Abstract

Post thawed frozen semen viability is one of the most important keys in artificial inseminationdepended on two major cell death mechanism, apoptosis and necrosis. It has been known that good fertilitydiary bull?s seminal plasma contains high concentration of osteopontin (OPN). Osteopontin also known ascell survival protein via inhibition to apoptotic cell death, and B-cell CLL/Lymphoma-2 (Bcl-2)expressionmostly related to the ability of cell survival. The aim of this study was to investigate the influence ofadditional OPN into frozen semen diluter on post thawed sperm Bcl-2 expression. Fresh semen collectedfrom ±4 year Friesian Holstein bull. Treatment group divided into four groups i.e.: control group (P0:without OPN supplementation), (P1: fresh semen with OPN supplementation 5 ?g/5.107 spermatozoa), (P2:fresh semen with OPN supplementation 10 ?g/5.107 spermatozoa), (P3: fresh semen with OPNsupplementation 20 ?g/5.107spermatozoa). Bcl-2 sperm expression was determined usingimunocytochemistry. The result of this study showed that there was no significant difference betweengroup P0 and P1 (p>0,05), but both group P2 and group P3 showed a significant difference with P0 (p<0,05).Nevertheless, there was no significant difference between group P2to group P3 on post thawed FriesianHolstein sperm Bcl-2 expression (p>0,05). The conclusion of this study is osteopontin supplement in frozensemen diluter is capable to increase post thawed Friesian Holstein sperm Bcl-2 expression.

Abstract

The aim of this study was to measure the effect of corn oil in feed on estrogen hormone level in blood, hierarchy follicular performance and productivity of Japanese quail. Thirty female Japanese quail were housed in caged and fed ad libitum, the birds were randomly divided into three grouped namely: Po, P1 and the (P0) was fed with commercial feed without corn oil, whereas the first test treatment (P1) was fed with commercial feed mixture containing 3% corn oil, and the second test treatment (P2) was feed with commercial feed mixture containing 6% corn oil along the study. Every day the egg production was recorded. After eight weeks, blood samples were collected from jugular vein to isolate the serum. Then all samples were sacrificed to observe the hierarchy follicular performance. Based on this study it was found that 3% and 6 % corn oil supplementation on feed (P1 and P2) did not cause any significant changes in serum estrogen concentration, and productivity, but had significant changes on hierarchy follicular. This result show that hierarchy follicular increasing due to ovarian activity optimizing induced by polyunsaturated fatty acid (PUFA)by consumed corn oil. Thus, the suplementation of corn oil can be an alternative to increase egg productivity of quail farm. ABSTRAK Penelitian ini bertujuan untuk mengukur pengaruh pemberian minyak jagung dalam pakan terhadap kadar estrogen serum darah, hirarki folikular dan produktivitas burung puyuh. Sebanyak 30 ekor burung puyuh betina dibagi menjadi tiga perlakuan. Perlakuan kontrol diberi pakan komersial tanpa suplementasi minyak jagung, sedangkan perlakuan P1 dan P2 diberi pakan komersial dengan suplementasi minyak jagung sebanyak 3% dan 6%. Setiap hari dilakukan pencatatan terhadap produktivitas. Setelah delapan minggu, dilakukan koleksi sampel darah, kemudian semua burung puyuh dikorbankan nyawanya untuk mengamati hirarki folikular yang terbentuk. Pada P1 dan P2 tidak telihat perbedaan yang nyata terhadap peningkatan kadar estrogen serum darah dan produktivitas dibandingkan dengan P0, akan tetapi terlihat perbedaan nyata pada hirarki folikular yang terbentuk. Hasil ini menunjukkan bahwa peningkatan hirarki folikular yang terbentuk terjadi karena peningkatan kinerja ovarium dipicu oleh polyunsaturated fatty acid (PUFA) yang berasal konsumsi minyak jagung. Dengan demikian, suplementasi minyak jagung dapat dijadikan alternatif untuk meningkatkan produksi telur dalam peternakan burung puyuh.

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak kedelai (Glycine max) terhadap jumlah pertumbuhan folikel ovarium pada mencit (Mus musculus). Sampel terdiri dari 20 ekor mencit yang secara acak dibagi menjadi lima kelompok. Kelompok kontrol (K) tidak diberi ekstrak kedelai. Kelompok P1 diberi ekstrak kedelai dengan dosis 0,05 mg/kgBB. Kelompok P2 diberi ekstrak kedelai dengan dosis 0,010 mg/kgBB. Kelompok P3 diberi ekstrak kedelai dengan dosis 0,015 mg/kgBB. Kelompok P4 diberikan ekstrak kedelai dengan dosis 0,020 mg/kgBB. Ekstrak kedelai diberikan selama 14 hari. Analisis data menggunakan uji One Way ANOVA dan dilanjutkan dengan uji Honestly Significant Difference (HSD). Hasil penelitian menunjukkan kelompok kontrol (K) memiliki perbedaan signifikan dengan kelompok P1, P2, P3 dan P4. Hasil rata-rata jumlah folikel primer tertinggi terdapat pada perlakuan 4 (P4) dengan dosis 0,020 mg/kgBB dan jumlah rata-rata folikel sekunder tertinggi pada perlakuan 3 (P3) dengan dosis 0,015 mg/kgBB. Penelitian ini dapat disimpulkan bahwa ekstrak kedelai yang diberikan pada mencit betina dapat meningkatkan jumlah pertumbuhan folikel ovarium dan jumlah rata-rata folikel tertinggi adalah pada perlakuan 3 (P3) dengan dosis 0,015 mg/kgBB.

Abstract

Penelitian ini bertujuan mengisolasi protein Insulin Like Growth Factor-I (IGF-I) complex untuk meningkatkan kualitas semen beku kambing pada waktu ekuilibrasi setelah penambahan protein IGF-I complex. Penelitian ini terdiri atas 2 tahap yaitu isolasi protein IGF-I complex dari plasma seminal kambing dan aplikasi terhadap prosesing pembekuan semen. Pada tahap pertama dilakukan identifikasi IGF-I complex dengan menggunakan gel native polyacrylamide gel electrophoresis dan isolasi IGF-I complex. Pada tahap kedua dilakukan aplikasi penambahan protein IGF-I complex pada prosesing semen beku. Semen dikoleksi dengan menggunakan vagina buatan dan kemudian disentrifus selama 5 menit dengan kecepatan 1800 rpm. Kemudian semen dibagi menjadi tiga kelompok. Pada kelompok I, II, dan III ditambahkan protein IGF-I complex masing-masing 0, 12, dan 618 ng/ 3X10 sperma. Selanjutnya dilakukan ekuilibrasi selama 1 jam dan dilanjutkan dengan evaluasi motilitas, viabilitas, dan membran sperma. Hasil penelitian menunjukkan perbedaaan motilitas, viabilitas, dan membran sperma yang signifikan (P 0,05) di antara tiga kelompok perlakuan. Dapat disimpulkan bahwa penambahan protein IGF-I complex dapat meningkatkan kualitas semen beku kambing pada fase ekuilibrasi.

Abstract

Penelitian ini bertujuan mengetahui pengaruh kondisi lingkungan dalam aspek pH, suhu dan kelembaban antara sarang alami dengan sarang semi alami terhadap presentase daya tetas dan masa inkubasi telur Penyu Lekang (Lepidochelys olivacea) di kawasan Pantai Boom Banyuwangi. Sampel dipilih dengan metode purposive sampling berupa 20 telur Penyu Lekang yang berasal dari satu induk yang sama. Telur Penyu Lekang yang di inkubasi pada sarang alami dan sarang semi alami masing-masing berjumlah 10 butir. Rancangan penelitian menggunakan metode deskriptif, dengan melakukan observasi terhadap kondisi lingkungan antara sarang alami dengan semi alami. Uji t independent digunakan untuk membandingkan presentase daya tetas. Analisis dilakukan dengan program SPSS. Hasil penelitian menunjukkan presentase daya tetas pada sarang alami berbeda sangat nyata dengan sarang semi alami, sedangkan masa inkubasi tidak dapat diambil data. Presentase daya tetas pada sarang alami lebih baik dibandingkan dengan sarang semi alami. Sarang semi alami memiliki pH yang lebih asam dibandingkan pH pada sarang alami. Simpulan penelitian ini terdapat perbedaan kondisi lingkungan pada aspek pH, suhu, dan kelembaban antara sarang alami dan sarang semi alami yang mempengaruhi masa inkubasi dan daya tetas telur Penyu Lekang.

Abstract

The purpose of this study was known the difference of reproductive performans Limousine Cross Breed that haved artificial insemination from diffrence straw from BBIB Singosari and BBIB Lembang covering Conception Rate, Service per Conception, and Calving Rate in period 2016th. This study uses survey research which data from primary and secondary. Primary data were obtained from interview with Limousine Cross Breed breeders and secondary data were obtained from artificial insemination officer?s record. The observed variables are Conception rate (CR),  Service per conception (S/C) and Calving rate  (CvR). Data were analyzed descriptive that average and Chi Square.Result of research showed no significant difference (p>0.05) on CR, S/C and CvR between Limousin cross breed cows that haved artificial insemination from BBIB Singosari and BIB Lembang.

Abstract

The purpose of this study was to know the effect of L-arginine on the histology of primary spermatocyte count in mice (Mus musculus) after high temperature exposed. The subjects of this study were 20 adult male mice, 8 weeks old with an body weight range from 20-40 grams. This research conducted by using Complete Randomized Design (RAL) with 4 treatments and 5 replications. The treatments consisted of, P0- = treatment with 1ml of aquabidest without high temperature exposed, P0 + = treatment with 1ml of aquabidest after high temperature exposed for 1hour per-day for 35 days, P1 = 1.3mg / day L-arginine dissolved in 1ml aquabidest and given orally after high temperature exposed for 1hour per-day for 35 days and P2 = 2.6mg / day L-arginine dissolved in 1ml aquabidest given orally after high temperature exposed for 1hour per-day for 35 days. Observations done by making histologic preparations of testicular organs and then calculated the total number of primary spermatocyte per treatment. The data of primary spermatocyte calculated and analyzed by using Analysis of Variance (ANOVA), followed by Duncan test. The result from data analysis showed that there was a significant difference (p<0.05) between P1 and P2 with control,between P1 and P2 the analysis did not show any significant  difference (p> 0.05).

Abstract

This study aimed to determine the relationship between heat detector reading results and the feature of vaginal swab preparations as a parameter for determining estrus in Peranakan Etawa goats. Five healthy non-pregnant goats weighing 20-30 kg, were synchronized using PGF2? (7.5 mg Dinoprost, Enzaprost) intramuscularly twice with 11 days interval. Vaginal cytology examination began the day after the second PGF2? injection. The heat detector (Draminski®, Drami?ski SA, Poland) is an estrus detecting device based on the amount of electric current flowing through the vaginal mucus. Spearman correlation analysis showed a relationship (p <0.05) between heat detector reading and estrus detection based on vaginal cytology. The heat detector readings were very significantly correlated positively (p <0.01) with the number of epithelial cells (coefficient correlation, r= 0.564), but negatively correlated with the number of cornified cells (r = ?0.854), and also significantly correlated positively (p <0.05) to the number of leukocytes (r = 0.487). The estrus stages were significantly correlated positively (p <0.05) only to the number of leukocytes (r = 0.918). Likewise, the number of epithelial cells was highly correlated negatively (p <0.01) only to cornified cells (r = ?0,643), and the number of cornified cells was significantly correlated negatively (p <0.05) only to the number of leukocytes (r = ?0.472). It could be concluded that the features of the proestrus, estrus and metestrus stages based on the goat vaginal cytology was correlated with the reading results of the heat detector.

Abstract

Duck is one of the most poultry that potential can be best developed for its productivity to be used for meat and eggs. The obstacles faced by farmers include productivity of low and not meat and duck eggs. Application of science and technology through the development of reproductive technology of Artificial Insemination (AI) by conducting cross-breeding. The application to cross ducks is very beneficial because in addition to being economical it is also efficient. AI duck is very easy to apply and fast, does not require a complicated and long process as in large livestock. Ducks from artificial insemination are produced in Turi local female ducks (Anas plathirinchos) with plump males (Cairina moschata). The reason for the crossing is because the duck man has a body size that is too large compared to the local duck, so it can produce offspring with rapid growth and large body size. The conclusion is that duck farming with IB in Tawangrejo Turi Lamongan is useful for increasing body weight and productivity.

Abstract

This study aimed to determine the influence of mixture diluter of chicken egg yolk and young coconut water on the integrity of plasma membrane and abnormalities of Sapudi sheep spermatozoa. The experimental design used in this study was a complete random method. The data was analysed using ANOVA, and if there is a significant difference then proceed with Duncan?s Multiple Range Test ( DMRT ) at significance level of 5%. Based on the result of the research, the influence of mixture diluter of chicken egg yolk and young coconut water was not significantly different ( p > 0,05 ) on the intact plasma membrane of spermatozoa on the first day until the fourth day. Interaction between treatment and storage duration had no effect on spermatozoa abnormality on the first day, second day and the fourth day. It can be concluded that mixture diluter of chicken egg yolk and young coconut water had a good effect on the plasma membrane and spermatozoa abnormalities up to the third day.
