Abstract

It has been known that phytoplasma inducing witches broom disease can infect various kind of leguminous plants in Indonesia, among others soybean. Information on the resistance of various kind of soybean varieties to the phytoplasma is still limited. Experiments were conducted to eveluate the resistance of ten soybean varieties/lines to the phytoplasma. The evaluation was carried out by inoculation method through Orosius argentatus Evans, with two days of acquisition feeding period, ten days latent period and two days inoculation feeding period. Observation was conducted on the presence of phytoplasma, incubation period, symptom and number & weight of seed. The result indicated that line Malang 3474 was resistant, Sindiro and Sriono were tolerant, while the other tested varieties/lines, i.e. Galunggung, Orba, Ringgit, Wilis, Malang 2999, Malang 2805 and Sicinang were susceptible

Abstract

ABSTRACTMeristem tip culture is culture of isolated meristem with 1-2 leaf primordia on suitable medium. This method is generally used to obtain free virus plant. Optimation of plant growth regulators (PGRs) was done to accelerate explant growth without callus formation and to avoid somaclonal variation in meristem tip culture. The aims of this study were to achieve the best combination of PGR for meristem tip growth and to evaluate meristem tip culture potential for Onion yellow dwarf virus (OYDV) elimination in shallot. This study used combination of PGRs 0.25 mg L-1 (2-ip, BAP, GA3, kinetin) with or without 0.1 mg L-1 IAA and medium without PGR. This research consisted of two experiments conducted separately. In experiment I, cv. Bima Brebes was used and experiment II cv. Tiron was used. Each experiment was arranged in completely randomized block design with single factor (PGR combination) that has 8 combination levels and 3 replications. The result showed that medium without PGR was the most efficient for meristem tip growth. Primary shoot was growing without callus formation. RT-PCR analysis showed that all of the tested samples were still infected by OYDV. Meristem tip culture method did not eliminate OYDV in both cultivars.Keywords: Auxin, cytokinin, GA3, OYDV, RT-PCRABSTRAKKultur meristem tip merupakan kultur meristem yang diisolasi 1-2 primordia daun dan pada media yang sesuai. Metode ini umum digunakan untuk mendapatkan tanaman bebas virus. Optimasi terhadap zat pengatur tumbuh (ZPT) dilakukan untuk mempercepat pertumbuhan eksplan tanpa disertai pembentukan kalus untuk menghindari terjadinya variasi somaklonal pada kultur meristem tip. Penelitian ini bertujuan untuk mendapatkan kombinasi ZPT terbaik bagi pertumbuhan meristem tip dan untuk mengevaluasi potensi kultur meristem tip dalam mengeliminasi virus Onion yellow dwarf virus (OYDV) pada tanaman bawang merah. Penelitian ini menggunakan 0.25 mg L-1 (2-ip, BAP, GA3, kinetin) dengan penambahan atau tanpa 0.1 mg L-1 IAA serta media tanpa ZPT. Penelitian ini terdiri atas 2 percobaan terpisah. Percobaan 1 menggunakan cv. Bima Brebes dan Percobaan 2 menggunakan cv. Tiron. Masing-masing percobaan disusun berdasarkan rancangan kelompok lengkap teracak (RKLT) dengan 1 faktor, yaitu kombinasi ZPT yang terdiri atas 8 taraf kombinasi dan 3 ulangan. Hasil yang didapat menunjukkan bahwa media tanpa penambahan ZPTmerupakan media yang paling efisien untuk pertumbuhan tunas meristem tip. Tunas utama tumbuh tanpa disertai pembentukan kalus. Hasil analisis RT-PCR menunjukkan bahwa seluruh sampel yang dideteksi masih terinfeksi OYDV. Metode kultur meristem tip belum dapat mengeliminasi virus OYDV pada kedua kultivar bawang merah.Kata kunci: Auksin, GA3, OYDV, RT-PCR, sitokinin

Abstract

Cowpea mild mottle virus (CPMMV) is one of important virus which infects soybean and become an endemic disease since the first time reported in Java and Sumatra. Research was conducted to study the host range of CPMMV and to evaluate response of new type soybean varieties to CPMMV specific isolate.  Host range study of CPMMV CR16 isolate was conducted by sap transmission method to 11 species of indicator plants, whereas evaluation of soybean response involved 10 varieties.  The resistance response of soybean variety was based on incubation period, and of either  the disease incidence or severity. Virus infection on test plants were confirmed by dot immunobinding assay (DIBA) using specific CPMMV antisera. The CPMMV CR16 isolate was able to infect systemically 8 plant species belongs to Leguminoceae, and Solancaeae; while the virus caused local infection on Amaranthaceae. All plant species infected systemically proved to become sources of inoculum for CPMMV when they used in back inoculation to soybean.  Response of soybean varieties to CPMMV CR16 isolate can be categorized into susceptible (Detam 1, Detam 2, Detam 3, Anjasmoro, Wilis), moderated (Detam 4, Malika, Dena 1) and resistant (Argomulyo and Grobogan).

Abstract

Infection of Onion yellow dwarf virus (OYDV) are reported causing problems in garlic production. Planting virus-free bulbs might help reduce viral disease incidence in the field. This research was aimed to develop method for eliminating OYDV from garlic bulbs using combination of electrotherapy (0, 5, 10, 15, and 20 mA each for 10 minutes) and thermotherapy (23, 28, 33, 38°C each for 4 weeks). Two garlic cultivars, i.e. Sangga Sembalun and Lumbu Hijau were used as seed bulbs for OYDV elimination tests. Virus infection was confirmed using transcription-polymerase chain reaction (RT-PCR).  The result showed that thermotherapy at 33 °Cwas the best method to eliminate OYDV in garlic although the efficiency was not the same for all cultivars. The efficiency reached up to 60% for cv. Lumbu Hijau, whereas for cv. Sangga Sembalun only reached up to 40%. Electrotherapy alone or in combination with thermotherapy were not able to produce OYDV-free plantlets.

Abstract

Severe mosaic symptoms was observed on papaya plants in Meusa village, Kutablang sub district, Bireun district and Lambaro Teunom village, Lembah Seulawah sub district, Aceh Besar district. Systemic mosaic was found in leaves, stem, twig, and fruit. Reverse transcription-polymerase chain reaction (RT-PCR) was conducted to detect the virus causing mosaic symptoms using universal primer for Potyvirus. Amplification of 320 bp DNA fragment was successfully obtained from leaves and fruits bearing severe mosaic symptoms. Nucleotide sequencing was proceeded using the DNA amplicon as the template. Further sequence analysis indicated that Potyvirus isolates from papaya in Aceh show the highest homology (92.7% to 94.7%) with Papaya ringspot virus (PRSV) from Philippines, Thailand, and Indonesia. This is the first report on PRSV infection on papaya in Nanggroe Aceh Darussalam province.   Key words: Potyvirus, RT-PCR, sequencing

Abstract

Plant biochemical resistance towards virus infection can be evaluated through measurement of salicylic acid accumulation and peroxidase concentration. A research was conducted to study biochemical resistance of chillipepper to infection of Pepper yellow leaf curl Begomovirus (PYLCV) the causal agent of pepper yellow leaf curl disease. Virus inoculation was done using insect vector, Bemisia tabaci. Salicylic acid accumulation and peroxidase concentration on 6 chillipepper genotypes (35C2, IPB C15, IPB C26, IPB C10, IPB C14, dan IPB C12) was measured 120 h after inoculation. The results showed that salicylic acid accumulation and peroxidase concentration were higher on plants after virus infection. Susceptible genotypes (35C2, IPB C15, IPB C26) tend to have higher salicylic acid accumulation and peroxidase concentration than those of resistant genotype (IPB C12). Positive correlation was observed between biochemical resistance (salicylic acid accumulation) and structural resistance (length of palisade meshophyll cell).Key words: Begomovirus, Pepper yellow leaf curl virus, peroxide enzyme, resistance mechanism, salicylic acid

Abstract

Ethyl Methane Sulfonate (EMS) may induce mutation leading to somaclonal variation if it is used at the appropriate combination of EMS concentration and exposure time. Variation in somaclonal might be valuable as a source of resistance to plant pathogens including plant viruses. This study was aimed 1) to determine the optimum EMS concentration and incubation time that may induce somaclonal variation in chilli pepper; and 2) to evaluate the resistance of the somaclone to ChiVMV infection. Shoot-tip explants of fi ve chilli pepper genotypes (Jatilaba, ICPN 12#4, PBC495, Helem, and Gelora) were treated with EMS at combination of different concentrations (0.25%, 0.5% 1.0% and control), and incubation time (15, 30, 60 min). Subsequently, each explant was grown in multiplication media (MS media + 5 mg L -1 BAP + 0.5 mg L-1 TDZ), rooting media (MS media + 1 mg L-1 NAA), and acclimatization media (mixture of soil : sand : compost 2:1:1 w/w). Our results showed that the higher EMS concentration and the longer incubation period the smaller the number of survive explants. The highest survival rate 20.4 % was achieved with 0.5% EMS in combination with 60 min incubation period. This treatment combination also showed induction of phenotypic variation. Two somaclonal plants derived from Gelora genotype, designated as somaclones K1 and K2, survived until fruit development and maturation. A total of 245 progenies of K1 and 243 progenies of K2, respectively were evaluated for their resistance to ChiVMV infection through mechanical inoculation using ChiVMV-Cikabayan isolate. Following the detection of ChiVMV using DAS-ELISA, it was confi rmed that 4.09% of the somaclonal progenies were resistance to ChiVMV. Resistance screening were conducted on 28-days-old pepper plants grown in 72-cell ?  ats by inoculating 5 mL of inoculum (contain 105 zoospore mL-1) to the base of each plant. Phytophthora capsici isolate used in this experiment was TG01, which then identi?  ed as race-3. The evaluation of pepper genotypes characteristics were conducted in ?  eld.  Eight characters demonstrated a wide genetic variability; those characters were resistance to Phytophthora blight, infection period, dicotomous height, canopy width, fruit weight, fruit length, fruit diameter and production. These characters along with fruit wall thickness, chlorophyll-a, chlorophyll-b, total chlorophyll had high heritability values. Using path analysis infection period, dicotomous height and fruit diameter are recommended to be simultaneously used as selection criteria for developing Phytophthora blight resistant pepper varieties. Abstract

Patchouli (Pogostemon cablin) is an economically important plant due to its capability to produce essential oil, i.e. patchouli oil. Pest and diseases may contribute to low production of patchouli oil in Indonesia. Aphids are commonly found associated with patchouli plants showing mosaic symptoms, indicated possible relationship between aphids and mosaic virus. Early diagnosis showed that mosaic disease on patchouli is caused by Potyvirus infection. A study was conducted to identify those aphid species and to examine relationship between the aphid species and mosaic virus. Two aphid species commonly found in patchouli plants in Indonesia are Aphis gossypii and Brachycaudus sp. Further study on the ability of A. gossypii to transmit Potyvirus revealed the possible non-persistent relationship, with 25 min optimum pre-acquisition starvation, 15 min acquisition period and 4 hours optimum inoculation feeding period, respectively.Key words: Aphis gossypii, Brachycaudus, nilam, Pogostemon cablin, Potyvirus

Abstract

Viral disease has been reported to cause significant effect on production of shallot (A. cepa) and garlic (A. sativum). The study was conducted to detect main viruses from leaves and bulbs of shallot and garlic using specific antibodies by ELISA method. Leaf and bulb samples was collected from West Java (Bandung, Bogor and Cirebon), Central Java (Brebes), and Yogyakarta (Bantul). Single as well as mix infection of GCLV, SLV, and Potyvirus was successfully detected using specific antibodies. The average percentage of virus infection in the crop ranged from 11.2?14.3% on shallot, and 14.3% on garlic; whereas in the bulb ranged from 11.2?13.3% on shallot, and 9.18% on garlic.
