Abstract

Kalanchoe is the biggest genera of Crassulaceae family and distributed in tropical and subtropical regions. This genera is found to be a rich source of biologically active natural products such as triterpenes, flavonoids and steroids. As a part of our continuing search for novel insecticidal compounds from Indonesian Kalanchoe plants, we examined Kalanchoe daigremontiana collected from Bandung region, West Java, Indonesia. The methanolic extract of the dried leaves of K. daigremontiana was concentrated and extracted with methylene chloride The methylene chloride extract exhibited an insecticidal activity toward silkworms. The methylene chloride extract was partitioned between n-hexane and methanol containing 10% water. The active lower layer was extracted with ethyl acetate. By using the insecticidal activity to follow the separations, the ethyl acetate fraction was separated by combination of column chromatography on Kieselgel 60 and ODS to afford two insecticidal bufadienolides 1 and 2. The structures of these compounds were elucidated based on spectroscopic analysis (UV, IR, NMR, 2D-NMR) and comparison with those related data previously reported. In this paper, the isolation, structural elucidation, and insecticidal activities against the third instar larvae of silkworm will be described.

Abstract

Myristica fatua merupakan salah satu tumbuhan obat asli Indonesia yang berpotensi untuk dikembangkan. Telah dilakukan penapisan fitokimia dan uji toksisitas menggunakan metode BSLT (Brine Shrimp Lethality Toxicity) terhadap ekstrak metanol, ekstrak aseton dan etanol 70%. Selanjutnya ekstrak metanol difraksinasi untuk memperoleh fraksi n-heksana, etil asetat, dan n-butanol dari daun Myristica fatua Houtt dan dilakukan uji toksisitas lebih lanjut. Hasil penapisan fitokimia menunjukkan bahwa daun M. fatua mengandung senyawa terpenoid, steroid, flavonoid, saponin, dan tanin. Potensi antikanker juga ditunjukkan terhadap ekstrak metanol dan fraksi etil asetat yang mempunyai toksisitas paling tinggi dari fraksi lainnya dengan LC50 masing-masing adalah sebesar 64,57 dan 83,75 μg/mL.

Abstract

Macaranga known locally as mahang-mahangan has uniquely ecological function, and also became a part of traditional medicine in Indonesia. Macaranga genus also known as a sources of terpenoid and phenolic (flavonoid) compounds which have biological activity as antioxidant and anticancer (cytotoxicity).There are few phytochemical investigations have been done on M. gigantifolia species. As a part of our continuing research of isolation anticancer compound from natural product, a geranylated flavonoid compound (macarangin) has been isolated from ethyl acetate fraction of Macaranga gigantifolia leaves using chromatography methods. The isolated compund (isolat MG) was elucidated to gain the chemical structure based on spectroscopic data (LC-MS and FT-NMR). Cytotoxicity test of this compound was tested against MCF 7 cell lines, showed that macarangin has a potential activity with IC50 value 119.12μg/mL. Key words: Macarangin, geranylated  flavonoid, Macaranga gigantifolia, MCF 7 cell lines.

Abstract

9-Methoxycanthin-6-one compound has been isolated from Pasak Bumi (E. Longifolia) roots. The isolation process using maseration, colum chromatography vacuum, and recrystallization techniques. E. Longifolia root maserated with n-hexane and methanol respectively. The methanol extract fractionated using column chromatography vacuum with gradient elution (hexane-etil acetate) to obtain 9 fractions. Fraction 5 further purification using recrystallization technique to obtain yellow crystal, identified by LC-MS and NMR spectroscopy as 9-Methoxycanthin-6-one.Keyword: E. Abstract

Mistletoes are used traditionally in Indonesiaas medicinal plant, one of this as anticancer. Cancer isone of degenerative diseases could be triggered by toomuch free radicals in the body, whereas antioxidantcould reduced the risk of degenerative diseases.Mistletoes reposted content flavonoids which known have antioxidant activity. Mistletoe potency as drugmaterial should be studied sa the utilization ofmistletoe could be developed. Antioxidant activitytest using "DPPH free radical scavenger" methodand toxicity test using BSLT (Brine Shrimp LethalityTest) method had been conducted on water andethanol extracts of mistletoe Dendropthoe pentandra(L.) Miq.) that grown on various trees (star fruit,mango, cananga, duku, sour-sop, kepel, mahkotadewa, and tea). The result showed that ethanol andwater extracte-ofD, petandra on all host plants haveantioxidant activity (IC50 value between 6.4 - 51.8ug/mL). The result of toxicity test showed that ethanolextract of mistletoe on cananga, starfruit, kepel andmahkota dewa host plants have toxicity effect againstA. salina larvae with LCso value below 1000 ug/mL,nevertheless ethanol extract on others host plants andwater extract didn It give toxicity effect against A.salina larvae (LC50 > 1000 ug/mL).Key words: mistletoe, D. petandra, DPPH, antioxidant,toxicity, degenerative disea.

Abstract

The Flavanone compound with anti diabetic activity was isolated from ethyl acetate extract of Artocarpus communis leaves using column chromatography techniques. The structure of the flavanone compound was elucidated on the basic of spectroscopic evidence and comparison to published values. This compound, 8-geranyl-4,5,7-trihydroxyflavone, showed strong anti diabetic activity on α-glucosidase inhibition assay with IC50 18.120 µg mL-1.   Keywords: Artocarpus communis, 8-geranyl-4,5,7-trihydroxyflavone, anti diabetic activity

Abstract

Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan senyawa flavonoid terprenilasi dari ekstrak diklorometana daun Artocarpus communis dengan menggunakan uji penangkapan radikal bebas1,1-difenil-2-pikril hidrazil. Ekstrak diklorometana daun Artocarpus communis difraksinasi dengan teknik kromatografi kolom pada silika gel, dan setiap fraksi diuji aktivitasnya. Senyawa aktif, 1-(2,4-dihidroksifenil)-3-[8-hidroksi-2-metil-2-(4-metil-3-pentenil)-2H-1-benzopiran-5-il]1-propanon, menunjukkan aktivitas antioksidan yang moderat dengan IC50 54.6 μg mL-1.

Abstract

Scopoletin was isolated from the methanol extract of leaves of Macaranga hispida. leaf extract was isolated using chromatography with a solvent gradient. Determination of chemical structurebased on spectroscopic data and compared with the reference. Scopoletin cytotoxicity test using MTT assay method against MCF-7 and T47D cells showed strong cytotoxic activity with IC50 of 51.5 and 66.5μg /mL.

Abstract

Tetrameles nudliflora is one of the endemic plants in Wallace region, especially in Mekongga Forest, Southeast Sulawesi, Indonesia. As part of our project in the field of natural drug discovery based on local bioresources, this study aimed to isolate secondary metabolite compounds from the ethyl acetate fractions of the leaves of T. nudiflora and evaluate their cytotoxic activities against breast carcinoma cancer lines, MCF-7. The methods used were various chromatography techniques such as vacuum column chromatography, gravitation column chromatography, thin layer chromatography, preparative thin layer chromatography, and re-crystallization using two different organic solvents. In the present study, two flavonoid compounds, kaempferol (1) and quercetin (2) were identified. The structures of 1-2 were determined using serial spectroscopic techniques, including mass spectrometry, one- and two-dimensional nuclear magnetic resonance. Moreover, using 3-(4,5-dimethylthiazol-2-yl)-2,5 diphenyl tetrazolium bromide (MTT) assay method, both compounds showed in vitro cytotoxic activity IC50 of 34.162 and 133.148 ?g mL-1, respectively against MCF-7 cell lines.
