Abstract

This study aim was to determine lactagogue effect of torbangun leaves to plasma levels of lactogenic hormone and gene expression of their receptors in mammary glands of lactation rats. Lactagogue activity was evaluated by volume of milk was produced by the rats treated with commercial milk booster contained ?katuk' leaves extract (AF), ethyl acetate fraction of torbangun leaves (EA), water extraction of torbangun leaves (AQ) and kaempferol (KP). Lactating rats (n=5) of Sprague dawley with six pups were fed with AF, EA, AQ, and KP in the amount of 50 mg/kg, 30 mg/kg, 80 mg/kg and 60 mg/kg body weight, respectively. The feed was given orally every two days and starting from day 2 after giving birth until day 28. The volume of milk was estimated by the increment pup weight after breastfed. The levels of serum lactogenic hormones were determined by ELISA methods. Moreover, in order to measure the gene expression of the lactogenic hormone's receptors in the mammary gland a real time - reverse transcription polymerase chain reaction (RT-PCR) method was performed. The results showed that ethyl acetate fraction of torbangun leaves (EA) (a) was not significantly stimulating the synthesis of serum prolactin and estradiol at day 14 and day 28 lactation period, (b) down-regulated the gene expression of estradiol receptor (ER?) at day 28, and (c) up-regulated the gene expression of prolactin receptor (PRLR) in mammary gland at day 14 and day 28. This study indicated that ethyl acetate fraction of torbangun leaves was induced milk production, within up-regulated the gene expression of prolactin receptor (PRLR) in the mammary gland of lactation rats.

Abstract

Identification of alternate bioactive compounds isolated from microbial endophytes from plants may lead to better solution against antibiotic resistance. Main objective of this research is to determine the bioactivity of antibacterial compounds produced by endophytic actinomycetes, which was isolated from stem bark of Neesia altissima Bl. at Halimun Salak Mount. Detection of bioactive compounds was assayed using Thin Layer Chromatography (TLC) by employing bioautography. Bioactivity was assayed using crude extract against five microorganisms using agar diffusion methods. Tetracycline was used as a positive control. The result of antibacterial activity from endophytic actinomycetes isolate revealed that the bioactive compounds were effective in inhibiting the growth of bacteria Bacillus cereus (1.25 mm), Salmonella typhimurium (5.75 mm), and Shigella flexneri (4.37 mm). Bioactivity of ethyl acetate extract from bioactive compounds of endophytic actinomycetes has an antibacterial activity against bacteria Bacillus cereus (13.00 mm) and Staphylococcus aureus (15.90 mm), at a concentration of 250,000 ppm. Each of TLC fraction was able to inhibit growth of B. cereus at a minimum concentration of 10,000 ppm. In conclusion, the bioactive compounds produced by endophytic actinomycetes can have inhibitory effect towards B. cereus. The category of that compounds are narrow spectrum. The bioactive compound isolated from endophytic actinomycetes may have other implications such as anticancer, antifungal, and antioxidant.

Abstract

The proviral insertion site in Moloney murine leukemia virus-1 (Pim1) kinase is overexpressed in many human cancer diseases. Inhibition of Pim1 kinase has been reported can suppress cell proliferation and induce apoptotic cell death, so that the protein could be chosen as drug target for the treatment of cancer. Citronellol and geraniol mainly contained in citronella oil that?s one of Indonesian natural products have bioactivity as antitumor agents, so that both of the compounds can be used as guiding compound to be developed as chemoprevention and chemotherapeutic agent compounds for cancer especially for leukemia cancer. In this research, sixteen citronellol and geraniol esters as an inhibitor of leukemia cancer designed were docked based on their interaction with leukemia receptor target of Pim1 kinase by using Molegro Virtual Docker (MVD). The result showed that the binding citronellol and geraniol esters to Pim1 kinase are more stable and better affinity  than the binding between citronellol and geraniol, indicated by rerank score were lower than rerank score of citronellol/geraniolPim1 kinase. The docking resulted in the four top-ranked compounds, namely, citronellyl caproate, citronellyl caprylate, geranyl caproate and geranyl caprylate with rerank score value from -182.560 to -189.822 KJ/mol. These compounds have hydrogen bonding with amino acid residues of Lys 67 and Asp 186 of an active site of Pim1 kinase. Based on Lipinski?s rule of five, among of the four top-ranked compounds, citronellyl caproate and geranyl caproate were predicted have potency as new chemopreventive and chemotherapeutic agents for leukemia anticancer candidates.

Abstract

In the laboratory scale, the effect of the concentration of H2SO4 catalyst (1.25-6.25) (% w/w) and the equivalent mol ratio between lauric acid and glycerol (1:1; 1:2.5; 2.5:1) on the synthesis of a-monolaurin has been studied. The a-monolaurin compound has been synthesized from lauric acid and glycerol was done by batch esterification on the free solvent system. The esterification by using 5% H2SO4 catalyst and equivalent mol ratio between lauric acid and glycerol 1:2.5 produced most monolaurin, and dilaurin in amount of 31.14 and 4.42%, respectively. The monolaurin and dilaurin are identified by thin layer chromatography (TLC), infrared spectrophotometer (FTIR), liquid chromatography-mass spectrometer (LC-MS), and Nuclear Magnetic Resonance (NMR) spectrometer. The spectral data of monolaurin was compared to spectral data of standard a-monolaurin.Telah dilakukan penelitian sintesis senyawa a-monolaurin dari asam laurat dan gliserol, dengan konsentrasi katalis H2SO4 (1,25 s.d. 6,25) (% berat) dan perbandingan molaritas reaktan asam laurat terhadap gliserol (1:1; 1:2,5; dan 2,5:1) sebagai parameter. Pada reaksi esterifikasi dengan konsentrasi katalis H2SO4 5%  dan perbandingan molaritas asam laurat terhadap gliserol 1:2,5 dihasilkan senyawa a-monolaurin terbanyak 31,14% sebagai hasil utama dan senyawa a,a?-dilaurin 4,42% sebagai hasil samping sintesis. Kedua senyawa hasil sintesis diidentifikasi dengan kromatografi lapis tipis (KLT), spektrofotometer infra merah (FTIR), spektrometer massa liquid chromatography-mass spectrometer (LC-MS), dan spektrometer resonansi magnetik inti (NMR). Karakteristik senyawa a-monolaurin hasil sintesis dibandingkan  dengan karakteristik a-monolaurin standar.

Abstract

The aims of the research were to investigate the potential medicine from ethyl acetate fraction of Coleus amboinicus L. for improving milk yield, body weight of rats and its effects on growth of the rat pups. Galactagogue activity was evaluated in terms of quantity of milk produced from the rats treated with ethyl acetate fraction of torbangun leaves (FEA), and commercial milk booster contained ?katuk? leaves extract (AF). Lactating rats (n=5) of Sprague dawley with six pups were fed with FEA and AF in the amount of 30 mg/kg and 50 mg/kg body weight, respectively. The feed were given orally every two days and starting from day 2 after giving birth until day 14. The performance of milk production was measured along the experimental period by weight-suckle-weight method. The rates of pups? growth were measured as the weight gain along experimental period. The rats fed with ethyl acetate fraction of torbangun leaves produced higher milk than AF and the control groups, while FEA did not affected the body weight of rats in comparison to the control groups. Ethyl acetate fraction of torbangun leaves was identified to increase milk production by 17%, while AF by 22%. The mean of yields produced by the rats during lactation period for FEA, AF and control were 7.12±1.16, 7.43±1.04 and 6.07±0.85 g/pups/day, respectively. The FEA administration showed gradually increased of rat pups growth and body weight gain significantly in pups 2-14 days old compared to the control group. The present study reveals the ethyl acetate fraction has the potency as a remedy for improving milk yield without influencing the rats.

Abstract

The Novel compound of analog UK-3A defined as HF-1 (6-hydroxy-N-phenylnicotinamide) and HF-2 (6-hydroxy-N-phenylpicolinamide)was successfully synthesized by amidation reaction of aromatic carboxilyc acids and primary amine by adding the activator of DCC (dicyclohexylcarbodiimide), catalyst DMAP (4-dimethyl aminopyridine, and DMSO (dimethyl sulfoxide) as the solvent. Reaction run for 24 hours in 55oC. Result shows yield of HF-1 as much as 52%, and for HF-2 is 16%. The analogue compounds structure of UK-3A were characterized by spectrophotometer FT-IR, LCMS, and NMR. Citotoxicity assay against Murine Leukemia cells of HF-1 and HF-2 by MTT (3-(4,5-dimethylltiazo-2-yl-) 2,5-diphenyltetrazolium bromide) assay. The result of bioassay showed IC50(inhibitory concentration)value 71 µg/mL and 63 µg/mL for HF-1 and HF-2 respectively. It informed that the analogue compounds have lower activity than UK-3A compound which has IC50 value 38 µg/mL.

Abstract

Tourism is one of the new industries which are able to accelerate economic growth and employment, boosting incomes, living standards and to stimulate other productive sectors. This prompted the Department of Culture and Tourism Rokan Hulu to create the attraction Airpanas Hapanasan with the aim to increase the budget and make the area one of the leading tourist attraction in the province of Riau. In increasing the number of visitors needed an effective marketing strategy to create attraction. The aim of this study was to determine how the marketing strategy undertaken by the Department of Culture and Tourism Rokan Hulu in increasing tourist arrivals area and outside the region. In addition, this study also aims to mengetehaui factors supporting and inhibiting factors in the implementation of marketing strategies.This study used qualitative research methods. Subjects in this study consisted of seven people and data collection techniques are grouped into three sections, namely; in-depth interviews, observation, and documentation. This study uses data analysis interactive model of Miles and Hubermen, using a technique that checks the validity of the data through the extension of participation and triangulation.Results of this study show that the marketing strategy of Culture and Tourism Rokan Hulu is decisive segmentation, perform targeting, promotion through print media and online media. Supporting factors in conducting marketing strategy is the attraction Airpanas Hapanasan an asset in the regions, the object of Hapanasan Airpanas tourists are also breeding butterflies, childrens playground as well as outbound, and the location of Rokan Hulu directly adjacent to West Sumatra. Inhibiting factor is the lack of cooperation with a third party / private sector in managing attractions Airpanas Hapanasan the minimnnya intensity of promotion by local governments in promoting attractions Airpanas Hapanasan the professionalism, knowledge in promoting regional tourism is still minimal and competition attractions same become an obstacle to the government in promoting the tourist attraction.Jom

Abstract

Peperomia pellucida L. Kunth known as “Katumpangan Air” is a potential medicinal plants used traditionally for uric acid, rheumatic, headache, stomachache, antimicrobial. This study aims to establish  the non-specific  and specific parameters of ethanol extract from Katumpangan air (EKA). EKA was collected from three different growth places: South Tangerang (6.833%), Bogor (7.8%) and Yogyakarta (13.125%). The results show that organoleptic extract characterization from three places were same: thick, blackish green brown, bitter and have a characteristic odor. The average content of water-soluble compounds was within range 7.39-13.29%, ethanol-soluble 15.33-16.68%, flavonoid total 3.807-4.244%. The range of moisture content was 12.25-16.34%, ash total 1.21-2.78%, acid insoluble ash 0.19-1.62%, drying shrinkage 21.62-24.98%, specific gravity 1.0010-1.0034 g/mL for the non-specific parameter. Microbial contamination testing results 0.61x103-1.13x103 coloni/g, the contaminant testing mold/yeast 0.1x102-1.7x102 coloni/g, while lead metal 0.15-0.18 mg/kg, cadmium 0-0.11 mg/kg and arsenic <0.005µg/kg.

Abstract

Abstract - The objective of this research is to improve the mine planning performance in PT Freeport Indonesia (PTFI). Currently metal forecasting often faces many scenarios and changes shortly because of some dynamic parameters and conditions. The new improved mine planning must be more powerful to handle it. To achieve the objective, the writer surveyed and analyzed the forecasting performance data in the past and then proceeded with a mapping of forecasting process to see how the parameters linked and conditions applied. The team observed that the current conventional method is too rigid for doing mine-mill model simulation. Adjusting the amount of ore delivery to mill needs extension until the Mine Scheduling or Sequencing, beyond than Cutoff grade adjustment. Furthermore, the geological type of ore mined needs fully integration to the mill-model. After the problem identified, by using the Goldratt?s analyses tool called Current Realty Tree (CRT), a team is constructed to analyze some undesired effects and then to seek its solution. Surveying some literatures and benchmarking to other mining companies are conducted to gain more tactical solutions.Finally, a new mine planning method were set up. It uses mining reserve database, filtering concept and bench percentage mining logical formula. This new method is well accepted and implemented to analyze mining scenario quickly. This new method is now applied side by side with convectional method.  This new design is available to produce a robust plan to support strategic planning group on optimizing the strategic mine planning goals. Keywords: Grasberg Surface Mine, Long Range Mine Planning, Forecasting, Mine Scheduling, Interactive Mine Planning.
