Abstract

Background: Phospholipase A2 (PLA2) is involved in inflammation and cell death following stroke, and inhibition of its activity may promote neuroregeneration. This study aimed to observe the influence of Acalypha indica Linn root extract towards relative cell viability and PLA2 enzyme level in post-hypoxic hippocampal tissue culture.Methods: Experimental in vitro study using 24 primary neuronal cell cultures obtained from Sprague Dawley rat exposed to hypoxia with 5% O2 / 5% CO2 / N2 balanced gas for 24 hours. Post-hypoxia, Acalypha indica Linn root extract was added at doses of 10, 15, and 20 mg/mL to three treatment groups. No treatment was given to the control group. Each group consists of six samples. After 72 hours of incubation, relative cell viability was measured using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) examination, and phospholipase A2 enzyme level was determined using ELISA.Results: PLA2 enzyme level of rat hippocampal tissue culture treated with Acalypha indica Linn root extract at 10, 15, and 20 mg/mL were significantly lower than that of control (5.55 ng/mL, 6.85 ng/mL, and 7.42 ng/mL vs 7.96 ng/mL, p < 0.05).Conclusion: Acalypha indica Linn root extract increases the relative cell viability and decreases the PLA2 enzyme level of post-hypoxic mouse hippocampal tissue with the optimal dose of the extract at 10 mg/mL. Abstract

This paper describes the development of a set of data and planning models based instrumentation applications seventeen plus patterns in BK teachers SMP / MTs in Bondowoso. The results of this study indicate that counselling activity plan-ning model using the approach pattern seventeen plus becomes very necessary. B.K. teachers in SMP/MTs of Bondowoso still do not have a wide range of data that should be held for the provision of counselling services. The teachers feel it is important to have variety of data sets and instrumentation applications for the smooth running of counselling services (score = 3.23). The evaluation of the implementation of set of data and applications instrument was still very low (score = 1.14). The planning model development activities and application in-strumentation data set found that B.K. teachers desperately need (score = 4.28). The assessment model development planning activities data set and instrumenta-tion applications that are promoted rated excellent (score = 4.47).Keywords: planning, data set, application instrumentation

Abstract

Sejak diciptakannya mobil dengan ruang penumpang tertutup, muncul pemikiran bagaimana caranya agar ruangan di dalam mobil tersebut tidak terasa panas dan pengap.Setelah berbagai cara dilakukan, muncullah ide yang paling efektif untuk mengurangi rasa panas, gerah, dan pengap di dalam mobil yakni dengan memasang AC (Air Conditioner).Air Conditioner atau yang lebih dikenal dengan AC adalah suatu alat yang berfungsi untuk mengatur suhu udara. Secara umum, cara kerja AC sama dengan cara kerja alat pendingin lain yaitu memanfaatkan proses perpindahan panas dan terjadi proses pendinginan. Untuk keperluan pemindahkan energi panas tersebut, dibutuhkan suatu fluida penukar kalor yang selanjutnya disebut refrigerant.Refrigerant yang digunakan pada AC mobil saat ini adalah R134a.Namun berdasarkan penelitian,refrigerant R134a memiliki potensi sebagai zat yang dapat menyebabkan efek pemanasan global.Oleh karena itu dalam penelitian ini kami bermaksud melakukan penelitian studi komparasi refrigerant R134a dengan refrigerant R600a.Ini dikarenakan refrigerant R600a sifat termodinamikanya lebih baik dibandingkan dengan Refrigeran 134a. Jenis penelitian ini adalah penelitian eksperimen.Penelitian ini bertujuan untuk mengetahui COP, tingkat konsumsi energi, dan besar nilai ekonomisasi pada refrigerant R134a dan R600a. Hasil dalam penelitian ini adalah untuk mengetahui: (1) COP dari masing-masing refrigerant, (2) pengaruh penggantian refrigerant R134a dengan refrigerant R600a pada tingkat konsumsi energi yang digunakan untuk menjalankan sistim, (3) nilai ekonomisasi pada penggunaan kedua jenis refrigerant tersebut, (4) pengaruh kecepatan blower evaporator pada sistim kerja AC mobil. Abstract

Salah satu jenis retribusi daerah adalah retribusi parkir. Setiap pemerintah daerah memiliki wewenang dalam menetapkan tarif retribusi parkir. Tetapi, sering sekali para juru parkir meminta lebih dari tarif yang sudah ditetapkan. Hal tersebut merugikan pengguna parkir, selain harus membayar lebih, pengguna parkir sering tidak mendapatkan karcis parkir. Belum lagi pengelolaan retribusi parkir yang tidak transparan menjadi salah satu penyebab rendahnya penerimaan retribusi parkir. Permasalahan ini dapat diminimalkan dengan menerapkan teknologi RESTful web services pada sistem pembayaran retribusi parkir yang terintegrasi dan transparan sehingga diharapkan mampu menjadi alternatif solusi mengatasi masalah pengelolaan retribusi parkir yang dialami oleh Pemkot maupun Pemda di Indonesia. Metode pengembangan sistem yang digunakan yaitu Extreme Programming (XP) dengan tahap : planning (perencanaan), design (perancangan), coding (pengkodean) dan testing (pengujian). Hasil aplikasi berbasis Android yaitu Markir sebagai sarana utama mengirimkan data pembayaran parkir dan OAuth 2.0 sebagai protokol otorisasi setiap  transaksi data yang dilakukan. Hasil pengujian aplikasi Markir dengan metode black-box testing, sistem telah terintegrasi dengan baik dan dilakukan berbagai tahap validasi seperti client credentials, access token dan scope akses fungsi yang tersedia pada API Markir sesuai standar protokol OAuth 2.0 sehingga transaksi data lebih aman jika dibandingkan menggunakan otorisasi API standar seperti basic-auth API ataupun API Key.

Abstract

Pada penelitian ini dirancang antena mikrostrip monopole planar untuk aplikasi UWB yang bekerja pada frekuensi 3,1-10,6  GHz. Penelitian ini dilakukan dalam dua tahap. Pada tahap pertama dilakukan optimasi rancangan antena melalui simulasi menggunakan perangkat lunak 3D Electromagnetic Simulator. Pada tahap kedua dilakukan fabrikasi dengan menggunakan bahan dasar substrat Roger RT-58880 dengan konstanta dielektrik 2,2 dan ketebalan 1,6 mm. Dari hasil simulasi didapatkan  frekuensi kerja antena antara 2,8 – 9 GHz dengan nilai return loss <-10 dB dan VSWR kurang dari 2, dan Bandwidth sebesar 6,3 GHz. Antena memiliki pola radiasi omnidireksional dengan gain pada frekuensi 3,1 GHz MHz sebesar 2,2 dBi dan impedansi input pada frekuensi 3,1 GHz sebesar 50,231Ω. Dari hasil pengukuran didapatkan rentang frekuesi kerja antena antara 2,6 – 9,5 GHz dengan nilai return loss di bawah -10 dB dan VSWR kurang dari 2, dan Bandwdith sebesar 6,9 GHz. Antena memiliki pola radiasi omnidireksional dengan gain  pada frekuensi 3,1 GHz sebesar 2,1 dBi dan impedansi input pada frekuensi 3,1 GHz sebesar 51,031  Ω. Dari hasil simulasi dan pengukuran dapat disimpulkan bahwa antenna ini dapat digunakan untuk aplikasi UWB.

Abstract

The main target of the research is to show that planning is the most important stages of management function, mainly to face exsternal environment that changed dynamically. The main topic on this research was focused on the certain elements from planning process and the process that have very closed relation with the problem solving and also the program development of counseling service at school. The Planning model for counceling activity by using seventeen plus  pattern approach to be very urgent to be done by guiding and counceling teachers in order the implantation is in line with the students’ need at the field.From the introduction study can be concluded that in general the Bk/guiding and counceling teachers at Junior High School/SMP at Bondowoso is still having not variant data that should be owned for conducting guiding and counceling service at school. In general, the guiding and counceling teachers SMP/Jumior High School at Bondowoso felt important and even very important for having data collection and instrumentation application for running on the duty for conducting guiding and counceling service at school, with average score: 3,23.Besides that, it can be explained that conducting evaluation of data collecting and the existing of instrumentation application nowadays, it can be concluded that it was still very low in general with the average score = 1,14. While, it was linked with the need of model development for data compilation activity planning and instrumentation application were found that guiding and counceling’s teachers very needed in general with the average score = 4,28. While it was linled with the assessment of model development of data compilation activity planning and instrumentation application that were promoted, it was found that guiding and counceling’s teachers gave very good assessment in general with the average score = 4,47.

SMP/MTs di Bondowoso. Hasil penelitian ini menunjukkan bahwa model perencanaan kegiatan konseling dengan menggunakan pendekatan pola tujuh belas plus menjadi sangat perlu dilakukan agar implementasinya sesuai dengan kebutuhan siswa. Guru BK SMP/MTs di Bondowoso masih banyak yang belum memiliki berbagai data untuk penyelenggaraan layanan konseling. Guru-guru tersebut sangat memerlukan himpunan data dan aplikasi instrumentasi untuk kelancaran menjalankan tugasnya (rerata = 3,23). Sementara evaluasi pelaksanaan himpunan data dan aplikasi instrumennya ternyata masih sangat rendah (rerata = 1,14. Guru-guru BK juga sangat membutuhkan (rerata = 4,28). Penilaian pengembangan model perencanaan kegiatan himpunan data dan aplikasi instrumentasi yang dipromosikan dinilai sangat baik (rata = 4,47)

Abstract

BACKGROUND Various products used in cellular therapy utilize tissue culture techniques requiring keratinocyte culture. An efficient and clinically acceptable keratinocyte culture system requires supplements with mitogenic activity. Geraniin is a phytochemical with the potential as a supplement for expansion culture of keratinocytes. The objective of the present study was to verify the mitogenic activity of geraniin on human keratinocytes. METHODS This was an experimental study using two samples of human foreskin obtained by circumcision of a male child. Epidermal keratinocytes were isolated from the foreskin samples and were divided into paired groups, comprising intervention and control groups. The intervention groups were cultured with geraniin supplementation, whereas the control groups with standard supplements, without the addition of geraniin. Mitochondrial activity of the cells was evaluated by means of the 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl-tetrazolium-bromide (MTT) proliferation assay. Absorbance values in each of the groups was measured at 450 nm. Data analysis was performed with the paired t-test. RESULTS Geraniin supplementation significantly increased the keratinocyte proliferation rates at dosages of 0.8 to 3.1 μM. An increase of 57% in the proliferation rate was obtained at a dosage of 1.6 μM, while at a dosage of 12.5 μM toxic effects were starting to appear. Geraniin presumably causes increased cellular energy status, resulting in increased proliferation rates. CONCLUSION The findings in this study provide evidence in support of the utilization of geraniin as a supplement for expansion culture of keratinocytes. Further studies may presumably identify the molecules acting as geraniin receptors and the intracellular mechanisms underlying the increase in proliferation rates.

Abstract

BACKGROUNDTransplanted allogeneic dermal fibroblasts retain stem cell subpopulations, and are easily isolated, expanded and stored using standard techniques. Their potential for regenerative therapy of chronic wounds should be evaluated. The aim of this study was to determine allogeneic fibroblast viability in the presence of peripheral blood mononuclear cells (PBMC).METHODSIn this experimental study, fibroblasts were isolated from foreskin explants, expanded in the presence of serum, and stored using slow-freezing. We used one intervention group of allogeneic fibroblasts co-cultured with PBMC and 2 control groups of separate fibroblast and PBMC cultures.Fibroblasts were characterized by their collagen secretion and octamer-binding transcription factor 4 (OCT4) expression. Viability was evaluated using water soluble tetrazolium-1 (WST-1) proliferation assay. Absorbances were measured at 450 nm. Data analysis was performed by student’s paired t-test.RESULTSDermal fibroblasts were shown to secrete collagen, express OCT4, be recoverable after cryopreservation, and become attached to the culture dish in a co-culture with PBMC. Co-cultured and control fibroblasts had no significantly different cell viabilities (p>0.05). Calculated viable cell numbers increased 1.8 and 5.1- fold, respectively, at days 2 and 4 in vitro. Both groups showed comparable doubling times at days 2 and 4 in vitro. PBMC did not interfere with allogeneic fibroblast viability and proliferative capacityCONCLUSIONSAllogeneic fibroblasts remain viable and proliferate in the presence of host PBMC. Future research should evaluate allogeneic human dermal fibroblast competency in clinical settings. Dermal fibroblasts are a potential source for cell therapy in chronic wound management.

Abstract

Background: Dimethyl sulfoxide (Me2SO) is a common cryoprotective agent widely used in cell preservation system. Me2SO is currently known to cause epigenetic changes which are  critical in stem cells development and cellular differentiation. Therefore, it is imperative to develop cryopreservation techniques that protect cellular functions and avert Me2SO adverse effect. Trehalose was able to protect organism in extreme condition such as dehydration and cold. This study aimed to verify the protective effect of trehalose preincubation procedure in cryopreservation.Methods: The study was conducted using experimental design. Thawed mesenchymal (CD271+) stem cells from YARSI biorepository were used for the experiment. Trehalose preincubation was performed for 1 hour, internalized trehalose was confirmed by FTIR-ATR measurement. Three groups consisted of (1) cryopreserved without trehalose preincubation, (2) cryopreserved with trehalose preincubation, and (3) did not undergo cryopreservation were evaluated after 24 hours in LN2 for viability in culture. The absorbance from each group was measured at 450 nm. The analysis performed using paired student t test.Results: Viability of thawed mesenchymal (CD271+) stem cells that undergo trehalose preincubation prior cryopreservation was significantly higher (p<0.05) compared to group without trehalose preincubation. Higher viability observed between group with trehalose preincubation compared with controlled group suggests protection to trypsinization. Mesenchymal (CD271+) stem cells incubated for 1 hour in 100 mM trehalose supplemented medium  results in 15%  trehalose loading efficiency.Conclusion: These findings confirm the protective effect of trehalose preincubation in cryopreservation. Future research should be directed to elucidate the trehalose internalization mechanism and eventually the protective mechanism of trehalose in mammalian cell cryopreservation.
