Abstract

Deoxyribonucleic acid (DNA) vaccine has recently been developed as an alternative vaccine against virus infection.This study was the first step of DNA vaccine development to protect cyprinids including common carp (Cyprinuscarpio) and fancy koi (Cyprinus carpio) from KHV (koi herpesvirus) infection in Indonesia. One of KHV glycoproteingenes, i.e. glycoprotein (GP) was ligated with Japanese medaka (Oryzias latipes) â-actin promoter to generatepAct/GP as a DNA vaccine. Fourty fish in body weight of 10-15 g/fish were individually injected by pAct/GP intomuscle in different dosage of 2.5 ?g, 7.5 ?g and 12.5 ?g/100 ?l phosphate buffer saline. Total RNA was extractedfrom the 12.5 ?g of pAct/GP-injected fish muscle at 24, 48 and 67 hours post-injection to analyze GP expression byRT-PCR method. Potential of pAct/GP as DNA vaccine was examined by injecting KHV into the 30-days-vaccinatedfish. Both of possitive and negative control fish group were not vaccinated. Possitive control fish group wereinjected with KHV, but negative control fish group were not. KHV-challenged fish were reared for 1 month, and thedeath fish were calculated daily. Result of RT-PCR analysis showed that GP gene expression were detected at 3 dpost-injection. Expression of GP in the vaccinated fish groups helped to improve their survival rate after challengedby KHV. All of fish without DNA vaccination had dead 17 days after KHV injection. The results demonstrated thatpAct/GP had high potency to be used as a DNA vaccine against KHV infection in cyprinids.

Abstract

This study was conducted to examine activity of medaka (Oryzias latipes) â-actin promoter (mBP) in common carp(Cyprinus carpio) as the first step towards development of common carp transgenic in country. Gene constructpmBP-hrGFP that consists of mBA promoter and humanized Renilla reniformis green fluorescent protein gene(hrGFP) was injected into cytoplasm of one cell stage of common carp by using microinjector. PmBP-hrGFPconcentration used for microinjection was 50 ?g/mL aquabides. Parameters observed were survival rate of embryo(SRe), hatching rate (HR) and expression of hrGFP gene. SRe was calculated before eggs hacthed, while hatchingrate (HR) was after all of eggs hatched. The activity of mBA promoter was analyzed by observation of hrGFP genetransient expression using a fluorescence microscope. The results of experiment showed that SRe (87,5%) andHR (79.2%) of control was respectevily higher than that of injected treatment (75.0% & 61.7%). Expression of hrGFPwas observed firstly at blastula (12 hours after fertilization) to 1-day-old larval stages (24 hours after hatching)with higher gene expression at blastula to late gastrula stages. Percentage of micronjected larvae expressinghrGFP at 6 hours after hatching reached 71.6 ± 6.7%. Conclusion was that mBA promoter could drove hrGFPexpression in common carp, hence it can be used to produce common carp transgenic by changing hrGFP withgenes correlated with important traits in aquaculture.

Abstract

The world market demand for fishery products is increasing even frequently not fulfilled. It required an effort to overcome the problems in fulfilling the demand for fishery products. Types of freshwater fish cultivated for balita fish at UD Suhada is a type of tilapia and goldfish. The purpose of this study is to examine the feasibility of balita fish agribusiness investment in UD Suhada; identify the internal and external factors that influence the development of agribusiness balita fish at UD Suhada; formulate and recommend appropriate alternative development strategies to be applied to agribusiness balita fish at UD Suhada in business development. Methods of data collection using purposive sampling method, is by deliberately selecting the sample to be studied as a respondent. Respondents were selected from the management and the workers UD Suhada. Financial analysis at 14 percent of interest rate and 5 years of project lifetime resulted in feasible decision with Rp1,105,005,110 of NPV; 4.85 of net B/C Ratio; 38.95 percent of IRR, 3 years and 5 month of PBP; and Rp1,522,035,876 or 32.725 kg of BEP. The combination of value IFE EFE values for 2.688 and 2.651 in the IE matrix shows that the position of the business in V (five) cells, that cell growth. Based on the SWOT analysis and QSPM analysis the strategic priorities is to create a diverse variety of products while maintaining the quality of balita fish, extending the range of distribution and marketing, in collaboration with researchers in developing the quality of balta fish products to face competition.Key words: balita fish, business feasibility, agribusiness, business development strategy

Abstract

Pecel Lele Lela Restaurant branches Pinangranti  is a restaurant that first presents the traditional Deep-fried Catfish food menu into a unique concept with modern restaurant management. This Research aims to (1) find out feasibility, (2) understand the marketing mix of services and assessments of consumer againt services marketing mix so far applied,  (3) analysis the  factors of internal and external environment that affect marketing strategies, and (4) establish  the  priority of alternative marketing strategies that can be  applied. Sampling technique in this research is purposive sampling. The method used in this research is descriptive method that focuses on invesment and business development aspect, as well as NPV, B/C, PBP, IRR, SWOT and QSPM analysis. Based on financial criteria feasibility on discount rate of 6% NPV Rp2.535.752.046. Net B/C 4,22. IRR 21,34% (IRR > 6%).The return on investment (PBP) which is based on the value of now with the discount rate of 6% are 1.81. It means that the restaurant pecel lele lela branch Pinangranti, East Jakarta worthy to be implemented.The resulted total scores value of both Internal Factor Evaluation (IFE) and External Factor Evaluation (EFE) were respectively 2,543 and 2,310 which puts the position on quadrant V, which hold and maintain. Abstract

Perairan Teluk Jakarta, Banten, dan Lada telah mengalami pencemaran, terutama tercemar oleh logam berat seperti Pb, Cd, Cr dan Hg. Penelitian ini bertujuan untuk mengetahui kandungan logam tersebut dalam gonad kerang hijau (Perna viridis) dan pengaruh akumulasi logam tersebut pada spermatogenesis kerang hijau. Parameter yang diamati adalah jumlah spermatogenium, spermatosit primer, spermatosit sekunder, spermatozoa, total se-sel kelamin, diameter, luas dan volume lumen folikel gonad kerang hijau, pada tingkat IV spermatogenesis. Hasil penelitian menunjukan bahwa gonad kerang hijau yang berasal dari Teluk Jakarta telah terakumulasi logam berat Pb = 359.75±272.41 ppb; Cd = 36.559±21.90 ppb; Cr = 504.21±448.64 ppb dan Hg = 0.0092± 0.0085 ppb. Logam Cd (13.13 ppb) dan Pb (0.077 ppm) ditemukan dalam gonad kerang hijau yang berasal dari Teluk Banten, tetapi tidak terdeteksi logam Cr dan Hg. Namun tidak ditemukan logam berat tersebut dalam gonad kerang hijau berasal dari Teluk Lada. Di Teluk Jakarta ditemukan korelasi antara kandungan logam Cd terhadap perkembangan jumlah sel spermatozoa (r = 0.64), total sel kelamin (r = 0.60, diameter (r = 0.57), luas (r = 0.71) and volume (r = 0.71) lumen folikel gonad. Logam Hg berpengaruh terhadap perkembangan spermatogenium (r = 0.68) dan spermatosit sekunder (r = 0.61), sedangkan logam Cr mempunyai pengaruh terhadap luas (r = 0.75) dan volume (r = 0.75) lumen folikel gonad.Kata kunci: akumulasi, logam berat, spermatogenesis dan kerang hijau.

Abstract

The one commodity that is highly prospective for cultivated on an industrial scale and households are catfish (Clarias sp.). The purpose of this study was (1) analyze the feasibility of catfish farming in dry land in the district of Gunung Kidul Yogyakarta Province, (2) analyze the potential of catfish farming in ponds tarp on dry land, (3) develop alternative business development in the district of Gunung Kidul Yogyakarta Province. The method used in this thesis is descriptive method that focuses on technical aspects and business cultivation, as well as a SWOT analysis. Based on the feasibility analysis has a promising prospect and deserves to be developed. To maximize revenue catfish farmers, a combination of strategies SO, WO Strategies and strategy ST is an appropriate strategy to be chosen by farmers to improve their business. Increase the number and extent of ponds and develop farming, applying methods of maintenance and cultivation of good, as well as expand market coverage ranging from individual consumers, restaurants and traditional markets to restaurants to modern markets to improve capital efficiency and increase profits for farmers catfish on dry land in the district of Gunung Kidul.

Abstract

The recent study has been conducted to develop testicular germ cell (TGC) transplantation as a tool for preservation and propagation of male germ-plasm from endangered fish species. In practice of TGC transplantation,recipient and donor cell may not be immediately available at the same time whereas the testis can not be survive longer when it is outside of the body. Therefore, preservation of testis tissue may be required before transplantation.The research was conducted to evaluate the viability of spermatogonia isolated from short term preserved testis. Testis was preserved in physiological NaCl solution at 4 oC for 6, 12, 24 and 48 hours. Testis were dissociated in 0.5% trypsin and 3% DNase 10 IU/8L in PBS (phosphate buffered solution) complemented with 5% FBS ( fetal bovine serum), 25 mM HEPES and 1mM CaCl2 to obtain testicular germ cell suspension. The testicular germ cells isolated from 24 and 48 hours preservation were performed in trypan blue staining dye (1:1) and the viability of spermatogonia were observed under microscope. The results showed that the viability of spermatogonia started todecrease significantly in 12 hours preservation (P 0.05) and up to 48 hours preservation, cell viability was as high as 54,48±8,33%. In conclusion, preserved testicular tissue at 4oC still produced viable spermatogonia that areallowed to use as the source of donor cell for testicular germ cell transplantation of giant gourami.

Abstract

The success of transgenic fish production has been achieved through eggs fertilization using electroporated sperms carrying exogenous DNA. This study was conducted in order to obtain the optimal electroporation condition for stripped catfish sperm. A plasmid containing green fluorescent protein (GFP) gene driven by carp β-actin promoter was transferred into sperm using electrophoresis method towards transgenic stripped catfish (Pangasionodon hypophthalmus) production. Electroporation was carried out using square wave shock with pulse length of 30 ms and pulse interval of 0.1 sec. Treatments are combination between voltage (50 V, 75 V, and 100 V) and pulse number (1 and 3). Exogenous DNA concentration used was 10 μg/mL of Tris-EDTA. Results showed that increasing the voltage from 50 to 100 decreased sperm motility, while pulse number did not affect sperm motility. Voltage of 50 gave the best motility of sperm, although sperm viability relatively similar between treatments and control except at 100 V with 3 pulses number. Further, electroporation-treated sperms were able to fertilize eggs. Higher hatching rate of eggs was obtained in electroporation treatment at 50 V with pulse number of 1 and 3. The persistence of transferred GFP was detected in electroporated and incubated sperms (control). However, GFP was only detected in larvae from eggs that were fertilized by electroporated sperm. Thus, electroporation could be applied to produce transgenic stripped catfish.

Abstract

Fluktuasi asimetri organ berpasangan merupakan salah satu metode sederhana yang dapat digunakan untuk menghitung keragaman fenotip pada ikan nila. Penelitian ini bertujuan untuk mengukur fluktuasi asimetri ikan nila 69 dari Danau Tempe (Sulawesi Selatan) dan ikan nila GIFT dari Sukamandi, Jatiluhur, dan Sukabumi. Ikan nila yang digunakan berukuran konsumsi (150—250 g/ekor) dan berasal dari masing-masing lokasi diukur 50 ekor sebagai ikan uji. Hasil penelitian menunjukkan bahwa ikan nila 69 dari Danau Tempe mempunyai nilai fluktuasi asimetri bilangan (Fan) dan besaran (Fam) gabungan terkecil yaitu 0,8 dan 2,84. Ikan nila dari Jatiluhur mempunyai nilai fluktuasi asimetri bilangan (Fan) dan besaran (Fam) gabungan tertinggi yaitu 4,43 dan 16,66.The asymmetric fluctuating of couple organs were used as simple method to calculate fenotype variation of nile tilapia. The objective of this study was to measure the asymmetric fluctuation nile tilapia 69 from Lake Tempe (South Sulawesi) and nile tilapia GIFT product from West Java (Jatiluhur, Cirata, Sukamandi, and Sukabumi). The results showed that nile tilapia from Lake Tempe had the smallest value of fluctuating asymmetry number (Fan) and magnitute (Fam), which are 0.8 and 2.84 prespectively. The biggest value of fluctuating asymmetry number (Fan) and magnitute (Fam), which are 4.43 and 16.66 respectively.

Abstract

The Cyca-DAB1*05 allele of major histocompatibility complex class II genes is recently suggested to have a link with the European common carp strain resistant to koi herpesvirus (KHV). In this study, a set of specific primers for Cyca-DAB1*05 was designed and applied as a marker to identify broodstocks of majalaya common carp strain subsequently used as a candidate resistant to KHV infection. From a total of 23 broodstock subjected to PCR analysis, two female and male fish, both having (P) and no Cyca-DAB1*05 (N), were selected and then diallelly mated. Disease resistance of progenies from 10 crosses was determined by a survival analysis in pond rearing and a laboratory challenge-test using cohabitation method. The results have revealed that the average survivals of PxP progenies for pond rearing and KHV challenge test were 86% and 100% higher (P < 0.05) respectively compared to that of NxN fish. Survival rate of PxN/NxP progenies was significantly lower (P < 0.05) than that of PxP fish. Furthermore, PCR analysis showed that almost 91% progenies of PxP crosses seemed to have a KHV resistant gene marker. Thus, this study suggests that the marker is associated with the KHV resistance in majalaya common carp strain, and farming of PxP progenies can be useful to increase common carp production.
