Abstract

Ikan kerapu cantik merupakan hibrida antara betina kerapu macan E. fuscoguttatus dan jantan kerapu batik E. microdon. Penelitian ini bertujuan untuk mengamati perkembangan dan tingkah laku larva ikan kerapu cantik agar bisa diambil langkah-langkah untuk perbaikan pada pemeliharaan larva sehingga dapat meningkatkan produksi. Penelitian dilakukan di Balai Besar Riset Budidaya Laut dan Penyuluhan Perikanan Gondol, Bali Tahun 2016. Larva dipelihara pada tiga buah  bak semen ukuran 10 m3  yang diisi telur dengan kepadatan 50.000 butir/bak, dipelihara hingga juvenil umur 40 hari (D40). Pemeliharaan larva diulang tiga kali dengan waktu yang berbeda, yaitu siklus 1: Januari-Maret; siklus 2 : Mei-Juli dan siklus 3 : September-Nopember. Pengamatan perkembangan dan tingkah laku larva dilakukan pada setiap siklus yang   dibagi menjadi 4 kelompok umur, yakni: 1) D1 sampai D10; 2). D11 sampai D20; 3). D21 sampai D30; dan 4). D31 sampai D40. Hasil menunjukkan larva ikan kerapu cantik mulai buka mulut dan makan pada D3, aktif berenang setelah tumbuh sirip punggung dan dada yaitu D4, sirip terus berkembang menjadi duri memanjang. D7 kondisi larva terlihat padat dilakukan penjarangan. Duri punggung dan dada mulai memendek, berubah menjadi sirip mulai D30, selanjutnya menjadi juvenil dilakukan grading untuk menekan kanibalisme, D40 dengan ukuran panjang total sekitar 3 cm larva ikan kerapu cantik seluruhnya menjadi juvenil. Perkembangan dan tingkah laku larva ikan kerapu cantik berpengaruh terhadap hasil produksinya.

Abstract

Samples of yellowfin tuna (Thunnus albacares) were taken from three locations Bali, North Sulawesi and North Maluku. The glucose-6-phosphate isomerase (GPI) was analyzed from liver using allozyme electrophoresis method. Polymorphism of GPI enzyme was observed and four alleles (A, B ,C, D) were found in Bali population, three alleles (A,B,C) were found in North Maluku and North Sulawesi populations. Heterozygosity values, from Bali, North Maluku and North Sulawesi were 0.419; 0.417; 0.143 respectively. Genetic distance between North Sulawesi and North Maluku were 0.029, and with Bali population was 0.353. These results indicated that North Maluku and North Sulawesi population were not separate by geographic barrier, therefore genetic distance of both populations was closed. However, those populations were genetically separated to the Bali population might be due to major hydrological barrier.

Abstract

Fertilization rate of the first generation of coral trout grouper, Plectropomus leopardus from wild broodstock in cultured tank was relatively low of 10-35%. Based on gonad histology analysis of 30 samples, only one male was found in early stage maturation. Therefore, it is needed to increase functional male production to ensure an increase level of egg fertilization. The experiment was conducted for 9 months using coral trout broodstock, from wild stock as control and selected F1 from culture as treatment. Broodstocks were reared in three tanks, each was 100 m3 in volume with 54 fishes/tank. All selected cultured fish (F1) in tank C were implanted with 17a-MT hormone at 50 mg/kg of body weight every month for 3 consecutive months. Wild stock fish in tank A and selected cultured fish (F1) in tank B were not implant as controls. The result showed that the number of male  increased 6 times in implanted by 17a-MT hormone compared to fish without implant only increased 2 times. Therefore, eggs fertilization and hatching rate were significantly increased by implanting F1 broodstocks with hormone17a-MT. Abstract

Characterization of three microsatellite loci are able to provide information about genetic variation or polymorphism of yellow fin tuna eggs and larvae produced by domestication of broodstock. Thirty samples of eggs and larvae were collected from different spawning day and analyzed. Amplification of those three loci were carried out with multiflexing technique and electrophoreted ABI Prism 310 Genetic Analyzer sequencer. From those three loci were used (Ttho-1, Ttho-4 and Ttho-7) showed that locus Ttho-7 had higher genetic variance (0.720-1.212) compared to others loci. Based on the result, it can be concluded that high allele frequency as showed at Ttho-7 could be used as marker for  parental analysis.

Abstract

Perbenihan kerapu tikus, Cromileptes altilvelis belum mencapai tingkat keberhasilan yang memuaskan. disebabkan oleh terbatasnya informasi tentang aspek biologi dan lingkungan larva kerapu yang tersedia. Untuk itu perlu dilakukan berbagai penelitian tentang aspek tersebut, seperti pengaruh lama penyinaran terhadap pertumbuhan dan sintasan larva kerapu tikus.

Selain pakan, faktor lingkungan merupakan kendala dominan dalam pemeliharaan larvakerapu bebek. Satu di antara faktor lingkungan yang diduga berpengaruh terhadap pertumbuhan dan sintasan larva adalah warna wadah.
