Abstract

ABSTRACT This study was conducted to evaluate the profile of 17ß-estradiol (E2) and vitellogenin (Vtg) in plasma and egg diameter during gonad maturity process of striped catfish Pangasianodon hypophthalmus. Blood samples were collected from immature striped catfish, male and female with different stage of gonad maturity (stage I, II, III, and IV) to measure the concentrations of E2 and Vtg. Gonad maturity development of striped catfish was observed based on egg diameter. Result showed that E2 concentrations were the highest (843.65 pg/mL) on female with maturity stage III, the lowest on the male (26.34 pg/mL), and immature female fish (29.37 pg/mL). The protein band of Vtg was obtained on the plasma of the mature female (stage I, II, III and IV) with a molecular weight (MW) between 140?180 kDa, but it was not obtained on immature female dan male striped catfish. The highest concentration of Vtg was found in the plasma of the female fish with maturity stage III (87.34 mg/mL), then on the stage II (74.83 mg/mL), I (68.58 mg/mL), and IV (33.45 mg/mL). It showed that egg yolk formation occurred in the female mature. The average egg diameter was 0.107 ± 0.052 mm, 0.318 ± 0.086 mm, 0.864 ± 0.099 mm, and 1.041 ± 0.058 mm on the maturity stage I, II, III, and IV respectively. The increase of egg diameter along with development of gonad maturity stage indicated that egg development occurred due to the process of vitellogenesis and the addition of egg yolk on oocyte. Keywords : egg diameter, gonad maturity, striped catfish , 17ß?estradiol, vitellogenin ABSTRAK Penelitian dilakukan untuk mengevaluasi profil estradiol-17? (E2), vitelogenin (Vtg) dalam plasma dan diameter telur pada proses pematangan gonad ikan patin siam (Pangasianodon hypophthalmus). Sampel darah untuk pengukuran konsentrasi E2 dan Vtg plasma diperoleh dari ikan patin siam betina yang belum matang gonad, ikan jantan, ikan betina dengan tahap kematangan gonad yang berbeda (tahap I, II, III dan IV). Perkembangan kematangan gonad ikan patin siam diamati berdasarkan diameter telur. Hasil penelitian menunjukkan bahwa konsentrasi E2 tertinggi (843,65 pg/mL) pada ikan betina dengan kematangan tahap III, terendah pada ikan jantan (26,34 pg/mL), dan ikan betina tidak matang gonad (29,37 pg/mL). Pita protein Vtg pada sampel plasma diperoleh dari betina matang gonad (tahap I, II, III dan IV) dengan berat molekul antara 140-180 kDa, tetapi tidak diperoleh pada ikan patin siam betina yang belum dewasa dan jantan. Nilai konsentrasi tertinggi Vtg ditemukan dalam plasma darah ikan betina dengan tingkat kematangan III (87,34 mg/mL) kemudian pada tahap II (74,83 mg/mL), I (68,58 mg/mL) dan IV (33,45 mg/mL). Hal ini menunjukkan bahwa pada ikan betina dewasa terjadi proses pembentukan kuning telur (vitelogenesis). Rata-rata diameter telur adalah 0,107 ± 0,052 mm, 0,318 ± 0,086 mm, 0,864 ± 0,099 mm dan 1,041 ± 0,058 mm pada tingkat kematangan I, II, III dan IV secara berurutan. Peningkatan nilai diameter telur seiring dengan perkembangan tahap kematangan gonad menunjukkan bahwa perkembangan telur terjadi karena proses vitelogenesis dan penambahan bahan kuning telur pada oosit. Kata kunci : diameter telur, 17ß-estradiol, kematangan gonad, patin siam, vitelogenin

Abstract

ABSTRACT This study aimed to evaluate the effects of Zinc (Zn) supplementation on the quality and quantity of striped catfish sperm. Experimental design for this study was a complete randomized design with five treatments and five replications. Male broods fed with Zn supplementation for eight weeks. The Zn supplemented into the fish diet at different concentrations (0, 50, 100, 150 and 200 mg/kg of feed). The results showed that Zn supplementation could improve the quality and quantity of striped catfish sperm. The treatments also showed significant effects on semen volume, sperm motility, sperm viability, and sperm concentration (P<0.05). Zn supplementation at a dose of 200 mg/kg feed demonstrated the best result has indicated by enhancement of quality and quantity of striped catfish sperm, increasing 51% of the volume, 11.6% of motility, 5.81% of viability, 54.1% of concentrations. The results suggested that Zn played an important role in improving reproductive performances of male striped catfish reproduction. Keywords: quality of sperm, a quantity of sperm, striped catfish, supplementation zinc                                                                                            ABSTRAK Penelitian ini bertujuan untuk mengetahui pengaruh suplementasi Zinc (Zn) terhadap kualitas dan kuantitas sperma ikan patinPangasianodon hypophthalmus. Penelitian ini menggunakan rancangan acak lengkap dengan  lima perlakuan danlima ulangan. Induk jantan diberi pakan dengan suplementasi Zn selama 8 minggu. Zn disuplementasikan dengan dosis berbeda (0, 50, 100, 150 dan 200 mg/kg pakan). Hasil penelitian menunjukkan bahwa suplementasi Zn dapat meningkatkan kualitas dan kuantitas sperma ikan patin sehingga berpengaruh signifikan terhadap volume semen, motilitas, viabilitas dan konsentrasi sperma (P<0,05). Suplementasi Zn pada dosis pakan 200 mg/kg menunjukkan hasil terbaik yang ditunjukkan oleh peningkatan kualitas dan kuantitas sperma ikan patin 51% volume; 11,6% motilitas; 5,81% viabilitas; 54,1% konsentrasi sperma. Hasil penelitian ini menunjukkan bahwa Zn memainkan peran penting dalam meningkatkan reproduksi ikan patin. Kata kunci: kualitas dan kuantitas sperma, Ikan patin, suplementasi Zn

Abstract

ABSTRACT Reproductive design for gonadal maturation process mostly related with some factors such as environmental signals, reproductive organs, hormonal and nutrition. This research was conducted on female Mutiara strain of North African Catfish, Clarias gariepinus by combining two kinds of materials administered to broodstock diet, namely oocyte developer (Oodev) which contains of PMSG hormone and antidopamin, and astaxanthin carotenoid. Research designs were divided into C (Control), A50 (astaxanthin 50 mg/kg feed), A100 (astaxanthin 100 mg/kg feed), Od0.5 (Oodev 0.5 mL/kg fish for two weeks), Od1 (1 mL/kg fish for 2 weeks), Od0.5A50 (combined Od0.5 with A50), Od1A50 (combined Od1 with A50), Od0.5A100 (combined Od0.5 with A100), and Od1A100 (combined Od1 with A100). This research was performed during twelve weeks of feeding. The Od1A100 treatment showed the best reproduction performance result compared to other treatment with highest hepatosomatic (HSI) and gonadosomatic (HSI) indexes (P<0.05), also fastest increase in egg diameters (P<0.05), shorter rematuration periods and highest proportion of mature broodstock. These results indicated that Oodev and astaxanthin could accelerate gonadal maturity in female broodstock of Mutiara catfish.Keywords: Broodstock, hormonal, reproduction, oocyte developer, astaxanthin  ABSTRAK Rekayasa reproduksi untuk proses pematangan gonad sebagian besar terkait dengan beberapa faktor seperti sinyal lingkungan, organ reproduksi, hormonal dan nutrisi. Penelitian ini dilakukan terhadap strain ikan lele Mutiara (Clarias gariepinus) betina menggunakan dua bahan yang dicampur pada pakan induk, yaitu oocyte developer (Oodev) yang mengandung hormon PMSG dan antidopamin, dan karotenoid astaxanthin. Eksperimen yang dirancang adalah K (Kontrol), A50 (Astaxanthin 50 mg/kg pakan), A100 (Astaxanthin 100 mg/kg pakan), Od0.5 (Oodev 0,5 mL/kg induk untuk 2 minggu), Od1 (Oodev 1 mL/kg induk untuk 2 minggu), Od0.5A50 (kombinasi Od0.5 dan A50), Od1A50 (kombinasi Od1 dan A50), Od0.5A100 (kombinasi Od0.5 dan A100), dan Od1A100 (kombinasi Od1 dan A100). Penelitian ini dilakukan dengan memberi makan dua belas minggu. Performa reproduksi terbaik didapat pada perlakuan Od1A100. Od1A100 memiliki indeks hepatosomatik (HSI) dan gonadosomatik (HSI) tertinggi (P <0,05), juga diameter telur paling cepat besar (p <0,05), periode rematurasi terpendek, dan proporsi induk matang gonad tertinggi. Abstract

  Tilapia fish farming using monosex male population has been reported to have 10% higher productivity compared to mix-sex culture. This study aimed to determine immersion dose of 17?-metiltestosterone (MT) that allowed higher male percentage, growth, survival, and measure testosterone level in fish body. The experiment was designed as three immersion doses, namely: 0; 1.8; and 5.4 mg/L MT, each with three replications. Immersion was conducted to 14-day-old larvae for four hours, with density of 100 fish/L of water. Testosterone levels in fish was measured using ELISA method, and sex identification was performed by histology method. The result showed that percentage of male fish was the same in the treatment of 1.8 mg/L and 5.4 mg/L, and 53?65% higher than the control without MT treatment (54% male). Growth and survival of fish until day 56 post immersion were the same. By ELISA analysis, the levels of testosterone in larvae just after immersion was similar in 1.8 mg/L and 5.4 mg/L treatments, decreased drastically on day 14 after immersion, and then the levels of testosterone to be similar with the control on day 28, i.e. about 2 ng/g. By PCR method with specific primer, sex reversed and normal males could be distinguished, and on day 71 the testosterone levels in sex reversed and normal males fish was also the same, 0.3 ng/g (P>0.05). Thus, sex reversal by immersion at a dose of 1.8 mg/L can be consider as a standard protocol for monosex tilapia production. Testosterone level in the body of MT-treated fish becomes the same to the control of less than one month post immersion

Abstract

Penelitian ini bertujuan menentukan frekuensi pemberian vaksin DNA koi herpesvirus (KHV) yang menghasilkan kelangsungan hidup tinggi pada ikan mas yang diinfeksi KHV. Frekuensi pemberian vaksin yaitu vaksinasi satu kali seminggu (K1), vaksinasi dua kali seminggu (K2), vaksinasi tiga kali seminggu (K3), ikan tidak divaksin dan diuji tantang KHV (K4), dan ikan diinjeksi dengan larutan phosphate buffered saline (K5). Pemberian pakan dilakukan 2 kali sehari secara satiasi.Mortalitas pada K1; K2; K3; dan K4 masing-masing adalah 33,33±5,77; 20,00±10,00; 6,67± 5,77; dan 43,33±5,77%, sedangkan kelangsungan hidup relatif (RPS) K1; K2; dan K3 masing-masing adalah 23,07±13,32; 53,84±23,07; 84,60±13,32%. Disimpulkan bahwa pemberian pakan mengandung vaksin DNA anti-KHV dengan frekuensi tiga kali dalam satu minggu efektif meningkatkan kelangsungan hidup ikan mas yang diinfeksi KHV.

Abstract

This experiment was conducted to ditermined phosphorus content in the feed of giant gouramy (Osprhonemus gouramy). Four semipurified diet containing isoprotein (41,0%) and isoenergy (10.0 kcal/g protein) but different in phosphorus content were prepared. Diet containing phosphorus at the level of 0,32, 0,63, 0,89 and 1,06% respectively. Fish weighing 21-25 g/ind was stocked at a density of 4 ind./aquarium. Fish was fed at a satiation level three times daily. Fish were reared for 40 days and sampling was done in every 10 day. Result of this study shows that growth performance of fish were increase with the increasing of phosphorus level in the diet till of level (0,89%). Make up of rate phosphor more than 1,06% in the diet will negating the growth. It can be concluded that giant gouramy require phosporus at the level of 0,63-0,89% for maximal growth. Key words : Phosphor, giant gouramy, Osphronemus goruamy, mineral   ABSTRAK Penelitian ini dilakukan untuk menentukan kebutuhan ikan gurame (Osphronemus gouramy) akan mineral fosfor. Pakan yang digunakan dalam penelitian ini adalah pakan semimumi dengan kadar protein 41,0% dan rasio energi- protein 10 kkal/g protein. Pakan yang digunakan memiliki kandungan fosfor berturut-turut sebesar 0,32, 0,63, 0,89 dan 1,06%. Ikan gurame yang digunakan berbobot awal antara 21-25 g/ekor dan panjang tubuh awal 8,97-13,45 cm/ekor, dipelihara pada akuarium berukuran 50x40x35 cm dengan kepadatan 4 ekor/akuarium. Ikan diberi pakan 3 kali sehari secara satiation selama 40 hari. Setiap 10 hari sekali dilakukan penimbangan bobot tubuh ikan. Hasil penelitian menunjukkan bahwa performan pertumbuhan ikan gurame meningkat dengan meningkatknya kadar fosfor dalam makanan sampai pada tingkat 0,89%. Peningkatan kadar fosfor diatas tingkat tersebut berdampak negatif terhadap pertumbuhan. Pertumbuhan maksimal didapat pada ikan yang diberi pakan dengan kadar fosfor 0,63-1,06%. Abstract

Asam humat (AH) terdiri atas AH alami (AHA) dan sintetik (AHS), namun efektivitasnya sebagai feed additive pada pakan ikan kakap putih belum dikaji. Tujuan penelitian ini untuk mengetahui efektivitas penambahan AH pada pakan yang mengandung kadmium (Cd) dari kerang hijau Perna viridis terhadap status kesehatan dan pertumbuhan ikan kakap putih, Lates calcarifer. Percobaan dirancang menggunakan rancangan acak lengkap (RAL) dengan lima perlakuan dan tiga ulangan. Perlakuan terdiri atas lima pakan uji mengandung AH yang berbeda, yaitu 0; 1.600; 10.000; dan 20.000 mg.kg-1 (AHA) dan sebagai pembanding menggunakan AHS sebesar 1.600 mg.kg-1 pakan. Benih ikan kakap putih (4,18 ± 0,25 g) dipelihara dalam akuarium ukuran 80 cm x 35 cm x 28 cm yang diisi air laut dengan sistem resirkulasi selama 70 hari. Ikan diberi pakan uji sesuai perlakuan tiga kali sehari sampai kenyang. Hasil penelitian menunjukkan penambahan AH baik jenis AHA maupun AHS pada pakan dapat menurunkan akumulasi Cd dalam daging, ginjal, dan hati; kedua jenis AH tersebut mampu mengeliminasi Cd di dalam daging ikan. Pada dosis 1.600 mg.kg-1 kedua jenis AH tersebut mampu meningkatkan performa pertumbuhan dan kelangsungan hidup ikan, namun pada dosis AHA > 1.600 mg.kg-1 pertumbuhannya relatif menurun. Pola respons pertumbuhan ikan bersesuaian dengan parameter hematologi, enzim pencernaan, dan status antioksidan di hati ikan. Kesimpulan penelitian ini, penambahan asam humat pada dosis yang sama (1.600 mg.kg-1) pada pakan, AHS lebih efisien dibandingkan AHA dalam hal meningkatkan pertumbuhan. Penambahan AH dari jenis asam humat alami dan sintetik dalam pakan uji dapat meningkatkan status kesehatan dan mengeliminasi Cd di dalam daging ikan. Penambahan AHA pada dosis tinggi pada pakan memberikan respons negatif terhadap status kesehatan, kelangsungan hidup, dan kinerja pertumbuhan ikan kakap putih.Humic acids (HAs) are available in natural and synthetic forms. HA has potential applications in aquaculture, yet its effectiveness as a feed additive in Asian seabass, Lates calcarifer has not well studied. The purpose of this study was to asses the effectiveness of the addition of natural and synthetic humic acids to reduce cadmium (Cd) concentration in green mussels Perna viridis used for Asian seabass feed and evaluate the fish health status and growth performance. The experiment was designed using a completely randomized design (CRD) with five treatments and three replications. Five test feeds containing different levels of humic acid, i.e., 0; 1,600; 10,000; and 20,000 mg natural humic acid kg-1 feed. As a comparison, a test feed was added with 1,600 mg synthetic humic acid kg-1 feed. Asian seabass juveniles (4.18 ± 0.25 g) were cultivated in seawater aquarium equipped with a recirculation system for 70 days. Fifteen aquaria of 80 cm x 35 cm x 28 cm were used as the culture tanks. The fish were fed with the experimental diet three times every day at the satiation level. The results showed that the addition of both HAs (natural, AHA and synthetic, AHS) in feed could reduce Cd level in the fish meat, kidneys, and liver. At a dose of 1,600 mg.kg-1, both HAs were able to improve the growth performance and survival of fish. However, at doses > 1,600 mg.kg-1, fish growth was relatively suppressed. Fish growth response patterns were concomitant with the hematological parameters, digestive enzymes, and antioxidant status in fish liver. This study concludes that the addition of AHS at 1,600 mg.kg-1 feed is more efficient in terms of increasing growth compared with the same AHA level. The addition of HA, either natural and synthetic humic acid in the feed, can improve the health status of Asian seabass and eliminate Cd in the fish meat. The addition of AHA at higher doses (> 1,600 mg.kg-1 feed) might cause a negative response to health status, survival, and growth performance of Asian seabass.

Abstract

ABSTRACTThis study was conducted to evaluate stress responses of transportation on red tilapia Oreochromis sp. which given feed containing chromium. Three isonitrogenous and isocaloric experimental feeds were prepared, these diets were control (without chromium), CrPic 1 mg/kg, and CrYst 2 mg/kg supplementation in feed, all group were arranged triplicate. Satiation feeding was done three times a day. After a 60-day feeding experiment, the experimental fishes were fasted and distributed in polyethylene bags (N=60 fish/bag) containing 3 L of water, subjected to condition of transport simulation for 13 hours. Survival rate, levels of plasma cortisol, blood glucose, superoxide dismutase (SOD), and malondialdehyde (MDA) enzyme were observed at before transportation, after transportation, one day, and two days after transportation. The result showed that chromium supplementation reduced the levels of plasma cortisol before and after transportation, one day, and two days after transportation. Also, it decreased blood glucose compared with control significantly before transportation and one day after transportation. The SOD enzyme concentration increased significantly after fish was fed with feed containing chromium for 30 days, while the MDA enzyme concentration increased significantly after two days of transportation. However, there was no significant difference in the survival of red tilapia between treatments. The best result was obtained in the treatment of fish which fed with feed containing chromium. CrPic supplementation 1 mg/kg and CrYst 2 mg/kg increased the body resistance in red tilapia by decreasing the negative effect of stress while transportation. Keywords: stress, transportation, red tilapia, chromium  ABSTRAKPenelitian dilakukan untuk mengevaluasi respons stres transportasi ikan nila merah Oreochromis sp. yang diberikan pakan yang mengandung kromium. Pada penelitian ini digunakan tiga jenis pakan, terdiri atas pakan tanpa suplementasi kromium (kontrol), pakan bersuplementasi kromium pikolinat (CrPic 1 mg/kg), dan kromium yeast (CrYst 2 mg/kg), semua perlakuan diulang sebanyak tiga ulangan. Setelah 30 hari pemeliharaan, ikan uji dipuasakan dan didistribusikan dalam plastik polietilen (N=60 ekor ikan/kantong plastik) yang berisi 3 L air, dilakukan dengan simulasi transportasi selama 13 jam. Parameter yang diamati pada penelitian ini adalah kelangsungan hidup, kortisol, glukosa darah, enzim superoksida dismustase (SOD), dan malondialdehida (MDA) saat sebelum transportasi, sesaat setelah transportasi, sehari, dan dua hari setelah transportasi. Hasil yang didapatkan adalah suplementasi kromium menurunkan konsentrasi kortisol secara signifikan sebelum transportasi, sesaat, sehari, dan dua hari setelah transportasi. Suplementasi kromium menurunkan glukosa darah secara signifikan pada saat sebelum transportasi dan sehari setelah transportasi. Konsentrasi enzim SOD meningkat secara signifikan setelah pemberian pakan bersuplementasi kromium selama 30 hari, sedangkan konsentrasi enzim MDA meningkat secara signifikan setelah dua hari transportasi pada ikan yang diberi pakan bersuplementasi kromium. Namun, tidak ada perbedaan yang signifikan pada kelangsungan hidup ikan nila merah antarperlakuan. Hasil terbaik diperoleh pada perlakuan ikan dengan suplementasi kromium. Suplementasi 1 mg/kg CrPic dan 2 mg/kg CrYst dapat meningkatkan daya tahan tubuh pada budidaya ikan nila merah dengan menurunkan pengaruh negatif stres akibat transportasi. Kata kunci: stres, transportasi, nila merah, kromium

Abstract

ABSTRACT The aim of this study was to determine the effect of various types of honey (longan honey, lychee honey, and cottonwoods honey) on sperm quality of Siamese catfish Pangasianodon hypopthalmus with different dilution ratios after the storage period which includes sperm viability, fertilization rate, and hatching rate. The best treatment was obtained on the aplication of longan honey as a sperm diluent at 1:50 dilution ratio, with sperm viability 96.33±0.58%, fertilization rate 97.10±0.70%, and hatching rate 93.44±2.39%. Keywords: Siamese catfish, Pangasianodon hypophthalmus, honey, sperm dilution ratio  ABSTRAK Tujuan penelitian ini adalah mengevaluasi pengaruh pemberian berbagai jenis madu (madu kelengkeng, madu leci, dan madu randu) terhadap kualitas sperma ikan patin siam Pangasianodon hypopthalmus dengan rasio pengenceran berbeda setelah masa penyimpanan, yang meliputi viabilitas sperma, tingkat fertilisasi, dan derajat penetasan. Abstract

Motile aeromonad septicaemia (MAS) is caused by the bacterium Aeromonas hydrophila. The use of plant (natural materials) such as meniran and garlic can be as an alternative way to inhibit the activity of A. hydrophila by their active substances which have potency as an antibacterial and immunostimulant. The aim of this study was to analyze the effectiveness of using a mixture of meniran and garlic in feed to control of A. hydrophila in catfish. There were split in two kind of doses namely, preventive (garlic:meniran=5 ppt:20 ppt) and curative (garlic:meniran=10 ppt:40 ppt). The preventive treatment was given for two weeks before challenging test. The curative treatment was performed on 2th-8th day after challenging test.  Challenging test was carried out by intramuscularly injecting of 0,1 mL A. hydrophila (108cfu/ml)  into the fish. The results indicated that preventive treatment with a mixture of extracts 5 ppt meniran and 20 ppt garlic was more effective in preventing infection of A. hydrophila than curative treatment.  Keywords: Phyllanthus niruri, Allium sativum, catfish, Aeromonas hydrophila.  ABSTRAK  Penyakit MAS (motile aeromonad septicaemia) disebabkan oleh bakteri Aeromonas hydrophila. Bahan alami seperti meniran dan bawang putih dapat digunakan sebagai alternatif untuk menghambat aktivitas bakteri ini.  Zat aktif yang dimiliki bahan ini berpotensi sebagai antibakteri dan immunostimulan. Tujuan dari penelitian ini adalah untuk menganalisis efektivitas penggunaan campuran meniran dan bawang putih dalam pakan untuk pengendalian penyakit MAS pada ikan lele. Perlakuan dibagi menjadi dua dosis, yaitu pencegahan (bawang putih:meniran=5 ppt:20 ppt) dan pengobatan (bawang putih:meniran=10 ppt:40 ppt). Perlakuan pencegahan diberikan selama seminggu sebelum uji tantang. Perlakuan pengobatan dilakukan pada hari ke 2 hingga hari ke-8 setelah uji tantang. Uji tantang dilakukan dengan menyuntikkan secara intramuskuler 0,1 ml A. hydrophila (108cfu/ml) ke ikan lele. Hasil penelitian menunjukkan bahwa perlakuan pencegahan dengan campuran ekstrak meniran 5 ppt dan bawang putih 20 ppt efektif dalam mencegah infeksi A. hydrophila dibandingkan dengan perlakuan pengobatan dengan campuran ekstrak meniran 10 ppt dan 40 ppt bawang putih.   Kata kunci: meniran, bawang putih, lele, Aeromonas hydrophila.
