Abstract

Background: This study aimed to analyze the cytotoxicity effect of γ-sitosterol isolated from “Kejibeling” (Strobilanthes crispus), a medicinal plant, on several cancer cell lines. The mechanisms of the effects were studied through the expression of cancer-caused gene, c-myc and apoptotic pathways.Methods: This in vitro study was done using human colon cancer cell lines (Caco-2), liver cancer cell lines (HepG2), hormone-dependent breast cancer cell lines (MCF-7) and the normal liver cell lines (Chang Liver). The cytotoxic effect was measured through MTT assay and the potential cytotoxic value was calculated by determining the toxic concentration which may kill up to 50% of the total cell used (IC50). Meanwhile, the cytotoxic mechanism was studied by determining the effect of adding γ-sitosterol to the c-myc gene expression by reverse transciptase-polymerase chain reaction (RT-PCR). The effect of γ-sitosterol through apoptotic pathway was studied by using terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay.Results: γ-sitosterol was cytotoxic against Caco-2, HepG2, and MCF-7 with IC50-values of 8.3, 21.8, and 28.8 μg/mL, respectively. There were no IC50-values obtained from this compound against Chang Liver cell line. This compound induced apotosis on Caco-2 and HepG2 cell lines and suppressed the c-myc genes expression in both cells.Conclusion: γ-sitosterol was cytotoxic against colon and liver cancer cell lines and the effect was mediated by down-regulation of c-myc expression and induction of the apoptotic pathways.

Abstract

BACKGROUND Plant-derived herbal compounds have a long history of clinical use, better patient tolerance and acceptance. They are freely available natural compounds that can be safely used to prevent various ailments. Plants have been the basis of traditional medicine throughout the world for thousands of years and are providing mankind with new remedies. The objective of this study was to determine the cytotoxicity of soursop (Anona muricata Linn) leaves and pearl grass (Hedyotis corymbosa (L.) Lam.) on the hormone-dependent human breast carcinoma Michigan Cancer Foundation-7 (MCF-7) cell line. METHODS This study used two types of solvents (water and ethanol) in the extraction process and two incubation times (24 hours and 48 hours) in the MTT assays to analyze the cytotoxic effects of both plants. RESULTS Preliminary results showed that the ethanolic extract of soursop leaves (SE) displayed cytotoxic effects against MCF-7 on 24- and 48-hour incubation times with IC50 values of 88.788 μg/ml and 14.678 μg/ml, respectively. Ethanolic pearl grass extract (PE) showed similar results, with IC50 values of 65.011 μg/ ml on 24-hour incubation time and 52.329 μg/ml on 48-hour incubation time against MCF-7 cell line. However, the water extract of both plants displayed lower cytotoxic effect against MCF-7 cell line. CONCLUSION The ethanolic extract of both plants displayed cytotoxic effect against MCF-7. Soursop (Anona muricata Linn) leaves have the strongest cytotoxic activity against MCF-7 breast cancer cell line.

Abstract

Oxidative stress has been known to contribute to Alzheimer’s disease. Acetylcholinesterase (AChE) enzyme may lead to Alzheimer’s disease as a neurotransmitter. Antioxidants may have protective activities against oxidative damage and Alzheimer’s disease. Acetylcholinesterase inhibitors also can be used in the treatment of various neurological disorders for management of Alzheimer’s disease. This study aimed to determine antioxidant and anticholinesterase effects of Garcinia picrorrhiza Miq. fruit extract (GpKar) and its compounds, xanthone. Antioxidant activity was measured by H2O2 scavenging inhibitory activity, while anticholinesterase activity was measured using modified Ellman method. GpKar has higher H2O2 scavenging inhibitory activity (IC50= 967.28 µg/ml) compared to xanthone (IC50= 1198.95 µg/ml). In the anticholinesterase inhibitory activity, GpKar has lower activity (IC50= 70.25 µg/ml) compared to xanthone (11.80 µg/ml). In summary, GpKar has higher antioxidant activity but lower anticholinesterase activity compared to its compounds, xanthone. However, GpKar has potency as antioxidant agent to prevent Alzheimer’s disease.

Abstract

ABSTRACT The premarital sex behavior is a problem and social phenomenon that is  increasingly prevalent in society. At a young age, the premarital sex should be a major concern to reduce pregnancy rates unintended, sexually transmitted diseases, school drop out, abortion and even a death. One effort to prevent the premarital sex behavior for students in schools with methods of providing knowledge about sexual behavior, reproductive health, an understanding of religious and the early marriage law. Evaluation of  indicators usingpre test and post test data that are  analyzed with paired t and regression test. The results show that after counseling, the knowledge of junior high school students about sexual behavior, reproductive health and religious knowledge increased significantly (p <0.05), while high school students of religious knowledge and early marriage law increased significantly.  Keywords: premarital sex; reproductive health; sexual behavior

Abstract

BACKGROUND: The number of obesity cases is still increasing worldwide and has reached an epidemic scale. Plants are known to have a protection role in the development of obesity, however their antioxidant and anti-obesity activities have not widely known. This study was conducted to assess the in vitro antioxidant and anti-obesity activities of different types of freeze-dried fruits cultivated in Indonesia, especially Canarium sp., Averrhoa bilimbi L. and Malus domestica.METHODS: Total phenolic content of freeze-dried fruits was identified by Folin-Ciocalteu method, while the total flavonoid content was measured by aluminium chloride colorimetric assay. To assessed the antioxidant activity, 2,2-diphenyl 1-picrylhydrazyl (DPPH) scavenging activity assay and 2,2?-azino-bis (3-ethylbenzothiazoline6-sulphonic acid) (ABTS) reducing activity assay were performed. The ?-amylase and lipase inhibitory activity assay were performed to assess the anti-obesity activity. For comparison, hydroxycitric acid (HCA) compound was also assessed with DPPH, ABTS, ?-amylase and lipase assays.RESULTS: A. bilimbi had the highest total phenol content (6.35 µg GAE/mg), meanwhile M. domestica had the highest total flavonoid content (2.06 µg QE/mg). A. bilimbi also showed the highest antioxidant activity both in DPPH and ABTS assay, with inhibitory concentration (IC50)=279.99 µg/mL and 631.78 µg/mL, respectively. The freeze-dried M. domestica had the highest anti-?-amylase activity (IC50=258.85 µg/mL), while Canarium sp. had the highest anti-lipase activity (IC50=118.66 µg/mL).CONCLUSION: Freeze-dried fruits demonstrate in vitro benefits toward obesity. A. bilimbi has potent antioxidant activity and is beneficial against obesity-related adverse health effect by relieving oxidative stress. M. domestica and Canarium sp. hamper the fat accumulation by reducing the carbohydrate absorption and dietary lipid.KEYWORDS: antioxidant, anti-obesity, Canarium sp., Averrhoa bilimbi L., Malus domestica, hydroxycitric acid

Abstract

Studi terhadap tanaman yang berpengaruh terhadap proses reproduksi masih menjadi skala prioritas. Salah satunya adalah Kola (Cola  nitida), yang dikenal mempunyai efek stimulan. Telah dilakukan studi untuk mengetahui pengaruh ekstrak daun Kola terhadap viabilitas, motilitas dan integritas membran spermatozoa manusia in vitro. Sampel sperma diperoleh dari 20 orang pria dengan kategori normozoospermia. Sampel semen dibagi menjadi 1 kelompok kontrol ( kontrol negatif) dan 2 kelompok perlakuan dengan ekstrak daun Kola (0.05% dan 0.025%). Hasil penelitian menunjukkan bahwa ekstrak daun Kola tidak berpengaruh terhadap viabilitas spermatozoa tetapi secara bermakna meningkatkan motilitas dan integritas membran spermatozoa (p 0.05). Hasil ini mengindikasikan bahwa ekstrak daun Kola berpengaruh terhadap kualitas spermatozoa manusia in vitro.Study on medicinal plants that have potential effect on  reproductive process is still important to be done. One of the plant is Kola (Cola nitida) that has a stimulant effect. This study evaluated  the effects of Kola  leaf  extract  toward viability, motility and membrane integrity on human sperm in vitro. Normozoospermia semens from  twenty volunteers were taken and divided into 3 groups: 1 group  was  treated  as control  and 2 groups were treated with Kola leaf extracts  (0.05% dan 0.025%). The results showed that Kola leaf extract  had no significant effect in viability but increased significantly (p 0.05) the motility and membrane integrity. It indicates that Kola leaf extract influences human semen quality in vitro.

Abstract

Previous study has been shown the potential anticarcinogenic effect of Strobilanthes crispus, a plant native to countries from Madagascar to Indonesia, on human liver cancer (HepG-2) cell lines. The objective of the present study was to investigate the mechanism of anticarcinogenic effectof S.crispus extract through apoptotic pathway. Exposure of HepG-2 cells to S.crispus extract resulted in induction of apoptosis in a dose-dependent manner as measured by Fluoresence Microscopy and Confocal Laser Scanning Microscopy analysis. These findings provide important new insight into the possible molecular mechanisms of the anticancer activity of S.crispus.

Abstract

Liver cancer is one among cancers with increasing incidence in the world. Cola fruit (Cola nitida) is a fruit that is rich in properties and has been known since the Dutch colonial era. This fruit contains ingredients such as those contained in tea and chocolate such as methylxanthine and its derivatives. This study aims to determine the content of antioxidants and cytotoxic effects of cola fruit extracts obtained from Indonesia and Malaysia on liver cancer cell lines. Antioxidant content of fruit extracts of cola was assessed using DPPH (2,2-diphenyl-2-picrylhydrazyl hydrate) and cytotoxic effects were studied using MTT (3 - (4,5-Dimethylthiazol-2-Yl)diphenyltetrazolium bromide -2.5) on human liver cancer cell lines (HepG2). The results showed that cola fruit from Malaysia contained high antioxidant with the IC50 value of 37.2 ?g/mL whereas IC50 of value of its Indonesia?s cola fruit was 66.0 ug / mL. The similar results have been shown in the cytotoxic test using HepG-2 liver cancer cell lines. Malaysia?s cola fruit extract has a smaller IC50 value of 6.5 ?g / mL while the fruit extract of Indonesia?s cola showed IC50 value of 39.5 ?g / mL. These values indicates that the fruit extract of cola is a potential anticancer activities especially on liver cancer. Further studies are required to clarify this hypothesis.

Abstract

The research was conducted to determine the anticarcinogenic properties of ?sisik naga?(Drymoglossum piloselloides Presl.), by the microculture tetrazolium salt (MTT) assay on the human breast carcinoma dependenthormone (MCF-7) cell lines. The preliminary results showed that the ?sisik naga? extract displayed the cytotoxic effects against MCF-7 with IC50 value of 83.63µg/ml. The antioxidative activity of the extracts which could contribute to their cytotoxic properties was also studied. The ?sisik naga? extract was found to have high antioxidant activity with IC50-value of 4.229 ppm. The strong cytotoxic properties of the ?sisik naga? extract could be due to its high antioxidant activity.
