Abstract

Abstract. The purposes of this research are for molecular characterization and hemagglutination activity test of flagellinprotein of Salmonella typhi. The research samples consist of 7 strains of S. typhi isolates from Central Java (5 strains from Semarang city, 1 strain from Salatiga and 1 strain from Magelang) and 2 strains of S. typhi from Yogyakarta (Doctor Sardjito Hospital and Bethesda Hospital). The undertaking procedures are: 1) PCR and sequencing of fliC genes using primer LPW 1856 and LPW 1857.2) Isolation and separation of flagellin protein using SDS-PAGE. 3) Hemagglutination Activity Test upon human erythrocytes of blood group A, B, AB and O.The results show that 8 strains of S. typhi have a fliC gene size of 1452 to 1488 bp including serovar H1-d, and 1 strain with the size of 1267 bp including serovar H1-J. Flagella protein resulted from SDS-PAGE protein consists of 1-2 major proteins and 1-3 minor proteins with a molecular weight of 16-116 kDa. The results of hemagglutination activity test of flagellin protein show that there are 3 strains of S. typhi (MG-1, SA02.2 and BET) which are able to agglutinate human erythrocytes of bloodgroup A, B, AB and O (2-64HA), 6 other strains show various hemagglutination activities varied

Abstract

Abstract. Plumbum (Pb) is a heavy metal affecting renal tubular epithelial cells and causing necrosis. This study aimedto demonstrate the effectiveness of rice leaf extract in degeneration and necrosis prevention of renal tubule epitheliumexposed to lead. Twenty eight rats were divided into a control and 3 treatment groups all exposed to Pb by 0.5 g/ kg/ day,but only the treatment groups had received rice leaf extract in varied doses for 8 weeks. Results from Kruskal-Wallis andsphearman rho tests showed that the number of normal cells, degeneration and necrosis were significantly different (0.03;0.05; 0.04) between control and treatment groups with a strong positive correlation of treatment (r = 0.97) in normal cellsand a strong negative one in cell degeneration and necrosis. (R = -0.92 and r = -0.93). As conclusion, rice leaf extractshowed ability in the prevention of kidney damage in rats exposed to lead.

Abstract

Background: Vegetable materials, such as rice, corn, bean, and soybean have metallothionein proteins contained in their roots, stems, leaves, flowers, and fruits. Of those vegetable materials, paddy leaves have the most numerous metallotionein proteins. However, the varieties which produce the highest metallotionein proteins have not yet been found. The roles of metallothionein proteins are to bind and to detoxify heavy metals. Objectives:  to analyze, identify, and formulate the metallothionein extractscontained in numerous varieties of paddy leaves. An experimental research method. Each vegetable material of numerous varieties of paddy leaves is made into simplicia and is then processed into infusion. ELISA method is used as a technique to measure the protein level of each infusion. The metallothionein protein levels contained in numerous varieties of paddy leaves are then identified to determine the one with the highest metallothionein protein level. Extract formula is taken from the highest metallothionein level. Results :the highest metallothionein protein level is contained in paddy leaves of IR begendit (28.832 ng) while the lowest one is in paddy leaves of IR-64 SS (0.358 ng). The extracts of Paddy leaves of IR begendit with the highest protein level are then formulated into metallothionein extracts. Conclusion: Analysis on metallothionein levels of numerous varieties of paddy leaves with the highest protein level is contained in paddy leaves of IR begendit (28.832 ng) while the lowest one is in paddy leaves of IR-64 SS (0. 358 ng).Keywords: paddy leaves, metallothionein

Abstract

Latar belakang: Differensial counting merupakan hitung jenis lekosit  dilakukan bersama-sama dengan pemeriksaan apus darah tepi. Bentuk preparat darah apus adalah simetris sehingga dimungkinkan distribusi yang sama atau hampir antara zona atas dan bawah. Penelitian ini bertujuan untuk mengetahui perbedaan antara zona atas dan bawah sehingga pembacaan pada zona IV sampai VI dapat dilakukan guna menghitung 100 sel lekosit.Metoda: Penelitian dilaksanakan di Laboratorium Patologi Klinik Unimus antara bulan April sampai Juni 2009. Jenis penelitian ini adalah analitik  dengan cara membandingkan hasil hitung jenis sel lekosit berupa basofil, eosinofil, staff, segmen dan limfosit pada zona atas dan bawah preparat darah apus. Sampel diperoleh secara purposif dari mahasiswa D III Analis Kesehatan sebanyak 10 orang berupa darah vena.  Masing-masing sampel dibuat preparat apus duplo menggunakan pengecatan Giemsa. Pembacaan preparat darah apus dilakukan pada zona atas dan bawah masing-masing sebanyak 100 sel lekosit. Dibandingkan rerata zona atas dan bawah, analisis data menggunakan analisis statistik dengan program komputer SPSS. Teknik analisis data untuk mengetahui perbedaan jenis-jenis lekosit yang berupa basofil, eosinofil, staff, segmen dan limfosit menggunakan analisis t test relatid / Wilcoxon.Hasil penelitian: tidak terdapat perbedaan hasil penghitungan jenis-jenis lekosit antara zona atas dan bawah.Simpulan: hitung jenis lekosit dapat dilakukan pada salah satu bagian yaitu atas atau bawah zona IV sampai VI. Abstract

The purpose of this study is to molecularly characterize 7 strains of flagellin proteins of Salmonella typhi isolates taken from Central Java (5 strains from Semarang, 1 strain from Salatiga, 1 strain from Magelang) and 2 isolate strains from Yogyakarta (1 strain from Doctor Sarjito Hospital, and 1 strain from Bethesda Hospital). Flagellin gene (fliC) amplification is conducted using PCR (primer LPW 1856 and LPW 1857). Flagellin proteins resulted from Alexan method (2009) is then modified while flagellin protein profiles are obtained with SDS-PAGE method. The results show that the size of flagellin gene (fliC) of S. typhi Isolates from Salatiga and the 8 other strains is respectively 1262bp and 1500bp. Flagellin proteins which are composed of 2-6 protein sub-units consist of 1-2 major proteins and 1-4 minor proteins with the sizes of 16-116 kDa.Keywords: Salmonella typhi, flagellin, Molecular Characterizations
