Abstract

Ekstrak hidroalkoholik kunyit direkomendasikan sebagai suplemen untuk prevensi atherosklerosis. Namun,penggunaannya dalam pengobatan tradisional dirasa kurang praktis dan kurang acceptable, sehingga perlu dibuat sediaan tablet hisap. Tujuan penelitian ini adalah untuk mengetahui pengaruh variasi kombinasi bahan pengisi manitol-amilum manihot terhadap sifat fisik granul, tablet dan rasa tablet.Formula optimum diperoleh dengan menggunakan metode simplex lattice design. Ekstrak air dibuat menggunakan penyari berupa akuades suhu 80OC sedangkan ekstrak alkohol menggunakan penyari etanol 96%. Kedua ekstrak dicampur  sehingga diperoleh ekstrak hidroalkoholik kunyit, kemudian digranul dengan metode granulasi basah, dibuat menjadi lima formula dengan kombinasi bahan pengisi manitol-amilum manihot. Hasil penelitian menunjukkan bahwa kombinasi bahan pengisi manitol-amilum manihot mampu mempengaruhi kekerasan tablet, waktu larut dan penerimaan rasa. Formula optimum yang didapat adalah campuran bahan pengisi manitol-amilum manihot dengan perbandingan 80% : 20%.

Abstract

Endophytic fungus IP-2 is the endophytic fungus isolated from the plant Artemisia annua L. This fungus is known to have activity as an inhibitor of heme polymerization. The aim of this study is to examine the suitable medium for the growth and production of the active metabolite and to determine the appropriate time for harvesting the active metabolite. Endophytic fungus was fermented in 4 types of media for 14 days. Sampling was taken every day to obtain the weight of the dry cells which were used for analysis of growth. The media obtained is used to determine their activities as heme polymerization inhibitor. Tests were carried out using the method developed by Bassilico et al (1998). Data of dry cell weight and the percentage inhibition of heme polymerization were analyzed in order to obtain the curve of growth and metabolite production. Both curves will be used to determine the proper time to harvest the active metabolite. The results showed that the PDB was the best medium for endophytic fungus IP-2 growth and was able to produce the active metabolite higher than most other media. In the PDB media, at day-7 showed that active metabolite produced at most so that proper harvest time metabolite is on the 7th day of fermentation.

Abstract

The hull of delima fruits (Punica granatum L.) are traditionally used to cure dysmenorhoe. Preliminary study indicated that extract of the hull was able to inhibit the growth of Candida albicans. Therefore this study was aimed to isolate and identify active compounds responsible for the activity from the hull of P. granatum. The study was initiated by extracting the powdered material with petroleum ether followed by methanol. Antifungal activity test (100 mg/ml) indicated that the petroleum ether extract was more active than the methanol extract (inhibition zone: 10.59 vs 6.92 mm). The pet. ether extract was triturated by n-hexane to give n-hexane insoluble and n-hexane soluble fractions. The latter that was active (inh. zone: 9.50 vs 0.00 mm) was fractionated by vacuum liquid colomn chromatography (vlc; SiO2, n-hexane with increasing amount of ethylacetate) to give 7 fractions (F1-F7). Fraction 2 (inh. zone: 9.05 mm) and 3 (11.05 mm) displayed antifungal activity, then F3 was subjected to contact bioautography to give 2 active compounds [Rf. 0.50 (major) and 0.10 (minor)]. Preparative tlc [SiO2 F-254 nm; nhexane: ethylasetate 4-1, developed 2x) of F3 was aimed to separate 2 active compounds. Due to limited amount of the minor compound, the MFC was applied only for the major compound (Rf. 0.50; 200 mg/ml). The structure identification was done by mean of spectroscopic methods (uv, ir, ms and nmr) to be a setrol type of compound having stigmastane skeleton, esterified by a long-chain fatty acid.Keywords: Punica granatum L., Candida albicans, active compound, terpenoid

Abstract

Anti-fungal activity of essential oil distilled from exctracts of Piper betle leaves against Candida albicans and the gas chromatography profiles has been studied. The leaves were collected from Kaliurang, Kulonprogo, and Gunung Kidul. This research was aimed to find out the influences of extract production process and the difference of starting material source regions to anti-fungi activities against Candida albicans. The gas chromatographic profile of essential oils distilled from sirih (Piper betle L.) extracts were also observed. The essential oils used in this research was collected from ethanol 95% extract of sirih dried leaves by distillation method. Anti-fungi activity was assayed using Sabaroud agar-diffusion method. The results showed that the essential oil from Kulonprogo has the highest activity. The gas chromatographic profile of each essential oil had been examined individually and the results showed that the composition of substances containing in each extracts were different. The study comparison of essential oils composition of Piper betle L. fresh herbs showed that the extraction method reduced the anti-candida potency of Piper betle L. leaves extract. The habitat of the starting materials influences the gas chromatographic profile of the essential oil.Key words: Piper betle Leaves Extract; anti-fungi, chromatographic profiles

Abstract

A research has been done to determine the toxicity of ethanolic extract of fruit, seed and leaves of makutadewa (Phaleria macrocarpa (Scheff) Boerl.) using Brine Shrimp Lethality Test method. Extract were made by maceration of the materials for 24 hours. The bioassay has been done using 48 hour old Artemia saline Leach. The effect of ethanolic extract was identified by determining and analysing the shrimp death percentage and LC50 of each extract using probit analysis.The result showed that these ethanolic extract were toxic to the shrimp. The ethanolic extract of seed was the most active with the LC50 of 1.60 x 102 g/ml, whereas that of the fruit has the LC50 of 30.42 g/ml. The thin layer chromatogram profile of the active extract suggested that the extract contains alkaloids.Key words : toxicity, makutadewa, brine shrimp lethality test

Abstract

Hydroalcoholic extract of turmeric (Curcuma longa L.) doses of 1.66 mg/kg containing 10% of curcuminoid was recommended as a supplement for prevention of atherosclerosis. This study aimed to formulate hydroalcoholic extract of turmeric into chewable which are more acceptable and practical. Mannitol-Lactose selected as a filler to make the chewable in this study. Optimum composition of filler of the mixture obtained by the Simplex Lattice Design method.Identification of turmeric rhizome has been done at Departemen of Plant Taxonomy, Faculty of Biology, Gadjah Mada University, Yogyakarta, Indonesia. The rhizome extraction carried out as re to research carried out by Quiles. The mixture between curcuminoid, ethanol and water extract made up to 25% level of curcuminoid level. Then, Extract was granulated with combination of filler mannitol-lactose to be five formulas, there are: FI (mannitol 100%), FII (mannitol-lactose 75%:25%), FIII (mannitol-lactose 50%:50%), FIV (mannitol-lactose 25%:75%), FV (lactose 100%). The granules then tested their physical properties such as flowability, compactibility, and water content. Furthermore, the granules tableted and physical tested including : weight uniformity, hardness, friability, absorption, flavor acceptance and soluble time. Determining the optimum formulation obtained from the physical properties of the granules and chewable physical properties based on SLD methods. Optimum formula of chewable then tested with kurkuminoid levels. The physical properties data of granules and chewable were analyzed statistically using student?s t-test.The result of this study was the optimum formula obtained from the mixture of mannitol-lactose fillers with a ratio of 90% mannitol: 10% lactose. The optimum formula of turmeric extract chewable produced meets the physical properties of the granules and chewable. Acceptance of flavor chewable optimum formula appreciation in the range of sweet and sour taste to sour. Percentage kurkuminoid levels in chewable after manufacture process was 76.42%.
