Abstract

Penelitian ini bertujuan untuk mengetahui keragaman genetik ikan batak dari beberapa lokasi yang berbeda. Ikan batak yang digunakan dalam penelitian ini dikumpulkan dari Sungai Tarutung, Sungai Bahorok, Sungai Aek Sirambe dan Sungai Asahan di provinsi Sumatera Utara, dan dari Kabupaten Sumedang, Jawa Barat. Penelitian ini menggunakan metode Random Amplified Polymorphism DNA (RAPD). Hasil penelitianmenunjukkan bahwa nilai rata-rata heterozygote adalah antara 0.0909 ? 0.1407. Populasi ikan batak dariTarutung dan Bahorok memiliki jarak genetik paling dekat dengan nilai 0.1272 dan populasi ikan batak dari Asahan dan Aek Sirambe memiliki jarak genetik paling jauh dengan nilai 0.3106. Dari dendogram jarak genetik terlihat bahwa terjadi dua pengelompokkan populasi, kelompok pertama terdiri dari populasi Tarutung, Bahorok dan Asahan, dan kelompok kedua terdiri dari populasi Sumedang dan Aek Sirambe.Kata kunci: ikan batak, Tor soro, RAPD.

Abstract

Indonesia dikenal sebagai pusat sumber daya udang air tawar, salah satu di antaranya adalah udang galah dari daerah aliran sungai (DAS) Bengawan Solo. Kegiatan penelitian bertujuan untuk mengidentifikasi secara genetik dan mendapatkan informasi tentang karakter reproduksi dan daya adaptasinya terhadap lingkungan dari udang galah Bengawan Solo. Identifikasi dilakukan melalui sekuensing daerah mtDNA CO-1. Pengamatan reproduksi dilakukan pada saat matang gonad pertama. Toleransi terhadap lingkungan terdiri atas uji salinitas, pH, suhu, dan oksigen terlarut (DO). Data sekuensing dianalisis dengan menggunakan program CLUSTAL OMEGA. Data reproduksi dianalisis secara deskriptif dengan menggunakan program Excel. Data hasil uji toleransi terhadap lingkungan dianalisis dengan menggunakan program SPSS 16. Sekuens mtDNA CO-1 udang galah Bengawan Solo mempunyai tingkat similaritas 98% terhadap Macrobrachium rosenbergii (KM234150). Proporsi basa A (27,62%), G (19,53%), T (27,34%), dan C (25,52%) menyusun 239 residu asam amino. Filogenetik berdasarkan jarak genetik mengelompokkan udang galah Bengawan Solo, GImacro, Siratu, Mahakam, dan KM234150 dalam satu grup. Induk udang galah umur 216 hari mencapai matang gonad yang pertama dengan ukuran panjang 15,32 ± 0,58 cm; bobot 48,58 ± 5,87 g (jantan) dan 13,86 ± 0,75 cm; bobot 29,04 ± 4,64 g (betina). Tingkat fekunditas yang dimiliki oleh induk betina adalah 834,67 ± 57,73 butir/g. Diameter telur berkisar 0,40-0,53 mm dengan bobot rata-rata 0,112 g. Sintasan larva hingga umur satu bulan adalah sebesar 50,56 ± 0,61%. Benih udang galah pada salinitas 5-25 ppt mempunyai tingkat sintasan 73,33%-86,67%; pH 4-8 dengan tingkat sintasan 66,67%-73,33%; suhu 20°C-34°C dengan tingkat sintasan 76,67%-96,67% dan tingkat oksigen yang dibutuhkan benih udang galah > 1,04 mg/L.Indonesia is known as a resource center for freshwater shrimp, one of which is the giant freshwater prawn from Bengawan Solo watershed. This research aimed to genetically identify and obtain information about the giant freshwater prawn?s reproduction and adaptation characters in a culture environment. Identification was made by sequencing the MtDNA CO-1 region. Reproductive observation was carried out when the first gonad matured. Environmental tolerance tests consisted of salinity, pH, temperature, and dissolved oxygen (DO) tests. Sequencing data were analyzed using the CLUSTAL OMEGA program. Reproduction data were analyzed descriptively using the Excel program. Data from the environmental tolerance tests were analyzed using the SPSS 16 software package. The mtDNA CO-1 sequencing result of Bengawan Solo prawns has a 98% similarity rate to Macrobrachium rosenbergii (KM234150). The proportions of base A (27.62%), G (19.53%), T (27.34%), and C (25.52%) have compiled 239 amino acid residues. Phylogenetic analysis based on the genetic distance has grouped Bengawan Solo, GImacro, Siratu, Mahakam, and M. rosenbergii-KM234150 in one group. The broodstock parent reached the first gonadal maturity at 216 days with an average body length and weight of 15.32 ± 0.58 cm; 48.58 ± 5.87 g for male and 13.86 ± 0.75 cm; 29.04 ± 4.64 g for female, respectively. The fecundity rate of the female parent was 834.67 ± 57.73 eggs/g body weight. Egg diameters ranged from 0.40 to 0.53 mm, with an average weight of 0.112 g. Larval survival was 50.56 ± 0.61%. Seed prawns subjected with: salinity tests between 5-25 ppt have survival rates between 73.33%-86.67%; pH tests ranged between 4-8 have survival rates between 66.67%-73.33%; temperature test between 20°C-34°C have survival rate between 76.67%-96.67%. The optimum oxygen level needed for giant prawn seeds > 1.04 mg/L.

Abstract

Sekitar 10,37 juta jiwa jumlah penduduk DKI pada tahun 2017. Kondisi ini memerlukan pasokan bahan pangan yang cukup besar dan terjamin. Salah satu sumber protein yang dapat dihasilkan dalam jumlah massal dan cepat adalah produk perikanan air tawar. Saat ini telah dikembangkan teknologi bioflok (BFT) yang hemat lahan dan air sangat cocok untuk dikembangkan didaerah perkotaan atau hunian padat penduduk yang juga merupakan pasar produk yang dihasilkan. Tingkat produktifitas BFT adalah 80-100 kg per m3 air untuk ikan lele, sedangkan ikan nila mempunyai tingkat produksi 16-23 kg/m3 air. Dengan masa pemeliharaan yang berkisar antara 2-4 bulan serta harga pokok produksi (HPP) sekitar Rp 14-17 ribu per kg, maka diperkirakan BFT ini akan menjadi suatu terobosan selain sebagai pendukung program ketahanan pangan dalam rangka pasokan protein yang cukup (misalnya dalam mengurangi masalah stunting) juga dapat digunakan sebagai mata pencarian alternatif yang lebih bermartabat di DKI.

Abstract

Overfishing, the use of unfriendly-environmental fishing gears, changes of environmental water conditions and diversity of habitats are threaten the continuity of Bujuk fish (Channa lucius) and generate the morphologycal diversity. Consequently, appropriate conservation efforts are needed to preserve the fish. The first phase of these efforts were done through morphological approach. The research was held in 2010 in Singkarak Lake West Sumatera Province, in floodplain, Pematang sub-district Lindung Mendahara Ulu Regency East Tanjung Jabung, Province of Jambi, and in floodplain Mentulik Regency Kampar Kiri Hilir Riau Province. The aims of the research were to analyze the morphological characteristic of the fish with Principal Component Analysis (PCA) and to determine the range of similarity between populations of the fish. Determination of the location of fish sampling was conducted by proposive sampling in which there were 30 samples for every research location. The measurement of the specimen was done by digital calipers on the left of the body, at 4 meristic characters and 19 morphometric characters. The data obtained were analyzed by One Way ANOVA and principal component analysis using Statistical software version 13. The results showed that based on the analysis of morphological characteristics of bujuk fish, populations that exist in Lake Singkarak were different from those of Pematang Lindung floodplain sub-district Mendahara Ulu Regency East Tanjung Jabung Jambi Province and Mentulik Regency floodplain Kampar Kiri Hilir Province Riau. The meristic difference was on the number of scales along the lateral line with a value of 0.783. While the main difference on morphometric characters were the snout length with a value of 0.848, and length of upper jaw with a value of 0.847. Thus, the study showed that the fish was isolated geographically into two groups, which were Jambi-Riau population and Singkarak Lake population.

Abstract

The objective of this research was to analyze changes in bioavailability of zinc (Zn) caused by addition of Na2EDTA and processing of fortified wheat flour. Bioavailability of zinc was tested by in vitro method as a simulation of human digestive track. The result showed that there was no significant difference in the bioavailability of zinc on wheat flour which is processed by frying (donuts), steaming (steamed bun), and baking (bread) (p>0.05). However, there was a tendency to increase the bioavailability of wheat flour processed by frying (donuts) and baking (bread) when compared to the raw dough. Raw dough had bioavailability of zinc 13.29%, after frying (donut) increase to 14.72%, and after baking (bread) increase to 17.06%. The addition of Na2EDTA also did not increase bioavailability of zinc significantly for the three types of processing (p>0.05), despite increased bioavailability of zinc on the donuts by 4.67%.

Abstract

Genetic variability of giant gouramy strains has been observed using RAPD Marker. Genomic DNA was extracted from giant gouramy fin clip, and amplified using primers OPA 1-20. There was no significant difference among three strains of giant gouramy analyzed. Based on two primers of RAPD (OPA 4 and 7), the highest variability was observed in Bluesafir with heterogeneity value of 0.3050 and followed by Paris (0.2832) and Soang (0.2360) respectively. The average of Nei?s genetic distance is 0.118, and the smallest on is between Paris and Bluesafir.

Abstract

African catfish, namely lele dumbo, is an economically important fresh water fish in Nusa Tenggara Barat. There are three strains of African catfish that are well distributed among farmers. Genetic information of three strain catfish stocks is needed in order to sustain their development program. Genetic variations of the three strains of catfish, i.e. Sangkuriang, Masamo, Paiton has been evaluated using RAPD markers. DNA genome was extracted using phenol-chloroform methods. DNA was amplified using 20 primers (OPA 1 – 20). The results showed that three of the 20 primers have good amplification products. There was no significant genetic differences among three strains of catfish analyzed (P>0.05). Genetic variation of three African catfish stocks ranges from low to middle value. The highest genetic variation is found in strain Masamo with heterozygosity of 0.273 (with 70% polymorphism loci), followed by Sangkuriang 0.189 (60%) and Paiton0.147 (40%). Hybrid candidates of Masamo-Sangkuriang have eterozygosity ranges between 0.256 - 0.306 (with 66.6 7 – 73.77% polymorphism loci). Strain Masamo and Paiton has more close genetic relationship than among the both and Sangkuriang.

Abstract

Directional-individual selection on tilapia growth had been conducted in West Nusa Tenggara Province. The base population had been constructed through di allele crossing between six tilapia strains i.e. NIRWANA, BEST, SULTANA, Citralada, JATIMBULAN and White Sleman. A total of 150 brood stock pairs were used for spawning with the ratio of female and male parent 1: 1 in net with size of 1x1x1 m. Two hundred larvae  produced from each spawn pairs were communally reared in a pond for three months. Directional-individual selection was conducted at the size of > 50 grams. The cut-off for minimum selected fish was based on the lowest individual weight of 10% top population. The average value of heritability on body weight was 0.251 for males and 0.258 for the female population. The total value of genetic gain of body weight for four generations was 51.68% for male population and 56.78% for the female population.

Abstract

Penelitian ini dilakukan untuk mengamati variasi genetik strain Bastar, Paris dan Blusafir dengan menggunakan penanda RAPD. DNA ikan gurame diekstraksi dari sirip, dan diamplifikasi secara random dengan menggunakan primer OPA 1-20. Tidak terdapat perbedaan secara genetik antara tiga strain yang diuji. Berdasarkan dua primer RAPD (OPA 4 dan 7), variabilitas tertinggi diamati pada Bluesafir dengan nilai heterogenitas 0,3050 diikuti dengan Paris dengan nilai 0,2832 dan Bastar dengan nilai 0,2360. Jarak rata Nei genetik adalah 0,118, dengan terendah yang teramati antara Paris dan Bluesafir.

Abstract

First generation (F1) of hatchery produced humpback grouper (Cromileptes altivelis) has been characterized genetically in order to serve the information of their status in related to their breeding strategy. PCR-RFLP method was used to detect the variation of mtDNA D-loop region of F1 population at BBPBL Lampung and BBAP Situbondo. The result of study showed that reducing of haplotype diversity had been arised from broodstock (0.8548) to F1 generation population (0.7473; 0.7273; and 0.6947, respectively).  Genetic divergence that had found between population BBPBL Lampung and BBAP Situbondo make it possible to do outbreeding in order to get its heterosis's effect. Keywords: mtDNA, haplotype diversity, genetic differentiation, Cromileptes altivelis   ABSTRAK Ikan kerapu tikus (Cromileptes altivelis) generasi pertama (F1) hasil domestikasi di hatchery telah dikarakterisasi secara genetik untuk menyediakan informasi status sehubungan dengan program pemuliaannya.  Metode PCR-RFLP digunakan untuk mendeteksi variasi sekuens D-loop mtDNA ikan kerapu tikus F1 yang diproduksi di BBPBL Lampung dan BBAP Situbondo.  Hasil penelitian menunjukkan bahwa telah terjadi penurunan keragaman haplotipe dari induk (0,8548) ke populasi generasi F1 (masing-masing 0,7473; 0,7273; dan 0,6947).  Adanya keragaman genetik antara populasi ikan kerapu tikus di BBPBL dan BBAP Situbondo memungkinkan dilakukannya outbreeding untuk mendapatkan efek heterosis.
