Abstract

Wualae (Etlingera elatior (Jack) R.M. Smith) adalah salah satu tumbuhan dari genus Zingeberaceae yang banyak tersebar di Sulawesi Tenggara yang secara empiris telah digunakan sebagai bahan pangan dan pengobatan tradisional. Tujuan penelitian ini untuk mengetahui aktivitas antibakteri ekstrak etanol buah E. elatior terhadap bakteri gram positif Staphylococcus aureus ATCC 25923, Bacillus subtilis FNCC 0060, dan bakteri gram negatif Pseudomonas aeruginosa ATCC 27853, Salmonella enterica ATCC 14028 menggunakan metode Cup-plate technique dengan pengukuran diameter daya hambat (DDH). Hasil pengujian menunjukkan bahwa ekstrak lebih aktif pada bakteri gram positif dibandingkan gram negatif. Penghambatan tertinggi ditunjukkan oleh konsentrasi 20 mg/mL dengan nilai DDH 5 mm. sehingga dapat disimpulkan bahwa ekstrak etanol buah E. elatior memiliki aktivitas antibakteri dengan DDH kategori lemah.

Abstract

The purposes of this study were to know the impact of different level of sea water inundation on the growth capacity and biochemical adaptation of the mangrove Rhizophoraceae family of Rhizophora mucronata and Bruguiera gymnorrhiza seedlings. The propagules of these mangroves were planted with different levels of sea water flooding, namely 90 cm, 60 cm, 45 cm and 30 cm. The growth of height and diameter of these mangrove seedlings were measured at early and termination of treatments, while their biochemical adaptations were analyzed in the Laboratory. The relatively growth rate RGR of high and diameter of these seedlings were analyzed, while statistical test was performed by anova. The RGR of high of the two mangrove seedlings were significantly different (P <0.05), though their RGR of diameter did not differ significantly (P> 0.05). The growth of height of R. mucronata seedlings was fastest at the lower level of inundation (30 cm) and the slowest at the higher level of submergence (90 cm). On the contrary, the height growth of B. gymnorrhiza seedlings was fastest at submergence of 90 cm, and the slowest at submergence of 30 cm. The flavonoids and vitamin C contents in both mangrove seedlings increased significantly (P <0.05) with increased inundation levels. Thus, although biochemical responses tended to be the same for both mangroves, but B. gymnorrhiza seedlings indicate  more able to grow and develop on rising sea levels than R. mucronata seedlings. This study provides important information for revegetation planning of degraded mangrove under a sea level rise scenario.Keywords: Biochemical adaptation, Sea water level, mangrove seedling growth, mangrove revegetation.

Abstract

Penelitian ini bertujuan untuk membuktikan efek dari pemberian ekstrak etanol akar rumput belulang (E. indica L. Gaertn) terhadap penurunan jumlah bakteri pada hepar mencit (Mus musculus) yang diinokulasi S. typhi.Penelitian ini adalah penelitian eksperimental laboratorium dengan rancangan the post-test only control group design. Dua puluh ekor mencit dibagi menjadi 4 kelompok, satu kelompok kontrol dan tiga kelompok uji. Kelompok I tidak diberi ekstrak etanol akar rumput belulang sebagai K. Kelompok II diberi ekstrak etanol akar rumput belulang dengan dosis 10 mg/ekor sebagai Uji I. Kelompok III diberi ekstrak etanol akar rumput belulang dengan dosis 20 mg/ekor sebagai Uji II. Kelompok IV diberi ekstrak etanol akar rumput belulang dengan dosis 40 mg/ekor sebagai Uji III. Semua kelompok diberi pakan standar pada hari ke-1 sampai hari ke-7. Pada hari ke-8 mencit diinokulasi S. typhi 1 x 10 6, setelah 2 jam mencit diberi ekstrak etanol akar rumput belulang pada kelompok uji dan Aquadest pada kelompok kontrol. Setelah 24 jam mencit didekapitasi dan diambil heparnya untuk menghitung jumlah koloni bakteri pada jaringan hepar. Rerata jumlah bakteri pada pengenceran ke-2 didapatkan kelompok K = 26.78, kelompok Uji I = 28.28, kelompok Uji II = 24.74, dan kelompok Uji III = 7.98. Uji Mann-Whitney U didapatkan perbedaan signifikan antara kelompok K dengan Uji III p=0.008, Uji I dengan Uji III p=0.007, dan Uji II dengan Uji III p=0.009(p=<0,05).Ekstrak etanol akar rumput belulang dapat menurunkan jumlah koloni bakteri pada mencit yang diinokulasi S. typhi pada tiap-tiap dosis.Kata Kunci : Akar rumput belulang, Koloni bakteri, Hepar, Salmonella typhi

Abstract

Imunomodulator merupakan bahan yang dapat mengembalikan ketidakseimbangan sistem imun. Spons Xestospongia Sp. diduga mengandung senyawa-senyawa aktif yang berperan sebagai agen imunomodulator. Penelitian ini dilakukan untuk mengetahui pengaruh pemberian ekstrak etanol Spons Xestospongia Sp. terhadap aktivitas fagositosis makrofag. Sebanyak dua puluh empat ekor mencit jantan galur Balb/C umur 2-3 bulan dengan berat badan 20-30 gram dibagi ke dalam 6 kelompok. Kelompok pertama mendapat pemberian ekstrak etanol Spons Xestospongia Sp. 100 mg/kgBB, kelompok kedua mendapat pemberian ekstrak etanol Spons Xestospongia Sp. 200 mg/kgBB, kelompok ketiga mendapat pemberian ekstrak etanol Spons Xestospongia Sp. 300 mg/kgBB dan kelompok keempat mendapat pemberian ekstrak etanol Spons Xestospongia Sp. 400 mg/kgBB. Kelompok kontrol positif mendapat ekstrak Phyllanthus niruri Linn. (Stimuno®) 0,13 mg/gBB dan kelompok kontrol negatif mendapatkan Na-CMC 0,5%. Ekstrak diberikan secara peroral sejak hari pertama hingga ketujuh. Pada hari kedelapan masing-masing mencit diinjeksikan bakteri Staphylococcus aureus (SA) 0,5 mL secara intra peritoneal. Aktivitas sel makrofag dihitung dari apusan cairan peritoneum mencit. Peningkatan dosis ekstrak etanol Spons Xestospongia Sp. meningkatkan jumlah aktivitas fagositosis makrofag dari 24,25 % (Na-CMC), 34,25% (100 mg/kgBB), 47,00% (200 mg/kgBB), 59,50 % (300 mg/kgBB) dan 62,75% (400 mg/kgBB). Hasil penelitian menunjukan bahwa ekstrak etanol Spons Xestospongia Sp. memiliki potensi sebagai imunomodulator pada dosis 300 mg/kgBB dan 400 mg/kgBB dengan efektivitas yang tidak berbeda bermakna dengan kontrol positif (Stimuno®) dalam meningkatkan aktivitas fagositosis sel makrofag berdasarkan hasil uji statistik post hoc TUKEY (sig. &gt; 0,05).

Abstract

Isolasi dan penentuan struktur senyawa metabolit sekunder dari ekstrak metanol spons laut Clathria sp serta uji toksisitasnya terhadap larva udang Artemia salina Leach telah dilakukan. Tahapan isolasi dimulai dengan ekstraksi menggunakan teknik maserasi dengan pelarut metanol dilanjutkan, selanjutnya dilakukan pemisahan dengan teknik kromatografi, meliputi kromatografi kolom vakum (KKV), dan kromatografi radial (KR). Struktur senyawa hasil pemurnian dianalisis dengan metode spektroskopi menggunakan FTIR dan NMR-1D (1H dan 13C-NMR) serta dibandingkan dengan data sejenis dari literatur. Isolat diperoleh berupa padatan berwarna putih 3β-(hydroxymethyl)-A-nor-5α-cholest-l5-ene. Hasil uji toksisitas akut isolat terhadap larva A. salina menunjukkan isolat tidak bersifat toksik dengan nilai LC50 sebesar 497,58 ppmKata kunci: Clathria, spons, steroid, toksisitas, A. salina

Abstract

Isolation and identification of secondary metabolite stem bark of Polygonum pulchrum and its antioxidant activity were done. Extraction was performed by maceration using methanol solvent. The separation and purification obtained by using vacuum liquid chromatography (VLC), radial chromatography (RC) and monitored by thin layer chromatography (TLC). Identification of isolated compounds determined by spectroscopy method i.e. FT-IR, 1D-NMR (1H, 13C-NMR and DEPT) and 2-D NMR (HMQC and HMBC). White solid compound was isolated and characterized as 5(E)-eicosenoic acid. Antioxidant was analyzed towards DPPH (1,1-diphenyl-picrilhydrazil). The compound showed that has antioxidant potency with IC50 value at 348.10 µM.Keywords: Polygonum pulchrum, 5(E)-eicosenoic acid, antimicrobial, antioxidant

Abstract

Ten resveratrol oligomers have been isolated from the stem bark of H. gregaria i.e. resveratrol lOC-glucopyranoside (1), heimiol A (2), balanocarpol (3), ampelopsin A (4), hopeaphuran (5), E-viniferin (6), parviflorol (7), a-viniferin (8), hopeaphenol (9), and vaticanoi B (10). The structures of these compounds were determined base on spectroscopic methods including UV, IR, 1-D NMR, 2-D NMR and comparison with the reported data. Implication of the compounds invention is not only strengthen conclusion that Hopea tends to produce resveratrol dimmers but also showed that resveratrol lOC-glucopyranoside (1) and heimiol A (2) are phytochemical markers of. H. gregaria.

Abstract

Five resveratrol oligomers which consist of two resveratrol dimers, balanocarpol (1) and ?-viniferin (2), ? resveratrol trimer i.e. ?-viniferin (3), and two resveratrol tetramers, vaticanol B (4), and hopeaphenol (5) have been isolated from acetone extract of the stem bark of Dryobalanops lanceolata. These compounds were isolated by vacuum liquid chromatography method. Furthermore, the compound structures were determined based on the spectroscopic evidence, including UV, IR, 1-D and 2D NMR spectra, and comparing with those related data reported previously.

Abstract

Six resveratrol dimers have been isolated from the stem bark of H. gregaria, ampelopsin A (1), balanocarpol (2), ε-viniferin (3), hopeafuran (4), heimiol A (5), and parviflorol (6). The structures of these compounds were determined based on spectroscopic evidence such as UV, IR, 1-D, 2-D NMR and comparison with the reported data. These compound inventions are strengthen conclusion that Hopea tends to produce resveratrol dimers. Biological activity of those compounds against murine leukemia P-388 cells showed that ε-viniferin (3) is the most active compound with IC50 value 5.1 ± 0.3 μg/mL.   Keywords: H. gregaria, resveratrol dimers, murine leukemia P-388 cells

Abstract

Nine resveratrol oligomers have been isolated from the stem bark of Hopea mengarawan i.e. diptoindonesin D (1), balanocarpol (2), ampelopsin A (3), hopeaphuran (4), heimiol A (5), parviflorol (6), a-viniferin (7), isohopeaphenol (8), and vaticanol B (9).The structures of these compounds were determined based on spectroscopic evidence such as UV, IR, 1-D, 2-D NMR and comparison with the reported data. Those compounds implied that Hopea tends to produce resveratrol dimers. Moreover, in subgenus level, besides ampelopsin A (3) and isohopeaphenol (9), resveratrol oligomers in H. mengarawan the same as in H.dryobalanoides. It indicated that H. mengarawan and H. dryobalanoid.es came from the same subgenus.
