Abstract

ABSTRACT           Introduction: Dengue is the most important mosquito-borne flavivirus disease. The number of Dengue cases in Indonesia in 2010 range from 150,000 cases with the deaths of around 1,317 people. Huge number of cases have made Indonesia was the first ranked as the state with the highest Dengue cases in the ASEAN region and the world's second ranking after Brazil. The drugs  or antibiotics that can be administered effectively to cure this disease has not been found yet. Many study have been done and some that have been reported include viral RNA synthesis inhibitors, protein inhibitors of NS3 helicase and protease and inhibitors that inhibit Dengue virus maturation. Curcumin have preventive activity against several viruses: vasicular stomatis ( VSV ), HSV 1 and 2, parainfluenza - 3, reovirus - 1, feline corona virus, feline herpes virus . Curcumin also known have ubiquitin proteasome inhibition system was able to decrease the production of Japanese ensefalitis virus. Objectives: This study aims to determine safe concentrations of curcumin against vero cells (cytotoxic test results) and know the Dengue-2 antiviral potency of curcumin. Methods: Including quasi-experimental study. The anti viral potency of curcumin seen from the result of immunocytochemistry Streptavidin Biotin Peroxidase Complex (SBPC).  Data were analyzed by ANOVA.Results: The results showed that secure concentrations from cytotoxic of curcumin against vero cells is 6.25 ppm. The calculation of positive rate from immunocytochemistry in vero cells infected by Dengue - 2 incubation 1 and 3 days were the result is significantly different than the control. Conclusion: The secure concentration of curcumin against vero cells was 6.25 ppm and curcumin was able to lower the positive rate due to Dengue-2 infection.Key Words : Dengue virus, Curcumin, Immunocytochemistry.

Abstract

Leptospirosis in Sukoharjo was discovered in 2014. Examination with RDT (Rapid Diagnostic Test) leptotek supported by clinical symptoms showed 6 positive cases of Leptospira, and until March 2015 one person was found to be Leptospira positive. The aim of this study was to identify rats as the main reservoir of leptospirosis, calculate the catching rate of rats and to detect the presence of pathogenic Leptospira in rats. This study was a cross-sectional survey, and was conducted in Pabelan village Kartasura Sukoharjo on May 2015. Polymerase Chain Reaction (PCR) assay was conducted in Bacteriology Laboratory Balai Litbang P2B2 Banjarnegara to detect leptospira in the kidney of the rats. Data were analyzed descriptively. Results of rats and shrew catching obtained Rattus tanezumi, Rattus norvegicus and Suncus murinus. The species most commonly found was balanced between R. tanezumi and S.murinus (46%). The trap success rate inside and outside the house are 1.50% and 5%, respectively. Result of laboratory test showed from 13 rats kidneys, two kidneys were found to be Leptospira positive and was from R.tanezumi and R.norvegicus.

Abstract

Aedes aegypti is a major vector for Dengue, a deadly disease causing death of millions of people in developing countries both in urban and rural populations. Ae. aegypti control using chemical insecticide was always carried out and lead to a widespread insecticide resistance. Therefore, mosquito biological control is needed to replace the usage of chemical insecticide. A chitinolytic bacteria, was isolated from shrimp?s waste (head and shell). The isolate showed chitinolytic activity as a transparent zone in colony inside the synthetic media, containing (w/v)- 0,3 % colloidal chitin, 1% pepton, 0,5% yeast extract, 0,1% NaCl, 0,1% K2HPO4, 0,05% MgSO4.7H2O, 0,001% FeSO4.7H2O, 0,001% ZnSO4.7H2O, and each of 0,0001% CuSO4.5H2O, MnSO4.nH2O and CaCl2.2H2O at pH 7 and  300C after 72 h of incubation. The isolate was identified as gram positive group based on gram staining. In the experimental method, four concentrations of chitinolytic bacteria (4%, 8%, 16% and 32%) was exposed to Ae. aegypti larvae. The result showed that chitinolytic bacterium degrades exoskeleton of third instar larvae of Ae. aegypti. Degradation of exoskeleton started on the 2nd days and occurred in thorax region. Probit analysis showed LC50 value was obtaninedat concentration of 2%.

Abstract

Rats could play a role as reservoir of diseases which caused by protozoa parasite such as toxoplasmosis. Toxoplasmosis is zoonotic disease which caused by Toxoplasma gondii, with Felidae (cat) as definive host. Toxoplasmosis in cat can be obtained from predator to small mammals like rat at surroundings. The aims of this study were to measure the trap success, identify of rat species, measure diversity index, and dominant species. In addition, we also analyse toxoplasmosis in rat and their role in toxoplasmosis transmission in Banjarnegara District. This study was descriptive, with purposive samples collection.  Rat survey was conducted by single live trap at main market, hospital and Semampir shopping complex. Toxoplasmosis determination was carried out by imunostic FELISA. Mean of rat?s trap success was 17%, which the highest rate at main market 17%. Rats species trapped were Rattus tanezumi and R. norvegicus, which R. tanezumi 98,32% as dominant species. Majority of rat?s sex species is female.  Diversity index of rat species at Banjarnegara District public places was low (<1). Rat?s toxoplasmosis was 3,57%, showed that rats could be had minimal role in toxoplasmosis transmission in Banjarnegara District.

Abstract

Treatment of Dengue patients today are to relieve symptoms because specific therapies and effective anti-viral drugs have not been found. Curcumin is known has anti-viral activity, anti-inflammatory, anti-oxidant, anti-parasitic and anti-cancer. Curcumin has a weakness that is sensitive to acidity (pH) and light, so curcumin analogues that ispentagamavunon-0 (PGV-0) was made in order to obtain better anti viral activity. The purpose of this study was to determine the effect of curcumin and PGV-0 on vero cells infected by Dengue-2. This research was conducted in the laboratory of parasitology Faculty of Medicine of Gadjah Mada University in January-September 2013. Experimental studies using RT-PCR test to determine the presence of Dengue-2 antigen. Dengue-2 virus propagated in C636 cellsand then infected in vero cells to further treated with curcumin and PGV-0. The incubation period of Dengue-2 infections performed for 1 and 3 days. The results of RT-PCR showed Dengue-2 antigen was seen in the 3-day infection period in the treatment of curcumin. This result indicates PGV-0 has potential antiviral better than curcumin.
