Abstract

Infectious Myonecrosis Virus (IMNV) adalah virus yang umum menyerang udang putih (Litopenaeus vannamei) dalam industri budidaya udang di dunia. Di Indonesia, penyakit myonecrosis pertama kali diketahui terjadi pada udang putih dari pertambakan di Situbondo, Jawa Timur, pada tahun 2006 dengan prevalensi 11.11% dan gejala klinis serupa dengan kejadian wabah myonecrosis di Brazil pada tahun 2002. Penelitian ini bertujuan untuk mengetahui prevalensi dan karakteristik molekuler IMNV di sentra budidaya udang vaname di Propinsi Banten. Sampel udang dikumpulkan selama periode Maret hingga Juni 2015. Sebanyak 24 sampel diperoleh dari 24 area pertambakan aktif kemudian di-pool dan diuji menggunakan metode Real Time Polymerase Chain Reaction (PCR). Hasil uji menunjukkan bahwa prevalensi IMNV secara keseluruhan di Propinsi Banten adalah sebesar 33,3% dengan rincian: Kota Serang 0%; Kabupaten Serang 0%; Kabupaten Tangerang 14,3% dan Kabupaten Pandeglang 100%. Analisis perbandingan genom IMNV pada ORF1parsial menunjukkan bahwa isolat lapang Banten, isolat Indonesia dan Brazil memiliki persentase kemiripan 97,4-100%. Analisis sekuens asam amino menunjukkan persentase kemiripan 97,6-100%. Analisis fi logenetik sekuens nukleotida menunjukkan bahwa telah terjadi diversifikasi genetik antara IMNV Indonesia dan Brazil dan antar isolat Indonesia sendiri.

Abstract

The study aimed was to know the influence of Piper crocatum exstract on the blood profile included total erythrocyte, total leukocyte, differential leukocyte (DL), phagocytosis indeks (IF) and survival rate of common carp (Cyprinus carpio) infected by Aeromonas hydrophila and the best concentration of P. crocatum for imunostimulatory common carp of infected A. hydrophila. The research was conducted at Research and Development of Freshwater Aquaculture, Bogor. The methods used in this research is experimental methods by using random design complete (RAL) with 4 treatments and three replicates. The research was conducted at Research and Development of Freshwater Aquaculture, Bogor. The treatments of different addition of P. crocatum exstract on fish feed, 0 gr/kg feed (treatment A), 5 gr/kg feed (treatment B), 10 gr/kg feed (treatment C), 15 gr/kg feed (treatment D). The treatnent was given 21 days. The research results obtained additional P. crocatum exstract significantly different effect (P0,05) on percentage of netrofil (2%) and survival rate of common carp (C. carpio) infected by A. hydrophila (40%), the best concentration of P. crocatum on feed for immunostimulant common carp of infected A. hydrophila, the result showed the best dose 5 g/kg feed

Abstract

Carp is a freshwater species that is easily cultivated. One of the constraints in cultivating carps (C. carpio) is diseases that ultimately caused mass mortality. The disease that generally affects carp is Motile Aeromonas Septicemia (MAS) caused by Aeromonas hydrophila. Soursop leaves is one of the herbs that can be used as an ingredient to control fish diseases because it contains chemicals that acts bacteriocide. The purpose of this research was to determine the influence and the best dose of soursop leaf extract in feed to the blood profile and survival of carp infected by A. hydrophila. The treatments of this research were addition of soursop leaf extract on feed with 4 different dosages and 3 replicates. The treatments were A (0 g/kg diet), B (5 g/kg diet), C (10 g/kg diet) and D (15 g/kg diet). The result showed that soursop leaf extract in feed of treatment B (5 g/kg diet) significantly different (P0.05) on total erythrocyte (0.83×106 cells/mm3), differential leukocyte include the percentage of monocytes (7.67%), neutrophils (8.67%), lymphocytes (83.67%), phagocytic activity (37%) and survival rate (50%). It can be conclude that soursop leaf extract at dosage 5g/kg was able to stimulate immune response of carp, it was characterized by an increased total leukocytes of carp infected by A. hydrophila.

Abstract

 Pengendalian penyakit bakterial yang umum dilakukan dengan pemakaian antibiotik atau  bahan kimia sudah tidak diperbolehkan lagi karena menimbulkan patogen yang resisten  terhadap bahan kimia tersebut, terlebih jika penggunaan tidak sesuai dengan anjuran yang diberikan. Dampak negatif terhadap kesehatan konsumen berupa residu antibiotik juga menjadi pertimbangan yang harus diperhatikan. Manipulasi terhadap populasi mikroba yang berada di perairan guna pencegahan sebelum terjadinya serangan bakteri yang bersifat mematikan perlu dilakukan sebagaimana konsep probiotik sebagai biokontrol. Tujuan penelitian ini adalah menguji kandidat probiotik dalam menekan atau menghambat bakteri patogen Aeromonas hydrophila. Penelitian ini dilaksananakan dalam dua tahap. Tahap pertama adalah tahap pengujian bakteri kandidat probiotik secara in vitro menggunakan metode zona hambat dan kultur bersama pada media agar.  Tahap kedua adalah uji tentang bakteri kandidat probiotik dengan patogen pada media budidaya. Hasil terbaik penelitian tahap pertama pada  uji kultur bersama antara kandidat probiotik B. firmus dengan A. hydrophila pada skala in vitro adalah dengan penambahan probiotik  B. firmus sebanyak 108 cfu/ml. Sedangkan pada penelitian tahap kedua didapatkan hasil berturut-turut perlakuan D dengan tingkat kelangsungan hidup (SR) mencapai 90%, perlakuan C dengan SR 75%, perlakuan A dengan SR 50% dan perlakuan K dengan SR 50%.  Controlling bacterial disease with the use of antibiotics or chemicals is no longer allowed as it results in pathogens that are resistant to the chemicals, especially when not in accordance with the recommendations provided. The negative impactsof the antibiotics residues on the consumers? health  also need to be considered. Manipulation of microbial populations present in the waters as preventation before the lethal attack of bacteria needs to be done which is in accordance with the concept of probiotics as biocontrol.The purpose of this study was to test the probiotic candidates in suppressing or inhibiting pathogenic bacteria Aeromonas hydrophila. This study was conducted in two stages. The first stage was to test a candidate probiotic bacteria in vitro using culture methods and inhibition zone on the media together. The second stage wasto test candidate probiotic bacteria to pathogens on the cultivation media. The best results in the first phase of the research is shared culture test between probiotic candidate B. FIRMUS with A. hydrophila on vitro scale is the addition of the probiotic B. FIRMUS 108 cfu / ml. While in the second phase of the research results obtained successively: treatment D with a survival rate (SR) reaches 90%, treatment C with SR 75%, treatment A with SR 50% and treatment K with SR 50%.

Abstract

ABSTRACT Streptococcosis is one of the diseases that often arise and can cause death by 100% in tilapia fish farming. This study aimed to determine the most optimal probiotics combination of cells Bacillus cereus P22, Bacillus subtillis ND2 and Staphylococcus lentus L1k through the feed for streptococcosis prevention. The experimental fish with initial body weight of (13.06±0.18 g), were randomly distributed at 25 fish per 60 L water, and reared for 21 days. The fish fed with supplemented feed (except controls) with FR 3%, about three times a day, for 14 days. The treatment of combination probiotics-supplemented feed on, K1 (P22 and ND2, density of 106 CFU/mL), K2 (ND2 and L1k, density of 105 CFU/mL), K3 (P22 and L1k, density of 105 CFU/mL), K4 (P22, ND2 and L1k, density of 106 CFU/mL), the negative and positive control treatment given feed without supplementation. On day 15, the experimental fish, (except negative control) were challenged with Streptococcus agalactiae via intraperitoneal injection 0.1 mL/fish (104 CFU/mL). The results indicated that the best combination was shown by K3 (the combination of Bacillus cereus P22 and Staphylococcus lentus L1k) with cells addition 1% (v/w). The result of K3 survival rate shown 89.33±6.11%, significantly different from the K + and K- wich were 66.67±4.62% and 88.00±8.00%. The highest increased immune value shown by K3 were wich phagocytic index value 64.00±2.65% and respiratory burst activity value 0.07±0.00 OD. Keywords: multispecies probiotics, Oreochormis niloticus, streptococcosis  ABSTRAK Streptococcosis merupakan salah satu penyakit yang seringkali muncul dan dapat menyebabkan kematian hingga 100% pada budidaya ikan nila. Penelitian ini bertujuan untuk menentukan kombinasi optimal dari probiotik Bacillus cereus P22, Bacillus subtillis ND2, dan Staphylococcus lentus L1k melalui pakan sebagai upaya pencegahan streptococcosis. Ikan uji berukuran (13,06±0,18 g), secara acak dipelihara dengan kepadatan 25 ekor/akuarium dengan volume air 60 L, dipelihara selama 21 hari dan diberi pakan yang disuplementasi probiotik (kecuali kontrol) dengan perkiraan FR 3% sebanyak tiga kali sehari, selama 14 hari. Perlakuan kombinasi probiotik yang disuplementasi pada pakan yaitu: K1 (P22 dan ND2, kepadatan 106 CFU/mL), K2 (ND2 dan L1k, kepadatan 105 CFU/mL), K3 (P22 dan L1k, kepadatan 105 CFU/mL), K4 (P22, ND2 dan L1k, kepadatan 106 CFU/mL), perlakuan kontrol negatif dan posistif diberi pakan tanpa suplementasi probiotik. Pada hari ke-15, ikan uji (kecuali kontrol negatif) diinjeksi dengan Streptococcus agalactiae dengan kepadatan 104 CFU/mL sebanyak 0,1 mL secara intraperitoneal. Hasil penelitian ini menunjukkan bahwa kombinasi probiotik multispesies terbaik adalah perlakuan K3 (kombinasi Bacillus cereus P22 dan Staphylococcus lentus L1k) dengan dosis 1% (v/w). Nilai kelangsungan hidup K3 didapat 89,33±6,11%, berbeda nyata dengan K+ (66,67±4,62%) dan K- (88,00±8,00%). Nilai respons imun dengan peningkatan tertinggi pada perlakuan K3 dengan nilai indeks fagositik 64,00±2,65% dan aktivitas respiratory burst 0,071±0,00 OD.   Kata kunci: probiotik multispesies, Oreochormis niloticus, streptococcosis

Abstract

This research aimed to know the toxicity of extracellular products (ECP) of Streptococcus agalactiae was tastedin cultured Nile tilapia (Oreochromis niloticus). Streptococcus agalactiae had two haemolytic types: ?-haemolyticand non-haemolytic type. Toxicity test of ECP to know the virulancy factor of S. agalactiae was still limited. It wasfound that after tested on 15 fish weighing 15 g through intraperitoneal injection 0,1 ml/fish, both bacteria causedchanges in swimming pattern, palatability, external and internal anatomy macroscopically and microscopically.Extracellular products of S. agalactiae non-haemolytic type (BHIA and BHI 24 h) and ?-haemolytic type (BHI 72 h)caused mortality 12 hours after injection and the mortality continued till day 7 th of culture. Whirling happened 96hours after injection with ECP S. agalactiae ?-haemolytic type (BHIA 72 h incubation) whereas injection with ECP(BHI 24 h) on 72 h after injection and continued untill day 7 th. Behavior disease signs caused by S. agalactiaeoccured on eyes. There were opacity, purulens, eye shrink, lateral and bilateral exopthalmia and haemorrhage oninfected-fish. Silver staining of sodium dodecyl sulphate-polyacrylamide gels to S. agalactiae revealed thatpredominant 51.8-69.6 kDa bands were present in BHIA ECP fraction. The 69.6 kDa was absent from the BHI ECP.Total protein on non-haemolytic S. agalactiae ECP are 28.18 ppm on BHIA medium and 13.64 ppm on BHI medium.Whereas ?-haemolytic S. agalactiae ECP are 2.73 ppm on BHIA medium and 8.18 ppm on BHI medium. Concentrationof protein in ECP was one of factor that caused non-haemolytic S. agalactiae more virulent than ?-haemolytic type.The conclusion from the research that ECP was virulent factor on ?-haemolytic and non-haemolytic S. agalactiaein fish which caused changes in behavior disease signs.

Abstract

One of the disease that often led to mortality in catfish resulting in harvest failure is Motile AeromonadsSepticemia (MAS) caused by infection of Aeromonas hydrophila. To avoid this situation, the preventionand control of disease is very crucial. An environmentally friendly approach acceptable in aquaculture isthe use of probiotics to control the pathogenic infection. African catfishes used in the study weighted of1,97±0,7g/fish, and were maintained in the aquarium size of 56x39x34 cm3filled with 30 liters of waterwith stock density of 25 fishes . Probiotic was given daily for 28 days. This research was consisted of 6treatments; (A) AH26 NAR103 CFU/mL, probiotics ND2 CefR 105 CFU/mL dan P23 CipR 105 CFU/mL ; (B)AH26 NAR103 CFU/mL, probiotics ND2 CefR 103 CFU/mL dan L1k TetR 103 CFU/mL; (C) AH26 NAR103CFU/mL, probiotics P23 CipR 104 CFU/mL dan L1k TetR 104 CFU/mL; (D) AH26 NAR103 CFU/mL, probioticsND2 CefR 105 CFU/mL, P23 CipR 105 CFU/mL dan L1k TetR 105 CFU/mL; (K+) AH26 NAR103 CFU/mL, without probiotics; (K-) Without the addition of probiotics and without AH26 NAR in the media. Eachtreatment has 3 replications. The results showed that the combination of probiotics ND2CefR103CFU/mLand L1k TetR103CFU/mLcan supressedA. hydrophila cell density up to 40% lower, induced immuneresponses (hematocrit level and respiratory burst activity) and increased the survival rate of catfishuntilthe end of the research.

Abstract

The main problem in catfish farms is mass mortality coused by Motile Aeromonas Septicemia (MAS) by Aeromonas hydrophila. Natural ingredients that are currently being developed is soursop leaf. It was because soursop leaf contains acetogenin compounds that wishes compared to other plant parts. An acetogenin compound has the benefit of as an antibacterial. The purpose of this research was to determine the influence and best dose of sourshoup leaf extract in feed to the blood profile and survival rate of catfish infected by A. hydrophila. The treatment in this study were the addition of soursop leaf extract on feed with different dosage and 3 replicates. The treatments were A (0 g/kg diet), B (5 g/kg diet), C (10 g/kg diet), D (15 g/kg diet). The result showed that soursop leaf extract in feed of treatment B on the best dose (5 g/kg diet) was total erythrocytes(1,51 x 106 sel/mm3), total leukocytes (8,00 x 103 sel/mm3), limphocytes (92%), monocyte (8%), neutrophils, phagocytic index (75%), and survival rate (93%). It can be concluded that dose of 5 g/kg diet was the best dosage of soursop leaf extract in the feed that was able to stimulate the immune response of catfish, it was characterized by an increased erythrocytes, total leukocytes, differetial leukocyte (limphocytes and monocyte) of catfish infected by A. hydrophila.

Abstract

Multilocus sequence typing (MLST) has greater utility for determining the recent ancestral lineage and the relatedness of individual strains. Group B streptococci (GBS) is one of the major causes of subclinical mastitis of dairy cattle in several countries. GBS also sporadically causes epizootic infections in fish. The aim of this study was to compare the evolutionary lineage of fish and bovine isolates in relation to the S. agalactiae global population as a whole by comparing the MLST profiles. Twenty S. agalactiae isolates were obtained from dairy cattle and fish. PCR products were amplified with seven different oligonucleotide primer pairs designed from the NEM316 GBS genome sequence. Clone complexes demonstrated that bovine and fish isolates were separate populations. These findings lead us to conclude that fish S. agalactiae is not a zoonotic agent for bovine. MLST could help clarify the emergence of pathogenic clones and to decide whether the host acts as a reservoir for another pathogenic lineage.

Abstract

The research with the aim to know the effectivity (yield gap) of the application of Streptococcus agalactiae vaccine (pure whole cell) in prevention of streptococcosis on tilapia (Oreochromis niloticus) culture has been carried out. The isolate of S.agalactiae – N14G was used as a master seed on vaccine production. Priming vaccination was administered by immersion method, and booster vaccination was taken th place two months latter by oral method. Challenge test at the lethal dose (LD50) against active bacteria was done at 14 days post booster vaccination, and observation was taken place for 14 days post artificial infection. The results of the research showed that the highest survival rate and relative percent survival (RPS) was found in group treated with Streptovac vaccine (S. agalactiae and A. hydrophila combination) (65.58% and 35.36%) followed by S. agalactiae vaccine (52.08% and 10.01%). The lowest survival rate was found in control group (46.75%). The result of confirmation effectivity of the vaccines by challenge test in the laboratory showed that the highest survival rate and relative percent survival (RPS) was found in S. agalactiae vaccine (50.00% dan 37.50%) followed by Streptovac vaccine (40.00% and 25.00%), and the lowest survival rate was found in control group (20.00%). Vaccination is better than the non vaccinated.
