Abstract

The evaluation of toxicity of a mixed herbal extract containing mahkota dewa fruits (Phaleria macrocarpa Scheff. Boerl) and pegagan leaves (Centella asiatica L. Urban) on Wistar-strain rats had been carried out by the determination of the urea and creatinine content in urine and plasma after feeding. Oral doses of 100 mg, 500 mg, and 2500 mg of the mixed extract/kg body Weight were administered for 16 consecutive weeks to three groups of rats. Each treated group consisted of 15 males and 15 females, and the control group was represented by 10 males and 10 females. Samples of urine and plasma of the treated groups were taken at the time right before treatment (Week zero) and at Sm, 16th, 18th Week, while those of the control were taken at zero Week, 8th, and 16th Week, respectively. The result showed that theurea and creatinine contents among the treated and control groups were not signihcantly different. It could be concluded that oral administration ofthe mixed extract by a dose up to 2500 mg/kg body weight for 16 Weeks did not influence the urea and creatinine contents both in urine and plasma of the treated animals. Based on this result, it could be assumed that the use of the mixed extract is safe.

Abstract

Penelitian ini bertujuan memferifikasi aktivitas ekstrak etanol pegagan atau Centella asiatica Urban (CA) local sebagai peningkat imunitas tubuh dengan parameter uji kapasitas dan aktivitas fagositosis makrofag peritoneum secara in vivo. Tikus jantan galur Sprague Dawley dibagi menjadi 4 kelompok, masing-masing terdiri dari 5 ekor. Kelompok dosis 1, dosis 2 dan dosis 3 diberi ekstrak 10, 20 dan 40 mg/200 g BB . secara oral selama 15 hari berturut-turut, dan kelompok kontrol mendapat pembawa. Satu hari setelah pemberian ekstrak yang terakhir, semua hewan coba dibunuh setelah diinduksi dengan bakteri Staphylococcus epidermidis (109 CFU per ekor) i.p. Aktivitas makrofag dan kapasitas makrofag peritoneum dihitung dengan dengan pewarnaan giemsa menggunakan mikroskop. Hasil menunjukkan bahwa terdapat korelasi positif antara peningkatan dosis dan aktivitas maupun kapasitas makrofag. Nilai aktivitas dan kapsitas makrofag pada kelompok yang mendapat ekstrak 40 mg/200 g BB secara statistik berbeda secara bermakna dibanding kelompok control (96% aktivitas makrofag dan 61 bakteri untuk kapasitas makrofag). Berdasarkan penelitian ini dapat ditarik kesimpulan bahwa ekstrak etanol CA berpotensi untuk dikembangkan sebagai bahan peningkat daya imunitas tubuh. Namun demikian, masih perlu dilakukan pengujian lebih lanjut untuk mengelaborasi kemampuan peningkat system imun secara lengkap dan detail.

Abstract

Drying process of Mengkudu (Morinda citrifolia L.) juice by spray drying method had been carried out. Two kinds of fillers were used in this process, i.e. 5%, 15%, 20%, and 25% of lactose monohydrate or 15% and 20% of maltodextrin, both in w/w %. The result obtained showed, that the dried powder containing 15 % of maltodextrin was better (dry fine and brown) than those with other concentration of fillers.

Abstract

The liver function as a detoxification create liver damage and can produce fibrosis condition both acute and chronic. One approach for the treatment of liver fibrosis is used antioxidant of natural materials. Extract of gambier was verified containing phenolic compounds of antioxidants. This research was aimed to evaluate gambier potency as hepatoprotective in animal models of liver fibrosis. Total 45 of male Sprague-Dawley rat were equally divided into nine groups. Consisted of D1 (received gambier 13 mg/200 g bw + CCl4), D2 (gambier 26 mg/200 g bw + CCl4), D3 (gambier 53 mg/200 g bw + CCl4), K+1 (poliphenol 25 mg/kg bw + CCl4), K+2 (mixtured medicine extract 302 mg/kg bw + CCl4), K- (CCl4), KG (gambier 53 mg/200 g bw), KP (coconut oil), KN (normal group). CCl4 (0.1 ml/kg bw) given twice a week. The all samples was administered orally (1 ml/200 g bb) for six consecutive weeks. The analyzed parameters were the activities of liver enzymes, malondyaldehyde and glutathione levels were measured in liver homogenates and histopathology were studied in liver sampels. The result showed that extract of gambier repaired the liver damage. Administered extract of gambier showed that gambier haspotential activity as a hepatoprotector significantly (p<0.05) compared with the negative control (K-). The hepatoprotective activity of gambier extract was more effective in D2 (26 mg/200 g bw).
