Abstract

Growth Hormone Receptor (GHR) gene is a member of the super familycytokine/hematopoietin receptor. GHR plays a role in mediating the biological activity of growth hormone on target cells that lead to the affection of the nature ofgrowth in beef cattle carcasses and the nature of milk production in dairy cows. This study was designed to identify the genetic diversity of GHR genes exon 10 in Bali cattle. Genotype variation of gene candidates was identified using PCR-RFLP technique. The results showed that mutation at position of 755bp exon 10 of GHR gene as determined by Maskur (2012) can be identified using MBO II restriction enzymes at position of 125bp or at position of 282bp base nucleotidescaused by transition mutation of  C/T and substitution of amino acid threonine to isoleucine.Genotype identification of  GHR gene exon 10 resulted in three individual genotypes in Bali cattle population namely CC, TT and CT with frequency of 0.1931; 0.5455 and 0.2614respectively. The result of X² test and heterozygosity observationas well as heterozygosity expectation indicate that the genotypedistribution on Bali cattle population in this study were not in Hardy-Weinberg equilibrium (Hardy-Weinberg Equilibrium/ HWE). The value of  GHR|MBO II gene Polymorphic Informative Content was 0.342051, might be used for genetic identifier, and was quite informative as gene identifier for linkage analysis in the population.

Abstract

A descriptive study using 30 female local Lombok ducks age ready to lay was carried out to know the local Lombok egg duck production and its quality raised intensively in Centre Lombok for making a salted egg. The observation was done in three months. Variable observed were eggs production (monthly production, egg weight, egg index), shell quality (texture, thickness, color), and internal egg quality (yolk index, yolk color, yolk and albumen weight ratio, egg spot). Data were analyzed by using simple statistic to get the mean and the deviation standard. The observation results were: monthly egg production in the first three months were 58,15 ± 10,98%; 60,25 ± 10,60%; 50,00 ± 15,73%; average egg weight and its index were 61,42 ± 2,73 gram and 0,74 ± 0,10. The shell surface was smooth and the thickness was 0,40 ± 0,09 mm. The shell color was greenish dark blue. The yolk index was 0,47, the yolk color varied between 10 and 14, the yolk albumen ratio was 1.04, and there was not an egg spot. The quality of the local Lombok egg ducks was good for making a salted egg.

Abstract

The purpose of this research was to identify polymorphism of leptin gene in Bali cattle and Kacang goat by PCR-RFLP technique. Material used was blood samples collected from 50 head of Bali cattle and 50 head of Kacang goat. Identification procedure was consisted of three stages namely extraction of DNA genom, PCR amplication of leptin gene (552bp on intron 2-exon 3) and RFLP method using enzyme  restriction (BsaA1). Primer sequences used were forward primer: 5'GTCTGGAGGCA AAGGGCAGAGT-3'and reverse primer: 5'CCACCACCTCTGTGGAGTAG-3'. Data analyzed were frequency of allele, genotype frequency, heterozygosity value, and value of Polymorphic Informative Content (PIC). The results of this study indicate that three genotypes (AA, AG and GG) were found on Bali cattle whilst only two genotypes (AA andAG) were on Kacang goat. The values of heterozygosity observation (Ho) and expectation heterozygosity (He) on Bali cattle were 0.08 and 0.48 respectively. Kacang goat showed the value of 0,14 (Ho) and 0.14 (He). The PIC value of Leptin-BsaA1gene on Bali cattle was 0.43 (moderate high), and on Kacang goat considered as low (0.13). These results showed than Leptin-BsaA1 in Bali cattle and Kacang goat may be used as a genetic marker for selection.

Abstract

The aims of this study were to identify the mutations of FecX gene in the local goats and to analyze its polymorphism as well as its influence on the prolific nature of the local goats in West Lombok Regency, Indonesia. The study was conducted in the Immunobiology Laboratory of Mataram University, using DNA isolated from 100 blood samples of local female goats which have given birth of once to three times. The methods used were PCR-RFLP method and the PCR products were digested with HinfI restriction enzyme (G|ANTC) then analyzed visually based on DNA banding patterns on 2% agarose gels. The frequency of allele and genotype obtained, were then analyzed through a comparison with the secondary data of litter size obtained from the local goat keepers information. The results showed that the gene mutation of FecXG produced two alleles: "wild-type" (+) sized of 110 bp and 31 bp, and the mutant allele (G) of 141 bp with the allele frequency of 0,965 and 0,035 respectively. Combinations of alleles in the gene BMP15 produced two genotypes, namely (a) genotype ++ (110 bp/110 bp) with a frequency of 0.93, with the average litter size of 1.59 ± 0.319, and (b) genotype G + (141bp/110 bp), with a frequency of 0.07 and with the average litter size of 1.65 ± 0.202. The results of this study indicated that mutation occurred in BMP15 gene, i.e. FecXG gene, the gene responsible for the prolificacy of animals studied. Furthermore there was a correlation between polymorphism of FecXG gene and the prolific nature of the local goats, which was predicted to lead the divergence in litter size of each local goat genotype

Abstract

This study aims to determine the morphometric characteristics of Bali cattle in Sumbawa Regency. The material used was Bali Cattleconsisting of 15calves, 48 young and 61adults. Sampling was carried outby purposive samplingby observing morphometric characteristics of Bali cattle.The results show that body size of male and female in the calf age group have height, body length, and hip height was different (P<0,05). In young age group just height parameters werenot different (P>0,05) and in adult age group just body length parameter was different (P>0,05).  Chest circumference has the highest correlation coefficient on the body weight followed by body lengthand hip heightwith thevalues ??of 0,88; 0,82; and 0,80respectively. The equation BW= -207,89 + 1,49BL+ 2,5CC ? 0,74HHand the degree of determination (R2) of0.80 was determined using regression analysis.Chest circumference and body length significantly (P<0.05)effected thebody weight whilst hip height not(P>0.05) and simultaneously influences body weight (P<0.05).

Abstract

This study aims to determine the morphometric characteristics of the Sumbawa x Bali (Sumbal) crossbred cattlein Sumbawa Regency. The material used wasSumbal Cattleconsisting of 14 calves, 20 young and 27 adults. Sampling was carried outby purposive samplingby observing morphometric characteristics of Sumbal cattle.The results show that body size of male Sumbal cattle wasnot different (P>0.05) from female in the calf and young age groups. Whereas in the adult age group, male waslower than female (P<0.05) in terms of hip height parameters. Chest circumference has the highest correlation coefficient on the body weight followed by height and body length with thevalues ??of 0,96; 0,83; and 0,80respectively.The equation BW= -173,104 ? 1,837H+ 0,052BL+ 4,493CCand the degree of determination (R2) of0.939was determined using regression analysis. Abstract

Penelitian dilakukan dengan tujuan untuk mengetahui pengaruh level penambahan tepung tapioka terhadap kualitas kimia sosis daging ayam dengan penambahan tepung tapioka. Rancangan penelitian yang digunakan adalah Rancangan Acak Lengkap Pola Searah dengan 4 perlakuan yaitu penambahan tepung tapioka: 0%, 10%, 20 % dan 30% dan setiap perlakuan terdapat 3 ulangan. Data hasil penelitian kualitas sosis dianalisa menggunakan Analisa Variansi dan dilanjutkan dengan uji jarak Ganda (Duncan). Hasil analisa varian menunjukkan bahwa penambahan tepung tapioka dalam proses pembuatan sosis daging ayam memberikan pengaruh yang nyata (P<0.05) terhadap kadar air dan protein, sedangkan terhadap kadar lemak dan abu tidak berpengaruh nyata (P>0,05). Penambahan tepung tapioka sebanyak 10% dalam pembuatan sosis daging ayam dapat memberikan nilai gizi (protein) yaitu 17,67±0,93% yang lebih tinggi dibandingkan dengan penambahan 20% dan 30%.

Abstract

Penelitian ini bertujuan untuk mengidentifikasi keragaman sifat kualitatif dan morfometrik induk ayam kampung berdasarkan tipe jengger di Pulau Lombok. Materi yang digunakan yaitu induk ayam kampung yang dipelihara secara tradisional sebanyak 417 ekor. Metode yang digunakan yaitu survei. Penentuan lokasi dilakukan secara purposif sampling dan  pengambilan sampel dilakukan secara random sampling. Data kualitatif meliputi tipe jengger, warna bulu, warna kulit, warna kaki. Data morfometrik meliputi tinggi jengger, panjang shank, panjang tibia, panjang femur, jarak antar tulang pubis, jarak antara tulang pubis dengan tulang dada dan bobot badan. Data dianalisis menggunakan analisis statistik sederhana. Hasil penelitian menemukan 4 (empat) tipe jengger walnut 36.45 persen, tunggal 30.45 persen, pea 24.46 persen dan rose 8.64 persen. Warna bulu yaitu hitam, keemasan, lurik dan putih. Warna bulu putih hanya ditemukan pada ayam jengger pea. Warna cuping telinga yaitu merah dan putih. Warna shank yaitu hitam, putih dan kuning. Warna kulit yaitu kuning dan putih. Morfometrik ayam kampung tertinggi dicapai ayam tipe jengger rose yaitu tinggi jengger 12.16 + 2.78mm, panjang shank 74.26 + 11.62mm, panjang tibia 88.47 + 11.87mm, panjang femur 82.11 + 12.45mm, jarak dua tulang pubis 35.95 + 7.32mm, jarak tulang pubis dengan tulang dada 54.74 + 9.86mm dan bobot badan 1658.19+339.63gram.
