Abstract

ABSTRACTInsulin resistance is a common condition that is believed to play a major role in the pathogenesis of the metabolic syndrome, obesity and type 2 diabetes mellitus, and heart disease. The purpose of this study was to determine the antidiabetic activity mellitus combination of purified extract of bitter herbs and artemisia herb in rats with type 2 diabetes mellitus insulin resistance compared with the effect of each extract.Test activity of purified extract of bitter herbs, herbal extracts purified artemisia and combinations thereof done by grouping test animals which have been declared insulin resistance is divided into 7 groups, Group I: a combination of purified extract of bitter herbs 390.9 mg / kg and 83.56 mg of the herb artemisia / kg; Group II: a combination of purified extract of bitter herbs 651.5 mg / kg and herb artemisia 41.7 mg / kg; Group III: purified extract of bitter herbs single dose of 651.5 mg / kg rat; Group IV : purified extract of bitter herbs single dose of 1303 mg / kg, po; Group V: purified extract of the herb artemisia single dose 41.78 mg / kg, po; Group VI: purified extract of the herb artemisia single dose 83.56 mg / KgBW , po; Group VII: positive control (given metformin dose of 45 mg / kg, po); Group VIII: negative control (CMC-Na was given 1%, po). Then the results were analyzed with SPSS 16 statistical test.The percentage decrease preprandial blood glucose levels of administration of a single dose of artemisia extract 83.56 mg / KgBW better than the combination of bitter and herbal28extracts artemisia, while the percentage reduction in postprandial glucose levels in the administration of a combination of extracts of bitter herbs bitter dose of 651.5 mg / kg BB artemisia herb and 41.7 mg / kg better than the extract of bitter herbs and herbal extracts artemisia singly.Keywords: bitter herbs, Artemisia herb, diabetes mellitus, glucose levels

Abstract

ABSTRACT Triglycerides (triacylglycerols) is the major lipid compounds in body fat and food deposits. Hypertriglyceridemia is a disorder characterized by an  increase of triglyceridesconcentration  in the blood. Tuber purse known as  Umbi Kimpul(Xanthosomaviolaceum Schott.) is believed as a medicinal plant. This study aims to determine the effect of hipotrigliseridemiaof purse tuber flour and the effective dose. To determine the effect of hipotrigliseridemia of purse tuber flour, 30 white male Wistar rats were used and they were divided into 6 groups. Group I (normal control) was  fed a standard food, group II (negative control) was given suspension CMC Na 0,5%, group III (positive control) was given suspension gemfibrozil 54 mg / kgbody weight of mice, group IV, V and VI were given pursetuber flour wtih graded dose at 270, 540 and 1080 mg / kgbody weight of mice. The Induction of hypertriglyceridemia was  a mixture of standard rat food with 15% lard and 5% egg yolk of duck, as well as fructose 1.8 g / kg body weight of mice. The Induction was conducted for 50 days, unless  the normal control group. Blood triglyceride levels were measured on days 0, 51 and 58. The triglyceride levels were calculated their decline percentagebefore statistically tested using SPSS statistical release16. The results showed a significant difference in the negative control group CMCNa suspension with purse tuber flour suspension. It is concluded that  the purse tuber flour suspension has an effect of hipotrigliseridemia. The effective dose of pursetuber flour as hipotrigliseridemia is 270 mg / kg body weight of mice. Abstract

ABSTRAK   Kedelai (Glycine max L. Merril)   tidak hanya digunakan sebagai sumber protein, tetapi juga sebagai pangan fungsional yang dapat mencegah timbulnya penyakit-penyakit degeneratif, dikarenakan kandungan isoflavon. Senyawa ini dikenal memiliki kesamaan molekul dengan estrogen. Penggunaan isoflavon sebagai alternatif hormon konvesional terapi telah meningkat dalam beberapa tahun terakhir, karena aktivitas estrogenik dan efek samping yang rendah.  Daidzein dan genistein merupakan  isoflavon yang banyak  terdapat dalam kedelai.  Penelitian ini bertujuan untuk mengetahui kandungan  daidzein dan genistein pada  kedelai  (Glycine max L. Merril) varietas Anjasmoro, Argomulyo dan  Gema yang dihasilkan dari Balai Penelitian Tanaman Aneka Kacang dan Umbi (Balitkabi) Malang. Penyarian senyawa aktif digunakan metanol hasil optimasi, selanjutnya dianalisis kandungan daidzein dan genistein dengan Kromatografi Cair Kinerja Tinggi (KCKT). Sistem KCKT yang digunakan dilengkapi kolom RP-C18 Sun Fire TMC-18 (150 mm x 4,6 mm,5µm),  detektor Photo Dioda Array (PDA), sistem elusi isokratik,  fase gerak  metanol-air yang mengandung asam asetat 0,1 % (53:47),  kecepatan alir 1,0 mL/menit. Hasil penelitian menunjukkan bahwa kandungan daidzein dan genistein pada kedelai (Glycine max L. Merril) varietas Anjasmoro, Argomulyo dan Gema diperoleh kadar daidzein dan genistein masing-masing adalah 18,69 mg/100g dan 23,67 mg/100g; 29,68 mg/100 g dan 22,15 mg/100 g;  14,15 mg/100 g dan 21,22 mg/100 g.
