Abstract

In the course of our continuing search for novel paralytic compound from Indonesian plants, the methanol extract of the stem bark of Erythrina variegata (Leguminosae) showed significant paralytic activity against the third instar larvae of silkworm (Bombyx mori). The purposes of this research were isolation and structural elucidation of paralytic compound from the stem bark of E. variegata. Using the paralytic activity following the separation, the methanol extract was separated by combination of column chromatography to yield prenylisoflavone, warangalone. The chemical structure of warangalone was identified based on spectroscopic evidence and comparison with the previous reported. The paralytic activity of warangalone showed weak activity against the third instar larvae of silkworm (B. mori).

Abstract

Phenolic compounds such as kaempferol (1), quercetin (2), and methyl caffeate (3) have been isolated from the ethyl acetate extract of Kalanchoe prolifera (Crassulaceae). Abstract

Pirdot plant(Saurauia vulcani. Kurth)is a plant that grows naturally in a tropical climate. During this decoction pirdot leaves plants are used by the people to treat diabetes disease. The purpose of this study was to isolate triterpenoid compound from ethyl acetate extract of pirdot leaves. The sample extraction was performed by total of maceration method using ethyl acetate as a solvent. The separation and purification of the compound was carried out by column chromatography method followed by stain pattern analysis with thin layer chromatography (TLC). Structure characterization of pure compound was determined using UV, IR, 1H-NMR, 13C-NMR, DEPT 135º, HMQC, HMBC, 1H-1H COSY and MS spectroscopies including compared with various literatures. Based on the analysis result, the pure isolate obtained was identified as a pentacyclic triterpenoid compound, 3-hydroxy acid, 12(13)-en,28-oleanolate acid with the molecular formula C30H48O3. The triterpenoid compound of pentacyclic oleanolic acid is the first reported from the pirdot plant (Saurauia vulcani. Kurth).

Abstract

A new antiplasmodial compound, 2-(3H-diazirine-3-yl)benzaldehyde (1), has been isolated from the Papuan marine sponge Xestospongia sp. The structure elucidation of compound 1 was determined by spectroscopic evidences including UV, IR, MS, 1D and 2D-NMR analysis. Compound 1 showed moderate antiplasmodial activity against Plasmodium falciparum with IC50 value of 1.08 x 10-6 mM. DOI:http://dx.doi.org/10.15408/jkv.v4i1.6735

Abstract

Erythrina plants, locally known as “dadap ayam”, are higher plant species and have been used as a folk medicine for treatment of cancer. To prove the effectiveness of the leaves and stem bark of E. variegata as an anti-cancer agent, the assay in this research was focused on in vitro  test  towards  breast  cancer  cell  T47D.  In  the course  of  our  continuing  search  for novel anti-cancer agent from Erythrina plants, the methanol extract of the leaves and stem bark of  E. variegata  showed  significant  anti-cancer  activity  against  breast  cancer  cell  T47D  in  vitro  using the Sulphorhodamine B (SRB) assay. By using the anti-cancer activity to follow the separations, the methanol extract was separated by combination of column chromatography. The chemical structure  of  an  anti-cancer  compounds  were  determined  on  the  basis  of  spectroscopic evidence  and  comparison  with  the  previously  reported  and  identified  as  an  erythrina  alkaloid (1)  and  isoflavonoid  (2).  Compounds  (1-2)  showed  anti-cancer  activity  against  breast  cancer cell  T47D  used  with  IC50  of    1.0  and  3.3  µg/mL,  respectively.  This  results  strongly  suggested that E. variegata is promising sources for anti-cancer agents.Keywords: Anti-cancer, Erythrina variegata, Leguminoseae

Abstract

Cancer is the second cause of death after cardiovascular diseases in the world. Anticancer prevention used can cause undesirable things. Flavonoids are secondary metabolites derived from natural products that are useful for anticancer treatment. This study was performed to observe the cytotoxic activity of alpinumiso?avone from Erythrina poeppigiana, toward cervical cancer (Hela), colon cancer (WiDr), and hepatoma cancer (HepG2) cells. The cytotoxic activity of alpinumiso?avone was tested using (3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyltetrazolium bromide) assay. The percentage of cell mortality was calculated and the IC50 was calculated using probit analysis. The result shown that alpinumiso?avone has antiproliferative e?ect to colon cancer (WiDr), cervical cancer (Hela), and hepatoma cancer (HepG2) cells with the value of IC50 are 5.63, 7.18, and 18.08 µg/ml, respectively. Based on the value of IC50 alpinumiso?avone is very cytotoxic to colon cancer WiDr cell.

Abstract

Two flavanoid compounds, catechin (1) and epicatechin (2), have been isolated from the stembark of Chisocheton pentandrus. The chemical structure of compounds1and2were identified byspectroscopic data including, UV, IR, NMR (1H, 13C, DEPT 135°, HMQC, HMBC, 1H-1H COSY) and MS and by comparing with previously reported spectral data. Compounds 1 and 2, were isolated in this plant for first time and showed no cytotoxic activity against MCF-7 breast cancer cells.

Abstract

The leaves of Erythrina variegata (Leguminosae) used tradisional plant of an antimalarial. In the course of our continuing search for novel an antimalarial compound from Erythrina plants, the methanol extract of the leaves ofE. variegata showed significant antimalarial activity in vitro toward Plasmodium falciparum in vitro using the lactate dehydrogenase (LDH) method. The methanol extract of the leaves of E. variegata showed against bothstrains of parasite with IC50of 6.8 ?g/ml against K1 and > 60 ?g/ml against 3D7, respectively. The methanol extract of the leaves of E. variegata was separated by using bioassay-guide fractionation. The n-buthanol fraction yieldedthe most activity, exhibiting equipotency against both strains of parasite with IC50of 5.1 ?g/ml against K1 and 13.5 ?g/ml against 3D7, respectively. Furthermore, by using the antimalarial activity to follow separation, the n-buthanol fraction was separated by combination of column chromatography to yield an active compound. The active compound showed antimalarial activity against both strains of parasite used with IC50 of 4.3 ?g/ml against K1 and 23.5 ?g/ml against 3D7, respectively. Its inhibition of the resistant strain (K1) was also much better compared to its inhibition of the sensitive strain (3D7), indicated that the leaves of E. variegata to be potential as antimalarial agents, but its lower potency compared to artemisinin and chloroquin.

Abstract

Erythrina plants known plants ?dadap? is a higher plant that grows in tropical and subtropical regions. E. poeppigiana plants was a source of secondary metabolites, which contain flavonoids. This study aims to isolate the flavonoid compounds from the leaves of  E. poeppigiana through the stages of extraction, fraction, separation and purification. E. poeppigiana leaves powder (2.5 kg) was extracted with methanol and partitioned with n-hexane and ethyl acetate. Furthermore, the separation of ethyl acetate of E. poeppigiana leaves fraction using a combination of column chromatographic was obtained pure compound (5 mg) in the form of a yellow amorphous  solid. The chemical structure of pure compound was based on the data spectroscopy (MS, UV, IR, 1H-NMR and 13C-NMR) and identified as the compound 3,3 ?, 4?, 5,7-pentahidroksiflavon or known as quercetin.

Abstract

Tumbuhan Erythrina variegata (Leguminosae) secara tradisional dikenal oleh masyarakat Indonesia sebagai obat antikanker dan antimalaria. Bagian tumbuhan ini yang biasa digunakan sebagai bahan pengobatan adalah daun dan kulit batang, tapi kandungan senyawa kimia aktif biologisnya belum banyak dilaporkan. Tujuan penelitian ini adalah untuk mengungkapkan senyawa aktif antikanker dan antimalaria secara in vitro yang terdapat di dalam daun dan kulit batang E. variegata. Penelitian dilakukan dengan cara ekstraksi metanol dan fraksionasi dari daun dan kulit batang E. variegata yang dipandu dengan uji antikanker secara in vitro terhadap sel kanker payudara T47D menggunakan metode Sulforodamin B (SRB) dan uji antimalaria secara in vitro terhadap Plasmodium falciparum 3D7 (sensitif klorokuin) dan K1 (resisten klorokuin) menggunakan metode laktatdehidrogenase (LDH). Selanjutnya dilakukan pemisahan fraksi etil asetat daun dan kulit batang E. variegata yang dipantau dengan uji antikanker dan antimalaria secara in vitro menggunakan kombinasi kolom kromatografi diperoleh tiga senyawa aktif (1, 2, dan 3). Struktur kimia senyawa aktif (1, 2, dan 3) ditetapkan berdasarkan data-data spektroskopi dan diidentifikasikan sebagai turunan triterpenoid pentasiklik glikosida(1); flavonoid, eristagallin A (2); dan steroid, (22E)-5α,8α-epidioksiergosta-6,22-dien-3β-ol (3). Senyawa (1) menunjukkan aktivitas antimalaria secara in vitro terhadap P. falciparum strain 3D7 (sensitif klorokuin) dengan IC50 1,8 µg/mL dan terhadap strain K1 (resisten klorokuin) dengan IC50 3,3 µg/mL. Senyawa (2) dan (3) menunjukkan aktivitas antikanker secara in vitro terhadap sel kanker payurada T47D dengan IC50 masing-masing 3,0 dan 3,2 µg/mL. Tumbuhan E. variegata mempunyai potensi sebagai bahan dasar obat herbal antikanker payudara dan antimalaria.
