Abstract

This research was aimed to study the influence of animal age and concentration of the acetic acidsolution on physical and chemical properties of pigskin gelatin. The experiment used CompletelyRandomized Design (CRD) with two factors. The first factor was animal age consisted of 3 levels (5, 7and 9 months). The second factor was concentration of acetic acid solution consisted of 3 levels (2, 4and 6 percents). The result showed that animal age had significant effect (P<0.01) on the yields, gelstrength, viscosity, protein content and pH value but interaction of animal age and concentration ofacetic acid had no significant effect (P>0.05) on the yields, gel strength, viscosity, protein content andpH value. It was concluded that pigskin gelatin from ages of 5, 7 and 9 months and acetic acidconcentration of 2, 4 and 6% had similar characteristics to the commercial gelatin, but the optimumproduction of gelatin was combination of pigskin gelatin from 7 months and of 2% acetic acid.

Abstract

The objective of the research was to identify the effect of the concentration of hydrochloric acid (HCl) on yield and the characteristics of gelatin from buffalo hide. A completely randomized design was used with various concentrations of HCl treatment 0.3M, 0.6M, 0.9 M, 1.2 M and 1.5M, and four times replication, followed by least significant difference test. The HCl concentration gave significantly effect (P<0.05) on yield, viscosity, pH, gel strength and redness of gelatin. The highest yield (29.17%) was obtained at a concentration of HCl 0.9M. The highest gel strength and viscosity at concentration of 1.2M, were 293.41g bloom and 22.17 cp, respectively. The pH of the gelatin were 5.08 to 5.52. The lightness level (L*) was 69.92-70.97, whereas the redness ranged (a*)  from 0.55 to 1.54 and the yellowness ranged (b*) from 17.54 to 19.59. The moisture, protein and ash concentrations of gelatin were 7.05±0.07-8.92±0.06%, protein content of 83.38±0.06 -91.11±0.03%, and the ash content 0.53 ± 0.04 to 1.23 ± 0.09%, respectively. Based on the spectra absorption of infrared were detected four areas amides: amide A (3402.43-3448.72 cm-1), amide I (1635.65 cm-1), amide II (1527.62 cm-1) and amide III (1242.16 cm-1). The differences in the concentration of HCl had impact on yield,characteristics and secondary structure of buffalo hide gelatin. The HCl concentration at 0.9M was the optimum condition to obtain the highest yield. The characteristics of buffalo hide gelatin after pretreatment using hydrochloric acid had met the standards of GMIA (2012).

Abstract

Latar belakang. Pemakaian ulang kateter penghisap telah biasa dilakukan di Indonesia, namun sejauh inibelum ada penelitian mengenai sterilitas dan keamanannya.Tujuan. Mengevaluasi sterilitas, sifat mekanik, dan permukaan serta kualitas matriks kateter penghisap yangdipakai ulang setelah diproses dengan dua jenis prosedur pengolahan yang berbeda.Metode. Kateter penghisap yang dipakai ulang setelah diproses dan disterilisasi menggunakan gas etilen oksida(EO), atau menggunakan sterilisasi pemanasan kering (kelompok B). Semua sampel dibersihkan dan didesinfeksidengan prosedur yang hampir sama. Kateter penghisap baru dipakai sebagai standar. Mikroba yang tumbuh padamedium kultur diidentifikasi. Semua sampel menjalani uji tarik dan kompresi. Analisis mikrostruktur dilakukandengan menggunakan mikroskop elektron (SEM) dan energi-dispersif spektroskopi sinar-X (EDX).Hasil. Kultur positif bakteri komensal pada 6 di antara 15 sampel pada kelompok A, dan 6 dari 17 sampelpada kelompok B. Terdapat perbedaan yang bermakna dari sifat mekanik sampel penelitian (p<0,05).Sampel dari kelompok A memiliki kekuatan yang paling rendah. Sampel dari kedua kelompok penelitianmengalami perubahan kelenturan dan keuletan dibanding standar. Analisis mikrostruktur menggunakanXPS dan EDX pada permukaan dalam ujung kateter penghisap yang dipakai ulang menunjukkan degradasikomponen matriks. Analisis SEM mendeteksi beberapa partikel tambahan dan rekahan. Analisis EDX padapartikel tambahan menunjukkan pengayaan sinyal na+ dan ca++. Secara keseluruhan, didapatkan tandakontaminasi serta kerusakan material.Kesimpulan. Kedua metode pengolahan ulang kateter penghisap memberikan hasil sterilitas yang sebanding.Sampel yang dipakai ulang mengalami penurunan kekuatan, menjadi lebih lentur, dan tidak ulet.Ditemukan tanda kontaminasi, perubahan sifat permukaan dan kerusakan matriks dari kateter penghisapyang dipakai ulang.

Abstract

It was concluded that pigskin gelatin from ages of 5, 7 and 9 months and concentration acetic acid 2, 4 and 6 % had similar physical and chemical peoperties with the commercial gelatin.

Abstract

Background Reusing endotracheal tubes (EITs) has been performed in Indonesia with no evidence of its safety. Objective To evaluate sterility, as well as the mechanical, surface, and matrix properties of reused EITs following 2 different reprocessing procedures.Methods Reused EITs were cleaned and disinfected, then sterilized by ethylene oxide gas sterilization (group A) or dry heat sterilization (group B). New EITs were used as the standard for comparison. Microbes were identified and microbial counts were determined as colony forming units (CFUs). Evaluation of mechanical properties was perfonned by a Universal Testing machine. All samples underwent tensile and compression tests.Load defonnation curves were recorded from F max and strain at F max. Microstructure analysis was done using X􀁅ray photoelectron spectroscopy (XPS), scanning electron microscopy (SEM), and energy-dispersive X-ray spectroscopy (EDX).Results Positive cultures of commensal bacteria were found in 2/12 samples in group A, and 5/17 samples in group B. T here was no statistically significant difference between them (P =0.07). Pseudomonas aeruginosa or other common pathogens were not found. Samples from both groups showed equal flaccidity, compared to the standard. Surface microstructure analysis of reused EITs With XPS and EDX showed degradation of the matrixcomponent. SEM analysis detected some large particles and fissures. EDX analysis on the large particles detected sodium and calcium signals. Altogether, signs of contamination and material damage were very strong.Conclusion Both reprocessing methods of reused EITs gave comparable results on sterility and mechanical behavior, but reprocessing may cause decreased surface and matrix quality.

Abstract

Bovine split hide is the subcutis layer with the low percentage of collagen so that the quality of the gelatin is different from that of the gelatin from cattle?s skin. This study aims to improve the characteristics of bovine split hide gelatin combined with soy protein isolate (SPI) using transglutaminase (TGase) enzyme as a protein cross-linking agent. The study was conducted using a completely randomized design with 3 x 3 factorial pattern consisting of three levels of a mixture of bovine split hide gelatin : SPI at the ratios of 90 : 10, 80 : 20, 70 : 30, and three concentrations of transglutaminase enzyme, i.e. 10, 20, and 30 U. The results showed that treatments significantly affected viscosity, gel strength, the moisture, ash, fat, and protein content of gelatin. Electrophoresis of gelatin protein showed bands distribution between 60-190 kDa. The amino acid profile of the gelatin was similar to that of collagen with a high level of aspartate, glutamate, cysteine, and proline. Morphology of gelatin was observed by Scanning Electronic Microscope (SEM) and showed a compact distributed collagen crosslink. The combination of gelatin bovine split hide and SPI at the ratio of 90 : 10 provides the best physicochemical characteristic.
