Abstract

The objective of this research was to examine the correlation between estrogen level and estrous cycle of rat fed with soybean flour and tempeh flour. A completely randomized design was applied in this study with 6 treatment groups and 5 replications. Rats in group 1, 2, and 3 were non-ovariectomized rats which were fed with pellet, 10 g soybean flour/100 g body weight/day, and 10 g tempeh flour/ 100 g body weight/day, respectively. The rats in group 4, 5, qnd 6 were ovariectomized rat which were fed with pellet, 10 g soybean flour/100 g body weight/day, and 10 g tempeh flour/100 g body weight/day, respectively. The rats were after fed with pellet, soybean and tempeh flour for 4 weeks, the estrous cycle phases of rats were examined based on the presence of vaginal epithelial cells and the number of qualitative vaginal epithelial cells, while the estrogen concentrations in serum were measured by radioimmunoassay method. The data was analyzed using analysis of variance and followed by Duncan multiple range test at 95% confidence. Non-ovariectomized rats given tempeh flour have longer estrus cycle than the non-ovariectomized rats which given soybean flour. The isoflavon contained in tempeh flour and soybean flour induced proliferation and cornification of vaginal epithelial cell. Feeding the soybean flour and tempeh flour to ovariectomized rats could optimize the estrogen hormone in initiating the estrus phase, in which of the tempeh flour addition had better effect than soybean flour. Correlation between the level of estrogen to the length of estrous cycle in both nonovariectomized rats and ovariectomized rats was positive, in which the higher the levels of estrogen the longer the length of estrous cycle.

Abstract

The aim of this research was to determine the anatomy and histology of pancreas of water monitor lizard (Varanus salvator). Two pancreas were used in this research (a male and a female). Water monitor lizard was euthanized using chloroform and subsequently necropsied to collect the pancreas. The pancreas was washed in 0.9 % NaCl solution. Anatomical observations were carried out including location, shape, and color of pancreas, also the length and weight measurements. Then, pancreas was fixed with 4% paraformaldehyde for 7 days to proceed to histological preparation and hematoxylin eosin (HE) staining. The results of the anatomical observations showed that pancreas of water monitor lizard have two lobes, dorsal lobe (splenic lobe) and ventral lobe (duodenal lobe). Dorsal lobe which was in oval form adhered with the spleen, and ventral lobe was located in the cranial of intestines with the form of long spherical. Both of pancreas lobes were white-yellow color with the length of 1.45±0.64 cm (dorsal lobe) and 7.00±0.42 cm (ventral lobe). Weight of dorsal lobe was 0.15±0.01 g and ventral lobe was 6.35±2.30 g. The histological observations found the acinar cells, ducts, and blood vessels in both of pancreas lobes. Additionally, the similar structure of Langerhans islet only found at the ventral lobe of pancreas. In conclusion, pancreas of water monitor lizard consists of two lobes which located differently. Ventral lobe is larger than the dorsal lobe. Both lobes have similar histological structure except the Langerhans islets are only appeared in the ventral lobe.

Abstract

The aim of this study was to observe cortisol levels and liver histopathology of broiler chicken that were treated with black cumin oil (BCO) under heat stress. A total of 15 broiler chickens were used in this study and divided into 5 groups, K- (without treatment), K+ (given heat stress), P1 (given heat stress and 0.56mL BCO/400kg body weight, P2 (given heat stress and 1.11 mL BCO/400 g body weight), and P3 (given heat stress and 2.22 mL BCO/400 g body weight). Heat stress was given for 5 hours with temperature range of 34-35° C for 7 days. Cortisol was measured using the cortisol enzyme-linked immunosorbent assay (ELISA) kit. Liver histopathology was stained with hematoxylin eosin and observed with electron microscope. The data were analyzed using one way analysis of variance (ANOVA). This study found that application of heat stress to broiler chickens increased cortisol levels and induced histopathological changes in the liver. The BCO administration reduced cortisol level significantly (P 0.05) in heat-stressed broilers. BCO administration also significantly  reduced (P 0.05) the degenerative changes in liver histopathology such as fat degeneration, hemorrhage and necrosis in broiler chickens under heat stress, but did not significantly influence the inflammatory cells infiltration. As conclusion, BCO administration to broiler chickens under heat stress can reduce cortisol levels and minimize histopathological changes in the liver.

Abstract

ABSTRAK. Penelitian ini bertujuan mempelajari pengaruh pemberian kombinasi pakan fermentasi dengan multi enzim pencernaan dan vitamin E dalam ransum komersial terhadap peningkatan kualitas semen ayam arab. Penelitian ini menggunakan rancangan acak kelompok (RAK) yang terdiri dari 4 kelompok perlakuan dengan 5 kali ulangan. 1) kontrol (P0) ayam diberikan ransum komersial 100 %, 2) perlakuan 1 (P1) ayam diberi 89,83% ransum komersial + 10% pakan fermentasi + 0,15% multi enzim + 0,02% vitamin E, (3) perlakuan 2 (P2) ayam diberi 79,83% ransum komersial + 20% pakan fermentasi + 0,15% multi enzim + 0,02% vitamin E, dan (4) perlakuan 3 (P3) ayam diberi 69,83% ransum komersial + 30% pakan fermentasi + 0,15% multi enzim + 0,02% vitamin E. Koleksi semen dilakukan pada hari ke 36 dengan metode masase. Data kualitas semen yang diperoleh dianalisis dengan menggunakan analysis of variance (ANOVA) satu arah yang dilanjutkan dengan uji Duncan. Hasil penelitian menunjukkan pemberian kombinasi pakan fermentasi dengan multi enzim dan vitamin E dalam ransum komersial berpengaruh secara nyata (P 0,05) terhadap peningkatan volume, pH semen, konsentrasi spermatozoa, motilitas, spermatozoa hidup, dan penurunan abnormalitas ayam arab, namun tidak berbeda nyata (P 0,05) terhadap warna, konsistensi dan gerakan massa spermatozoa. Dapat disimpulkan pemberian kombinasi pakan fermentasi dengan multi enzim dan vitamin E dalam ransum komersial dapat meningkatkan kualitas semen ayam arab. (Effect addition of fermented feed with the multi-enzyme digestion and vitamin E in commercial feed on improvement the quality of semen arab chicken)ABSTRACT. The study aimed to determine the effect of fermented feed combination with the multi enzymes digestion and vitamin E addition on the quality of semen arab?s chicken. This research used a randomized block design (RAK) consisting four treatments with five replications : P0: 100% Commercial feed (524), P1: 89.83% commercial feed + 10% fermented feed + 0.02% viamint E + 0.15% multi enzyme, P2: 79.83% commercial feed + 20% fermented feed + 0.02% vitamin E + 0.15% multi enzyme, P3: 69.83% commercial feed + 30% fermented feed + 0.02% vitamin E + 0.05% multi enzyme. Semen sample was collected at 36th  day using a massage method. Data obtained were analyzed using analysis of variance (ANOVA) and continued by Duncan test. The results showed that addition of combination of fermented feed with digestive multi enzymes and vitamin E has significantly effect (P 0,05) on increases of the volume, pH, spermatozoa concentration, percentage of spermatozoa motility, spermatozoa life, and decreased the percentage of spermatozoa abnormalities. This research can be concluded that the addition of  fermented feed combination with digestive multi enzymes and vitamin E able to improve the quality of arab chicken semen.

Abstract

This study aimed to determine the bone growth of broiler chickens fed with additional probiotic Aspergillusniger (AKBIS Prob). The organ taken was the femur bone. A total of 24 broiler chickens reared with additional feed on 4 treatments and 3 repetitions with a combination of additional feed namely P0 (commercial feed), P1 (commercial feed and 2% AKBIS Prob), P2 (commercial feed and 4% AKBIS Prob) and P3 ( commercial feed and 6% AKBIS Prob). The sampling of the femur bone was carried out on chicken 21 and 35 days old and observed histologically after stained with hematoxylin-eosin (HE). The results showed from the examination of macroscopic growth that the femur growth increased more at the age of 35 days compared to the age of 21 days in the treatment group P1, P2 and P3 who were given probiotic Apergillusniger (AKBIS Prob) where the amount of Ca and P required for growth can be fulfilled. On microscopic examination, the measurement parameters were the number of active osteoblasts and osteoblast that looked more passive on the 35th day of treatment, compared to the age of 21 days. While the number of osteoclasts was more at age 21 than at age of 35 days in the treatment group who were given probiotic Aspergillusniger (AKBIS Prob)as additional food.

Abstract

Penelitian mengenai pengaruh ekstrak batang Cissus quadrangula Salisb. (sipatah-patah) terhadap tingkat proliferasi dan diferensiasi sel-sel tulang tikus (Sprague Dawley) prepuber umur empat minggu menggunakan sistem kultur in vitro. Sel-sel tulang dikultur dalam medium dulbecco?s modified eagle?s medium (DMEM) yang diberi tambahan newborn calf serum (NBCS) 10%, non essential amino acid (NEAA) 10%, NaHCO3, ITS 1 µl/ml (mengandung insulin 5 ?g/ml, transferin 10 ?g/ml, selenium 5 ?g/ml; Sigma St Louis USA), dan 50 µg/ml gentamisin (mDMEM), dengan dan tanpa ekstrak Cissus quadrangula (CQ). Penelitian terdiri atas lima perlakuan yakni kontrol positif (mDMEM + deksametason 10-8 M), kontrol negatif (mDMEM), dan tiga konsentrasi CQ: mDMEM + CQ 0,3 mg/ml; mDMEM + CQ 0,6 mg/ml; dan mDMEM + CQ 1,2 mg/ml. Kultur dilakukan dalam inkubator CO2 5%, pada suhu 37° C selama tujuh hari. Parameter yang diamati adalah konsentrasi sel, komposisi, diameter osteoblas, dan diameter osteosit. Konsentrasi sel dihitung menggunakan hemositometer Newbauer. Komposisi osteoblas dan osteosit ditentukan berdasarkan pengamatan morfologi di bawah mikroskop cahaya. Diameter sel diukur menggunakan eyepiece micrometer. Data dianalisis menggunakan analisis varians dan uji Duncan. Hasil penelitian menunjukkan bahwa pemberian ekstrak Cissus quadrangula Salisb. pada konsentrasi 0,3 mg/ml; 0,6 mg/ml; dan 1,2 mg/ml secara signifikan dapat meningkatkan proliferasi sel tulang; dan pada konsentrasi 0,6 mg/ml mampu menginduksi diferensiasi osteoblas menjadi osteosit (P 0,05). Disimpulkan bahwa pemberian ekstrak Cissus quadrangula pada konsentrasi 0,6 mg/ml ke dalam medium kultur dapat meningkatkan proliferasi dan diferensiasi osteoblas.

Abstract

Cissus quadrangularis Salisb is a medicinal plant which has been used traditionally to cure bone fracture by Acehnese. This study was carried out to investigate the administration effects of Cissus quadrangularis Salisb extract (CQSE) on the histology of lumbar bone in ovarectomized rats. There were 16 female rats (Rattus norvegicus) as samples and were then divided into 4 groups of treatment, with 4 replications each. K0 consisted of ovarectomized rats without CQSE supplementation, whereas K1, K2 and K3 consisted of ovarectomized rats administrated with 300, 500 and 700 mg/kg body weight CQSE for 60 days, respectively. On the day 61st, all rats were euthanized with chloroform and their fourth lumbal bones were collected for histological preparation. Parameters observed were densities of active osteoblast, passive osteoblast and osteoclast. The results of this study showed that density of active osteoblast of rats in K3 (27.25 ± 2.75) was significantly higher (P 0.01) than rats in K0 (1.75 ± 1.25), K1(4.00 ± 1.41), and K2 (8.50 ± 7.40), respectively. There was no significant difference (P 0,05) of passive osteoblast density among rats in K0 (16.50 ± 6.13), K1 (41.00 ± 6.48), K2 (44.50 ± 16.34), and K3 (39.00 ± 15.23). Osteoclast density of rats in K0 (85.25 ± 25.91) was significantly higher (P 0,05) than that of rats in K1 (52.50 ± 16.66), K2 (51.00 ± 7.16), and K3 (50.75 ± 9.20). In conclusion, consecutive administration of 700 mg/kg body weight of CQSE extract for 60 days might increase active osteoblast density in ovarectomized rats.

Abstract

This study aimed to u determine the potency of Aceh Arabica coffee In rejuvenation of aging skin in rat. This study was conducted in vivousing completely randomized design (RAL), which consisted of 4 treatment groups as follows: premenopausal rats as negative control (KON),premenopausal rats with administration of distilled water as placebo (PLS), premenopausal rats with administration of ethinylestradiol 9x10-3mg/day/200 g BW (EST), and premenopausal rats with coffee extract administration of 300 mg/day/200 g BW (KOP). Each treatment groupconsisted of 5 rats. The parameters observed were skin collagen, water content/skin hydration, and skin ribonucleic acid (RNA) level. The datawere analyzed using analysis of variance (ANOVA) and Duncan test using SAS 9.13 software. The result showed the average skin collagen andRNA levels were higher in premenopausal rats with administration of coffee extract and ethinylestradiol (P 0.05) than control rats. On the otherhand, average of water level in premenopausal rats with coffee extract and ethinylestradiol administration were the same as control rats. It can beconcluded that administration of Aceh Arabica coffee extract can repair skin quality, which is indicated by the increase of collagen and RNAlevel on aging skin in rat.

Abstract

This report was aimed to describe the behavioral changes of catfish as observed before two different earthquakes hit the Province of Aceh, Indonesia in year of 2012. A number of 5 African?s catfish (Clarias gariepinus) were placed in an aquarium located in Gapang, Sukakarya Sub-district of Sabang Town, Aceh. Their behavior activities were observed manually at three different times (08.00 AM, 01.00 PM, and 05.00 PM) every day from 1st March up to 11th April 2012. The result of observation showed that the activity of catfish was different between all of them from 1st March 2012 up to morning observation on 11th April 2012. Most of those activities was not showed as very active behavior. However, their behavior was very active in the same pattern when observed at noon and afternoon times on 11 April 2012. On this date, there were two consecutive earthquakes happened at magnitude of 8 Richter Scale at 03.38 PM and 05.43 PM. The epicenter of this earthquake was in Simeulue, with the distance of more and less of 390 km from observation station of catfish in Gapang. It was noted that this abnormal behavior possibly showed as an indication of their ability to detect the increase of pre-seismic activity at 2.38 hours and 43 minutes before the two earthquakes. Perhaps, it is the result of their response to the increase of pre-seismic electromagnetic wave. Further continuous observation and research is needed to ensure the role of African?s catfish (Clarias gariepinus) as abio-sensor for the increase of seismic activity.

Abstract

The objective of this study was to investigate the pattern of carbohydrate distribution of accressory sex glands in male muntjac (Muntiacus muntjak muntjak) in the velvet antler stage. In macroanatomy observation, muntjac accessory sex glands consist of the ampulla, vesicular gland, pars diseminate prostat, and bulbourethral gland. The observation of neutral carbohydrate distribution has done by periodic acid Schiff (PAS) staining. In addition, acid carbohydrate was observed by using alcian blue (AB) pH2,5 staining. Result of this research showed that neutral and acid carbohydrate scattering in each accessories sex gland with different pattern. Neutral carbohydrate with medium (++) and high (+++) intensity reaction was found in the ampulla, vesicularis gland, and bulbourethral gland and strong in prostat gland. The acid carbohydrate with low (+) and medium (++) intensity reaction was observed in the ampulla, low in vesicularis gland, and negative in prostat and bulbourethral glands. In conclusion, the neutral carbohydrate was found dominantly on surface of all accessory sex glands of male muntjac in velvet antler stage with highest concentration and intensity of colour in prostat gland.Key words: carbohydrate, accessory sex glands, muntjac (Muntiacus muntjak muntjak)
