Abstract

Penelitian bertujuan mengetahui pengaruh sari seduh teh hitam terhadap pengekspresian PPAR ã sel adiposa jaringan lemak visera Rattus norvegicus strain Wistar. Duabelas ekor tikus (Rattus norvegicus strain Wistar) jantan umur 6-8 minggu, berat 200 gram, diberi 4 macam perlakuan, yaitu A (diet tinggi lemak + SSTH 0 g/hari), B (diet tinggi lemak + SSTH 0,015 g/hari), C (diet tinggi lemak + SSTH 0,030 g/hari) dan D (diet tinggi lemak + SSTH 0,045 g/hari) selama 90 hari. Setelah masa perlakuan, tikus dibedah dan diambil lemak viseranya. Jaringan tersebut kemudian dibuat preparat dengan metode parafin. Jumlah sel yang mengekspresikan PPAR ã dianalisis dengan menggunakan pewarnaan immunohistokimia dan untuk mengkonfirmasi bentuk sel adiposa digunakan pewarnaan Hematoxylene&Eosin. Antibodi primer yang digunakan adalah anti PPAR gamma poliklonal antibody rabbit IgG dan antibodi sekunder biotin-goat-anti rabbit IgG. Hasil penelitian menunjukkan jumlah sel adiposa yang mengekspresikan PPAR ã mengalami penurunan seiring dengan penambahan dosis SSTH. Hal tersebut mengindikasikan SSTH dapat menurunkan pengekspresian PPAR ã pada sel adiposa jaringan lemak visera. Kata kunci: jaringan lemak visera, PPAR ã, diet tinggi lemak, sari seduh teh hitam

Abstract

Rosela memiliki anthocyanin sebagai antioksidan polifenol yang berpotensi mencegah progresivitas penyakit akibat stres oksidasi. Diet aterogenik dan stres oksidasi memicu serangan radikal bebas pada komponen lemak dan protein sehingga terjadi perlemakan hepar dan peningkatan ekspresi ADMA (Asymmetric dimethylarginine). Penelitian ini bertujuan mengetahui pengaruh pemberian ekstrak rosela strain Hibiscus sabdariffa terhadap perlemakan hepar dan ekspresi ADMA hepar pada tikus Rattus norvegicus yang diberi diet aterogenik. Studi eksperimental menggunakan post test only control group pada Rattus norvegicus strain Wistar jantan yang dibagi dalam empat kelompok, yaitu kelompok kontrol positif (diet aterogenik), kelompok1 (ekstrak rosela 150mg/kgBB/hari + diet aterogenik), kelompok 2 (ekstrak rosela 300mg/kgBB/hari + diet aterogenik), dan kelompok kontrol negatif (tanpa pemberian diet aterogenik dan ekstrak rosela). Hasil penelitian menunjukkan bahwa ekstrak rosela mampu menurunkan perlemakan hepar yang berdasar pemeriksaan histopatologi berupa jumlah sel yang mengalami perlemakan lebih sedikit (p=0,000). Ekspresi ADMA dari gambaran imunohistokimia menunjukkan ekspresi yang lebih sedikit pada kelompok dengan diberi ekstrak rosela (p=0,000). Dapat disimpulkan pemberian ekstrak rosela menghambat perlemakan hepar akibat diet aterogenik dan ekspresi ADMA akibat diet aterogenik pada tikus, dengan dosis optimum rosela 300 mg/kgBB/hari. Kata Kunci: Diet aterogenik, ekspresi ADMA hepar, perlemakan hepar, rosela

Abstract

Shigella dysentriae is the most common organisms which causes bacillary dysentery in developing countries with high mortality. Pomegranates (Punica granatum) have beenused since a long time ago in folk medicine for several purposes. The antibacterial activity was linked to the presence of major phytocompounds like flavonoids, triterpenes, phenols, and ellagitannin. The aim of this experimental study wasto evaluate the antibacterial effect of Pomegranate?s (Punica Granatum) rind extract against Shigella dysentriae  and to determine the relationship between concentrations of Pomegranate?s rind extract with the growth of Shigella dysentriae in vitro. Antimicrobial effect was determined by tube dilution method. This consists of determination of  Minimum Inhibitory Concentrations (MICs), and Minimum Bactericidal Concentrations (MBCs). The concentrations of Pomegranate?s (Punica Granatum) rind extract were 0.8%, 1%, 1.2%, 1.4%, 1.6%, and 1.8%. At low extract concentration (0.8%)bacterial growth was delayed, while at higher extract concentration (1.8%) bacterial growth was elimanated (ANOVA, p =  0.000, Regression R = -0.901)

Abstract

Metabolic syndrome is a group of several risk factors consist of hypertension, dyslipidemia, glucose intolerance disorder, obesity, and has a relationship with CVD pathomechanism. WHO was report in 2003 more than 300 million adult in the world suffered from obesity. The role of adipose tissue in infectious diseases and adipocyte quality changes after exposure to pathogen has not been revealed. On the other hand, the prevalence of infectious diseases in developing country is still high. Previous studies proved a relationship between infection of Toxoplasma gondii and tissue dysfunction. Profilin is a membrane component of Toxoplasma gondii that able to stimulate expression of proinflammation cytokines through TLR-11 upregulation, and predicted able to trigger the adipocyte dysfunction. The aim of this study are to knows the effect of profilin exposure in vitro on subcutaneous adipocyte to the expression of IL-6 and TNF-α. The measurement of IL-6 and TNF-a levels was done by ELISA technique. The result showed profilin exposure doses 5, 15 and 25 ng / ml increasing IL-6. Profilin exposure cause adipocyte dysfunction through the increasing proinflammatory adipocytokine levels as pathomechanism of metabolic syndrome associated with adipose tissue.

Abstract

Differentiation of pre-adipocytes into adipocytes, is a complex process that involves coordination of  expression of specific genes and proteins associated with each stage of differentiation. This process is regulated by several signaling pathways. CCAAT element-binding proteins (C/EBP)  ?and  ?and sterol response element-binding protein 1 (ADD1/SREBP1) are active during the early stages of the differentiation process and induce the expression and/or activity of the peroxisome proliferator-activated receptor  ?(PPAR?), a pivotal coordinator of adipocyte differentiation. Recently, many biological effects of black tea have been reported and these effects may be partially related to its antioxidant  properties. The recent study showed that black tea flavonoids inhibited adipocyte differentiation. The aim of this study was to investigate the role of theaflavin, a flavonoids in black tea, in inhibiting adipocyte differentiation through inhibition of PPAR?expression. Preadipocytes were incubated in media containing standard-differentiated stimulator: Insulin, dexamethasone, IBMX and FBS. After 24 hours, preadipocytes culture media wasadded with theaflavin 50, 100, or 200  µM. The cells were harvested after TF incubation for 24 hours and then the expression of PPAR?was identified by immunocytochemistry and Western blot.
