Abstract

Broccoli (Brassica oleracea L. var. italica Plenck.) is a vegetable crop belongs to Brassicaceae family.  Broccoli has high nutrition, high in fiber and contains isotiacyanate that has anticancer activity.  Broccoli market in Indonesia, particularly in modern supermarkets, increases 15-20% per year, however, productivity was still low, therefore effort to increase broccoli production in Indonesia need to be investigated. Field trial was conducted at Batur Village, Kintamani District, Bangli Regency, Bali, to find out cultivars that were adapted in tropical region. Introduced cultivars being trialed were ?Alborada?,?Belstar?, ?Fiesta?, ?Sarasota?, ?Bay Meadows?, ?Castle Dome?, ?Liutenant?, ?Iron Man?, ?Marathon?, ?Green Gold?, ?Imperial?, ?Green Magic? and ?Lucky? as control.  Variable observed were vegetative growth, curding period, and productivity of each cultivar.  This study employed Randomized Completely Block Design with 3 replicates (plots) and 8 plants each plot.  Data were analyzed using ANOVA (Analysis of variance) on Costat program, followed by Duncan?s Multiple Range Test (DMRT) on 1% level.  Results show that each cultivar varied in adaptability in tropical region. ?Castle Dome? has the highest productivity, while ?Fiesta? was the lowest. Abstract

The effect of temperature and length of storage on vitamin C concentration of chili (Capsicum frustescens) was observed on this study. Samples were taken from farmer?s field around Gianyar Regency of Bali. The chilies were treated with temperature such as 10ºC, 20ºC, and 29ºC (room temperature) and stored for 5, 10, and 15 days. Jacobs method was applied for vitamin C determination. The highest vitamin C content was obtain from control (59,9 mg/100 ml). On the other hand, the lowest was from chili that stored at room temperature for 15 days (23,6 mg/100 ml). For weight loss, the highest of loss was from chili which stored at room temperature for 15 days.

Abstract

Research was done to elucidate the role of Ca2+ intracellular in stomatal movement induced by auxin. Two types of auxin (IAA and NAA) increased stomatal opening and the increase was concentration dependent. The addition of EGTA, Ruthenium red and Procaine as the modulator of Ca2+ concentration , inhibited stomatal opening induced by auxin. EGTA and procaine significantly inhibited somatal opening at 100 uM and 1 mM. Ruthenium red worked at lower concentrations which were 10 uM, 50 uM and 100 uM on LAA-induced stomatal opening, while on NAA-induced stomatal opening, Ruthenium red had an effect at 10 uM, 50 uM, 100 uM dan 1 mM. It is suggested that Ca2+ involves in stomatal opening induced by LAA and NAA as signaling agent.

Abstract

      Tanaman aren tergolong dalam keluarga palmae.  Secara ekonomis, pohon aren sangat bermanfaat dimana daunnya digunakan untuk keperluan upacara adat di Bali, nira dapat diproses untuk minuman khas Bali yang dikenal dengan nama ?tuak?, dan buahnya sering dicampur dengan es dan gula.  Perbanyakan tanaman aren dapat dilakukan dengan teknik kultur jaringan.  Media kultur (MS atau WPM) dapat ditambahkan hormon auksin dan sitokinin untuk mempercepat induksi tunas atau akar.  Hormon 2.4-D 4 ppm ditambahkan ke media untuk induksi kalus, sedangkan hormon NAA 1.5 ppm tergolong  auksin untuk induksi akar dan hormone BAP 1.5 ppm yang tergolong sitokinin untuk induksi tunas.  Pada umur 6 minggu setelah kultur eksplan berupa apokol, dapat terlihat massa kalus pada media dengan tambahan 2.4-D, sedangkan tambahan NAA dan BAP tidak menunjukkan pertumbuhan dari eksplan apokol.  Setelah diamati secara anatomi, eksplan apokol nampak dorman dan menunjukkan struktur perkembangan yang sama untuk semua perlakuan, dimana belum ada diferensiasi.

Abstract

  Keanekaragaman flora menjadi kekayaan hayati bagi kehidupan di alam. Penelitian bertujuan mengeksplorasi jenis dan manfaat tumbuhan di sekitar desa Mincidan agar dapat digunakan sebagai dasar pendidikan dan pengembangan ekowisata.   Metode deskriptif kualitatif dengan survei dan wawancara dengan penduduk desa dilakukan untuk melengkapi data yang dapat dikembangkan dalam rencana konservasi tanaman.  Ragam tanaman yang dijumpai di sepanjang rencana jalur trekking sepanjang 1,5 km tergolong pada tanaman obat, hias, buah, dan upakara.   Berdasarkan hasil survey, dapat diidentifikasi 102 jenis tumbuhan, 71 genera yang termasuk dalam 38 familia. Tanaman tersebut dapat dimanfaatkan untuk bahan upacara keagamaan dan obat tradisional.

Abstract

Dendrobium heterocarpum Lindl. is a native orchid in Indonesia. Its population continue to decline due to unsustainable exploitation. One solution to increase its population is via in vitro technique propagation. This research aimed to determine effect of hormone addition (Zeatin and NAA) to induce shoot multiplication of D. heterocarpum Lindl. grown in Murashige and Skoog (MS) basic medium. The research was conducted in Tissue Culture Laboratory, Department of Biology, Faculty of Mathematics and Science, Udayana University from November 2015 until March 2016. The experiment employed Completely Randomized Design (CRD) with 20 combinations of treatments, with three replicates each treatment. The treatment werer addition of Zeatin and NAA, with five concentrations of Zeatin (0, 0.1, 0,5, sweet corn and hybrids corn as the natural Zeatin); four NAA concentrations (0, 0.05, 0.1 and 0.5). Variable recorded include such as the number of shoots, roots and explant?s vigor. The quantitative data which include number of leaves and shoots after 12 weeks, were analyzed using Analysis of Variance (ANOVA); and if significantly different, tested further using Duncan Multiple Range Test (DMRT) at 5% level. Z5N3 medium revealed significantly different on amount of leaves and shoot height and also the best medium on amount of roots. Best hormone combination that can induce multiplication of new shoots are medium Z4N3 and Z5N1.

Abstract

Vanda is one of the most cultivated orchid species.  In nature, Vanda is endangered because it was exploited in large quantities. Orchid genetic viability can be maintained by embryo culture. The study aimed to investigate the growth of Vanda helvola seeds during 12 weeks after cultured. Media Murashige and Skoog (MS) added with tomato juice 100 g/L was used as culture medium. The results showed that embryo could grow on MS medium on stadium 6th. Protocorm started to form at 9 weeks after cultured. At 12 weeks, percentage of protocorm formed reach almost 24.1%. Tomato juice improved the growth of protocorm of  V.helvola. Key words: Medium, protocorm, Vanda helvola

Abstract

Tanaman tebu adalah keluarga poace yang berpotensi menghasilkan nilai ekonomi tinggi karena manfaatnya sebagai produk unggulan dalam menghasilkan gula serta bioetanol yang banyak digunakan oleh masyarakat luas. Untuk itu perlu upaya pemuliaan tanaman yang cepat untuk memenuhi kebutuhan pasar. Teknik kultur mikrospora merupakan aplikasi bioteknologi yang efisien dalam menghasilkan tanaman haploid. Teknik ini belum berhasil diaplikasikan pada tanaman tebu sehingga dalam penelitian ini bertujuan untuk memicu embriogenesis mikrospora tebu dengan aplikasi kultur mikrospora pada varietas bululawang. Metode yang digunakan yaitu dengan stress perlakukan perendaman mannitol 0.3M sebelum isolasi kultur pada lama perendaman yang berbeda. Analisis data menggunakan uji anova dan uji duncan. Hasil yang diperoleh yaitu Perlakuan stress yang diberikan berhasil memicu embriogenesis mikrospora dengan lama perendaman anther optimum pada 7 hari pada mannitol 0.3M

Abstract

Cabai rawit (Capsicum annuum) merupakan komoditi pertanian yang digunakan dalam berbagai jenis produk makanan. Pemanfaatan pupuk anorganik berdampak buruk terhadap lingkungan sehingga diperlukan agen hayati, seperti Trichoderma viride dan fungi mikoriza arbuskula (FMA) untuk memperbaiki kondisi tanah. Tujuan penelitian ini mengobservasi pemberian kombinasi T. viride dan FMA Glomus sp. Terhadap produksi cabai rawit serta mengetahui dosis kombinasi optimum dalam meningkatkan produktivitas cabai rawit. Penelitian dilakukan di laboratorium Taksonomi Tumbuhan/Mikologi dan green house Program Studi Biologi, Fakultas MIPA, Universitas Udayana. Penelitian menggunakan Rancangan Acak Kelompok dengan lima perlakuan yaitu (A) tanah steril (Kontrol negatif); (B) tanah steril dan 50 spora Glomus sp.; (C) tanah steril dan 2 g pupuk anorganik CPN15-0-14 KNO3 Merah; (D) tanah steril, 5 mL T. viride dan 50 spora Glomus sp.; (E) tanah steril, 10 mL T. viride dan 50 spora Glomus sp. Setiap perlakuan memiliki lima ulangan yang terdiri dari tiga unit tanaman. Pengamatan dilakukan terhadap tinggi tanaman, jumlah buah, berat basah dan berat kering buah, berat kering tajuk, berat kering akar dan infeksi FMA. Hasil penelitian menunjukkan bahwa penambahan pupuk anorganik menghasilkan nilai tertinggi pada hampir seluruh parameter yang diamati kecuali pada persentase kolonisasi FMA. Perlakuan E menunjukkan hasil yang lebih optimum dalam produksi cabai rawit meski belum mampu menandingi pupuk anorganik.

Abstract

Broccoli is a high value vegetable crop in Indonesia, however production is low due to limited number of suitable cultivars, so, breeding hybrid broccoli for warm climate is important.  The first step in hybridization is providing homozygote parent plants which can be done efficiently via microspore culture.  The objectives of this study were to determine 1) bud size that produce uninucleate microspore stage appropriate for culture; 2) pollen viability, 3) microspore development, in three broccoli cultivars (?BL 10001?, ?Royal Green? and ?Green Magic?). Various bud size (1 ? 5 cm) was squashed and observed microscopically to determine bud size containing uninucleate microspore.  Pollen viability was determined by IKI staining and pollen germination method.  Chromosome number was counted on root tips using squash method with aceto-orcein stain. Various heat treatment schemes were conducted to induce microspreo development.  Result showed uninucleate microspore derived from 2-3 mm and 3-4 mm bud length of ?BL-10001? and ?Royal Green? was responsive for microspore development in culture.  Pollen viability varied among cultivars, 78-87% on IKI method and 15-16% on germination test. Microspore culture showed different embryogenesis response; pollen-like structure was produced by ?BL 10001?.
