Abstract

Plant callus extracts are potential to be developed as ingredient in skincare products. Tomato callus is supposed to contain protein-derivatives and or other components such as secondary metabolites that play a role in skin regeneration. Therefore the production of calli is important to be studied for callus sustainable supply. This research aims to obtain optimum medium for callus induction and to analyze tomato callus development anatomically. In vitro culture response was assessed in tomato plant (Solanum lycopersicum L. ?Permata?) for optimum callus induction. Seeds were grown on ¼ MS medium for 10-15 days. Hypocotyl was excised and cultured on MS medium + 2 mg/l 2,4-D for 15 days as the explants for callus induction. Callus was transferred to MS medium with 8 variations of PGRs including the combination of BAP + NAA, and 2,4-D. Both fresh and dry weight was measured every 5 days over 60 days to establish the growth kinetics and growth efficiency of callus. Anatomic characters of calli were examined through paraffin-embedded method. The result showed of MS medium supplemented with 2.0 mg/l NAA and 0.2 mg/l BAP is optimum for tomato callus induction, based on highest number of the absolute growth rate on fresh weight (73.77% per day), dry weight (3.84% per day), and callus initiation time (5.56 days) achieved by the medium. Cells in the ground tissue of tomato hypocotyl are competent to be dedifferentiated into a callus. This research results were expected to find out suitable methods for tomato callus production in preparation for skincare uses.

Abstract

Abstrac : The results showed that students' initial observations akelas VII Civics have interest in studying the low category with a percentage of 37,63%. However, in the first cycle increased after given action in the form of internet usage. Interests of learners in the first cycle reaches a percentage of 57,63% with moderate category. In the first cycle is already noticeable activity shown by learners. For example PPKn accessing material through the search engines and also collect assignments via email. Learners've listened to every task and a good teacher's explanation regarding the use of the internet as well as a description of the material PPKn.Pada second cycle not only the use of email and also www alone in learning activities but also introduced Mesia PPKn chat and discussion forums. The existence of these types of services, students who can not be present in the classroom can still follow the activities and also collect assignments via email. Learners provide comments on the discussion forum and also perform FAQ using online chat. The use of Internet technology is able to increase the interest of learners. In this second cycle students' learning interest of class VII A SMPN 2 Bawen on learning PPPKn reach a percentage of 81,08% in the high category.Abstrak : Hasil penelitian menunjukan bahwa Observasi awal siswa akelas VII PPKN memiliki minat belajar kategori rendah dengan presentase sebesar 37,63%. Namun pada siklus I mengalami peningkatan setelah diberikan tindakan berupa penggunaan internet. Minat belajar peserta didik pada siklus I mencapai persentase sebesar 57,63% dengan kategori sedang. Pada siklus I ini sudah mulai terlihat aktivitas yang ditunjukkan oleh peserta didik. Sebagai contoh mengakses materi PPKn melalui search engine dan juga mengumpulkan tugas melalui email. Peserta didik sudah mendengarkan setiap tugas dan penjelasan guru baik mengenai penggunaan internet maupun penjelasan mengenai materi PPKn. Pada siklus II tidak hanya penggunaan email dan juga www saja dalam kegiatan pembelajaran PPKn namun juga dikenalkan mesia chating dan juga forum diskusi. Adanya jenis layanan tersebut, peserta didik yang tidak dapat hadir di dalam kelas tetap bisa mengikuti kegiatan dan juga mengumpulkan tugas melalui email. Peserta didik memberikan komen di dalam forum diskusi dan juga melakukan Tanya jawab melalui fasilitas chat. Penggunaan teknologi internet ini mampu meningkatkan minat belajar peserta didik. Pada siklus II ini minat belajar peserta didik kelas VII A SMPN 2 Bawen pada pembelajaran PPPKn mencapai persentase sebesar 81,08% dalam kategori tinggi.Kata Kunci : Minat Belajar, peserta didik, teknologi internet, PPKN

Abstract

Recently, the research to find alternative sources of carbohydrates as a replacement for rice has been developed. Walur is one of the carbohydrate sources that can be explored because it can be grown in any area with out special treatment. However, walur has limitation for direct consumption, because it contains calcium oxalate. The purposes of this study were to determine the chemical character (macronutrient content), calculate glycemic index and get the proper washing techniques to elimin ate calcium oxalate of walur. Macro nutrients content studied in this research include carbohydrates, fats, protein, star chand crude fiber. Analysis of macronutrients has been chemically done, while the glycemic index was measured by in vivo using glucose as a standard. Elimination of calcium oxalate was conducted by washing the fresh walur tubers using a solution of0.01NHCl-NaOH, 5% NaCl, and solution of lemon-lime. The content of oxalate before and after washing was analyzed by permanganometry method. The results showed that walur containing 4.34 ±0.07% of reducingsugar,3.24 ± 0.06 % of not-reducing sugar, 11.27±0.40 % of crude fiber,0.03±1.05 % of starchand0.57±0.01 % of protein. Qualitative analysis of fatty acids showed that hexade canoicacid, octade cadienoicacid, and the acide icosatetranoic were detected in high concentrations. The glycemic index valueof walur was relative lylow, about of 16.9. In addition, washing technique using a solution of lemon-lime was the most excellent technique and can reduce the oxalate content up to61.82%.Fromthis research, it can be concluded that walur can be used as food substitute esrice after washing treatment using lemon-lime solution to remove the calcium oxalate content.

Abstract

Indonesia kaya akan umbi lokal yang semakin hari semakin dilupakan oleh generasi sekarang sebagai sumber bahan pangan yang sehat. Seiring dengan makin meningkatnya penyakit degeneratif terkait tingginya asupan glukosa, perlu digali kembali kekayaan lokal umbi-umbian dan pembuktian kandungan karbohidrat sehat di dalamnya yang dapat dimanfaatkan sebagai pangan di masa depan. Penelitian ini bertujuan untuk mengetahui indek glikemik (IG)  umbi-umbian yang diberikan secara oral pada tikus. Umbi yang diteliti antara lain, Uwi (Dioscorea alata), Gadung (Dioscorea hispida) dan Talas (Colocasia esculenta). Tikus dikelompokkan menjadi 9 kelompok masing-masing terdiri 4 ekor. Umbi diberikan dalam bentuk serbuk yang disuspensikan ke dalam Na CMC 0,1%  dengan dosis 2,5 g/kg berat badan (BB), kontrol positif berupa glukosa pemberian oral dosis 2,5 g/kg BB. Dilakukan pengambilan sampel darah hewan uji pada jam ke 0 (sebelum pemberian senyawa uji atau kontrol), 0,5; 1 dan jam ke-2 setelah pemberian senyawa uji.  Serum darah selanjutnya ditetapkan kadar glukosanya menggunakan GOD-PAP kit. Data berupa kadar glukosa darah hewan uji versus waktu dianalisis menjadi Area Under Curve (AUC) kadar glukosa versus waktu (0-2 jam). Perhitungan IG dilakukan dengan membandingkan AUC (0-2jam) senyawa uji terhadap AUC (0-2jam) glukosa. Hasil penelitian menunjukkan bahwa ketiga umbi yang diteliti memiliki nilai IG rendah (14-22). Umbi lokal dapat dikembangkan sebagai sumber karbohidrat yang sehat.

Abstract

God only created two types of people, men, and women. There is no third type. And the existence of both of them is to complement each other. So to unite the two God formed an institution called family. That's where they will unite as husband and wife. They have their respective roles. The existence of a wife is a helper for her husband. And this is where a wife of noble character is needed to be a helper. What is a wife of noble character? Through the exposition of the book of Proverbs 31: 10-31 it will answer. A wife of noble character is a wife who has all the parts of a respectable character, and an extraordinary power to carry out all her duties and obligations, especially in her household, she is able to make a decision that can be useful for herself and her entire family.

Abstract

Indonesia has a wide range of rice cultivars and pigments. This rice can be used as a source of phytochemical compounds for cancer prevention. This research aims to analyze the cytotoxic activities of the ethanolic extract of black rice bran of 4 local cultivars i.e. ‘Cempo Ireng’, ‘Woja Laka’, ‘Toraja’ and ‘IR­64’ (white rice) on  cancer cells and to determine the compounds groups of those extracts. First step, rice bran was extracted with ethanol. This extract was applied to Raji (a human Burkitt Lymphoma cancer), HepG2 (a human liver cancer), and Vero (a nonhuman cell line) cells in order to measure the cytotoxic activities by using MTT assay. To determine descriptively the compounds groups of phenolics, flavonoids, terpenoids, steroids, and alkaloids the thin layer chromatography method was performed. The IC50 value was analyzed quantitatively by using probit analysis. Results showed that the IC50 values of ethanolic extract of rice bran ‘Woja Laka’, ‘Toraja’, ‘Cempo Ireng’ and ‘IR 64’ on HepG2 cells were 857.23±99.19; 1,896.55±83,8; 1,494.47±87.81 and 727.89±145,97 µg/ml respectively. The IC50 on Raji cells were 816.61±85.31; 1,079.93±28.31; 1,627.82; ±119.82, and 769.33±61.43 µg/ml respectively. The IC50 on Vero cells were 1,295.2±37; 1,232.07±165.51; 1,874.14±169.56, and 724.4±122.79 µg/ml respectively. The ethanolic extracts of rice bran from four cultivars contain phenolics, flavonoids, terpenoids, and steroids. However, alkaloids could not be detected. The variety of rice cultivars indicates the variation of cytotoxic activities on cancer cells. The ethanolic extracts of rice bran from those four rice cultivars contain similar kinds of organic compounds groups but vary in the Rf values.

Abstract

Senyawa aktif Gamma Amino Butyric Acid (GABA) merupakan asam amino yang terdapat di otak mammalia dan berperan sebagai neurotransmiter.  Selain pada sistem saraf pusat, GABA juga terdapat dalam konsentrasi tinggi di sel β pankreas serta memiliki potensi sebagai antidiabetes. GABA dapat disintesis oleh hewan, bakteri asam laktat dan tumbuhan.  Penelitian dilakukan dengan tujuan mendeteksi keberadaan GABA dalam bagian tumbuhan yaitu daun Artocarpus altilis.  Deteksi dilakukan dengan Kromatografi Lapis Tipis (KLT) menggunakan fase diam plat silika, fase gerak BAW(3:1:1), dan GABA murni sebagai pembanding.  Larutan GABA dibuat dengan pelarut etanol:air (7:3).  Hasil penelitian menunjukkan GABA terdeteksi pada ekstrak daun Artocarpus altilis.

Abstract

One of the urban stories from Lampung was called Warahan. Its an ancient story telling model with a moral meaning. This also one of a lesson items for students in Junior High School of SMP Negeri 1 Kotabumi, North Lampung. Considering how importance to give a moral value for students, this research was to investigate a Contextual Teaching and Learning (CTL) methods to understand a moral values from Warahan for students. Data has been taken from some kinds of techniques where the students were put in some discussion groups dan giving them a writing test. This experiment was found a significant progress with Contextual Teaching and Learning (CTL) methods where students were giving a chance to improve themselves dan learning by doing comparing with a reality.Salah satu jenis sastra daerah Lampung berbentuk cerita yang berisi pesan moral adalah Warahan. Materi ini terdapat di dalam mata pembelajaran Bahasa Lampung yang diajarkan di SMP Negeri 1 Kotabumi Lampung Utara. Mengingat pentingnya penanaman pesan moral melalui pembelajaran di sekolah, maka penelitian ini mencoba mengkaji model pembelajaran kontekstual yaitu Contextual Teaching and Learning (CTL) dalam meningkatkan pemahaman siswa terhadap materi warahan. Sebagai penelitian eksperimen, data diperoleh dengan cara membagi siswa dalam kelompok dan memberikan tes tertulis. Penelitian ini menemukan bahwa terdapat pengaruh yang signifikan dengan model pembelajaran CTL yang merupakan suatu model pembelajaran yang memberikan fasilitas kegiatan belajar siswa untuk mencari, mengolah, dan menemukan pengalaman belajar yang lebih bersifat konkret (terkait dengan kehidupan nyata) melalui keterlibatan aktivitas siswa dalam mencoba, melakukan, dan mengalami sendiri.Kata kunci: Contextual Teaching Learning (CTL), Memahami Warahan, Pesan Moral.

Abstract

The chemical contents and health benefits of black rice bran of some rice cultivars have been investigated. However, there has been little research on the ‘Cempo Ireng’ cultivar from Sleman, Yogyakarta. The aim of this present study was to determine the anthocyanin, antioxidant activity, and macro- and micronutrients contents of black rice bran from this local cultivar. The anthocyanin in the black rice bran was extracted using the maceration method with methanol as a solvent. The extract obtained was separated through a preparative thin layer chromatography (TLC) of silica GF254 and a mobile phase composed of n-butanol, acetic acid, and water. Two fractions were collected and analyzed for the anthocyanin content. The preparative TLC spots were separated for further detection and measurement of cyanidin 3-O-glucoside using HPLC followed by LC-MS. The antioxidant activity of the fractions were measured using the DPPH free radical scavenging method. The results showed that the anthocyanin in fraction 1 was identified as cyanidin 3-O-glucoside (66.1 ± 10.6 µg/g). The IC50 of fractions 1 and 2 were 200.96 and 218.36 µg/mL, respectively. Analysis of the macro- and micronutrients revealed that the black rice bran of ‘Cempo Ireng’ had nutrient contents comparable with other rice cultivars. Therefore, this local black rice bran can be used as a source of antioxidants and macro-- and micronutrients.
