Abstract

"Gemusis" fish cultivation group is one of the aquaculture business groups that exist in Tendeki Village, Matuari District, Bitung City, North Sulawesi Province. The width of  pond owned is around 3 ha with an average production of 2 tons per year. The main problem in this group is the difficulty in obtaining superior seeds, expensive feed prices and often the fish that are kept stricken with disease, resulting in less production and even crop failure. Alternative solutions to problems in partner problems are accompanied by two approaches, namely the classical and individual approaches. The classical approach is carried out when delivering the material. An individual approach is taken at the time of practice. Lectures in class in the form of delivery of how to make tilapia seed with sex reversal methods for the maintenance of monosex, making low-quality quality feed made from local raw materials and producing maggot as natural food, as well as how to increase fish immunity by giving immunostimulants to fish feed. Demonstration and practice with techniques for making cheap feed ingredients made from local raw materials. In order for the fish cultivation activities carried out by the partners according to the activity plan, monitoring and assistance will be carried out after the lecture and practice activities. The results obtained showed the group are able to cultivate fish with monosex system, produce feed using local raw materials, maggot cultivation as a natural feed, and can treat fish diseases using mmunostimulant.

Abstract

The aims of this research were to determine the total mercury concentration of Cardinalfish (Sphaeramia Orbicularis) and the waters in aquaculture area of Pintu Kota, North Lembeh, Bitung, and its water quality paramaters (temperature, salinity, pH). The research was conducted from June to Nopember 2019 in the cultivation area of tiger grouper fish in which the fish and seawater were sampled and analyzed in the laboratory. Determination stations were done by purposive sampling which refers to the physiographic lacation wherever possible in order to represent or describe these waters. Station 1 represented the sea transpartion port of Lembeh Island (no cultivation), Station 2 where the cultivation A, Station 3 where the cultivation B. The results showed the concentration  of the mercury obtained at Station 1 ranged from 0.00756 -0.00789 ppm, Station 2 ranged 0.00394-0.00418 ppm, and Station 3 ranges from 0.00506-0.00564 ppm. Whereas the concentration of mercury in seawaters samples were detected at Station 1, 2, and 3 were < 0.003 ppm. The temperature ranged between 26.2 ? 26.7°C, pH 7 and salinity 30-33 ppt. In general, the existence of the phisycal chemical parameters of water was in good measurement after comparing with the water quality standard of Kep. MENLH No. 51/2004. Mercury concentrations in all samples in Pintu Kota waters of North Lembeh as compared to WHO/FDA standard are still in good condition but have been already contaminated by mercury.

Abstract

The objective of research was to isolate probiotic bacteria from catfish intestine and to examine its effects on the growth and resistance of carp against Aeromonas hydrophila.  The probiotic isolated from intestine was grown on MRS agar.  Fish used in this research was juvenile of carp with an average weight of 5.5 g obtained from Freshwater Aquaculture Board at Tatelu Village, North Sulawesi Province.  Before running the experiment, the juveniles were adapted for one weeks in 15 glass aquaria at a density of 15 individuals each.  After acclimatization, the fish were fed diet supplemented with probiotic at different concentrations namely 1x109 , 1x108 , 1x107,  1x106 cfu/mL for three weeks. The fish were fed two times a dayat 08.00 am and 17.00 pm with a dose of 5% of body weight per day. At the end of feeding, the fish were challenged with A. hydrophila.  Data collected included average growth rate, absolute growth, feed efficiency, and food conversion ratio.  Research results found that the addition of probiotic into feed was  able to increase growth and feed efficiency and reduce food conversion ratio as well (p<0.01). The best growth, feed efficiency and food conversion ratio were obtained in fish fed diet added with probiotic bacteria at 1x108 cfu/mL.  The highest survival rate was also observed in fish probiotic diet containing 1x108 cfu/mL.  As conclusion, probiotic bacteria isolated from catfish intestine was potential to improve growth, feed efficiency, reduce food conversion ratio and increase resistance of fish against bacterial pathogen.Keywords: probiotic, carp, Aeromonas hydrophila, feed efficiency, food conversion ratioABSTRAKPenelitian ini bertujuan untuk mengisolasi bakteri probiotik dari usus ikan lele dan menguji pengaruh pemberian probiotik dari usus ikan lele terhadap pertumbuhan dan resistensi ikan mas terhadap infeksi Aeromonas hydrophilla. Sumber probiotik diisolasi dari usus ikan lele menggunakan media MRS (de Man Rogosa Sharpe) dan bakteri Aeromonas hydrophilla ditumbuhkan pada media TSA (Tryptic Soy Agar). ikan uji diambil dari Balai Budidaya Air Tawar Tatelu , Provinsi Sulawesi Utara . ikan diaklimatisasi selama seminggu dalam 15 akuarium dengan kepadatan 15 ekor/akuarium dan berat awal rata-rata 5,5 g. Setelah diaklimatisasi ikan diberi pakan yang ditambahkan bakteri probiotik sebagai perlakuan dengan konsentrasi berbeda yaitu 1x109 , 1x108 , 1x107,  1x106 cfu/mL selama empat minggu sebanyak 5% /berat tubuh/hari dengan frekuensi pemberian pakan dua kali sehari yaitu jam 08.00 pagi dan jam 17.00 sore. Setelah diberi perlakuan maka diuji tantang dengan bakteri Aeromonas hydrophilla.  Data yang dikumpulkan terdiri dari laju pertumbuhan harian, pertumbuhan mutlak, efesiensi pakan, konversi pakan dan uji tantang terhadap bakteri patogen. Hasil penelitian mendapatkan bahwa penambahan bakteri probiotik dalam pakan mampu meningkatkan pertumbuhan, efesiensi pakan dan menurunkan konversi pakan (p<0.01). Pertumbuhan, efesiensi pakan dan konversi pakan yang terbaik dicapai pada ikan yang diberi pakan dengan penambahan bakteri probiotik 1x108 cfu/mL.  Kelangsungan hidup ikan mas yang diuji tantang dengan bakteri patogen yang paling tinggi dicapai pada ikan yang diberi pakan 1x108 cfu/mL. Sebagai kesimpulan bahwa bakteri probiotik yang diisolasi dari usus ikan lele berpotensi meningkatkan pertumbuhan, efesiensi pakan, menurunkan konversi pakan serta meningkatkan sistem kekebalan tubuh pada ikan mas.Kata Kunci : Probiotik, Ikan Mas, Aeromonas hydrophilla, Pertumbuhan, Efesiensi Pakan, Konversi Pakan
