Abstract

Tanaman kelapa sawit memiliki kandungan polisakarida dan polifenol yang tinggi. Kontaminasi polisakarida dan polifenol menyebabkan sulitnya proses isolasi RNA dari jaringan tanaman kelapa sawit. Penelitian ini dilakukan untuk mengoptimasi beberapa protokol isolasi RNA tanaman kelapa sawit yang efektif dan efisien. Penelitian dilaksanakan selama tiga bulan, pada bulan Oktober-Desember 2018 di Laboratorium Biokimia dan Biologi Molekuler, Pusat Penelitian Bioteknologi dan Bioindustri Indonesia (PPBBI), Jalan Taman Kencana No. 1 Bogor 16128. Penelitian dilakukan dengan melaksanakan teknik isolasi RNA menggunakan tiga protokol, yaitu modifikasi Cetyl Trimethyl Ammonium Bromide (CTAB), kit isolasi RNA RNeasy Plant Mini Kit (Qiagen), dan kit isolasi NucleoSpin RNAPlant (Macherey-Nagel). Sampel yang digunakan adalah daun dan akar tanaman kelapa sawit berumur kurang dari tiga bulan dengan bobot 0,1 gram dan 2,5 gram yang disesuaikan untuk tiap protokol. Variabel yang diamati adalah konsentrasi (ng/µl), kemurnian (rasio A260/A280 dan A260/A230), dan pita RNA pada elektroforesis gel agarosa. Hasil penelitian menunjukkan, RNA total hasil isolasi protokol NucleoSpin RNAPlant (Macherey-Nagel) memiliki kualitas paling tinggi. Konsentrasi RNA total daun dan akar kelapa sawit yang didapatkan melalui protokol NucleoSpin RNAPlant (Macherey-Nagel) sebesar 338 ng/µl dan 184,4 ng/µl dengan rasio A260/A280 RNA total daun dan akar kelapa sawit sebesar 2,13 dan 2,18 serta rasio A260/A230 sebesar 2,09 dan 2,20. Hasil elektroforesis gel agarosa menunjukkan integritas yang bagus dari RNA total hasil isolasi RNeasy Plant Mini Kit (Qiagen) dan NucleoSpin RNAPlant (Macherey-Nagel), namun terdapat kontaminasi dan smear pada RNA total hasil isolasi CTAB modifikasi 1 dan 2. Kata kunci: kelapa sawit, isolasi RNA, spektrofotometer, elektroforesis gel agarosa

Abstract

A connected graph G is primitive provided there exists a positive integer k such that for each pair of vertices u and v in G there is a walk of length k connecting u and v. The smallest of such positive integer k is the exponent of G. A primitive graph is said to be odd primitive graph if it has an odd exponent. It is known that if G is an odd primitive graph then G contains two disjoint odd cycles. This paper discusses exponents of a class of primitivegraphs containing of exactly two disjoint odd cycles. For such graphs we characterize the odd and even primitive graphs.

Abstract

The purpose of this study was to describe the efforts that students made in developing speaking skill. Descriptive qualitative was used in this study; the subjects were 65 students at year 2015 studying at English Education department, Syiah Kuala University, Banda Aceh. The instrument used in this study was questionnaire. The questionnaire questions were adapted from the activity scale from Pace Kuh. The questionnaire revealed that speaking experience such as communicating by using English with friends and family was the highest effort chosen by the students because it was very comfortable to perform speaking among the closest people. Club and organization was the lowest effort chosen by the student. The students took part in organization just to spend their spare time. The students thought that joining in an organization did not have contribution in developing their speaking skill. The implication of this study was to motivate the students to make some efforts to develop their speaking skill. Focusing in certain effort will help the students improve their speaking skill mastery. In addition, for the suggestion, the students should create activities that promote English communication in order to develop their speaking skill.

Abstract

Tujuan dari sistem transmisi adalah menyalurkan daya listrik dari pusat pembangkit ke pusat beban secara aman, effisien, andal dan ekonomis. Untuk memberikan pelayanan yang andal, sistem tenaga listrik harus tetap utuh dan mampu mengatasi berbagai macam gangguan yang mungkin terjadi. Dengan demikian merupakan suatu hal yang sangat penting bahwa sistem harus direncanakan agar dalam keadaan normal maupun dalam keadaan gangguan, sistem tetap mampu menyalurkan energi listrik kepada konsumen. Tugas akhir ini menjelaskan tentang kontingensi yang disebabkan oleh terlepasnya saluran transmisi yang terjadi pada sistem interkoneksi 150 KV Aceh. Akibat terputusnya saluran transmisi menyebabkan perubahan tegangan pada bus, dan overload pada saluran transmisi, sehingga diperlukan perbaikan untuk mengatasi masalah ini dengan sebuah simulasi analisis kontingensi menggunakan ETAP 12.6 untuk menemukan solusi dari masalah yang ditimbulkan akibat terputusnya saluran transmisi. Dari hasil simulasi diketahui bahwa ada tiga bus yang tegangannya turun melewati batas operasi yang diijinkan akibat kontingensi pada saluran penghantar, yaitu Banda Aceh, Lhokseumawe dan Panton Labu dengan tegangan pada bus Banda Aceh (134.8 KV), Lhokseumawe (134.6 KV) dan Panton Labu (133.8 KV). Dan juga diketahui bahwa ada satu saluran transmisi yang mengalami pembebanan kritis akibat adanya kontingensi, yaitu saluran antara Lhokseumawe - Arun dengan arus nominal sebesar 84 %. Untuk mengatasi masalah tegangan turun dan pembebanan lebih tersebut maka dilakukan solusi yaitu pelepasan beban (load shedding). Hasil dari perhitungan analisis kontingensi dapat digunakan secara optimal untuk perencanaan operasi sistem pembangkitan energi listrik, dengan begitu perencanaan sistem dapat mempengaruhi keandalan dan keamanan dari sistem interkoneksi.

Abstract

This study examines the fresh fruit bunch (FFB) yield and the effect of fruit storage on the quality of palm oil in Bangun Bandar Estate, Dolok Masihul, North Sumatra, with special reference to the physicochemical parameters. The quality of palm oil is mostly determined by the color, free fatty acids (FFAs), moisture, impurities, carotene content, and deterioration of bleachability index (DOBI). The palm fruits were stored in a refrigerator for 0, 7, 14, 21, and 28 days after harvesting before analyzing their carotenoid content, DOBI, FFAs, moisture, and impurities. The results showed that the potency of the crude palm oil (CPO) types, dura, pisifera, and tenera, were 23.5, 26.2, and 23.6%, respectively. The carotenoid content and DOBI of the palm oil types, dura, pisifera, and tenera, were decreased significantly by lengthy storage of the fruit. By contrast, lengthy storage of the fruit significantly increased the FFA, moisture, and impurity contents of these palm oil types, indicating that this reduced the quality of the palm oil. Oil processed from FFB has excellent quality with a high carotene content and DOBI, and a low concentration of FFA, moisture, and impurities. The present study contributes to palm oil stability and nutritional value.

Abstract

This study aimed to analyze the effect of leadership functions, work environment, and work motivation on personnel performance at the police station Pasaman Barat. Type of research is causative. The study population was all personnel at the police station Pasaman Barat. Sampling was based on stratified random sampling. Analysis of data used path analysis and hypothesis testing using the F test and t test. The results showed that: (1) The function of leadership a significant effect on work environment, (2) The function of leadership and work environment have a significant effect on work motivation, (3) The function of leadership, work environment, and work motivation have a significant effect on performance. (4) The function of leadership have a significant effect on performance through work environment, (5) The function of leadership have a significant effect on performance through motivation, and (6) work environment significant effect on performance through work motivation. This suggests that the function of leadership, work environment, and work motivation has a significant effect, directly or indirectly, to the performance of personnel at the police station Pasaman Barat.
