Abstract

The research was conducted to study the biodegradation capability on palm oil sludge substrate by Phanerochaete  chrysosporium   with increasing urea  as  nitrogen  source. The  experiment was  designed using Completely Randomized Design (CRD) with 5 treatments and 4 replications, namely : U-0 = 0,0 %  Urea,    U-1=      0,5  %  Urea,    U-2  =  1,0  %  Urea,    U-3  =  1,5  %  Urea  and  U-4  =  2,0  %  Urea. The observed variables were the amount of spore, C/N ratio, pH, crude fiber, crude protein, lignin, cellulose and enzyme activities. The result of this study showed that added urea were significantly (P<0.01) crude fiber, crude protein, lignin, cellulose and enzyme activities. The use of urea on the first level amounting to  1.5  %  in palm  oil  sludge  can  help  Phanerochaete  chrysosporium   to  reduce  crude  fiber to (30,71%),  lignin (29.89  %),  cellulose (36.42  %) and it  can  increase  the  content  of crude  protein    to    (34.50  %), reduction sugar and enzyme activities.

Abstract

This study aims to test the activity of cellulase enzyme (CMCase) from indigenous bacteria "Bacillus Sp YLB1" as bioactivator of palm oil based feed. The treatment consists of various combinations of substart (feed), namely: P1 = 80% palm leaves + 10% palm kernel cake + 10% sludge; P2 = 60% palm leaves + 20% palm kernel cake + 20% sludge and P3 = 60% palm leaves + 30% palm kernel cake + 30% sludge. The parameters observed were cellulase enzyme activity (CMCase) on various combinations of palm oil waste and fermentation time. The results showed that cellulase enzyme (CMCase) production of Bacillus sp YLB1 bacteria during fermentation fluctuated ie 0.143 Units / ml; 0.372 Units / ml; 0.588 Units / ml; 1.013 Units / ml; 0.906 Units / ml; 1,065 Units / ml and 1,198 Units / ml.The activity of cellulase enzyme (CMCase) on substrate P1 (0.460 Unit / ml) is lower than P2 (0.897 Unit / ml) and P3 (0.908 Units / ml). From the results of this study can be concluded that cellulase enzyme activity indigenous bacteria Bacillus sp YLB1 influenced by the combination of substrate and fermentation time. The P3 substrate fermented with Bacillus sp YLB1 for 7 days gave a more optimal result than the other treatments.   Keywords: Cellulase enzyme, indigenous bacteria, substart (feed)

Abstract

ABSTRACTThis study aimed to explore the optimization of the concentration of oil palm trunks andthermostable amylase enzyme preformance produce highest sugar (glucose total and reducingsugars) as well as the degree of polymerization. The design used in this study was a completelyrandomized design (CRD) factorial using 2 factors: factor A which consists of four levels ofamylase enzyme that A1: 250 units / kg, A2: 500 units / kg, A3: 750 units / kg and A4: 1000 units/ kg. Factor B is the concentration of oil palm trunk yitu B1: 20% (200 g / 1 liter) B2: 40% (400g / 1 liter) and B3: 60% (600 g / 1 liter) were repeated 3 times. The results showed that there is ahighly significant interaction (P <0.01) in the levels of amylase enzyme (factor A) with theconcentration of oil palm trunks (factor B) on reducing sugar, siqnificantly interaction (P <0.05)on total sugars and degree of polymerization (DP) . This research can be concluded that the besttreatment at a concentration of 60% oil palm trunks and levels 250 unit/kg of an enzymeconcentration that produces of total sugars 7.86 mg/ml, reducing sugar 15.69 mg /ml and degreeof polymerization 0.50Keywords : amilase, total gula, gula pereduksi, derajat polimerisasi.

Abstract

Propinsi Sumatera Barat memiliki pertanian relatif luas dengan bahan pakan seperti jerami padi, Rumput Gajah dan kelompok legume yang dapat dimanfaatkan untuk kebutuhan ternak sapi potong. Luas keseluruhan Kota Padang adalah 694,96 km², dan lebih dari 60% dari luas tersebut, sekitar ± 434,63 km² merupakan daerah perbukitan yang ditutupi hutan lindung, sementara selebihnya merupakan daerah efektif perkotaan. Sedangkan keadaan topografi kota ini bervariasi, 49,48% luas wilayah daratan Kota Padang berada pada wilayah kemiringan lebih dari 40% dan 23,57% berada pada wilayah kemiringan landai. Wilayah timur ini terdiri dari beberapa kecamatan yaitu mulai utara ke selatan berturut- turut dari Kecamatan Koto tangah, Pauh, Kuranji, dan Lubuk kilangan. Daerah Pauh seperti desa Lambung Bukik yang termasuk ke dalam program forum Pertides (perguruan tinggi untuk desa) yang bekerjasama yang diawali MOU Rektor tahun 201 dengan Kemendes PDTT) untuk mempersiapkan dan melaksanakan program membantu desa membangun. Sebagian pendapatan mereka diperoleh dari beternak dan bercocok tanam. Secara umum, baik usaha pertanian maupun peternakan masih dilaksanakan secara tradisional, sehingga tidak mengherankan apabila hasil yang diperoleh pun relatif rendah. Ternak sapi hanya dikandangkan atau ditambatkan pada malam hari, sedangkan siang harinya dilepas untuk mencari makanan &nbsp;dipadang rumput atau dilahan tidur sekitar desa. Baru sedikit &nbsp;upaya untuk memelihara ternak secara intensif dengan mengandangkan dan memberikan makanan secara cukup dan teratur. Rendahnya produksi ternak selain disebabkan oleh kurangnya pengetahuan peternak dalam cara pemeliharaan ternak yang benar, juga karena kurangnya pakan baik hijauan maupun mahalnya harga konsentrat. Dengan meningkatnya populasi ternak tentu membutuhkan hijauan yang lebih banyak dan mencukupi sepanjang tahun. Namun, penyediaan hijauan tersebut mengalami hambatan yang cukup serius. Salah satunya, adanya musim kemarau yang menyebabkan menurunnya produksi hijauan. Oleh karena itu usaha pengembangan ternak sapi potong akan lebih menguntungkan apabila dapat mencari alternatif pengganti hijauan konvensional dengan penggunaan silo sebagai tempat fermentasi pakan. Pemanfaatan hasil ikutan pertanian (seperti jerami padi) dan tanaman pangan lainnya sebagai pakan ternak diharapkan dapat menjawab permasalahan di atas. Hal ini dimungkinkan karena pemeliharaan ternak sapi pada umumnya terintegrasi dengan usaha tani lainnya khususnya tanaman pangan (sawah) sehingga hasil ikutan pertanian akan tersedia sepanjang tahun. Oleh sebab itu, perlulah semacam sentuhan teknologi pakan dengan pemanfaatan agroindustri by product seperti jerami yang terbukti available setiap saat. Pelaksanaan pengaplikasian teknologi amoniasi jerami padi dilapangan dengan pemberian langsung amoniasi yang sudah dismpan dan diber kotoran ayam. Partisipasi dan motivasi kelompok petani peternak dalam mengikuti serangkaian kegiatan pengabdian sangat tinggi. Karena selama ini belum pernah dilakukan pembinaan yang berkaitan dengan aspek teknis serta manajemen dalam pemeliharaan sapi potong. Peternak sudah tahu bagaimana manajemen dalam penggemukan sapi potong seperti pemberian konsentrat seperti bungkil kelapa, bungkil kedele, tepung ikan, ampas tahu dan dedak serta pemberian mineral premix.

Abstract

Pediocin N6 powder is a bacteriocin the heat resistant derived from isolates Pediococcus pentosaceus strain N6. These were isolated from water source heat Rimbo Panti West Sumatra. Pediocin N6 powder has high antimicrobial activity, so the potential to be used as biopreservatif on meat and food processing industry which involves heating. Toxicity test was conducted to determine the effects of the toxic effect of a single dose of oral Pediocin N6 powder in test animals male white mice to determine the LD50 and see changes in body weight of mice for 15 days of treatment. Changes in body weight of mice was determined by using non factorial experiment in a completely randomized design consists of 4 treatments and 5 replications. The test animals were divided into 4 groups and each treatment consisted of 5 mice. The treatments tested consisted of Pediocin N6 powder 5000 mg/kg body weight, 10,000 mg/kg body weight, 15,000 mg /kg and 20,000 mg /kg body weight of mice. The test results showed that the Pediocin N6 powder up to a dose of 20 000 mg/kg in a single oral dose administration, there are no death of mice to 15 days of treatment. Based on the LD50 value of a single oral dose can not be calculated, based on it can be stated LD50 value pseudo Pediocin N6 powder greater than 20,000 mg /kg in male mice. The average changes in body weight of mice at a dose of Pediocin N6 powder treatment of up to 20,000 mg/kg every 2 days weighing from day 1 to day 15 of 2.1 gr. Based on these tests Pediocin N6 powder safe used as industry biopreservatif on meat and food processing involves heating.Key word: Toxicity Test, Pediocin N6 Powder, White Mice, Biopreservatif on Meat
