Abstract

Penelitian ini bertujuan mengetahui motilitas dan kemampuan memfertilisasi sperma kucing yang berasal dari epididimis yang disimpan pada suhu 4° C. Epididimis disimpan dalam media phosphate buffer saline (PBS) pada suhu 4? C selama 1, 3, dan 6 hari. Viabilitas spermatozoa diamati dengan pewarnaan hoechst-propidium iodine. Fungsi biologis spermatozoa dievaluasi melalui teknik kultur in vitro dengan fertilisasi mikro dan perkembangan embrio di dalam media kultur CR1aa. Hasil penelitian menunjukkan persentase spermatozoa hidup pada hari ke-1, 3, dan 6 penyimpanan masing-masing adalah 81,0; 71,7; dan 70,7% (duktus deferens), 84,0; 81,2; dan 63,2% (kauda epididimis), 84,0%; 75,0; dan 74,7% (korpus epididimis). Persentase pronukleus (PN) yang didapat dengan teknik intra cytoplasmic sperm injection (ICSI) menggunakan spermatozoa epididimis pada hari ke-1, 3, dan 6 hari penyimpanan masing-masing adalah 8,0; 10,0; dan 5,9%. Preservasi epididimis pada suhu 4? C dalam PBS dapat digunakan untuk fertilisasi dan menghasilkan embrio kucing secara in vitro.

Abstract

This study aimed to analyze MSP-1 gene in mice erythrocyte infected with Plasmodium berghei using polymerase chain reaction (PCR). The result showed the existence of 462 bp DNA band which was assumed to encode MSP-1 protein with molecular weight of 19 kDa that can only be found in erythrocyte infected with Plasmodium berghei. BLASTn analysis showed that PbMSP-1 obtained in this study have 100% similar identity with mRNA of PbMSP-1 partial sequence (462 bp), 93% similarity with PbMSP-1 complete sequence (5750 bp and 5376 bp), and 87% similarity with PbMSP-1 incomplete sequence (333 bp).

Abstract

Malaria is one of the most severe public health problems worldwide. It is a leading cause of death and disease in many developing countries, where young children and pregnant women are the groups most affected. Malaria disease caused by Plasmodium parasite have symptoms that typically include fever, fatigue, vomiting and headaches. In severe cases, it can cause yellow skin, seizures, coma or death. The present study is aimed to monitoring parasitemia level and percentage of parasite morphology as parasitaemia progresses. This research used Plasmodium berghei NK strain obtained from National University of Malaysia which originally from MR4, USA. Design used in this research was completely randomized design, with 2 treatments which were mice without infection and mice with infection of P. berghei parasite. Method used in this research was staining method of thin smear of blood using Giemsa stained and observed by microscopic. Parameter of observation were morphology and development of P. berghei at different level of parasitaemia (10%, 20%, 30% and 40%). The result showed from microscopic examination of blood slides prepared from the study animals indicated that the ring form was dominant stage obtained at all different stages of infection followed by trophozoite stage. Meanwhile schizont stage was the lowest stage obtained at all different stages observed.

Abstract

Abstract. Flamboyant [Delonix regia (Boj. ex Hook) Raf.] leaf contains flavonoid compounds that are expected to have immunostimulatory effect. This research was done to determine the effect of flamboyant leaf extract on immune response by accessing the activity of immune cells and capability test the extract as immunostimulant in mice. Leaf extraction was done by maceration using methanol in the Laboratory of Biology of Chemistry Department, Faculty of Mathematics and Natural Sciences of Syiah Kuala University whereas animal treatment and testing were carried out Micro-technique Laboratory of Biology Department of the same faculty. This research used 20 male mice strain Swiss-Webster aged 7-8 weeks were randomly assigned to 4 treatment groups with five replications each. Group 1 (P0) was untreated control; group 1-3 were mice administration flamboyant leaf extract 250 mg/kg BW (P1), 500 mg/kg BW (P2), and 750 mg/kg BW (P3) per oral. The treatments were given for 14 days after one week of adaptation period. Blood samples were collected before and after extract treatment and used for leukocyte count analysis. Phagocytosis activity was accessed by carbon clearance assay on day 15. At the end of the study, all mice were sacrificed for spleen weight analysis. Data obtained was analyzed by Analysis of Variance followed by Tukey test (Leukocyte count and spleen weight) or regression analysis (carbon clearance). The results showed a flamboyant leaf extract administration resulted in increased leukocyte counts that were significantly different (p 0.05) between treatment groups.  Phagocytosis test indicated the extract had moderate to strong immunostimulatory effect whereas spleen weight analysis did not show any difference among treatment groups. In conclusion, flamboyant leaf methanol extract was able to increase immune cells and had potential immunostimulatory activity in mice.Keywords: Delonix regia, immunostimulant, leukocytes, lymphocyte proliferation.

Abstract

Abstract. Temperurate is one of the important parameter in the embryo development of fish; However, to date no information this effect on the Betta splendens. Hence, this study aims to determine the effect of temperature on egg hatchability and survival of betta fish larvae. Samples of betta fish used in this study are similar in size and from the same type of broodstock. A completely randomized design used with four treatments and three replications. The parameters observed were the process of egg development, fertilization rate, hatching rate, hatching duration, survival rate, and water quality (pH, DO, temperature). The results showed that the relation between temperature and fertilization rate was not significantly different, whereas the calculated F value (0.23) ? F table 0.05 (4.07). The relation between temperature and hatching rate has a very significant effect, F value (18.91) ? F table 0.05 (4.07). The relation of temperature to hatching time showed the same difference, and the relation of temperature to survival of betta fish (Betta spelendes) larvae is significant with F ¬ values (39.98) F table 0.05 (4.07). Water quality parameters during the study were pH 7 - 7.5, DO 5.0 - 7.7 ppm and temperature at 27 - 29 oC.Keywords: Betta fish eggs, temperature, embryogenesis Abstrak.  Suhu adalah salah satu faktor menentukak proses perkembangan embrio pada ikan, namun pengaruh ini belum pernah diamati pada ikan cupang Betta splendens. Oleh karena itu, penelitian ini bertujuan untuk mengetahui pengaruh suhu terhadap perkembangan embrio, daya tetas telur dan kelangsungan hidup larva ikan cupang. Induk ikan cupang yang digunakan memiliki ukuran yang sama dan dari induk jenis yang sama. Adapun rancangan yang digunakan dalam penelitian adalah rancangan acak non factorialdengan 4 perlakuan 3 kali ulangan. Parameter yang diamati adalah proses perkembangan telur, tingkat pembuahan telur, tingkat penetasan telur, lama penetasan telur, tingkat kelangsungan hidup dan kualitas air pH, DO, suhu ruangan. Hasil penelitian menunjukkan bahwa hubungan suhu terhadap tingkat pembuahan telur tidak berbedanyata, dimana nilai F hitung (0,23) ? F tabel 0,05 (4,07). Hubungan suhu terhadap tingkat penetasan telur berpengaruh sangat berbedanyata, dimana nilai F hitung (18,91) ? F tabel 0,05 (4,07). Hubungan suhu terhadap lama penetasan menunjukkan berbeda sama, dan hubungan suhu terhadap kelangsungan hidup berpengaruh nyata terhadap kelangsungan hidup larva ikan cupang (Betta spelendes) dengan nilai F hitung (39,98) F tabel 0,05 (4,07). Parameter kualitas air selama penelitian rata-rata pH 7 ? 7,5, DO 5,0 ? 7,7 ppm dan suhu ruangan 27 ? 29 oC.Kata kunci: Telur ikan cupang, suhu, embriogenesis

Abstract

Abstract. Kawan fish (Poropuntius tawarensis) is an endemic fish found in Danau Laut Tawar, Central Aceh, Indonesia. This species has been threatened by ecological partubation, unfrindly fishing practices and pollution.  Cryopreservation is one of the ways to maintain the presence of these fish. Cryoprotectant (CP) is a critical material in the cryopreservation and DMSO is a common CP used in cryopreservation. Hence, the aim of the present study was to determine the optimum DMSO concentration for kawan fish sperm. The completely randomized design with 6 treatments and 3 replications were used in this study. The tested treatment was the difference of DMSO concentration, namely; 0, 3%, 6%, 9%; 12%, and 15% DMSO was combined with 5% egg yolk. The ratio of sperm to diluent is 1: 20. The cryotubes containing diluented sperm were evaporated at 5 cm from the surface of liquid nitrogen for 10 min, then stored in a liquid nitrogen container at -1960C for 2 weeks, then thawed and analyzed for the quality. The results showed that fresh sperm of kawan fish had motility of 48.67%, pH 7, milky white, with moderate consistency. The assessment of mass movements shows that the sperm has good quality. The ANOVA test showed that the addition of DMSO in diluents gavee significant effect on sperm motility, fertility and hatchability rates of fish eggs (P 0.05). The highest percentage of sperm motility and fertilization rates of fish eggs were found at concentration of 6%, respectively with the value of 46.67% and 45.67%, respectively. The highest percentage of hatching rate was also found in similar concentration of DMSO with the value of 19.33%. %. The DNA integrity test using the electrophoresis gel method showed that there was damage to DNA fish sperm after freezing, the the lower damage was found at 9% and 12% DMSO. It is concluded that the optimum concentration of DMSO for kawan fish sperm is at 6% of DMSO. Key words: kawan fish (Poropuntius tawarensis), cryopreservation, DMSO, DNA integrity Abstrak. Ikan kawan (Poropuntius tawarensis) merupakan ikan endemik yang terdapat di Danau Laut Tawar, Aceh Tengah, Indonesia. Menurut IUCN (International Union for Conservation of Nature), ikan ini termasuk ikan yang terancam punah oleh sebab kerusakan lingkungan, penangkapan tidak ramah lingkungan dan polusi. Salah satu cara untuk menjaga keberadaan ikan tersebut adalah dengan penerapan metode kriopreservasi sperma. Oleh karena itu, penelitian ini bertujuan untuk menentukan konsentrasi DMSO optimum dan melihat kerusakan DNA yang terjadi pada sperma ikan kawan(Poropontius tawarensis) pasca pembekuan.Metode yang digunakan dalam penelitian ini adalah Rancangan Acak Lengkap (RAL) dengan 6 perlakuan dan 3 ulangan.Perlakuan yang diuji adalah perbedaan konsentrasi DMSO dengan konsentrasi 0; 3%; 6%; 9%; 12% dan 15%.DMSO tersebut dikombinasikan dengan 5% kuning telur. Perbandingan sperma dengan pengencer adalah 1 : 20. Semua cryotube yang berisi sperma dan pengencer diuapkan pada jarak 5 cm dari permukaan nitrogen cair selama 10 menit, selanjutnya, disimpan dalam kontainer nitrogen cair bersuhu -1960C untuk disimpan selama 2 minggu. Hasil penelitian menunjukkan bahwa sperma segar ikan kawan memiliki nilai motilitas sebesar 48,67%, pH 7, berwarna putih susu, dengan konsistensi sedang. Penilaian gerakan massa menujukkan bahwa sperma tersebut berkualitas baik. Hasil uji ANOVA menunjukkan bahwa penambahan DMSO dalam pengencer berpengaruh nyata terhadap motilitas, fertilitas dan daya tetas telur ikan kawan (Poropontius tawarensis) (P 0,05) setelah pembekuan. Selanjutnya, uji lanjut Duncan menunjukkan bahwa persentase motilitas sperma dan pembuahan telur ikan kawan tertinggi terdapat pada penambahan DMSO dengan konsentrasi 6%, masing-masing sebesar 46,67% dan 45,67%. Persentase penetasan telur tertinggi juga dijumpai pada perlakuan 6% DMSO, dengan nilai 19,33%. Hasil uji integritas DNA menggunakan metode elektrofresis gel menunjukkan bahwa terdapat kerusakan pada DNA sperma ikan pasca pembekuan, Kerusakan yang terendah terdapat pada konsentrasi DMSO 9% dan 12%. Namun secara umum, disimpulkan bahwa konsentrasi optimum untuk kriopreservasi ikan kawan adalah 6% DMSO.Kata kunci: ikan kawan (Poropuntius tawarensis), kriopreservasi, DMSO, integritas DNA

Abstract

Aktivitas dan kapasitas makrofag merupakan faktor penting dalam menjaga sistem imun dalam tubuh. Beberapa senyawa berasal dari tumbuhan seperti daun flamboyan [Delonix regia (Boj. ex hook.) Raf.] dilaporkan banyak mengandung senyawa flavonoid yang berpotensi sebagai imunostimulan dan dilaporkan berpengaruh terhadap peningkatan aktivitas dan kapasitas fagositosis makrofag. Penelitian ini mengkaji tentang potensi  daun flamboyan sebagai agen imunostimulan pada peritonium mencit (Mus muculus) yang diinfeksi dengan bakteri staphylococcus aureus. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) yang terdiri dari lima perlakuan dan lima ulangan yaitu kontrol negatif (akuades) (P0), kontrol positif (Stimuno) (P1), ekstrak daun flamboyan pada dosis 250 mg/kg bb (P2), 500 mg/kg bb (P3) dan 750 mg/kg bb (P4). Mencit diberikan ekstrak selama 10 hari secara intubasi esofagus dan pada hari ke 11 diinfeksi dengan bakteri Staphylococcus aureus secara intraperitoneal. Jumlah makrofag yang aktif memfagositosis dan jumlah bakteri yang di fagosit menjadi parameter pengukuran. Hasil menunjukkan ekstrak daun flamboyan pada dosis 750 mg/kg bb (P4) memiliki nilai rata-rata aktivitas dan kapasitas fagositosis makrofag yang signifikan lebih tinggi berbanding P0, P1, P2, dan P3 (P < 0,05). Dari hasil ini disimpulkan bahwa ekstrak daun flamboyan pada dosis 750 mg/kg bb mampu mempengaruhi dan meningkatkan aktivitas dan kapasitas fagositosis makrofag.

Abstract

The whole plant Acalypha indica L. has been used as a traditional herbal medicine for generations by the society. However, the benefits of the whole plant Acalypha indica L. as a herbal medicine have not been known much, especially as an antifertility plant. It is assumed that the compunds contained within the ethanol extract of the whole plant Acalypha indica L. woulf affect upon one’s reproduction.  This research aimed to know the effect of the ethanol extract the whole plant Acalypha indica L. on pregnancy and fetal with obeserved on prenatal fetal (Mus musculus). It was carried out in july 2011 - february 2012 at Microtechnique Laboratory of  FMIPA Unsyiah. Completely randomized design was applied of the research, consisted of 4 treatments and 5 repetition of each treatment. The treatments were P0 (0 mg/ kg BW (control)), P1 (300 mg/ kg BW), P2 (600 mg/ kg BW), and P3 (900 mg/ kg BW) the ethanol extract of the whole plant  Acalypha indica L. that was given once a day for 7 days. The parameters were number of pregnancy, the average of fetal bodyweight, the number of fetal in uterine, and macroscopic defect. The data was analyzed by analysis of variance and chi-square, also continued by Tukey test. The result showed significant difference of number of pregnancy, the average of fetal bodyweight, and the number of fetal in uterine. The ethanol extract of the whole plant Acalypha indica L. extract could decrease the number of  pregnancy, the average of fetal bodyweight, the number of fetal in uterine on P1 (600 mg/ kg BW) and P2 (900 mg/ kg BW), also could not caused macroscopic defect on fetal.

Abstract

Aceh swamp buffaloes(Bubalus bubalis) are decreasing their population and genetic quality. This study was conducted to determine the influence of lactose and glycerol cryoprotectants on spermatozoa of Aceh swamp buffaloesafterthawing.This study used completelya factorial randomized design with nine treatments, and five replications.Fresh semen of the Aceh swamp buffalowere diluted by using a combination extender lactose cryoprotectants 0 mM (L0), 60 mM (L60), 120 mM(L120) and glycerol 3% (G3), 5% (G5), 7% (G7) with the equilibration of 4 hours.The results showed that the combination of cryoprotectants L120G7 influenced significantly (P < 0.05) on the quality of spermatozoa of the Aceh swamp buffalo(B. bubalis)after thawing.The percentage of sperm motility L120G7 (42.60 1.14);viability L120G7 (55.00 0.71);acrosome integrity L120G7 (52.00 0.71); and plasma membrane integrity L120G7(53.20 1.48). The combination of lactose cryoprotectants 120 mM (L120) and glycerol 7% (G7) was the best combination to maintain the quality of spermatozoa of swamp buffalo. This finding could be used to define a policy for the spermatozoa storage of Aceh swamp buffalo to artificial insemination (AI).

Abstract

Seurukan fish (Osteochilus vittatus) has many advantages, besides the fish also has disadvantages which are the slow growth, so the temperature shock of triploidization technique was expected to solve the problem. The objective of the present study was to obtain an effective temperature to increase of triploidization success of seurukan fish (Osteochilus vittatus). The experimental method and completely randomized design model were used in this study. Five levels of temperature shocks at three replicates were tasted: 4? (cold), 6? (cold), 28? (normal), 35? (heat) and 37? (heat). The sperms and eggs were fertilized in the plastic jar then a total 100 of fertilized eggs (zygotes) were taken randomly 3 minutes after fertilization and soaked in respective temperature for 90 seconds, and then incubated in incubation jars at the water temperature of 28-29?. The results showed that the temperature shock gave the significant effect on the hatching and the success of triploidization success (P?0.05), but did not give the significant effect the fertility and survival rates (P>0.05). The triploid fish can be achieved using cold and heat shock, but the higher triploid fish was recorded at 37? was the best temperature recommended for triploidization of Seurukan fish.
