Abstract

 Triple negative breast cancer (TNBC) is still associated with grave prognosis, especially compared to other breast cancer subtypes. Advances in medical science have improved our understanding on the biological nature and heterogeneity of TNBC, explaining the efficacy variability of existing chemotherapeutic drugs on TNBC patients. Complexity of TNBC has led to wide variation of TNBC treatment across the globe, resulting in unsatisfactory treatment outcome. This issue is further complicated by the absence of TNBC treatment guideline in many countries, including in Indonesia. This review discusses systemic treatment options for TNBC while taking account its molecular heterogeneity. Specific consideration is made for Indonesia, not only for current clinical practice, but also for future improvements. Immunotherapy, especially programmed cell death 1 (PD-1/PD-L1) inhibitor, has recently shown promising result in TNBC patients. It can be concluded that TNBC is heterogenous and treatment option should be tailored based on its molecular profile.

Abstract

ABSTRAK   Temu kunci (B.pandurata) mengandung senyawa-senyawa yang berpotensi antikanker. Penelitian ini bertujuan untuk mengkaji efek kemoprevensi ekstrak etanolik rimpang temu kunci pada karsinogenesis kulit terinduksi ultra violet. Ekstraksi serbuk rimpang B. pandurata dilakukan dengan metode maserasi  menggunakan etanol. Hewan uji yang digunakan adalah mencit Balb/C betina umur 28 hari yang dicukur rambut punggungnya, dikelompokkan menjadi 5 kelompok perlakuan, yaitu kelompok kontrol dan empat  kelompok   perlakuan ekstrak etanolik temu kunci yang diberikan secara oral. Induksi karsinogenesis dengan paparan UV dosis 0,167 J/cm2/hari, sebanyak 60 paparan (5 kali paparan/minggu). Efek penghambatan karsinogenesis kulit  dipelajari pada tingkat insidensi dan multiplisitas kanker. Hasil uji menunjukkan bahwa ekstrak etanolik temu kunci mampu menurunkan angka insidensi dan multiplisitas kanker kulit pada mencit Balb/C terinduksi UV. Abstract

ABSTRACT---In this paper, we analyze and compute distribution of thermal neutron flux in core of Kartini?s Reactor using ?Swadaya? detector, both for axial (function of z) and radial (function r) direction. First, ?Swadaya? detector is calibrated by activation plate method, which the output from detector is current. The result of calibration is thermal neutron flux (?) dependent on current (I) that can be written as ? = 4,57x1020I - 2,74x1011. Distribution for neutron thermal flux for axial direction and radial direction respectively ? = 5,10 x 1012 + 5,99 x 1013 and ? = 1.08 x 1012 + 1,00 x 103 which in reactor core ? has largest value in center position for radial direction.   Keywords: flux neutron distribution, nucleus reactor, Swadaya detector

Abstract

Lymphocyte is leukocyte component that difficult to culture in vitro. Several viruses need lymphocytes as host cell in order to proliferate and growth in this media such as Epstein-Barr virus (EBV). This virus was associated with malignant disease like nasopharyngeal carcinoma (NPC). The objectives of this research are to develop and to prepare lymphocyte cell culture as material source of DNA for molecular analysis of virus. Peripheral blood was collected from NPC patients which is histopatologically and serologically positive of EBV. Lymphocytes were separated from the other blood components using ficcol-histopaque. Lymphocytes were diluted using RPMt medium then they were cultivated into 96 microwell plate with concentration of 106 cell/ml. The medium consist of 10% FBS, RPMI, Penstrep and FK 506. Culture of lymphocytes were incubated in 5% CO2 at 37°C. The lymphocyte cultures developed and grew confluendy during the first week. Only B cells whith EBV would be well establish. After 50 cell generations, lymphocytes were transformed and immortalized to be lymphoblastoid cell fine (LCL).

Abstract

Cancer is a disease, a public health problem, which is found in the world as well as in Indonesia. Ingeneral, some of cancer theraphies are ineffective, characterized by the resistance performance of cancer cell line,the exposed normal cell and by the side effects. Nowadays, studies to fi nd the specifi c and safely anti-cancerdrugs were increased by the time. Several studies revealed that Curcuma mangga Val. Rhizome contains somesecondary metabolites, essential or non-essential oil, which has cytotoxic activities to the cancer cells. Basedon these anti-cancer potentials, this study has several aims to recognize anti-cancer selectivity and molecularmechanism by inducting apoptosis and inhibiting myeloma cell proliferation. To C. mangga Val. essential oil,immunocyto chemical test was performed to determine the expression of p53, caspase-9, Bcl-2, H-Ras proteinwhile TUNEL test was performed to determine the number of apoptosis cells.The results of this study shown that anti-cancer molecular mechanism of C. mangga Val. essential oil tomyeloma cell line was performed by increasing apoptosis; by increasing the expression of pro-apoptosis p53,caspase-9 protein and reducing protein which is increasing proliferation Bcl-2 and H-Ras.

Abstract

Minyak atsiri rimpang Curcuma mangga telah diketahui bersifat toksik terhadap beberapa jenis sel kanker. Toksisitas minyak atsiri dipengaruhi oleh komposisi senyawa penyusunnya. Komposisi minyak atsiri dipengaruhi oleh berbagai faktor ekologi dan metode isolasi. Penelitian ini bertujuan untuk mengetahui pengaruh lokasi tumbuh, umur tanaman dan variasi jenis destilasi terhadap komposisi senyawa minyak atsiri rimpang C. mangga. Untuk mempelajari pengaruh lokasi tumbuh diambil rimpang C. mangga yang berasal dari beberapa daerah di Daerah Istimewa Yogyakarta. Pengaruh umur tanaman terhadap komposisi senyawa dalam minyak atsiri digunakan rimpang C. mangga umur 1, 2, 3, 7, 10, 11 dan 12 bulan, sedangkan jenis destilasi yang digunakan adalah destilasi uap dan uap air. Analisis komponen senyawa minyak atsiri digunakan Kromatografi Gas-Spektrometer Massa (KG-SM). Hasil penelitian menunjukkan bahwa rimpang yang berasal dari daerah dataran rendah memiliki jenis senyawa yang lebih banyak daripada dataran tinggi dan dimungkinkan karena curah hujan di dataran rendah lebih kecil daripada dataran tinggi. Minyak atsiri rimpang C. mangga optimum apabila tanaman dipanen pada saat umur 8-10 bulan dan jenis destilasi berpengaruh terhadap komposisi minyak atsiri.  Namun begitu, senyawa utama minyak atsiri rimpang C. mangga untuk semua variasi perlakuan adalah sama yaitu β-osimen, β-pinen, β-mirsen dan p-sineol.

Abstract

Finding anticancer drugs from natural resources still proceeds. Oleandrin isolated from Nerium indicum Mill. inhibited the growth of mieloma cell line in vitro better than that of vincristine sulphate. This study was aimed to determine the cytotoxic effect of oleandrin on various human cancer cell lines. Cytotoxic test of oleandrin on seven human cancer cell lines was done by SRB-method. The analysis was conducted by comparing the ID50 of oleandrin with that of doxorubicin and cisplatin as positive controls. This result indicated that oleandrin possessed the best cytotoxic effect on breast cancer (MCF7) with ID50 at 8.85 nM. Keywords : Oleandrin, cytotoxicity, human cancer cells, ID50

Abstract

Oleandrin is one of cardenolida compound isolated from an active fraction of Nerium indicum Mill leaves. (fam. Apocynaceae), which have cytotoxic effect on several human cancer cells, and also to normal cells in vitro. Another compound which was potential as renal anticancer has also been isolated from N. indicum. However, its chemical structure has not been discovered. The aim of this study was to identify the potential compound as selective to renal cancer present in the leaves of N. indicum.The potential compound was isolated from the active fraction using Preparative TLC and structure elucidation was done by using spectroscopic methods (UV, IR, MS and NMR).Base on this spectra and by comparison with oleandrin data, it was indicated that the potential compound as Renal anticancer was as 16,17-dehydrodeacetyl-5a-oleandrin.Key word: N.indicum, oleandrin, cytotoxic, renal anticancer, spectroscopic

Abstract

The 16,17-dehydrodeacetyl-5α-oleandrin was isolated from an active fraction of Nerium indicum Mill leaves (fam. Apocynaceae). This compound was cytotoxic against various cancer cells, and selective on A498 cells (Renal cancer). However, the apoptosis mechanism was still unknown yet. Therefore, the aim of this study was to know the apoptotic mechanism of 16,17-dehydrodeacetyl-5α-oleandrin on A498 cells by FITC labeled annexin V and immunucytochemical assays. The detection of apoptotic mechanism on A498 cells was performed with FITC-conjugated annexin V using Flow Cytometry. The p53 protein expression were detected using immunocytochemical. Treatment with 16,17-hydrodeacetyl-5α-oleandrin (3.88 x 10-4 mM) using FITC-annexin V increased the percentage of the dead cells in the 24th and 48th hours incubation period. The 16,17-dehydrodeacetyl-5α-oleandrin (1,94x10-4 and 3,88x10-4 mM) raised significanly p53 protein expression (p<0,05). The percentage of the p53 protein expression increased throughout the time of samples incubation.Key words: 16,17-dehydrodeacetyl-5α-oleandrin,fluorescein isothiocyanate-annexin V, immunocytochemically, A498 cells.

Abstract

Mahkota Dewa [Phaleria macrocarpa (Scheff). Boerl.] is used traditionally to treat cancers in Indonesia. Extract methanol of the leaves displayed a small LC50 value (63.16 μg/ml) on BST (Brine Shrimp Lethality test) assay; therefore a phytochemical study of this extract was undertaken. A new benzophenoic glucoside was isolated from the methanol extract and identified as 4,5-dihydroxy,4’-methoxybenzophenone-3-0-β-D-glucoside (Phalerin) based on its spectroscopic data. Phalerin was cytotoxic to myeloma cell line (NS-1) (in vitro) having IC50 of 83 μg/ml or 1.9 x 10-1 mM.Key words: Phaleria macrocarpa, Phalerin, myeloma
