Abstract

It is well known that soybean has beneficial health effects. There are lot of active compounds in soybean, like protein and anti nutrition factors (ANF). Trypsin inhibitor and lectin, two kinds of ANF have an adverse effect on the morphology and function of digestive tract in animals. b-conglycinin in soybean protein, has been proven  has reducing body weight effect through increasing cholecystokinin (CCK) level. The aim of this study was to measure plasma CCK level and the histological changes of jejunum in Wistar rats after treatment with protein extract of Willis raw soybean (PEWS), protein extract of Detam 1 raw soybean (PEDS) and protein extract of Detam 1 tempeh (PEDT) for 14 days. This study was also to ascertain whether b-conglycinin and ANF contribute  to  reducing body weight by giving PEWS, PEDS, and PEDT to 4 groups of 6 rats for 14 days. We observed food intake, body weight, CCK level, and histological profile of jejunum. As a conclusion, PEWS, PEDS, and PEDT treatment to Wistar Rats  for 14 days caused increasing CCK plasma level and jejunum villi atrophy. The reducing body weight is caused not only by b-conglycinin but probably by ANF as well.
SOYBEAN IS A GOOD SOURCE OF PROTEIN. IT HAS TWO MAJOR FRACTIONS, B-CONGLYCININ (7S) AND GLYCININ (11S). B-CONGLYCININ’S FUNCTION WAS KNOWN TO SUPPRESS FOOD INTAKE, AND THIS EFFECT MAY BE DUE TO STIMULATING ENDOGENOUS CHOLECYSTOKININ (CCK) RELEASE. Abstract

Soybean is a good source of protein. It has two major fractions, b-conglycinin (7S) and glycinin (11S). b-conglycinin?s function was known to suppress food intake, and this effect may be due to stimulating endogenous cholecystokinin (CCK) release. The aims of this study were to determine the highest content of total b-conglycinin and b-conglycinin sub unit-b level obtained from two varieties of soybean i.e. Wilis and Detam 1 varieties using different preparation and extraction methods. These two soybean varieties were prepared into tempeh. Then the seed and tempeh were extracted using Deak and Panthee methods. There were six extracts analysed using sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS PAGE) and coomassie brilliant blue (CBB) staining. The result was shown that Detam variety and raw seed contained the highest total b-conglycinin level. And Panthee method was the best method for extraction of total b-conglycinin, while Deak method was the best method for extraction of b-conglycinin subunit-b.

Abstract

BACKGROUND: Hypercholesterolemia is a major risk factor for coronary heart disease. Pharmacological treatment of  hypercholesterolemia are known to have side effects, hence the need for an alternative medicine, like herbal medicine, such as soybean as an example. Research objective is to compare the effects of ethanol extract, protein extract, and ethyl acetate fraction of soybean tempeh Detam-1 on the total serum cholesterol level of male mice.METHODS: This study was a laboratory experimental study with Completely Randomized Design (CRD) method. Twenty five balb-C strain male mice were allocated into five groups, namely: the negative-control (aquabidest), the positive-control (simvastatin), ethanol extract, protein extract and ethyl acetate fraction of the soybean tempeh Detam-1 and all were fed with high cholesterol containing food for 31 days. Starting on the 18th day all groups were treated for 14 days. The observed parameter was the total serum cholesterol level. The data of the study results were analyzed with ANOVA, followed by Tukey HSD test.RESULTS: There were significant differences between treatment groups and the negative-control (p=0.004). The reduction of the total cholesterol level of soy ethanol extract was 35.11±15%, the ethyl acetate fraction of soybean tempeh 26.48±17%, and soy protein extract was 15.33±33.1%, and no significant difference was noted in the positive-control group (simvastatin). CONCLUSION: The ethanol extract of soybean seed, protein extract of soybean seed, and ethyl acetate fraction of soybean tempeh Detam-1 have the same effect as do simvastin, and ethanol extract of soybean seed is the most effective substance for lowering total serum cholesterol in Balb-C male mice.KEYWORDS: hypericholesterolemia, ethanol extract, protein extract and ethyl acetate fraction, soybean Detam-1

Abstract

Latar Belakang Biji kedelai Detam 1 dan daun jati Belanda berefek menghambat kenaikan berat badan, akan tetapi dikhawatirkan mempengaruhi organ hepar. Tujuan penelitian Tujuan penelitian untuk mengetahui efek ekstrak etanol biji kedelai Detam 1 (EEKD), ekstrak etanol daun jati Belanda (EEJB) dan kombinasinya terhadap penghambatan kenaikan berat badan dan gambaran histopatologis hepar pada tikus Wistar yang diberi pakan tinggi lemak (PTL). Metode penelitian Penelitian merupakan eksperimental laboratorium dengan rancangan acak lengkap bersifat komparatif. Sebanyak 40 ekor tikus Wistar jantan dibagi secara acak menjadi 8 kelompok perlakuan, masing masing terdiri dari 5 ekor. Selanjutnya diberi perlakuan selama 28 hari, semua kelompok kecuali kelompok kontrol negatif (KN), tetap diberi PTL. Pada hari ke-29, seluruh tikus dikorbankan dan semua hepar tikus, kecuali kelompok Orlistat (K6), dibuat sediaan histopatologis dengan pewarnaan Haematoxylin Eosin (HE). Hasil Penghambatan kenaikan berat badan terjadi pada semua kelompok perlakuan, kelompok K3 (EEKD 10 mg : EEJB 20 mg) menunjukkan penghambatan kenaikan berat badan yang paling baik dan potensinya setara dengan kontrol positip (KP) atau Orlistat. Pada semua kelompok perlakuan (K1, K2, K3, K4 dan K5) terjadi perubahan struktur arsitektur dan inflamasi di daerah portal namun tidak menyebabkan bengkak keruh dan degenerasi lemak. Kesimpulan: Pemberian kombinasi EEKD 10 mg : EEJB 20 mg menunjukkan penghambatan kenaikan berat badan yang paling baik. EEJB sediaan tunggal menyebabkan perubahan gambaran histopatologis hepar paling buruk pada tikus Wistar jantan yang diinduksi pakan tinggi lemak. Abstract

Soybean Detam 1 and Jati Belanda leaves are potential alternative medicines for treating dyslipidemia. The purpose of this study is to explore  the potential of the combination of Ethanolic Extract Soybean Detam 1 (EEKD) and Jati Belanda (EEJB) in lowering LDL cholesterol and triglycerides compared to single extract and Simvastatin. Using a real laboratory experimental method with CRD, a total of 36 male Wistar rats were divided into 6 groups and administrated for treatment of 28 days. The results showed a significant decrease of LDL cholesterol as well as triglyceride in NC  as compared to P1, P2, P3 (p <0.05) and no significant difference with PC (p> 0.05). The highest suppression of LDL cholesterol between P1 with P2 and P3 is (p <0.05) while the triglyceride is between P1 and P2 (p <0.05). This study concludes that the combination of EEKD 10 mg, 20 mg EEJB, EEKD 30 mg and EEJB 30 mg is significantly lowering LDL cholesterol and triglyceride. The combination of EEKD 10 mg, 20 mg EEJB is the highest compression of LDL cholesterol and triglyceride, and has an equivalent effect with simvastatin in lowering LDL cholesterol and triglycerides in male Wistar rats induced with high fat diet. Keywords: LDL cholesterol, triglycerides

Abstract

The combination of Detam 1 soybean and Jati Belanda leaves proved to have effect of weight loss. Since it will become longterm using, toxicity effectvresearch is needed.  This study was conducted to determine the effect of subchronic combination of ethanol extract of soybean Detam 1 (EEDS) and Jati belanda (EEJB) on kidney function, with parameters: urea and creatinine, macroscopic and histopathological of kidney. This study were conducted on 120 Wistar rats (60 males, 60 females), 90 days (treatment group) and 120 days (satellite group). Rats were divided into six treatment groups. Test results of urea and creatinine levels of all treatment groups was lower than control group (p <0.05). The treatment turned out to have a good effect on kidney function. Kidney weight of all groups showed no significant difference with the control group (p> 0.05). Histopathological score interpretation’s result of low dose group male and female kidney rats were not disturbed. In conclusion, the combination of EEDS and EEJB  have a good effect on kidney function, did  not lead to changes in organ weight and at low doses did not cause disturbance of  renal histopathology. Key words: combination of soybean jati Belanda, subchronic treatment, kidney function, kidney’s  weight, kidney’s histopathology

Abstract

Dental caries or tooth decay is a result of the production of acid from fermentation ofbacteria that destroy tooth tissue. It can be prevented by maintaining the stability of salivaryvolume and acidity. Chewing xylitol gum is beneficial for increasing salivary volume andacidity. The aim of the study is to determine the effect of chewing xylitol gum on salivary volumeand acidity. The study is a quasi experiment conducted to 30 males aged between 17-25 yearsold. Pre and post treatment’s salivary volume are measured with measuring cup. Pre and posttreatment’s salivary acidity are measured using a pHmeter with 0,1 accuracy. Data areanalyzed with paired “t” test (a = 0,05). Average salivary volume after chewing xylitol gumincreased very significantly from 0,26 mL to 13,37mL (p=0,000). Average salivary acidity afterchewing xylitol gum increased very significantyly from 6,47 to 7,36(p=0,000). The researchconcludes that chewing xylitol gum increases salivary volume and acidity.Keywords : dental caries, xylitol, salivary volume, salivary acidity

Abstract

Premenstrual syndrome (PMS) is complex of symptoms that occur during the luteal phase of the menstrual cycle. This study aimed to observe blood calcium levels differences between the patient and not the PMS, list the average intake of calcium / day among subjects, statistically analyze the relationship between calcium intake and blood calcium levels, and examine the effect of calcium supplementation on blood calcium levels and symptoms of PMS. We examined blood levels of calcium, analytical survey method using a cross-sectional study, experimental methods and PMS symptom questionnaires. The results showed that in the group of subjects SPM average blood calcium level, and the mean intake of calcium / day were lower than in non-PMS and showed a fairly correlation between the intake of calcium per day with blood calcium levels in SPM subjects. There is a significant relationship between increased calcium blood calcium levels and symptoms of PMS. The conclusions: 1. there are differences in blood calcium levels and 2. intake of calcium per day between patients with PMS and not PMS, 3. Calcium intake is significantly associated with calcium level in the PMS and 4. calcium supplementation affects blood calcium levels and reduce symptoms of PMS.Keywords : average calcium intake, blood calcium, calcium supplementation, Premenstrual Syndrome (PMS)

Abstract

Salah satu pilihan pengelolaan obesitas adalah memperlambat absorbsi asam lemak dengan cara menginhibisi enzim lipase dalam saluran pencernaan. Inhibitor lipase pankreas dilaporkan telah berhasil dalam pengelolaan terapi obesitas. Senyawa aktif dalam Jati Belanda dan kedelai dapat berfungsi sebagai inhibitor lipase. Tujuan penelitian ini adalah mengetahui aktifitas lipase dari ekstrak etanol kedelai Detam 1 (EEKD), daun Jati Belanda (EEJB), kombinasi serta hasil fraksionasinya menggunakan metode standar dan modifikasi. Uji aktifitas lipase terhadap EEKD, EEJB, kombinasinya serta fraksi etil asetat, air dan n-heksana menggunakan kit Lipase Liquicolor (HUMAN®) dengan prinsip kolorimetri, menggunakan spektrofotometer 580 nm. Metode modifikasi menggunakan 96 well plate, microplate reader 630 nm. Penelitian ini menyimpulkan bahwa aktifitas inhibitor lipase tertinggi didapatkan pada sampel kombinasi EEKD:EEJB perbandingan 1:2, baik menggunakan metode standar maupun metode modifikasi; fraksi yang mengandung aktifitas inhibitor lipase paling tinggi adalah fraksi air kedelai Detam 1 (KD-1) dan air Jati Belanda (JB) pada 100 ppm, serta aktifitas lipase dalam fraksi air JB lebih tinggi dari fraksi air KD-1.
