Abstract

The use of appropriate diluent is intended to ensure the physical and chemical requirements ofspermatozoa so that the quality, particularly capacitation can be maintained. This study aimed todetermine the quality of Limousin cattle sperm after sexing with density percoll centrifugation methodusing Andromed diluent and Cauda epididymal Plasma 2 (CEP-2) plus 10% egg yolk. Observations ofspermatozoa quality including : motility, viability, percentage of abnormal, concentrations, membraneintegrity and state of capacitation. The semen used were obtained from Limousin cattle at the Center forArtificial Insemination(BBIB) Singosari Malang, with motility criteria e? 70%. Data were analysed usinga paired T-test in order to determine differences in the use of thinning treatments at each layer. Theresults showed that both diluent, andromed and CEP-2 diluent plus 10% egg yolk were able to maintainthe quality of spermatozoa either in the upper and lower layers

Abstract

Tujuan dari penelitian ini adalah mengetahui pengaruh penambahan kuning telur dalam pengencer CEP-2 terhadap motilitas dan viabilitas spermatozoa sapi Limousin. Penelitian menggunakan rancangan acak kelompok dengan 10 kali ulangan dari 10 ejakulat dari sapi yang berbeda. Spermatozoa disimpan selama delapan hari. Motilitas spermatozoa diamati dengan mikroskop cahaya (200x) pada suhu 37? C. Viabiltas spermatozoa diamati dengan pewarnaan eosin-negrosin menggunakan mikroskop cahaya (400x). Hasil penelitian menunjukkan suplementasi kuning telur pada pengencer CEP-2 berpengaruh terhadap motilitas dan viabilitas spermatozoa sapi Limousin selama penyimpanan pada suhu 4-5° C. Konsentrasi kuning telur terbaik adalah 20% dalam mempertahankan motilitas (44,25±3,92%) dan viabilitas (87,46±5,40%) spermatozoa sapi Limousin setelah penyimpanan delapan hari pada suhu 4-5° C.

Abstract

This study aims to determine the effect of the substitution of bovine serum albumin (BSA) with cattle blood serum and egg white in the diluent of Cauda epididymal Plasma 2 (CEP-2) on sperm quality of Limousin cattle during cooling at 3-5? C. The research material used was rejected Limousin bull sperm (motility of 50-60%) from Artificial Insemination Centre Singosari, Malang. This research was a laboratory experiment using a randomized block design which was composed of six treatments with 10 replications, those were T0 as controls ((90% CEP-2 with BSA + 10% egg yolk); T1 (83.84% CEP-2 + 6.16% cattle blood serum + 10% egg yolk); T2 (81.84% CEP-2 + 8.16% cattle blood serum + 10% egg yolk); dan T3 (90% CEP-2 + 0,4% egg white + 10% egg yolk); T4 (90% CEP-2 + 0.8% egg white + 10% egg yolk); and T5 (90% CEP-2 without BSA + 10% egg yolk). Parameters measured were the percentage of motility, viability, and abnormality of sperms. Results of research after 48 hours of storage showed that the percentage of sperm motility in T0, T1, T2, T3, T4, and T5 were 40.50±5.90, 36±36.16, 34.00±6.58, 40.50±3.69, 38.50±3.37, and 38.50±4.12, respectively, while the percentage of sperms viability were 75.16±4.30, 70.50±2.88, 73.80±2.80, 74.80±3.30, 75.13±3.13, and 74.03±4.13, respectively, and the percentage of sperms abnormality were 10.14±2.34, 10.62±1.34, 11.33±2.00, 10.94±2.82, 10.02±1.95, and 10.78±1.96, respectively. Abstract

ABSTRAK. Pejantan untuk program inseminasi buatan adalah pejantan unggul yang diseleksi berdasarkan libido dan produksi spermatozoa. Penelitian bertujuan untuk menganalisa hubungan antara tingkah laku seksual dengan produksi spermatozoa sapi Brahman. Materi penelitian adalah 3 pejantan sapi Brahman yang ditampung menggunakan vagina buatan. Metode penelitian adalah observasional dengan mengamati waktu reaksi dan lama ejakulasi. Observasi laboratorium dengan mengamati volume, konsentrasi, dan motilitas individu untuk menghitung total spermatozoa (TS) dan total spermatozoa motil (TSM). Pengamatan dilakukan sebanyak 10 ulangan. Data dianalisa secara deskriptif serta hubungan antara tingkah laku seksual dan produksi spermatozoa dihitung dengan korelasi Pearson. Hasil penelitian menunjukkan terdapat variasi tingkah laku seksual dan produksi spermatozoa pada sapi Brahman. Individu Cipta mempunyai waktu reaksi dan lama ejakulasi paling singkat serta produksi spermatozoa paling rendah. Hubungan signifikan antara waktu reaksi dengan TS serta TSM pada individu Bizzyard dengan koefisien determinasi (R2) sebesar 14 % dan 20%. Hubungan sangat signifikan antara lama ejakulasi dengan TS serta TSM pada individu Cipta dengan R2 sebesar 46% dan 58%. Kesimpulan penelitian adalah terdapat hubungan antara tingkah laku seksual dengan produksi spermatozoa pada sapi Brahman, dengan koefisien korelasi (r) tertinggi sebesar 0,72 dan R2 sebesar 58% antara lama ejakulasi dan total spermatozoa motil pada individu Cipta.  (The relationship between sexual behavior and semen production in Brahman bulls) ABSTRACT. Bulls for artificial insemination must be selected based on libido and sperm production. The study aimed to analyze the relationship between sexual behavior with sperm production on Brahman bulls. The material was 3 Brahman bulls collected using artificial vagina. The method was observational by observing reaction time and ejaculation duration. Laboratory observations by observing volume, concentration, and motility to calculate total sperm (TS) and total motile sperm counts (TMSC). Observations were replicated 10 times. The data were analyzed descriptively, the relationship between behavior and sperm production was examined using Pearson correlation. The results showed that there were variations in sexual behavior and sperm production in each bull. Cipta have the shortest reaction time and ejaculation duration and the lowest sperm production. A significant relationship between reaction time and TS and TMSC in Bizzyard with coefficient of determination (R2) of 14% and 20%. A very significant between ejaculation duration with TS and TMSC in Cipta with R2 of 46% and 58%. In conclusion, there was a relationship between sexual behavior and sperm production in Brahman bulls, with the highest correlation coefficient (r) of 0.72 and R2 of 58% between ejaculation duration and the total motile sperm counts in Cipta.

Abstract

Effect of thawing technique to quality frozen semen spermatozoa in bali, madura and PO cattleABSTRACT. The experiment was conducted from November 2011 to January 2012 in BBIB Singosari. The study aims was to determine the effect of thawing techniques on the quality frozen bovine sperm Bali, Madura and PO cattle. This material used 90 frozen straw of three cattle from the three breed. The method is an experiment with three treatments and 10 replications for frozen to thawing in temperature water is 5 0C of duration 60 seconds, 15 0C duration 30 seconds and 37 0C duration 15 seconds. Variables observed in this study were sperm quality are motility, viability and abnormality. Data in percentage transformed in to arcus sinus were analyzed using Completely Randomized Block Design (CRBD) with replication as blocks of 10x. The results showed the thawing at 37 0C duration 15 seconds to highly significant effect (P 0,01) viability and motility and significant effect (P 0.05). The abnormalities, no significant effect (P 0,05). Breed cattle not significant effect to quality. It can be concluded that thawing techniques impact the motility and viability in frozen bovine semen spermatozoa Bali, Madura and PO cattle. Thawing using 37 0C temperature water with duration of 15 seconds is the best method to obtain the quality of spermatozoa post thawing frozen sperm.

Abstract

The purpose of this research was to determine the success of AI using sexed semen results based on NRR, S/C, CR, and rectal palpation were also determine.  The total number of 81 acceptors divided into 3 treatment groups, such as P0 (non- sexed semen) ; P1 (Frozen semen Sexed using White Yolk Sedimentation),  P2 (Frozen semen sexed using Percoll Gradient Centrifugation) . The results showed the NRR percentage of P0, P1 and P2 were between the treatments are P0 of 74.07 ± 12:00%; 65.43 ± 5.66% and 64.19 ±  8:56% respectively. Furthermore the highest of CR was in P0 44%,   P2 25.91% and P1 18.51%. Conclution of this research was the highest percentage of pregnancy was 59.25 %(P0) ,  P1 51.85% and the lowest was in 44.44%. The S/C was in (P0) 2.31 follow by 3.00 in P2 and 3.33 in P1.   Key words : artificial insemination , frozen semen sexed, the percentage of Gestation and feed

Abstract

Penelitian ini bertujuan untuk mengetahui perbedaan produksi semen beku pada berbagai bangsa sapi potong dan untuk mengetahui produksi semen pada masing-masing  individu pada bangsa yang sama. Penelitian dilakukan di Balai Inseminasi Buatan Daerah Ungaran Jawa Tengah Indonesia. Materi yang dipergunakan dalam penelitian ini adalah catatan produksi semen yang terdiri kualitas semen segar dan jumlah straw semen beku pada bulan Januari sampai Desember tahun 2014  sebanyak  39 ekor sapi  yang terdiri dari 18 ekor sapi Simental, 8 ekor sapi  Limousin, 8 ekor PO dan 5 ekor sapi Brahman. Pengencer yang dipergunakan adalah skim kuning telur.  Metode yang dipergunakan adalah studi kasus dengan rancangan acak kelompok. Variabel yang diamati adalah Volume semen, Konsentrasi, Motilitas individu, total spermatozoa dan total spermatozoa yang motil. Hasilnya menunjukkan volume sapi Limousin yang terbanyak = 7,2 + 1,3 ml, diikuti oleh  oleh sapi simental = 6,8 + 1,6 ml,  sapi PO = 6,1 + 1,2 ml dan yang paling sedikit adalah sapi Brahman = 4,2 + 1,8 ml. Rata-rata volume semen pada berbagai bangsa terdapat perbedaan yang nyata (P<0,05). Rata-rata persentase motilitas spermatozoa sapi  Simental =  57,8 + 13,05 %, Sapi  Limousin = 61,17 +1.37%, sapi PO = 63,5 + 6,62 %  dan  Brahman = 44,8 + 25,33 %, Rata-rata persentase motilitas pada berbagai bangsa tidak menunjukkan perbedaan yang nyata (P>0,05). Rata-rata Total spermatozoa dan total spermatozoa yang motil pada sapi Simental adalah 9.320,3 + 2.461,2 and 5.532,9 + 2.314,2 ; Sapi Limousin = 11.266,9 + 1.159,6 dan  7.908,9 + 3.851,2; Sapi PO = 8.995,1 + 2.860,8 dan  5.651,6 + 2.418,4 , Sapi  Brahman =  6.189,3 + 22,6 dan  3.053,3 + 4.356,6. Rata-rata kedua parameter ini tidak menunjukkan perbedaan yang nyata pada berbagai bangsa sapi  (P>0,05). Rata-rata produksi semen beku per hari pada sapi  Simental 280,67 + 68,57 straw ; sapi Limousin 315,6 + 44,32 straw ;  sapi PO 275,15 +70,61  straw  dan sapi   Brahman 225,18 + 59,74 straw. Rata-rata produksi semen beku tidak menunjukkan perbedaan yang nyata pada berbagai bangsa sapi  (P>0,05). Volume semen, motilitas individu dan produksi semen beku harian masing-masing  individu pada bangsa yang sama pada sapi Limousin, Simental, PO dan Brahman terdapat perbedaan yang sangat nyata (P<0,05). Keyword: Sapi Limousin , Sapi Simental, Sapi PO dan Sapi Brahman

Abstract

The purpose of this study was to determine the performance of reproduction in dairy cattle, the views of service per conception, days open, and calving interval. The material used in this study is the implementation of recording data or artificial insemination in 100 dairy cows females have at least twice to parturition known calving interval. Samples were selected randomly within the same district in Batu. The method used in this research is a case study. The data is taken primary and secondary data. Primary data retrieval is done by direct observation (observation), which includes several variables, where the variables include: the identity of the breeder, the number of cattle, cages and feeding and drinking. As for the secondary data obtained by recording the card Artificial Insemination (AI) owned by farmers. Sampling was done by purposive sampling and random with the following conditions: sampling was done by purposive sampling with the following conditions: minimal yearling heifer parturition twice and had a complete recording AI, including, the date AI and the date of parturition. Results from this study is the performance of reproduction in the dairy cooperatives Batu showed unfavorable results, average - the average total value of S/C of parity 1 s / d 6 is, 1.93 ± 1.18. As for the average - the average total value of DO and CI parity 1 s / d 5 is, 171.70 ± 132.20 days and 450.92 ± 132.83 days. In addition, the assessment of S / C, DO and CI at different parity using statistical tests showed that the S / C, DO and CI at various parity was not significantly different (P> 0.05).It concluded that reproduction display AI results in the KUD BATU less than the maximum and the difference between parity does not affect the value of the S / C, DO, and CI.

Abstract

The purpose of the study was to determine the correlation between cow weight and birth weight of Brahman Cross calves with different sex. The materials of the study were one hundred Brahman Cross cows which had been purposively selected. The variables of the study were cow body weight, calves birth weight, and sex. Data were analyzed using Pearson Correlation test (Product-Moment), Linear Regression and t Test. The study found that the birth weight diferred significantly (P <0.05) among male and female calves at the first and third parities. Also, there was significant correlation (P <0.05) between cow body weight and calves birth weight at the first parity. However, this cow body weight only influenced 22-25% on birth weight so that the study suggests to considering other factors such as environment and animal feed. Abstract

The purpose of this study was to evaluate artificial insemination program on the reproduction performance of Ongole crossbred and Limousin crossbred cattle. The materials of the study were Ongole crossbred and Limousin crossbred cattle. Descriptive analysis was used to determine service per conception (S/C), days open (DO), calving interval (CI), conception rate (CR) and calving rate (CvR). Meanwhile, t-test was used to analyse differences among those variables. The study showed that the value of S/C, DO and CI between Ongole crossbred and Limousin crossbreed cattle differed significantly (P<0.05). The average S/C, DO, CI of Ongole crossbred cattle were 1.3±0.32; 156.9±29.33 days; 430±43.72 days respectively. In addition, Ongole crossbred cattle had calving rate (CvR) as well as conception rate (CR) was 74%. Meanwhile Limousin crossbed cattle had 1.5±0.39 of S/C; 172.9±19.21 days of DO; 451.3±19.61 days of CI. Moreover, calving rate (CvR) and conception rate (CR) of Limousin crossbed cattle was 52%. The study concluded that reproduction performances of Ongole crossbred cattle were better than that of Limousin crossbred cattle.   Keywords : days open, service per conception, calving interval, cattle
