Abstract

In  managing the koi herpesvirus (KHV) outbreaks as a routine national program in Indonesia, testingbased on biotechnology, such as  immunopathologic immunohistochemical approach(es) using antibodythat is safe, rapid  and accurate need to be applied. This will hopely assist the Government of Indonesianin improving and enhancing the sustainability of national animal proteins program. The present studywas aimed to develop and apply the immunopathologic immunohistochemistry of streptavidin biotin (IHCSB) for detection of KHV in the apparently normal carps. The gills from 48 common carps  (Cyprinuscarpio) that appear to be healthy were prepared for  DNA-based KHV  by IHC SB.  Common carps werecollected from fish farms which had an outbreak of KHV in 2008-2009 in Yogyakarta.  All fish werenecropsied. The gills were processed histopathologically and then stained for IHC SB with monoclonalantibody anti-KHV. We demonstrated that all of the fish gills were positive for KHV antigen. Thus, it isconcluded that method is useful and consistent, very sensitive and rapid, and is a reliable method to beapplied for field condition to detect antigen KHV in the gills of normal, healthy looking carps.   In addition,and more importantly, the fish can act as a source of KHV (carriers for KHV) and may result in the spreadof diseases among susceptible fish.

Abstract

Prior studies have focused on the determinants of cash holdings. The purpose of this study is to investigate the relationship between cash holding and stock liquidity in Indonesia. This study employs panel data analysis for a sample of 115 manufacturing firms listed on the Indonesia Stock Exchange over the period from 2010 to 2014. This study considers share turnover as the proxy for the stock liquidity. The results show that stock liquidity has a positive and significant effect on cash holding. This finding is in line with Chen et al. (2012), Nyborg and Wang (2013), and Tehrani et al. (2014).

Abstract

The right issue announcement may lead the market to react and affect the stock performance. This study aims to analyze the effect of the right issue on the abnormal return. This research uses a sample of 18 firms listed in the Indonesia Stock Exchange from 2014 to 2016. This study employs one sample t test and wilcoxon test. The results show that there is no abnormal return in the days surrounding right issue announcement. The results also find that there is no difference in abnormal return between after and before the right issue announcement. This research implies that the existing stockholders and investors need to consider carefully before deciding to invest in go public companies do the right issue.

Abstract

Right issue is the subsequent stock offerings that give priority to the existing shareholders to buy new shares at a specified price and time. There are several reasons why a firm does right issue, for example, to raise firm?s capital, investment expansion or to pay the debt. The objective of this study is to analyze the effect of the right issue on the abnormal return of the company. This research uses secondary data from the Indonesia Stock Exchange from 2014 to 2016. A sample of 18 firms met the criteria selection. The results show that there is no abnormal return in the days surrounding right issue announcement. The results also find that abnormal stock return in the days after right issue announcement is not lower or equal to the days before right issue announcement.

Abstract

Abstract            The objective of this research is to determine the effects of turmeric (Curcuma domestica Val) extract administration toward heart histopathologic lesions of male albino Wistar rats + 3 months old exposed by cigarette smoke. Twenty male Wistar rats were divided randomly into four groups of five each: Group I was given neither cigarette smoke exposure nor turmeric extract administration (control) (KI), Group II was given cigarette smoke exposure of 9 cigarettes/ day (KII), Group III was given 1 ml turmeric extract 0.25% (KIII) and Group IV was given cigarette smoke exposure of 9 cigarettes/ day and 1 ml turmeric extract 0.25% (KIV). After 2 months of treatment, necropsy was conducted to all rats and the heart samples were collected to be processed histopatologically and stained routinely with hematoxylin-eosin. The results of the present study indicated that the cigarette smoke caused cardiomyopathy in KII marked by the histopathologic lesions such as vacuolation, homogeneous eosinophilic cytoplasms, edema and necrosis of myocardia, also congestion and mild hemorrhages (KII). Whereas, rats in Groups KI, KIII and KIV had normal histologic structures of myocardia. It is concluded, that turmeric extract could be a preventive herbs toward cardiotoxic effects of cigarette smoke. Further research has to be conducted to determine the anti cardiomyopathy mechanism of turmeric rhizome due to cigarette smoke exposure.

Abstract

Terbitnya Undang-undang Nomor 32 Tahun 2004 Tentang Pemerintahan Daerah, merupakan konsekuensi konstitusional dari amandemen UUD 1945, khususnya pasal 18 ayat (5) yang menetapkan: “Pemerintah daerah menjalankan otonomi seluas-luasnya, kecuali urusan pemerintahan yang oleh Undang-undang ditentukan sebagai urusan pemerintah pusat”. Amanat UUD 1945 diatas, ditindaklanjuti dengan menuangkannya dalam Undang-undang Nomor  32 Tahun 2004 tentang Pemerintahan Daerah, sebagaimana yang telah diubah dengan Undang-undang Nomor 12 Tahun 2008 tentang Perubahan Kedua Atas Undang-undang Nomor 32 Tahun 2004,  khususnya pasal 10, 13, dan pasal 14. Ketentuan Pasal 10 ayat (3) menetapkan rincian urusan pemerintahan yang menjadi wilayah kewenangan pemerintah pusat, sementara ketentuan pasal 13 dan 14 menetapkan rincian urusan pemerintahan yang menjadi hak, kewajiban dan tanggung jawab pemerintah daerah  propinsi, kabupaten dan kota, yang salah satunya adalah tentang pengendalian lingkungan hidup. Berpedoman pada ketentuan di atas, maka tanggung jawab pengendalian lingkungan hidup menjadi salah satu urusan wajib pemerintah daerah. Sedangkan menurut Undang-undang Nomor 23 Tahun 1997 tentang Pengelolaan Lingkungan Hidup, tanggung jawab pengelolaan dan pengendalian lingkungan hidup, sepenuhnya menjadi wewenang Pemerintah Pusat.Key word : pengelolaan lingkungan hidup, otonomi daerah

Abstract

Newcastle Disease (ND) atau yang dikenal dengan ?Tetelo? masih menjadi masalah di peternakan unggas komersil, meskipun telah dilakukan vaksinasi ND secara rutin, namun wabah ND masih tetap terjadi. Penelitian ini bertujuan untuk mengetahui patogenesitas molekuler dan genotipe virus ND berdasarkan analisis sekuen fragmen gen F, serta melihat hubungan kekerabatan antara isolat dalam penelitian dengan isolat ND di Indonesia sebelumnya serta strain vaksin pada peternakan ayam yang menerapkan vaksinasi ND secara berkala. Penelitian ini menggunakan primer yang didesain dengan konsensus fragmen gen F dari GenBank dan desain dengan aplikasi amplifX pada posisi 91-800 nt dengan panjang 710bp. Urutan basa pada primer kemudian di cek dengan BLAST primer dan di uji spesifisitas dengan beberapa virus penyakit unggas yaitu vaksin infectious bronchitis (IB) 120, virus vaksin infectious laryngotracheitis (ILT), virus vaksin avian influenza (AI), dan virus vaksin infectious bursal disease (IBD). Delapan sampel paru diperoleh dari delapan peternakan ayam komersial di Yogyakarta, Semarang, Jakarta, Magelang dan Muntilan. Tiga sampel diisolasi pada telur ayam berembrio dan diidentifikasi menggunakan uji HA dan HI. Lima sampel lain dilakukan ekstraksi secara langsung dari gerusan organ paru. Sampel di identifikasi dengan metode reverse transciptase-polymerase chain reaction (RT-PCR) untuk mendeteksi gen F menggunakan  primer forward 5? TCT CTT GAT GGC AGG CCT CTT G ?3 dan reverse 5? CCG CTA CCG ATT AAT GAG CTG AGT?3 dengan panjang produk 710 bp. Hasil penelitian menunjukkan bahwa seluruh sampel positif virus ND. Analisis hasil sekuen dengan menggunakan perangkat lunak MEGA v.7 didapatkan susunan asam amino penyusun cleavage site 112RRQKR?F117 dan 112RRRKR?F117yang menunjukkan bahwa virus ND tersebut digolongkan strain velogenik. Berdasarkan analisis pohon filogenetik isolat ND-Layer/GK-SR1/2013, ND-Lay/Pullet-80/ 27/16 (N), ND-Bro/Lingga 2L/24/2 (N), dan ND-Lay/Smg-P/2015 merupakan genotip VIIi, sedangkan 3 isolat ND yaitu ND-Bro/Yog-P/2015, JKT/P1/2016 (Jakarta), dan JKT/P2/2016 (Jakarta) merupakan ND genotip VIIh. Jarak genetik isolat yang diteliti dengan virus ND Indonesia yang pernah dilaporkan sebelumnya pada fragmen gen F posisi 91-798 berkisar 0,4 ? 9,6 % dengan tingkat homologi mencapai 90,4 ? 99,5%.

Abstract

Aeromonas hydrophila causes a disease that often infects fish and is known as Motile Aeromonas Septicaemia (MAS), Hemorrhagi Septisemia, Ulcer disease or Red-Sore disease. The   aims of this study were to develop polyclonal antibody of  Aeromonas hydrophila in the rabbits to   confirm the diagnosis of Aeromonas hydrophila  in the fish by immunohistochemistry staining method. Preparation of polyclonal antibodies was performed on the rabbits used to Aeromonas hydrophila bacteria that have been tested biochemically by intravenous and intraperitoneal injection. Doses of Aeromonas hydrophila  bacteria were 109 CPU/ml  of 0.5 ml at first injection, 1 ml at second injection, 2 ml at thirth injection and 3 ml at fourth injection. Blood serum collection was performed at week 5 after injection from  an  ear and intracardial vein. The result of antibody titer was 28 = 1024 which measured by   tube test. Furthermore, polyclonal   antibody was used to immunohistochemistry  staining with 400x dilution. The results of the staining showed that an immunopositive reaction in the liver, skin,lien,  gill, kidney, and heart of fish to Aeromonas hydrophila antibody. The research conclution was polyclonal antibody from rabbit can be used to accurately confirm the diagnosis of Aeromonas hydrophila  based on antigen and antibody reaction.

Abstract

Indonesia is among the developing countries that are struggling to develop in the field of industrial development. The logical consequence of any development process, especially industrial development is the emergence of associated impacts that greatly affect the durability and sustainability of the environment. Developing the equitable industry in order to create public welfare is important. However, maintaining the security and preservation of the environment is also very important, because it is only with the availability of a good environment and healthy living that human beings can perform daily living. The availability of good and healthy environment is the constitutional responsibility of the government, as well as part of the human rights of all citizens which must be given by the State. Efforts to create a good environment and healthy living will be effective if controlled by State government and institutions who understand the objective conditions on the ground. Abstract

Abstract An increasing number of smokers who tend to rise rapidly from year to year has become a major focus of public health issues. Exposure to environmental tobacco smoke, containing toxic chemicals can trigger acute or chronic inflammation and oxidative stress in the eyes. To overcome this condition, phytotherapeutic and/or phytopreventive approaches which are efficatious, safe and cheap is needed. One of spices from Indonesia which is greatly possible to be promoted as a herbal medicine, is a phytochemical curcumin in turmeric. This present study was conducted to identify and determine the effects of turmeric extract (Curcuma domestica Val.) as phytotherapeutic and/or phytopreventive in the eyes lesion of Wistar rats induced by cigarette smokes. Twenty male Wistar rats at 3 months old were divided randomly into four groups of five each. Those were the control group (KI), the group given 9 cigarettes smoke/day (K II), the group given turmeric extract 1 ml 0, 25% /day (K III) and the group given 9 cigarettes smoke/day and 0, 25% turmeric extract 1 ml /day (K IV) for 60 days. Water and feed was given ad libitum. At the end of the experiment, all rats were euthanized with ether, necropsied andeyes balls were collected, fixed formalin 10% and stained with routine hematoxylin-eosin. The results of the present study indicated that 9 cigarettes smoke/day increased mitotic activity and caused hyperplasia in the corneal epithelial cells (K II), and addition of turmeric extract 0, 25% 1 ml per oral for 60 days prevented both mitotic activity and hyperplasia in the corneal epithelial cells (K IV). Extract of turmeric (Curcuma domestica Val.) has been proved to have herbal therapeutic and/or preventive effects againts the pathotoxicological cigarette smokes. Further study needs to be done to more evaluate pathogenesis of the eyes lesions and phytotherapeutic and/or phytopreventive effects of turmeric extract as well.
