Abstract

Penelitian ini bertujuan mengetahui peran zink pada kesembuhan luka dalam kondisi berat badan berlebih. Dalam penelitian digunakan 20 ekor tikus Sprague Dawley jantan berumur tiga bulan (150-200 g). Tikus dibagi secara acak menjadi empat kelompok (A, B, C, dan D), masing-masing kelompok terdiri atas lima ekor. Kelompok A dan B diberi pakan normal, sedangkan kelompok C dan D diberi pakan lemak tinggi. Setelah dua bulan perlakuan pakan, seluruh tikus dioperasi untuk membuat luka irisan pada kulit, kemudian luka ditutup dengan jahitan. Luka jahitan pada kelompok A dan C diberi aplikasi topikal unguenta vaseline album (tanpa zink), sedangkan luka jahitan pada kelompok B dan D diberi aplikasi topikal unguenta zink 10% dengan bahan dasar vaselin album. Lima hari setelah operasi, seluruh tikus diambil darahnya untuk dianalisis terhadap jumlah leukosit total kemudian diikuti pengambilan jaringan kulit untuk pemeriksaan histopatologis dan penghitungan jumlah leukosit di jaringan. Hasil penelitian menunjukkan adanya interaksi yang signifikan antara pakan dan aplikasi topikal terhadap jumlah leukosittotal dalam darah (P 0,05). Jumlah leukosit di jaringan mengalami penurunan setelah aplikasi topikal zink, meskipun hasilnya tidak signifikan (P 0,05). Analisis histopatologis menunjukkan epidermis sudah tertutup pada seluruh tikus perlakuan, namun demikian sebagian tikus masih menunjukkan dermis yang terbuka, yaitu kelompok A sebanyak 60%, kelompok B sebanyak 60%, kelompok C sebanyak 60%, dan kelompok D sebanyak 40%. Dapat disimpulkan bahwa proses kesembuhan luka setelah pemberian topikal zink pada tikus yang diberi pakan lemak tinggi menunjukkan hasil yang baik, yaitu ditandai dengan penutupan lapisan epidermis dan dermis, serta menurunnya jumlah leukosit di jaringan.

Abstract

On Timor island, Nusa Tenggara Timur, faloak barks (Sterculia quadrifida R.Br.) has been used empirically to restore stamina. Faloak bark ethanolic extract proved to have immunomodulatory activity in vitro, which can increase macrophage phagocytosis activity. This research aimed: (i) to determine the immunomodulatory active fraction of faloak bark ethanolic extract, (ii) to determine the total flavonoid contents of faloak extract and fractions, and (iii) to evaluate the correlation of the total flavonoid contents of those extract and fractions with their macrophage phagocytosis activity. The simplisia powder is macerated with 96% ethanol. The extract was dissolved in methanol:water (9:1v/v) was then subsequently partitioned with n-hexane, ethyl acetate, and water to obtain n-hexane fraction, ethyl acetate fraction, water fraction, and insoluble fraction. Faloak extract and fractions at concentration 62,5; 125; 250; 500μg/mL were tested for their effect on the peritoneal macrophage phagocytosis of Balb/c mice in vitro by the latex beads method. Phagocytosis capacity and phagocytosis index were analyzed using one-way anova and post hoc Tukey HSD test with 95% confidence level. The results showed that ethyl acetate fraction had the highest macrophage phagocytosis capacity and the highest total flavonoid content compared to other fractions. The highest macrophage phagocytosis capacity of ethyl acetate fraction at concentration of 250 μg/mL was 51,94±4,67%, this value was significantly different from cell control (7,50±1,29%), negative controls of 0,0625% dimethylsulphoxide (6,25±0,36%), as well as positive control of 200 μg/mL echinaceae extract syrup® (9,97±0,33%). The total flavonoid content of ethyl acetate fraction determined by aluminum chloride method was 4,290±0.029 mg of quercetin equivalent/g fraction. There was a positive and strong correlation between the total flavonoid content of these extract and fractions with their macrophage phagocytosis capacity (Pearson correlation coefficient of 0,781) and showing linear relationship y=4,721x+19,663; R2=0,61.

Abstract

Osteoporosis and hypercholesterolemia are prevalent condition in menopausal women. The common therapy to prevent the estrogen degrading condition is Hormone Replacement Therapy (HRT). However, HRT possessed various risks. Curcubita pepo L. seed (pumpkin seed) contains lignan secoisolariciresinol and lariciresinol which exhibit estrogenic effect. The aim of this study is to determine the estrogenic effect of Ethanolic Extract of Pumpkin Seeds (EEPS) through in silico and in vivo study. In silico study were conducted by molecular docking of lignan which is secoisolariciresinol and lariciresinol with Estrogen Receptor (ERα and Erβ). In vivo study conducted by using ovariectomized Sprague dawley female rats as a model of postmenopausal women. Blood lipid profile, bone density, and uterine weight were assayed after thirty days. Molecular docking score of secoisolariciresinol and lariciresinol were similar to estradiol. In vivo study found that EEPS increase bone density and uterine weight percentage while also improve the blood profile. In conclusion, these result showed that EEPS is potential to be developed as an osteoporosis and hypercholesterolemia prevention agent. Keywords:  Lime peel, Limonene, Aromatic candles, Repellent

Abstract

Gynura procumbens (Lour) Merr., has been shown to suppress lung cancer development in mice and breast cancer development in rat when the extract was given at initiation stage. The aim of this research is to examine the potential of ethanolic extract of G. procumbens to suppress DMBAinduced breast cancer development at early development (post initiation I) and late development (post initiation II). Sprague Dawley Rats were used in this research and were grouped as indicated treatment. Ethanolic extract of G. procumbens was administered into 2 levels of doses, namely 250, 750 mg/kgBW. Tumor development was examined by palpation every week and terminated at week 16th after the end of DMBA treatment. The result showed that extract treatment at the dose of 250, and 750 mg/kgBW at the post initiation I could not reduce tumor incidence but suppressed of tumor multiplicity. However, the treatment at the post initiation II, the extract could not reduce neither incidence nor multiplicity. In conclusion, ethanolic extract of G. procumbens performs potential effect to suppress breast cancer development at the dose of 250 mg/kgBW when administered at the early stage of carcinogenesis.Key words : Carcinogenesis inhibition, G. procumbens, breast cancer, post initiation

Abstract

Curcuma zedoaria Rosc. rhizomes has been used as a traditional cancer medicine since a long time ago, but the so far researches performed on the pharmacological mechanism were lacking. This present research has been done to determine the effect of ethanolic extract of Curcuma zedoaria Rosc. rhizomes on lung cancer of female mice previously induced by Benzo[a]pyrene (B[a]P).Newborn mice were induced by B[a]P and then were separated between male and female mice. On 30th day after birth, female mice were given the ethanolic extract of Curcuma zedoaria Rosc. rhizomes and divided into five groups. Three groups were given 200, 500 and 750 mg/kgBB extract, a group was given solvent (DMSO) as negative control, and another group was given nothing as positive control.The results of this present research shown that the ethanolic extract of Curcuma zedoaria Rosc. rhizomes has proved to posses inhibition effect on lung cancer growth in female mice, at dose of 250 mg/kgBB (49,63%), 500 mg/kgBB (73,33%) and 750 mg/kgBB (77,78%).Keywords : Newborn Mice, Benzo[a]piren, Curcuma zedoaria Rosc.

Abstract

Gynura procumbens (Lour) Merr., empirically, used to prevent cancer development and has been proven to be able to suppress lung cancer development. The aim of this research is to examine the potential of ethanolic extract of G. procumbens to suppress DMBA-induced breast cancer development. Sprague Dawly Rats were used in this research and were grouped as indicated treatment. Ethanolic extract of G. procumbens was administered into 3 levels of doses, namely 250, 500, and 750 mg/kgBW. Tumor development was examined by palpation every week and terminated at week 16th after the end of DMBA treatment. The result showed that extract treatment at the dose of 250, 500, and 750 mg/kgBW reduced tumor incidence by 60%, 30 %, and 20 % respectively. The doses of 500 and 750 mg/kgBW exhibited strong suppression of tumor multiplicity, where as the dose of 250 performed less potential suppression. In conclusion, ethanolic extract of G. procumbens performs chemopreventive effect to suppress breast cancer development at the dose of 250 mg/kgBW.Key words : chemopreventive, Gynura procumbens (Lour) Merr, breast cancer.

Abstract

Antiangiogenesis (inhibition of new blood vessels formation) has become a strategy to inhibit cancer development lack of nutrition and oxygen supply. The aim of the present research is to investigate antiangiogenesis effect of ethanolic extract of Gynura procumbens (Lour.) Merr. Leaves in situ using chick embryo chorioallantoic membrane (CAM). Eight to 9 days old fertilized chicken eggs were treated with b-FGF (angiogenesis inductor) and extracts. Eggs were then incubated for 3 days in order to observe its angiogenesis response (new blood vessels converged toward the implant).The results showed that the ethanolic extract of G.procumbens could inhibit angiogenesis in a dose-dependent manner. Doses 10, 20, 40, 80 ug gave angiogenesis response of (in percent) 82.32 ± 6.33; 68.38 ± 6.24; 56.48 ± 11.61; 41.43 ± 7.46 (p<0.05), respectively. These results indicate a potential antiangiogenic effect of the extract.Key words: antiangiogenic, CAM, G.procumbens.

Abstract

The  objective  of  this  study  is  to  determine  the  hepatoprotective  effect  of  GVT-0  (one  of  curcumin  analogues)  against liver  damage  in  rat-induced  D-galactosamine  (D-GalN)/lipopolysaccharide  (LPS)  as  a model  of fulminant  hepatitis.  In  the study D˗GalN/LPS  elevated  serum  GPT  activity  that  indicate  a particular occurrence of liver damage due to depletion of UTP and UDP-glucuronic acid. Administration of GVT-0 (10 mg/kg) showed decreased  enzyme  activity  of  SGPT/SGOT  but  had  no  effect  on serum  ALP  and  total  bilirubin  levels,  whereas  at  doses  of  20 and 40  mg/kg,  the  protective  effect  of  GVT-0  was  decrease.  The glutathione  content  in  the D-GalN/LPS  (0.76  ±  0.07)  mol/g  liver content was found lower than  controls  (0.90  ±  0.03)  mol/g  liver. Administration  of  GVT-0  dose  of  10,  20  and  40  mg/kg  restored glutathione content returned  to normal  levels. The results showed that treatment of GVT-0 showed no effect on TBARS and catalase activity.  Treatment  of  D-GalN/LPS,  indicating  the  trend  of increased  TNF-α,  although  statistically  not  significant,  while  the administration  of  GVT-0  showed  a  tendency  to  decrease  the concentration  of  TNF-α.  All  findings  of  the  results  indicated  that the GVT-0 mainly lower dose (10 mg/kg) showed hepatoprotective action  in rat  model  of fulminant  hepatitis induced  by D-GalN/LPS. The  results  indicated  that  the  mechanism  of  hepatoprotective effect  of  GVT-0  is  not  via  antioxidant  properties  of  GVT-0. However,  further  studies  are  necessary  to  explain  the  molecular mechanism of hepatoprotective effect of GVT-0.Key  words:  Gamavuton-0,  hepatoprotective,  fulminan  hepatitis, D˗galactosamine/LPS

Abstract

Inhibitory Phases Effect of The Lung Cancer And Acute Toxicity of Chang Sheuw Tian RanLing Yao Capsule Extracts in House mice (Mus musculus) and Rat (Ratus tanezumi). Effortsto find anticancer agents have been developed nowadays, some of them are focused in traditionalherbs. One of the available products in the market that claims effective to cure cancer isthe Chang Sheuw Tian Ran Ling Yao, PT. Daun Teratai extract containing CAPSULE (CSTRLYextract). The aim of this study is to examine of confession of some people which are usingthe useful of medicine CSTRLY extract capsule through inhibitor laboratory effect of theCSTRLY extract in the initiation and post initiation phases of the lung cancer in mice and ratsthat had been induced by eather Benzo[?]pyrene (BP) or Dimetilbenz[?]antrazene (DMBA)and to clarify the potency of acute toxicity and specific toxic manifestations of thephytopharmaca.The results showed that the CSTRLY extract can reduce the cancer incidence caused bycarcinogen, BP and DMBA. Moreover, the extract can also inhibit the cancer growth in themice and rats, especially in the early post-initiation phase. Further, the histopathologicalevaluation showed that up to the highest dose level that technically could be administrated tothe animals (12500 mg/kg bw), no animal death was occurred. Furthermore, the ADG values formale and female rats indicated no significant different (P > 0.05) that relative to the controlgroup. No animals were shows physical symptom as a toxic manifestation. It’s indicated thatthe phytopharmaca no influenced to somatomotor and nervous system. Within the dose rangeadministrations, no detectable morphological toxic effects or histophatological changes of theliver, spleen, heart, and lungs were observed. the acute toxicity value of Chang Sheuw TianRan Ling Yao Capsule was very low (or minimal almost non-toxic with LD50 > 12500 mg/kg bw)and the spectrum of toxic effects of the phytopharmaca were considered negligible.Key words: Ekstract, CSTRLY, mice and rat, BP, DMBA, carsinogenesis, lung cancer
