Abstract

Stichopus vastus is one of sea cucumber resources utilize which has commercial economic value, locally known  as “gametes”. Because this species is continuously exploited, while the nature of life has not been known, one of them especially its growth.  Aspect of growth is an important parameter which is used as a basis for evaluating these resources, hence this resource is well managed and can be utilized in a sustainable manner. The study aims to: (1) estimates the growth characteristics mortality rate, recruitment and potential use of sea cucumber of  S.  vastus in Karimunjawa. The study is expected to give beneficial to the species of sea cucumber resource management strategy at  Karimunjawa waters, Jepara. The analysis of the growth parameters of S. vastus obtained values ​​of growth coeficient (K) of 0.55 year-1 and length infinity L ∞ value of 315.80 mm. Therefore the obtained values ​​of K tend to be close to zero value, it indicates the nature of growth tends to slow and with a relatively long life.  Life span of  S. vastus is 5.41 years, which can achieve body lengths of 283.06 to 296.91 mm, and the monthly growth rate ranged from 9.0 to 12.37 mm. The results of the analysis of mortality showed that S. vastus has value of total mortality (Z) of 0.98,  natural mortality (M) of 0.298, and catch mortality (F) of 0.682, with the rate of exploitation (E) of 0.6963. This suggests that the rate of exploitation is above the threshold standard set by the Government of 0.5 (BRKP, 2004). The analysis of the S. vastus the recruitment showed that the higest percentage recruit occurred in May-June is 17.16 - 18.33%. This is presumably due to a population increase of the spawning process in those months, although the value of the addition is not very significant.   It is suggested the need for regulations regarding: (a) the catchment season which is based on the tendency of the reproductive patterns of S. vastus, in Karimujawa, (b) restrictions on the size of the catch, (c) catch quotas, (d) the methods that are environmentally friendly catch, (e) permits the ship / boat used to catch sea cucumbers business. To support the success of regulation, it is necessary to strengthen community institutions through the management of sea cucumber resource-based society, with the Co-management approach.

Abstract

Salah satu upaya yang dilakukan untuk mengobati serangan bakteri A. hydrophila yaitu menggunakan bahan alami seperti ekstrak daun ketapang. Penelitian ini bertujuan untuk mengetahui kemampuan ekstrak daun ketapang sebagai antibakteri A. hydrophila, mengetahui pengaruh penggunaan ekstrak daun ketapang (T. cattapa) terhadap kelulushidupan dan histologi ikan mas yang diinfeksi bakteri A. hydrophila. Metode yang digunakan dalam penelitian ini adalah metode eksperimental dengan menggunakan Rancangan Acak Lengkap dengan 4 perlakuan dan 3 kali ulangan. Perlakuan yang digunakan dalam penelitian tersebut adalah perlakuan A (0 ppm), B (500 ppm), C (1000 ppm) dan D (1500 ppm). Ikan  mas yang digunakan sebanyak 120 ekor, kemudian disuntik bakteri A. hydrophila dengan kepadatan 107 CFU/mL secara intramuskular. Perendaman ekstrak daun ketapang dilakukan pada hari ke 2 pasca penyuntikan selama 15 menit. Nilai rata-rata kelulushidupan tertinggi hingga terendah berturut-turut yaitu 66,67% (perlakuan D), 55,67% (perlakuan C), 40% (perlakuan B) dan 3,33% (perlakuan A). Hasil penelitian menunjukkan bahwa pada uji sensitivitas ekstrak daun ketapang mampu memberikan efektivitas antibakteri secara in vitro. Perendaman dengan ekstrak daun ketapang menunjukkan hasil pengaruh nyata (P<0,05) terhadap kelulushidupan ikan mas. Pengamatan gambaran histologi hati diketahui adanya nekrosis, kongesti, degenerasi vakuola dan melanomakrofag. Perendaman ekstrak daun ketapang mampu mengobati ikan mas yang diinfeksi bakteri A. hydrophila pada dosis 1500 ppm. One of the effort to heal bacterial A. hydrophila infection is using natural ingredient such as ketapan leaves. The aims of this research were to determine the ability of ketapan leaves extract as an antibacterial of A. hydrophila, the soaking  effect of ketapan leaves (T. cattapa) extract toward the survival rate and histology of Carp infected by A. hydrophila. This research used experimental method with Completely Randomized Design 4 treatments and 3 replication. These treatments were 0 ppm (treatment A), 500 ppm (treatment B), 1000 ppm (treatment C) and 1500 ppm (treatment D). The experimental Carps used were 120 fish and injected by A. hydrophila with density of 107 CFU/mL intramuscularly. The soaking of ketapan leaves extract was carriedout 15 minutes and done on the  2nd day post-infection. The results showed that the average survival rate from the highest to the lowest were 66.67% (treatment D), 55.67% (treatment C), 40% (treatment B) and 3.33% (treatment A) respectively. The result of this research showed that on antibacterial sensitivity test of ketapan leaves extract was able to make an antibacterial effect from in vitro test. The soaking of ketapan leaves extract showed a significant effect (P<0.05) toward the survival rate of Carp. The results of histological observation of the liver showed necrosis, congestion, vacoula degeneration and melanomacrofage. The soaking of ketapan leaves extract could heal Carp infected by A. hydrophila at dosage of 1500 ppm.

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh penambahan serbuk daun binahong (Anredera cardivolia) pada pakan terhadap respon imun, kelulushidupan dan status kesehatan udang windu (P. monodon) yang diinfeksi V. harveyi. Penelitian dilakukan dengan metode eksperimen mengunakan Rancangan Acak Lengkap (RAL) dengan 4 perlakuan dan 3 ulangan. Udang uji yang digunakan berukuran berat 9 g. Udang dipelihara dalam media bervolume 10 L pada akuarium yang berukuran 30 cm x 40 cm x 60 cm. Udang uji dipelihara selama 31 hari, yaitu 7 hari aklimatisasi, 14 hari pemberian pakan perlakuan dan 10 hari paska uji tantang. Uji tantang dilakukan pada hari ke 14 setelah perlakuan. Perlakuan A yaitu dengan konsentrasi binahong sebanyak 0 g/100 g pakan, perlakuan B dengan konsentrasi 4 g/100 g pakan, perlakuan C dengan konsentrasi 8 g/100 g pakan dan perlakuan D dengan konsentrasi 12 g/100 g pakan. Hasil penelitian menunjukan ada pengaruh terhadap respon imun yang ditunjukan dengan bertambahnya jumlah total hemosit (THC) dengan hasil tertinggi ditunjukan oleh perlakuan B yaitu 2.640 x 107 sel/mL pada pemeriksaan hari ke 12 (H4). Peningkatan jumlah total hemosit terbaik ditunjukan oleh perlakuan B sebelum di infeksi V. harveyi. Kelulushidupan tertinggi di tunjukan oleh perlakuan D dengan SR sebesar 83 %. Dapat disimpulkan bahwa konsentrasi terbaik adalah perlakuan B (4 g/100g). The aims of this study are to determine of addition binahong powder A. cardivolia in feed to the immune response, health status and survival rate of tiger prawn (P. monodon) that were infected by V. harveyi. The study was conducted by using the experimental method with completely randomized design (CRD), 4 treatments and 3 replications. The verage weighth of experimental shrimp was 9 g. Shrimp was maintained in aquarium 30 cm x 40 cm x 60 cm with 10 L media with stoking density 8 shrimp per aquarium. Cultivation period was 31 days with 7 day acclimatization, 14 days treatment and 10 days after challenge by V. harveyi. Challenge test performed on day 14 after treatment. The A. cardivolia consentration in each treatment were (A) 0 g/100 g of feed, (B) 4 g/100 g of feed, (C) 8 g/100 g of feed and (D) 12 g /100 g of feed. The results showed that there was an effect of the treatment on immune response that it shown by increasing total hemocytes count number (THC) with highest result shown in treatment B, 2,640.107 cell/mL on 12th(H4) day examination and the higest result in survival was shown in treatment D with 83% survival rate. It can be concluded that the best treatment is B (4 g/100 g).

Abstract

Parasit Octolasmis sp. adalah jenis parasit yang menginfeksi kepiting bakau dan menempel pada insang. Parasit ini menyebabkan kerusakan pada organ tubuh dan terganggunya pertumbuhan serta menurunkan sistem pertahanan tubuh kepiting. Salah satu upaya yang dilakukan untuk mengobati serangan parasit Octolasmis sp. yaitu menggunakan bahan alami seperti ekstrak daun sirih. Penelitian ini bertujuan untuk mengetahui pengaruh perendaman ekstrak daun sirih terhadap kelulushidupan dan penurunan infestasi parasit Octolasmis sp. serta kandungan senyawa aktif daun sirih. Metode penelitian yang digunakan adalah metode eksperimental dengan 4 perlakuan dan 3 kali ulangan. Dosis ekstrak daun sirih yang digunakan pada penelitian ini adalah A (0 ml), B (3 ml/L), C (6 ml/L), dan D (9 ml/L). Kepiting bakau yang digunakan sebanyak 36 ekor, dan dipelihara dalam 9 akuarium dengan kepadatan 4 ekor/akuarium. Rata-rata kelulushidupan kepiting bakau yaitu 88,89±0,19 (perlakuan B), 77,78±0,38 (perlakuan C), 66,67±0,42 (perlakuan D), 44,45±0,62 (perlakuan A). Hasil penelitian menunjukkan bahwa prevalensi Octolasmis sp. menurun karena perendaman ekstrak daun sirih; kepiting bakau menujukkan respon pasif dan pasca perendaman dari ekstrak daun sirih tidak memiliki pengaruh yang nyata (P<0,05) terhadap kelangsungan hidup kepiting yang terinfeksi Octolasmis sp. namun ekstrak daun sirih menyebabkan tingkat kelulushidupan tinggi pada kepiting bakau yang terinfeksi. Parasite Octolasmis sp. is a type of parasite which infects mud crabs and stick to the gills. This parasite causes damage to the gills body organs decrease of growth and defense system of crabs. An attempt was made to treat the parasitic  Octolasmis sp. infected crab by using natural ingredients namely betel leaf (Piper betle linn) extract. This study aims to determine the effect of soaking of betel leaf extract to survival  and decrease parasitic infestation of Octolasmis sp. and the content of active compound of betel leaf. Experimental research of varian betel leaf extract use caused out in consentration of 4  treatments and 3 replications. The dose of betel leaf extract used in this study was A (0 ml), B (3 ml/L), C (6 ml/L), and D (9 ml/L). The mud crabs used with a totally 36 tails, and maintained in 9 aquariums with a density of 4 tails / aquarium. The mean survival rate of mangrove crabs was 88.89 ± 0.19 (treatment B), 77.78 ± 0.38 (treatment C), 66.67 ± 0.42 (treatment D), 44.45 ± 0.62 (Treatment A). The result experimental that prevalence of Octolasmis sp. declined along with the incause of betel leaf extract concentration; experimental crabs exhibited passive food response and in active past treatment for several insure soaking of betel leaf extract also didn’t have significant effect (P <0,05) to survival of mud crabs (Scylla serrata) infected by Octolasmis sp. However the betel leaf extract was able incause survival rate of infected mud crab.

Abstract

Permintaan akan ikan budidaya terutama ikan mas (Cyprinus carpio) yang semakin tinggi, masyarakat menerapkan sistem budidaya intensif bahkan super intensif. Kondisi ini tentunya akan menimbulkan kendala, salah satunya meningkatnya peluang terserangnya penyakit pada beberapa ikan. Upaya pengendalian penyakit bakterial pada budidaya ikan, sampai saat ini masih mengggunakan bahan kimia. Tujuan penelitian ini adalah untuk mengetahui sensitivitas isolat bakteri terhadap obat beredar serta bakteri yang menginfeksi ikan mas (C. carpio) Isolasi bakteri dilakukan pada 20 ekor ikan mas yang berasal dari Kedung Ombo dan Magelang dengan organ target hati, ginjal, dan luka pada media GSP. Uji sensitivitas dilakukan dengan mengkultur bakteri dengan kepadatan 108 CFU/mL ditebarkan di media TSA dengan jumlah 100 µl pada petridish dan diratakan menggunakan L glass kemudian didiamkan agar bakteri uji meresap ke dalam media. Paper disk steril diletakkan di atas media TSA yang sebelumnya telah direndam di dalam obat uji. Dosis obat yang digunakan sesuai yang dianjurkan dalam kemasan. Hasil isolasi diperoleh 29 isolat, kemudian dilakukan uji sensitivitas terhadap obat uji. Hasil penelitian menunjukkan bahwa semua isolat bakteri terhadap obat beredar A, B, C, dan D bersifat resistance, kecuali SN3, SN5, SN6, SN8, SN9, SN11, SN14, SN16, dan SN23 intermediate terhadap obat D. Karakterisasi bakteri dilakukan terhadap isolat bakteri terpilih (SN6, SN11, SN19, SN24, dan SN26) secara morfologi yang selanjutnya di uji biokimia dengan hasil Pseudomonas putida (SN6), P. anguilliseptica (SN11), Aeromonas sobria (SN19), A. caviae (SN24), dan P. pseudoalcaligenes (SN26). Common carp (Cyprinus carpio) is one of aquaculture product that have economic value. Intensive and super intensive system have improved carp production. However, this condition  caused disease problems in fishes. The Efforts to control bacterial diseases in fish culture, at the moment still rely on chemical treatments. The aims of this research were to determine the susceptibility of the bacterial isolation to the different marketed drugs. Bacteria were isolated from  20 common carp sample from Kedung Ombo and Magelang and obtained from liver, kidney, and fish wound growth in the GSP media. In vitro study using diffusion susceptibility test was apllied at bacterial density 108 CFU/mL in100 µL  TSA.  The bacteria was spread by L glass evenly prior to the disk placement that previously being soaked in the various drugs test. The bacterial isolation gained 29 isolates. Further test demonstrated that various marketed drugs (A, B, C, and D) were resistance to isolated bacteria except isolates SN3, SN5, SN6, SN8, SN9, SN11, SN14, SN16, and SN23 were intermediates to drug D. Identification of isolates (SN6, SN11, SN19, SN24, and SN26) revealed 5 species namely Pseudomonas putida (SN6), P. anguilliseptica (SN11), Aeromonas sobria (SN19), A. caviae (SN24), and P. pseudoalcaligenes (SN26).

Abstract

Aeromonas hydrophila merupakan bakteri opportunis yang sering menyebabkan penyakit pada ikan air tawar, salah satunya pada ikan tawes. Ekstrak buah mengkudu mengandung senyawa antibakteri yang dapat digunakan sebagai alternatif pengobatan penyakit yang disebabkan oleh  A. hydrophila pada ikan tawes. Penelitian ini bertujuan untuk mengetahui pengaruh dari perendaman ikan tawes yang diinfeksi A. hydrophila dengan ekstrak buah mengkudu. Ikan tawes yang digunakan sebanyak 120 ekor dengan panjang rata-rata 7,23±0,98 cm/ekor yang diinfeksi bakteri A. hydrophila sebanyak 0,1 mL secara intramuscular dengan kepadatan bakteri 106 CFU/mL. Hewan uji dilakukan pengobatan dengan perlakuan yang menggunakan Rancangan Acak Lengkap (RAL). Perlakuan tersebut yaitu perlakuan A (0 mg/L), B (15.000 mg/L), C (30.000 mg/L) dan D (45.000 mg/L) dan tiap perlakuan dilakukan pengulangan sebanyak 3 kali. Perendaman menggunakan ekstrak buah mengkudu dengan metode short bath selama 20 menit dilakukan setelah gejala klinis dari A. hydrophila telah muncul. Hasil penelitian menunjukkan perendaman ekstrak buah mengkudu berpengaruh nyata (P<0,05) terhadap gejala klinis dan kelulushidupan ikan tawes. Gejala klinis pasca penyuntikan A. hydrophila yang menunjukkan adanya warna tubuh memudar, opacity, haemorage pada permukaan tubuh serta tukak dan mulai mengalami pemulihan pasca perendaman ekstrak buah mengkudu yaitu berenang aktif, haemorage pada tubuh memudar, tukak pada tubuh mulai menutup. Kelulushidupan terbaik (70%) terlihat pada dosis 15.000 mg/L dibandingkan dengan perlakuan lainnya yang masing-masing yaitu 30.000 mg/L (53,33%), 45.000 mg/L (50%) dan 0 mg/L (30%). Konsentrasi ekstrak buah mengkudu 14.000 mg/L merupakan konsentrasi optimal untuk mengobati ikan tawes yang diinfeksi bakteri A. hydrophila. Aeromonas hydrophila is an opportunistic bacterial that often causes disease in freshwater fish, and one of them is on the java barb. Noni fruit extract contains antibacterial compounds that can be used an alternative treatment for diseases caused by A. hydrophila in the java barb. This study were aims to determine the immersed effect of noni fruit extract  to combat A. hydrophila. Java barb used were 120 fish with average length 7,23 ± 0,98 cm/fish infected with A. hydrophila as much as 0,1 mL intramuscularly with density of 106 CFU / mL. The experimental animals were treated in the treatment by using Completely Randomized Design (CRD). The treatments A (0 mg / L), B (15,000 mg / L), C (30,000 mg / L) and D (45,000 mg / L) and each treatment was replicated 3 times. Immersed with noni fruit extract was carried on by short bath method for 20 minutes immersed was done when clinical signs of A. hydrophila appeared. The results demonstrated that the used of noni fruit extract exhibited real effect (P<0.05) againts clinical symptoms and survival rate of java barb. Clinical symptoms in post injection of A. hydrophila that indicate the existence of a body color fades,  opacity, haemorage on the surface of the body also the ulcer and began to suffer submergence post crash noni fruit extracts namely active swim, body haemorage is faded, ulcers on the body begins to shut down. The results also demonstrated the best survival rate (70%) at concentration of 15,000 mg/L compared with the other treatments which each 30,000 mg/L (53.33%), 45,000 mg/L (50%) and 0 mg/L (30%). Concentration 14.000 mg/L of noni fruit extract is optimal concentration to treat java barb infected bacteria A. hydrophila.

Abstract

The study aimed was to know the influence of Piper crocatum exstract on the blood profile included total erythrocyte, total leukocyte, differential leukocyte (DL), phagocytosis indeks (IF) and survival rate of common carp (Cyprinus carpio) infected by Aeromonas hydrophila and the best concentration of P. crocatum for imunostimulatory common carp of infected A. hydrophila. The research was conducted at Research and Development of Freshwater Aquaculture, Bogor. The methods used in this research is experimental methods by using random design complete (RAL) with 4 treatments and three replicates. The research was conducted at Research and Development of Freshwater Aquaculture, Bogor. The treatments of different addition of P. crocatum exstract on fish feed, 0 gr/kg feed (treatment A), 5 gr/kg feed (treatment B), 10 gr/kg feed (treatment C), 15 gr/kg feed (treatment D). The treatnent was given 21 days. The research results obtained additional P. crocatum exstract significantly different effect (P0,05) on percentage of netrofil (2%) and survival rate of common carp (C. carpio) infected by A. hydrophila (40%), the best concentration of P. crocatum on feed for immunostimulant common carp of infected A. hydrophila, the result showed the best dose 5 g/kg feed

Abstract

Penelitian ini bertujuan untuk mengkaji pengaruh penambahan serbuk lidah buaya (Aloe vera) dalam pakan terhadap kelulushidupan dan profil darah ikan mas pasca infeksi bakteri Aeromonas hydrophila serta mengetahui penambahan dosis serbuk lidah buaya yang terbaik. Ikan uji yang digunakan adalah ikan mas (Cyprinus carpio) berukuran 8,55±0,71 cm dan bobot 10,18±2,24 g. Metode penelitian ini adalah eksperimen dengan 4 perlakuan dan 3 ulangan yaitu perlakuan A (0 g/kg pakan), B (30 g/kg pakan), C (60 g/kg pakan) dan D (90 g/kg pakan). Pemberian pakan selama 14 hari sebelum uji tantang. Uji tantang dilakukan dengan menyuntikkan suspensi A. hydrophila dengan dosis 107 sel/mm3 sebanyak 0,1 mL secara intramuscular. Pengamatan dilakukan selama 10 hari pasca infeksi A. hydrophila, pengamatan tersebut meliputi gejala klinis, kelulushidupan dan profil darah. Hasil penelitian menunjukkan bahwa gejala klinis ikan pasca infeksi diantaranya berenang tidak normal (lamban atau vertikal), respon makan menurun, produksi lendir berlebih, peradangan, timbul ulcer, sirip punggung geripis, sisik lepas, daging rusak. Total eritrosit hari ke-4 pasca infeksi mengalami penurunan disemua perlakuan. Total leukosit ikan mas pada hari ke-1 pasca infeksi semakin meningkat. Kadar hematokrit pasca infeksi pada perlakuan B, C dan D lebih tinggi dibandingkan dengan perlakuan A. Kesimpulan yang diperoleh adalah penambahan serbuk lidah buaya memberikan pengaruh sangat nyata terhadap kelulushidupan ikan mas pasca infeksi bakteri, penambahan serbuk lidah buaya dalam pakan dapat meningkatkan nilai total leukosit, eritrosit dan hematokrit lebih banyak dibandingkan dengan ikan yang tidak diberi pakan dengan tambahan serbuk lidah buaya dan dosis serbuk lidah buaya 30 g/kg pakan merupakan dosis terbaik. The aims of this research was to determine the effect of Aloe vera powder in the feed to againts survival rate and blood profile of carp that infected by Aeromonas hydrophila  and to know the addition of A. vera powder dose is best. The fish samples used were carp (C. carpio) with 8.55 ± 0.71 cm lenght and the average weight are 10.18 ± 2.24 g. The method in this research was experimental with 4 treatments and 3 replications. The treatment were A (0 g/kg of feed), B (30 g/kg of feed), C (60 g/kg of feed) and D (90 g/kg feed) and feeds for 14 days. Challenge test was done by injecting a suspension of A.hydrophila at a dose of 107 cells/mm3 as much as 0.1 mL intramuscularly. The observations were done at 10th days post-infection by A. hydrophila, these observations clinical symptoms, survival and include blood profile. The results showed clinical sign experienced after infected  that fishes were swimming in a slant condition (whirling), decreased feed response, inflammation, ulcers arise,necrosis, and scales loose. The results showed that the total erythrocytes at  4th days decrease in all treatments. Total leukocyte carp at the 1st day increased. Total hematocrit in treatment, B, C and D post-infection were higher than treatment A. The conclusion of this research highly significant effect (P > 0.05) to survival rate carp infected by  A. hydrophila and A. vera powder addition in feed able to increase the total erythrocytes, leukoist and total value compared with more hematokrit fish not given feed with the addition of A. vera powder and were dose of A. vera powder 30 g/kg feed gave a better survival rate of carp infected by bacteria A. hydrophila.

Abstract

ABSTRAK Kendala utama budidaya ikan kerapu adalah tingginya tingkat kematian terutama pada benih sebagai akibat serangan bakteri dan patogen. Penelitian ini bertujuan untuk mengkaji pengaruh penggunaan ekstrak daun jeruju (Acanthus ilicifolius) terhadap kelangsungan hidup ikan kerapu macan  (Epinephelus fuscogutattus) yang diinfeksi Vibrio alginolyticus. Ikan kerapu macan (E. fuscogutattus) sebanyak 120 ekor diberikan perlakuan selama 40 hari dengan pemberian pakan yang ditambahkan ekstrak daun jeruju (A. ilicifolius). Uji tantang dilakukan dengan menyuntikan suspensi bakteri V. alginolyticus secara intramuskular. Prosedur pengamatan dilakukan selama 10 hari pascainfeksi meliputi gejala klinis, kelulushidupan serta gambaran darah ikan kerapu macan. Hasil penelitian menunjukkan gejala klinis ikan sakit diantaranya bergerak lamban, berenang tidak teratur (erratic swimming), berenang di permukaan, warna tubuh ikan menjadi gelap, timbulnya luka di bagian punggung serta geripis pada bagian sirip ikan dan pembengkakan pada bola mata (exopthalmia). Pemberian ekstrak daun A. ilicifolius pada pakan menunjukkan hasil tidak berpengaruh nyata (P>0,05) terhadap kelulushidupan dan jumlah total eritrosit ikan kerapu macan pascainfeksi, akan tetapi penambahan ekstrak daun A. ilicifolius menunjukkan hasil berpengaruh nyata (P<0,05) pada hari ke-8 dengan nilai tertinggi 9,39x104 sel/mm3 serta berpengaruh sangat nyata (P<0,01) terhadap aktifitas fagositik hari ke-4 dengan nilai tertinggi 65%. Dosis terbaik dalam penelitian ini adalah penambahan ekstrak daun A. ilicifolius sebanyak 5 gr/kg pakan. ABSTRACT This research was aimed to find out the effect of using leaf extract Acanthus ilicifolius toward survival rate of tiger grouper (Epinephelus  fuscogutattus) infected by Vibrio alginolyticus. 120 fish E. fuscogutattus was treated 40 days with the added leaf extract A. ilicifolius in the feeding process. The test was done by injecting of bacterial suspensions V. alginolyticus in the intra-muscular of the fish. Observation procedure was performed for 10 days after the infection such as clinical symptoms, survival rate and blood picture of the fish. The results showed clinical symptoms of sick fishes i.e. abnormally swiming activity, erratic swimming, swimming on the surface, dark colouration of the skin, a wound on the back and fins, and exopthalmia. The test results showed that extract of A. ilicifolius leaf not significantly different (P>0.05) on survival rate and total erythrocyte of E. fuscogutattus post-infection, but the addition of leaf extracts A. ilicifolius indicate results significantly different (P<0.05) on day 8 with the highest score of 9.39 x104 cells/mm3 and differed significantly (P<0.01) on  phagocytic activity day 4 with a highest score of 65%. The best dosage in this research was 5 gr/kg of feed.

Abstract

Gurami (O. gouramy) adalah salah satu jenis ikan air tawar yang dipilih pembudidaya untuk dipelihara. Namun kendala yang sering dialami dalam membudidayakan kultivan ini adalah serangan penyakit yang disebabkan oleh bakteri E. tarda. Penggunaan antibiotik dalam jangka panjang dapat berdampak negatif, diantaranya dapat menimbulkan resisten terhadap bakteri dan dapat mencemari lingkungan. Penggunaan bahan alami mulai berkembang untuk pengobatan ikan yang terserang penyakit, salah satu bahan alami yang digunakan yakni ekstrak jahe merah (Z.officinale var. Rubrum). Penelitian ini bertujuan untuk mengetahui gejala klinis, pertumbuhan, serta pengaruh perendaman ekstak jahe merah sebagai terhadap kelulushidupan ikan gurami yang diinfeksi bakteri E. tarda. Ikan gurami yang digunakan sebanyak 120 ekor dengan ukuran 7 - 9 cm dan diinfeksi bakteri E. tarda dengan kepadatan 108 CFU/ml secara intramuskular. Perendaman ekstrak jahe merah dilakukan pada hari ke 3 pasca infeksi setelah muncul gejala klinis seperti timbulnya luka (ulcer), pendarahan (haemorhagic), dropsy dan geripis pada ekor, sirip dada dan punggung. Perendaman ekstrak jahe merah berpengaruh nyata (P<0,05) terhadap kelulushidupan ikan gurami yang diinfeksi bakteri E. tarda. Nilai rata – rata kelulushidupan terendah hingga tertinggi berturut – turut yaitu 3,33% (perlakuan A), 26,67% (perlakuan B), 40,00% (perlakuan C) dan 56,67% (perlakuan D). Gourami (O. gouramy) is one of freshwater fish that has choosen as cultured fish by farmers. However, there are problems in culturing this cultivan, one of them is disease caused by E. tarda. Long-term use of antibiotics have a negative impact, which could cause resistance to bacteria and contaminate environment. Red ginger extract (Z. officinale var. Rubrum) as natural ingredients could be used as alternative treatment for this disease. This research aimed to determine clinical sign, growth rate and effect of short bathing of red ginger extract on survival rate of gourami that were infected with E. tarda bacteria. Gourami used were 120 fish with 7 - 9 cm in size and infected with E. tarda with density of 108CFU/ml intramuscularly. Immersion in red ginger extract was conducted on 3rd day post infection after the clinical sign was showed such as ulcer, haemorhage, dropsy, and erosion at caudal, pectoral and dorsal fin. Short bathing in red ginger extract significantly (P <0.05) affected the survival rate of gourami were infected with E. tarda but did not affect growth rate. Average values of the survival rate respectively are 3,33% (treatment A), 26.67% (treatment B), 40.00% (treatment C) and 56.67% (treatment D).
