Abstract

Makanan fungsional bertujuan untuk mencegah terjadinya penyakit melalui target fungsional tertentu didalam proses fisiologi dan metabolisme tubuh, diantaranya adalah melalui proses immunomodulasi. Pada penelitian ini dilakukan percobaan aplikasi ekstrak dan serbuk simplisia rumput laut Halimeda sebagai makanan fungsional untuk memodulasi sistem pertahanan non spesifik pada udang vannamei. Penelitian dilakukan dengan metode eksperimental laboratoris menggunakan rancangan acak lengkap. Halimeda ditambahkan dalam pakan sebanyak 1%. Pakan diberikan sebanyak 5% berat badan per hari yang diberikan dalam tiga kali (pagi, sore dan malam). Parameter sistem pertahanan non spesifik udang diamati melalui penghitungan jumlah total hemosit dan aktivitas fagositosis hemosit. Selama penelitian juga dilakukan pengukuran kualitas air (salinitas, pH dan suhu) secara harian. Hasil penelitian menunjukan bahwa pemberian ekstrak Halimeda memberikan pengaruh dengan meningkatkan jumlah total hemosit 22,43% (hari ke-8) dan 96,24% (hari ke-12). Tetapi pemberian serbuk simplisia memberikan pengaruh lebih baik dengan meningkatkan jumlah total hemosit sebesar 76,18% (hari ke-8) dan 170,11 % (hari ke12). Hasil pengamatan aktivitas fagositosis menunjukan bahwa pemberian ekstrak dan serbuk simplisia Halimeda memberikan pengaruh meningkatkan aktivitas fagositosis pada pengamatan hari ke-12 yaitu berturut turut sebesar 35,75% dan 48,38% sehingga dapat disimpulkan bahwa Halimena mampu memodulasi sistem pertahanan non spesifik pada L. vannamei. Kata kunci : Halimeda, makanan fungsional, sistem pertahanan non spesifik, L. vannamei Functional foods should benefically affect one or more target functions in the body, among others is through immunomodulation process. This research was conducted to determine the effect of hot-water extract and powder simplicia of Halimeda sp. as functional foods to modulate non-specific defense system in vannamei shrimp. Research was carried out by laboratory experimental methods using a complete randomized design. Halimeda was incoorporated in the feed at concentrations of 1%. The feed was given as much as 5% of body weight per day given in three times (morning, afternoon and evening). Parameters of non-specific defense system of shrimp were observed by counting the total number of hemosit and hemosit phagocytosis activity. During the study also measured water quality (salinity, pH and temperature) daily. The results showed that administration of Halimeda extract increased total hemocyte count of 22.43% (at day 8) and 96.24% (at day 12). While administration of powder simplicia increased total hemocyt count  of 76.18% (at day 8) and 170,11% (at day 12). Phagocytosis activity parameter indicate that administration of extracts and powders simplicia Halimeda increased phagocytosis activity on the observation of 12 days consecutive for 35.75% and 48.38% and could be concluded that Halimeda sp. was able to enhance non-specific defence of  L. vannamei Key words : Halimeda, functional food,  non-spesific defence system, L. vannamei

Abstract

Infeksi bakteri patogen menjadi masalah kesehatan dunia. Antibiotik untuk penanggulangan infeksi menjadikan bakteri resisten karena pemakaiannya tidak sesuai dosis. Permasalahan ini dapat diatasi salah satunya dengan pencarian sumber antibiotik baru termasuk diantaranya pada rumput laut. Penelitian ini bertujuan untuk menentukan aktivitas antibakteri ekstrak metanol G. verrucosa terhadap Escherichia coli, Bacillus cereus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Vibrio anguillarum, dan Vibrio alginolyticus. Penelitian dilakukan dengan metode eksperimental laboratoris. Sampel rumput laut hasil budidaya Balai Besar Perikanan Budidaya Air Payau (BBPBAP) Jepara dikeringkan kemudian dimaserasi dengan metanol selama 2x24 jam. Aktivitas antibakteri ekstrak diuji pada konsentrasi 100 µg/disk, 50 µg/disk, 25 µg/disk, dan 10 µg/disk kemudian dilakukan uji fitokimia. Hasil uji aktivitas antibakteri menunjukkan bahwa ekstrak metanol G. verrucosa tidak memiliki aktivitas antibakteri. Uji fitokimia menunjukkan ekstrak mengandung flavonoid, saponin, steroid, dan triterpenoid. Pathogenic bacterial infection become a world health problem. Antibiotics for prevention of infection make bacteria resistant due to the incorrect dosage. One of these problems can be overcome by finding a new source of antibiotics including seaweed. This study aims to determine the antibacterial activity of methanol extract of G. verrucosa against Escherichia coli, Bacillus cereus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Vibrio anguillarum, and Vibrio alginolyticus. Laboratory experimental method was used in this study. Seaweed samples from the cultivation at Brackish Water Aquaculture Center (BBPBAP) Jepara were dried and then macerated with methanol for 2x24 hours. The antibacterial activity of the extract was tested at concentration of 100 µg / disk, 50 µg / disk, 25 µg / disk, and 10 µg / disk then phytochemical tests were carried out. The results of antibacterial activity test showed that G. verrucosa methanol extract did not have antibacterial activity. The phytochemical test showed that seaweed extract contained flavonoids, saponins, steroids, and triterpenoids

Abstract

Blue crab is one of the economically marine commodities and have high potential export. During this blue crab requirement derived from nature catches because in blue crab aquaculture not yet good branch out. To low survival rate of blue crab caused by bacterium vibrio disease, in addition to the content of nitrogen and phosphate can also affect the blue crab medium. This research as a purpose to test the effect of probiotic bacillus aplication to water quality and the total number of bacteria in the blue crab medium. the research was conducted with 2 stages, that is isolation and selection bacillus bacteria stages as probiotic and testing phase the effect of probiotic bacillus on water quality and the total number of bacteria. The result showed that probiotic bacillus with density 106 cfu/ml to reduce the content of ammonia (NH3), nitrite (NO2) and the total number of bacteria. While probiotics with density 105 cfu/ml to reduce the total number of bacteria in the blue crab medium, but have not been able to reduce the content of ammonia (NH3) and nitrite (NO2).

Abstract

Munculnya permasalahan lingkungan akibat plastik sintetik mendorong perlunya penelitian bahan kemasan mudah terurai (bioplastik). Salah satu bahan yang tersedia di alam dan berpotensi sebagai pembentuk bioplastik adalah alginat karena mampu membentuk gel. Bioplastik dari alginat memiliki sifat yang tidak fleksibel sehingga perlu ditambah gliserol sebagai plasticizer. Penelitian ini bertujuan untuk mengetahui pengaruh konsentrasi alginat dari Sargassum sp. terhadap sifat fisik dan mekanik bioplastik dengan penambahan gliserol dan CaCl2. Materi penelitian ini adalah alginat hasil ekstraksi Sargassum sp. yang diambil dari Perairan Pantai Sundak, Gunung Kidul. Penelitian ini menggunakan metode eksperimental laboratoris yang terdiri dari empat perlakuan konsentrasi alginat (0,5%, 1%, 3% dan 5%)  dengan tiga pengulangan.  Sifat bioplastik yang diuji meliputi sifat fisik (ketebalan, ketahanan air, kehilangan berat) dan sifat mekanik (kuat tarik dan elongasi). Data yang diperoleh dianalisis menggunakan analisis sidik ragam (ANOVA). Hasil penelitian menunjukkan semakin tinggi konsentrasi alginat akan meningkatkan  ketebalan, ketahanan air dan kuat tarik bioplastik namun menurunkan elongasi dan kehilangan berat bioplastik. Konsentrasi alginat berpengaruh terhadap sifat fisik (ketebalan, ketahanan air, kehilangan berat) dan sifat mekanik (kuat tarik  kecuali elongasi), konsentrasi alginat terbaik yang memenuhi Japanese Industrial Standard dan standar SNI 7188.7:2016 adalah 0,5% The emergence of environmental problems due to synthetic plastics has led to the need to research biodegradable packaging materials (bioplastics). One of the ingredients available in nature and has the potential to form bioplastics is alginate because it is capable of forming gel. Bioplastic from alginate has inflexible properties so it needs to be added by glycerol as a plasticizer. This research aims to determine the effect of alginate concentration from Sargassum sp. on the physical and mechanical properties of bioplastics with the addition of glycerol and CaCl2. The method of this research is alginate from Sargassum sp. taken from Sundak Beach Waters, Gunung Kidul. This research used a laboratory experimental method consisting of four treatments of alginate concentration (0.5%, 1%, 3% and 5%) with three repetitions. The properties of the bioplastics tested included physical properties (thickness, water resistance, weight loss) and mechanical properties (tensile strength and elongation). Data obtained were analyzed using variance analysis (ANOVA). The results showed that the higher the concentration of alginate would increased thickness, water resistance and tensile strength of bioplastics but decreased the elongation and weight loss of bioplastics. The alginate concentration affects the physical properties (thickness, water resistance, weight loss) and mechanical properties (tensile strength and except elongation), the best alginate concentrations that meet the Japanese Industrial Standard and SNI 7188.7: 2016 are 0.5%

Abstract

ABSTRAK : Rajungan (Portunus pelagicus) memiliki nilai ekonomi tinggi, dimana berbanding lurus dengan penangkapan yang meningkat. Tingkat pemanfaatan yang tidak mengindahkan ukuran dan kondisi rajungan dapat mempengaruhi struktur ukuran dan stok rajungan di suatu perairan. Penelitian ini bertujuan untuk melakukan kajian morfometri, rasio jenis kelamin dan tingkat kematangan gonad rajungan yang didaratkan di Kelurahan Pacar, Kabupaten Rembang. Metode yang digunakan dalam penelitian bersifat deskriptif dan eksploratif. Pengamatan rajungan dilakukan pada salah satu pengepul di Kelurahan Pacar. Rajungan yang diteliti berdasarkan morfometri, rasio seksual dan tingkat kematangan gonad selama 30 hari. Informasi ini dapat dijadikan pedoman dalam menentukan kebijakan mengenai pengelolaan perikanan rajungan yang berkelanjutan. Hasil dari penelitian diketahui bahwa ratio kelimpahan rajungan jantan sebesar 45% (1342 ekor); rajungan betina 55% (1658 ekor) dari 3000 ekor sampel rajungan. Rasio perbandingan jantan dan betina 0,80:1 berdasarkan uji chi square. Berdasarkan distribusi ukuran lebar karapas rajungan di perairan Kelurahan Pacar adalah berkisar antara 60?179 mm dan distribusi berat sebesar 23?251 gram. Distribusi tingkat kematangan gonad pada perairan Kelurahan Pacar adalah 558 ekor pada TKG 1; 761 ekor pada TKG 2; dan 339 pada TKG 3. Data menunjukkan bahwa pola pertumbuhan rajungan yang ada di perairan Kelurahan Pacar adalah allometrik negatif yaitu pertumbuhan lebar karapas lebih cepat bertambah daripada berat tubuhnya baik rajungan jantan maupun rajungan betina.ABSTRACT : Blue swimming crab (Portunus pelagicus) has a high economic value, which is causing the crab catch to rise as well. The catch rate that does not consider the size and maturity of sea crab can affect the size structure and population stock of sea crab in waters. This research to study morphometry, sexual ratio and maturity level of the gonads landed in Pacar Village, Rembang Regency.The method used in this study descriptive and exsplorative. Observation of blue swimming crab samples was carried out in one of the collectors in the village. Blue swimming crab was observed based on morphometry and sexual ratio for 30 days. The results of the study revealed that the abundance of male blue swimming crab was 45% (1342 male blue swimming crabs) and the abundance of the female blue swimming crabs was 55%  (1658 female blue swimming crabs) of 3000 blue swimming crab samples. The ratio of male and female   0.80 ; 1 based on the chi square. Based on the size distribution of the width of the blue swimming crab carapace in the waters of Pacar Village it range from 60-179 mm and the distribution of weight 23-251 grams. The distribution of gonad maturity level in the waters Pacar Village is 558 female blue swimming crab at TKG 1; 761 female blue swimming crab at TKG 2; and 339 female blue swimming crab at TKG 3. Data is showed that the blue swimming crab growth pattern in Pacar Village waters was negative allometric both male and female blue swimming crab.

Abstract

The shrimp fishery has a high economic value. Data from the Ministry of Maritime Affairs and Fisheries (2011) states that the group of marine crustaceans in Indonesia has high potential with production reaching 381,288 tons / yr. From these results more than half of its cultivation. Aquaculture has the risk of which is the onset of the disease caused by viruses, fungi, bacteria, fungi and parasites that cause high levels of mortality and loss of quality of meat. One way to overcome these problems is to use feed supplementation. This study aimed to determine the effect of supplementation of extracts G. verrucosa on feed on shrimp growth (weight and length), specific growth rate, survival rate of shrimp, feed conversion ratio, and Vibrio bacterial populations. The research was conducted through laboratory experimental method to the study design completely randomized design (CRD) with 3 treatments and each treatment with three replications. The treatments tested were feed by supplementing 0.5%, 1% and 2% respectively supplementation performed 3 times repetition. Feed obtained by extraction G. verrucosa, that is by G. verrucosa put in a pot submerged Stainless steel up to boiling and boil for ± 2 hours and obtained extracts G. verrucosa, then performed using a rotary solvent evaporation process to obtain dry extracts G. verrucosa. Further supplementing the feed. Results showed that the extract of G. verrucosa able to improve and control the growth of white shrimp vibrio bacteria populations in the intestines white shrimp (L. vannamei). In this study the condition of seaweed extract supplementation G. verrucosa at a concentration of 2% were able to increase the weight of shrimp growth of 131.43% and a growth of 32.50% shrimp length than controls during 28 days of treatment, as well as being able to control populations of bacteria vibrio, vibrio that reduces the total amount of 78.18% compared to controls at the third week.

Abstract

Penelusuran bakteri laut yang berasal dari perairan pulau panjangyang mempunyai aktivitas antagonis terhadap beberapa jenis bakteri Vibrio penyebab vibriosis pada ikan dan udang bertujuan untuk mendapatkan kandidat bakteri sebagai agen pengendali hayati penyakit vibriosis. Tolok ukur seleksi pada penelitian ini adalah kemampuan menghambat pertumbuhan bakteri vibrio pathogen dan bersifat non patogenik terhadap ikan dan udang. Bakteri diisolasi dari sedimen dan air laut, rumput laut, lamun, karang keras dan karang lunak . Uji aktivitas antagonis terhadap bakteri patogen (V. harveyi, V. fluvialis, V. alginolyticus, V. parahaemoliticus, V. anguilarum) dilakukan dengan metode tusukan dan overlay. Hasil uji aktivitas antagonis didapatkan 15 isoiat aktif terhadap V. harveyi, 25 Isolat aktif terhadap V. parahaemoliticus, dan 9 isolat aktif terhadap V. anguilarum, dan 6 isolat aktif terhadap V. Fluvialis, dan tidak ditemukan isoiat aktif terhadap V. alginoliticus. Kata kunci: Vibrio-antagonistic bacteria, agen pengendali hayati, vibriosis.

Abstract

Often the cut side of the plate pieces on the lever scissors is not good (distortion occurs). This is due to the incompatibility of cutting variables, such as clearance and cutting angle of both scissors knife. The extent to which the effect of the clearance and the cutting edge of the scissor knife on the cutting of the steel plate is to be studied to obtain the appropriate cutting parameter as reference in the slashing work of the plate with the lever scissors.This research is aimed to know the proper clearance and cutting angle for various variations of steel plate thickness. The thickness of the steel plate as a specimen of the test specimens was of a size 1.61 mm, 2.63 mm, 3.85 mm, 4.95 mm and 5.39 mm, respectively. While the variation of cutting angle of scissor knife include: 75o, 77,5o, 80o, 82,5o and 85o.The test results showed that cutting of 1.61mm thick steel plate with 0.5 mm clearance and 77,5o cutting angle resulted in good cuts with 45% incision depth. On steel plates with thickness of 2.63mm and 3.85mm, the best cut results at 1mm clearance and cutting angle of scissors knife 77,5o, where the depth of incision successively reached 35% and 25%. For thick plates 4.95 mm and 5.39 mm, the best results of the incision depth were respectively 20% and 18% on clearance 2 mm and cutting angle 82,5o.

Abstract

Strategi penyehatan ikan secara terpadu merupakan salah satu upaya yang paling efektif dalam pengendalian penyakit serta perlindungan lingkungan pada budidaya akuatik.  Pengembangan probiotik menduduki peran fungsional yang penting bersamaan dengan pengembangan vaksin dan immunostimulan. Penelitian ini bertujuan untuk menseleksi bakteri probiotik yang akan dikembangkan sebagai materi dasar untuk mengkonstruksi konsorsium probiotik dengan target fungsional di saluran pencernaan ikan kerapu. Salah satu kriteria seleksi ditetapkan berdasarkan desain konstruksi konsorsium gut probiotik yaitu kemampuan menghasilkan senyawa antibakteri terhadap bakteri pathogen dan kemampuan menghasilkan enzim pencernaan diantaranya adalah enzim proteolitik ekstraseluler. Penelitian ini dilakukan dengan metode eksplorasi dan eksperiment laboratoris. Ikan kerapu macan secara aseptic diambil saluran pencernaannya, kemudian dihancurkan menggunakan mortar. Penanaman bakteri dilakukan dengan metode pour-plate pada medium nutrient agar. Deteksi aktivitas antibakteria dilakukan dengan metode difusi agar menggunakan paper disk sedangkan deteksi produksi enzim proteolitik ekstraseluler dilakukan dengan metode tusukan pada media yang diperkaya dengan skim milk. Hasil seleksi berdasarkan kriteria seleksi yang telah ditetapkan diperoleh 8 isolat bakteri potensial untuk dapat dikembangkan sebagai konsorsium probiotik. Ke 8 isolat ini mempunyai kemampuan untuk  menghasilkan senyawa antibakteri yang aktif terhadap 4 jenis vibrio (V. parahaemolyticus, V. vulnivicus, V. harveyii, dan V.anguilarum) serta menghasilkan enzim proteolitik ekstraseluler yang diperlukan untuk mencerna senyawa yang bersifat protein yang merupakan komponen utama pakan ikanKata kunci: Probiotik, pengendalian penyakit, antibakteri, enzim proteolitik, saluran  pencernaan, ikan kerapu The strategy to healhty fish in an integrated manner is one of the efforts that is the most effective to environmentally friendly disease control. The development of probiotics occupied the functional role that is important along with the development of the vaccine and immunostimulant. This research aimed to select the candidates of probiotic bacteria that will be developed as basic material to construct the probiotic consortium with the functional target in the digestion tract  of the grouper fish. The one of selection criteria was appointed based on the construction of the gut probiotic consortium is the capacity to produce the antibacterial compound against the pathogen, and the capacity to produce proteolitic enzymes. The research was carried out with the exploration and experimenal laboratory methods The intestine was removed from the fish of the tiger grouper, afterwards was destroyed by mortar. The planting of the bacteria was carried out with the pour-plate method in nutrient agar medium. Antibacterial activity was detection by agar diffusion method using paper disk, while the detection capability to produce extracellular proteolitic enzymes was carried out by using enrichment media with skim milk. The Results of selection obtained eight bacterial isolates that can be developed potentially as the consortium of gut probiotic. Abstract

Lactid Acid bacteria are capable of fermenting sugars or carbohydrates to produce lactic acid in large numbers. The characteristics of lactic acid bacteria in general are cells reacted positively to the Gram  stain, catalase react negatively and do not form spores. And fermentation of glucose would result Iaktat acid. Isolation of lactic acid bacteria carried in the intestines of three species of shrimp that Tiger Shrimp (Penaeus monodon), shrimp Seker (Metapenaopsis sp.), Guts White shrimp (Penaeus merguensis). Selection of bacteriocin -producing lactic acid bacteria were in a qualitative and semi-quantitative (pitting) in MRS medium. Selected isolates were identified by morphological characteristics, biochemical, physiological, A number of 209 isolates were isolated from three species of shrimp. The results of the qualitative selection there were 54 isolates produced bacteriocins. The results of the quantitative selection of 54 isolates produced 24 fine isolates.  From the 24 isolates there are 2 isolates seed SFE-7 (33) and P12A (25). Characteristics of seed isolates had morphological characteristics of spherical shape, cell arrangement tetrad, gram positive, negative motility. While the results of the biochemical approach to gram-negative, catalase negative,  homofermentative (no gas in the fermentation of glucose), can ferment D-Galactose, D-Glucose, D-mannose, D-lactose. Physiological approaches to isolate growth SFE-7 (33) and P12A (25) can be generated that both isolates able to live in the range of 4 ° C to 50 ° C, in the range of pH between 4-10, and able to grow at levels of 5-10% NaCl .
