Abstract

ABSTRACTThe yellow sap is produced naturally in mangosteen organ except in the root. The yellow sap contaminated the aryl and rind if the epithelial cell walls rupture due to deficiency of calcium (Ca). Calcium is one of structural component of cell walls, whereas naphthaleneacetic acid (NAA) has its role in improving cell division and cell elongation. Interaction of Ca and NAA can improve sink strength and capacity because the newly formed cells need Ca to construct wall structure. This experiment aimed at finding out the effect of Ca and NAA applications in reducing the contamination of yellow sap in mangosteen. The experiment was conducted by using factorial random block design consisting of 2 factors and 3 replications. The first factor was Ca dosage (0 and 4.8 kg/tree), and the second factor was NAA concentration (0, 200, 400 and 600 ppm). The results showed that application of 4.8 Ca/tree and 200 ppm NAA as much as 5 ml / fruit effectively improve the content of Ca pectate in pericarp, reduced the percentage of yellow sap contamination on the fruit segment, aryl and rind to 0% and 12.3% respectively compared to control (17.8% on fruit segment, 36.8% on aryl and 56.1% on rind).Key words: aryl, Ca pectate, cell wall, middle lamela.ABSTRAKGetah kuning adalah getah yang dihasilkan secara alami pada setiap organ manggis, kecuali pada akar. Getah kuning akan keluar dan mencemari aril serta kulit jika dinding sel epitel pecah karena kekurangan kalsium (Ca). Kalsium adalah komponen dinding sel, berperan dalam struktur dan permeabilitas membran sedangkan asam naftalenasetat (NAA) berperan penting dalam meningkatkan pembelahan dan pembesaran sel. Interaksi keduanya dapat meningkatkan kapasitas sink buah karena sel yang baru terbentuk membutuhkan Ca dalam menyusun struktur dinding sel. Percobaan bertujuan untuk mengetahui pengaruh aplikasi Ca dan NAA dalam menurunkan cemaran getah kuning manggis. Percobaan menggunakan rancangan acak kelompok faktorial 2 faktor dengan 3 ulangan. Faktor ke-1 yaitu dosis Ca (0 dan 4.8 kg Ca/pohon) dan faktor ke-2 yaitu konsentrasi NAA (0, 200, 400 dan 600 ppm) dengan volume semprot 5 ml perbuah. Hasil percobaan menunjukkan bahwa aplikasi 4.8 kg Ca/pohon dengan NAA 200 ppm sebanyak 5 ml/buah efektif meningkatkan kandungan Ca pektat perikarp dan menurunkan persentase buah tercemar getah kuning menjadi 0% pada juring dan aril serta 12.3% pada kulit dibandingkan dengan perlakuan kontrol (17.8% pada juring, 36.8% pada aril dan 56.1% pada kulit buah).Kata kunci: aril, Ca pektat, dinding sel, lamela tengah

Abstract

ABSTRACTSomatic embryo induction medium was reported to be genotype dependent for soybean. This study was aimed to obtain the optimum medium for embryo somatic induction and proliferation, and to regenerate somatic embryo of five soybean genotypes. Five soybean genotypes (Tanggamus, Anjasmoro, Yellow Biloxi, CG-22-10, and SP-10-4) were used in this study. The research was divided into four steps: (1) embryogenic callus induction of  five soybean genotypes, (2) embryogenic callus proliferation of five soybean genotypes, (3) optimation of embryo somatic induction on five soybean genotypes and (4) embryo somatic regeneration of five soybean genotypes. The induction experiment showed that based on number of embryogenic callus, the best somatic embryo-induction medium was 3% sucrose+ NAA 5 mg L-1+2,4-D 5 mg L-1+ Vitamin B5. Embryogenic callus number for each genotype tested was increased on proliferation media of 3% sukrosa + 2,4-D 5 mg L-1 + NAA 5 mg L-1+ Vit B5, and Yellow Biloxi gave the highest number of proliferated somatic embryos compared to other genotypes. Increasing number of globular somatic embryo of all genotypes was obtained from the optimation of somatic embryo induction media being used, and Tanggamus genotype gave the highest number of globular somatic embryo which followed by Yellow Biloxi genotype. Tanggamus and Yellow Biloxi genotypes were also successfully formed the four steps of somatic embryos (globular, heart, torpedo, and cotyledonary stages), but in regeneration medium of MS0 and media MS + sukrosa 10 g L-1 + GA3 2 mg L-1 + BAP 4 mg L-1 + Vit B5 only Tanggamus genotype was regenerated into plantlet.  Keywords: 2,4-D, NAA, somatic embryos, induction, proliferation

The study was conducted in the greenhouse of the University Farm, Bogor Agricultural University from August  to October 2009.  The experiment was carried out as a Factorial experiment in a Completely Randomized Design with three replications. The ?  rst  factor was sorghum genotypes consisted of Numbu (tolerant) and B-75 (sensitive), and the second factor was combination of lime and phosphorous  fertilization consisted of: no lime-no P (R1), no lime-low P (R2), no lime-suf?  cient P (R3), lime-no P (R4), lime-low P (R5) and lime-suf?  cient P (R6). The results showed that all  variables were signi?  cantly affected by the interaction of  media and genotype, except for length of shoot and total P uptake.  Under low P and Al toxicity, sorghum variety Numbu showed a higher nutrient ef?  ciency than B-75 with ability of forming greater  dry mass. The sensitive genotypes showed a higher nutrient uptake but a lower P  nutrient use ef?ciency under   Al toxicity and P de? ciency.  Total P uptake and P nutrient use ef?  ciency were highly correlated with root growth and dry material accumulation. Abstract

Melastoma malabathricum L. grows well in acid soil with high level of soluble aluminum. One of the important proteins in the detoxifying acid and aluminum stress is a plasma membrane H+  -ATPase protein encoded by PMA gene. The objective of this research was to isolate and clone the cDNA fragment of MmPMA encoding plasma membrane H+ -ATPase from M. malabathricum L. By reverse transcription, total cDNA had been synthesized from the total RNA as template. The fragment of MmPMA  cDNA  had been successfully isolated by PCR by using total cDNA  as  template and PMA primer designed from conserved region for corresponding gene. This fragment had been successfully inserted into pGEM-T Easy and the recombinant plasmid was successfully introduced into E. coli DH5". Nucleotide sequence analysis showed that the length of MmPMA fragment is 806 bp encoding 268 amino acids. Local alignment analysis based on nucleotide of mRNA showed that MmPMA fragment was 81% identical to part of PMA of Sesbania rostrata, Juglans regia, and Prunus persica. Based on deduced amino acid sequence, MmPMA was 94% identical to part of PMA of Juglans regia; 93% to PMA of S. rostrata, and Arabidopsis thaliana. MmPMA fragment has phosphorylation intermediate domain (DKTGT) and ATP binding domain (KGAP, DPPR, MITGD, and GDGVN).   Keywords: isolation, Melastoma malabathricum L., MmPMA fragment, sequencing

Abstract

  Plant root has various responses for adaptation under low P condition. This study aims to observe information on root morphology responses from sorghum inbred lines under low P condition. This experiment used a randomized complete block design nested with two factors, namely genotype and P fertilizer dosage. The genotype consisted of 11 sorghum inbred lines (RIL-F8), B-69 (susceptible check), Numbu (tolerant check), and two national varieties (WHP and Kawali). Fertilizer dosage used a dose of sufficient P (66.76 kg ha-1 P2O5) and a dose of low P (33.38 kg ha-1 P2O5). The result showed there were differences in root morphological response between inbred lines (RIL-F8) sorghum under low P conditions. The tolerant lines had a higher character of the longest root length (41.33-61.87 cm), root volume (8.87-12.87 mL), root diameter (0.88-1.02 mm), and proteoid root total (2.4-4.0) as compared to sensitive lines In low P condition. These characters associated with the high biomass of tolerant lines in low P conditions, therefore it could be used as an important character for the selection of root adaptation in low P conditions. Abstract

The research on adaptation selection for utilization in phytoremediation of soil contaminated by crude oil using four type plants, such as Helianthus annuus, Paspalum conjugatum, Sorghum bicolor, and Tagetes erecta were conducted. The adaptability of four types of plants  on crude oil contaminated soil at total petroleum hydrocarbon (TPH) level at 0, 3, and 6% were observed and evaluated to their morphological and anatomical responses. Parameters observed were vegetative growth of plants including growth precentage, plant height, number of leaves, root length, root dry weight, shoot dry weight, root/shoot ratio, total dry weight, and stomatal density for 9 weeks cultivation in screen house. The results show that increasing in TPH level caused in significant reductions on morphological of four plants, such as percentage of plant growth, plant height, number of leaves, root length, root dry weight, shoot dry weight, and total dry weight. In contrast, the increasing in TPH level caused to increasing in root/shoot ratio. The four types of plants studied were effective to be used as plants for phytoremediation of petroleum contaminated soil. The plants of P. conjugatum and S. bicolor are recommended as phytoremediators for further studies.

Abstract

water deficiency. Genetic engineering at the level of transcription factors (TF) is particulary a promising strategy in developing drought tolerant rice cultivar. HD-Zip genes are TF that function in plant adaptation to some environmental stresses including water deficit. The recombinant plasmid pC1301H Oshox6 which contained HD-Zip Oshox6 gene was placed under a drought inducible promoter called LEA promoter, gusA and hpt genes were driven with CaMV promoter. The aim of research was to obtain indica rice transgenic plants of Batutegi and Kasalath cultivars using pC1301H Oshox6 plasmid. Recombinant plasmid was transformed into immature rice embryos using Agrobacterium tumefaciens. Kasalath cultivar showed a better capacity to form embryogenic calli compared to Batutegi. Transformation efficiency of Batutegi is lower (1.5 - 0.3%) than Kasalath (2.2-28.3%). Regeneration efficiency is 25-83.3% and 7.7-100% for Batutegi and Kasalath, respectively. Number of putative transformant plantlets of Batutegi and Kasalath are 63 and 48 plantlets, respectively. Southern blot analysis (using hpt probe) on 12 independent lines of each Batutegi and Kasalath cultivars showed different gene copy number, ranging from one to four copies of gene.   Keywords: agrobacterium tumefaciens, LEA promoter, HD-Zip Oshox6, rice

Abstract

The efectiveness of selection depends on the gene action cotrolling the character.  The presence of epistatic gene action will delay the genetic gain in self pollinated crops like soybean. The objective of this research was to obtain information on genetic control of soybean tolerance to aluminum toxicity on nutrient culture. This research was conducted at green house of  the University Farm IPB Cikabayan, Bogor, from April to June 2013. This research used 120 seedlings of F2 population  generated from  crossing between Argomulyo (Al-sensitive) with Tanggamus (Al-tolerant). Seedlings were grown in nutrient culture for 14 days with Al concentration of 1.5 mM and pH 4.0, to observe the vegetative growth under aluminum stress condition. The result showed that mean of roots length, shoot length, shoot to root ratio, wet and dry weight of root and shoot exceeded  both parents mean value with moderate to high heritability. All observed characters had continue distribution pattern, and controlled by many genes. Duplicate epistasis additive gene action controlled only root length while other characters were controlled by complementary epistasis gene action.Keywords: gene action, heritability, nutrient culture, selection, tocixity

Abstract

The aim of this study was to characterize full length sequence of JJ3 isolated from shade tolerant soybean. The characterization of full length sequence involved analyses of structure of full length sequence, open reading frame (ORF) or coding sequence (CDS), and homology. The analyses of JJ3 full length sequence structure were done using GeneScanW program, coding sequence (CDS) or open reading frame (ORF) using ORF Finder program, and the homology of JJ3 full length sequence were done using BLAST and clustalW programs. Results showed that the full length sequence of JJ3 consisted of single exon and polyA. Single exon was a coding sequence (CDS) 633 bp produced open reading frame (ORF) with 210 amino acid deduction comprised of 136 conserved amino acids and 77 varied amino acids. The conserved domain of JJ3 produced functional protein, PsaD photosystem I (PSI) subunit, related to photosynthetic transport electron in PSI reaction center. The full length of JJ3 homolog to psaD, a gene encoding protein subunit photosystem I (PSI) related to mechanism of plant adaptation to shade stress, in wood tobacco (Nicotiana sylvestris), rice (Oryza sativa), tomato (Lycopersicon esculentum), barley (Hordeum vulgare), spinach (Spinacia oleracea), Arabidopsis (Arabidopsis thaliana), and potato (Solanum tuberosum).Key words: full length sequence, gene, homology, shade tolerant, soybean
