third semen ejaculation. After collecting, volume, pH, color, consistency, concentration, motility, mortality, and abnormality of spermatozoa were immediately measured. This data was analyzed by analysis of variance (ANOVA) two-way pattern, followed by Duncant test. The result showed that the evaluation of bull semen at 3.0-4.0 years old and 4.0-5.0 years old affect significantly (P<0.05) on semen volume and spermatozoa concentration and had no contribute significantly on motility, mortality and abnormality of spermatozoa,while ejaculation frequency effect significantly (P<0.05) on semen volume, concentration, and abnormality spermatozoa, but had no effect significantly on motility and mortality (P>0.05). It is concluded that aged and ejaculation frequency effect significantly on quality of aceh bull fresh spermatozoa.Key words: ejaculation frequency, quality of spermatozoa, aceh bull, age

Abstract

This research aimed to determine the effect of spermatozoa X and Y separation using swim-up method on the quality of etawah crossbreed goat’s spermatozoa. This research used 6 male etawah crossbreed goats aged between 18-24 months, which were collected the semen once a week using electroejaculator. Immediately after semen collection, the quality of semen was examined, and then grouped into three treatment groups. Group 1 was control group (P0) refers to the semen was not separated by swim-up method. Group 2 (P1) was the semen separated by the swim-up method for 5 minutes. Group 3 (P2) was the semen separated by swim-up method for 10 minutes. Each treatment was repeated six times. Parameters for semen quality examined were the percentage of spermatozoa motility, live spermatozoa, and abnormal spermatozoa. Data were analyzed by analysis of variance (ANOVA) one way pattern, followed by Duncans multiple test. The results showed that the mean ± SD percentage of sperm motility on each treatment groups (P0; P1; P2) was 81.33±3.44, 89.67±3.21, and 90.00±3.10 %, respectively. The percentage of live spermatozoa on each group was 86.50 ± 2.07, 92.33 ± 2.08, and 91.83 ± 1.72%, respectively. The percentage of abnormal spermatozoa on each group was 13.00±2.53, 7.33±2.52, and 7.67±2.16%, respectively. Results of this research showed that the percentages of motility, live spermatozoa, and abnormal spermatozoa after separation by swim-up method were significantly different (P<0.05) compared to without separation. It can be concluded that the separation of spermatozoa using swim-up method can improve significantly (P<0.05) the percentage of live and motility of spermatozoa and decrease the percentage of abnormal spermatozoa of etawah crossbreed goats.Key words: Etawah crossbreed, spermatozoa quality, swim-up method

Abstract

ABSTRAK. Penelitian ini bertujuan mempelajari pengaruh pemberian kombinasi pakan fermentasi dengan multi enzim pencernaan dan vitamin E dalam ransum komersial terhadap peningkatan kualitas semen ayam arab. Penelitian ini menggunakan rancangan acak kelompok (RAK) yang terdiri dari 4 kelompok perlakuan dengan 5 kali ulangan. 1) kontrol (P0) ayam diberikan ransum komersial 100 %, 2) perlakuan 1 (P1) ayam diberi 89,83% ransum komersial + 10% pakan fermentasi + 0,15% multi enzim + 0,02% vitamin E, (3) perlakuan 2 (P2) ayam diberi 79,83% ransum komersial + 20% pakan fermentasi + 0,15% multi enzim + 0,02% vitamin E, dan (4) perlakuan 3 (P3) ayam diberi 69,83% ransum komersial + 30% pakan fermentasi + 0,15% multi enzim + 0,02% vitamin E. Koleksi semen dilakukan pada hari ke 36 dengan metode masase. Data kualitas semen yang diperoleh dianalisis dengan menggunakan analysis of variance (ANOVA) satu arah yang dilanjutkan dengan uji Duncan. Hasil penelitian menunjukkan pemberian kombinasi pakan fermentasi dengan multi enzim dan vitamin E dalam ransum komersial berpengaruh secara nyata (P 0,05) terhadap peningkatan volume, pH semen, konsentrasi spermatozoa, motilitas, spermatozoa hidup, dan penurunan abnormalitas ayam arab, namun tidak berbeda nyata (P 0,05) terhadap warna, konsistensi dan gerakan massa spermatozoa. Dapat disimpulkan pemberian kombinasi pakan fermentasi dengan multi enzim dan vitamin E dalam ransum komersial dapat meningkatkan kualitas semen ayam arab. (Effect addition of fermented feed with the multi-enzyme digestion and vitamin E in commercial feed on improvement the quality of semen arab chicken)ABSTRACT. The study aimed to determine the effect of fermented feed combination with the multi enzymes digestion and vitamin E addition on the quality of semen arab?s chicken. This research used a randomized block design (RAK) consisting four treatments with five replications : P0: 100% Commercial feed (524), P1: 89.83% commercial feed + 10% fermented feed + 0.02% viamint E + 0.15% multi enzyme, P2: 79.83% commercial feed + 20% fermented feed + 0.02% vitamin E + 0.15% multi enzyme, P3: 69.83% commercial feed + 30% fermented feed + 0.02% vitamin E + 0.05% multi enzyme. Semen sample was collected at 36th  day using a massage method. Data obtained were analyzed using analysis of variance (ANOVA) and continued by Duncan test. The results showed that addition of combination of fermented feed with digestive multi enzymes and vitamin E has significantly effect (P 0,05) on increases of the volume, pH, spermatozoa concentration, percentage of spermatozoa motility, spermatozoa life, and decreased the percentage of spermatozoa abnormalities. This research can be concluded that the addition of  fermented feed combination with digestive multi enzymes and vitamin E able to improve the quality of arab chicken semen.

Abstract

This study aimed to evaluate the genetic potential of local meat chickens (LMC) to get the candidate of parent stock through the development of the selection method applied, as well as economical and practical approach and adjusment of nutritional requirement, especially protein and energy balance. Research was divided into 2 groups, where the female and male parent was reared for 2 months and the offspring (F1) was reared for 4 months and selected. The parent was mated by the method of artificial insemination (AI). Method of selection was used on the orientation of the meat chicken production parameters with a positive correlation with the gene expression Diet treatments were formulated by optimal nutrition standards in order to drive maximum primary production of local meat chicken, contained 17, 19, and 21% of protein. The result showed that feeding diet with 19% protein can stimulate optimum growth and food conversion of ALPU male, whereas female growth LMC in line with the increasing availability of protein in the diet, wherein feeding 21% of protein result in optimal growth until age 4 months.
